O Lentinula edodes é um alimento de qualidade elevada, sendo rico em proteínas, vitaminas e sais minerais e pobre em calorias e gorduras. No entanto, seu valor nutricional varia em função da linhagem cultivada, do processamento após a colheita, do estágio de desenvolvimento do basidioma e do substrato utilizado. Assim, este trabalho teve como objetivo caracterizar bromatologicamente os basidiomas de oito linhagens de L. edodes (LE-95/01, LE-95/02, LE-95/07, LE-96/17, LE-96/18, LE-98/47, LE-98/55 e LE-96/13), inteiros ou em partes (estípete e píleo), produzidos em toras de E. grandis. Verificou-se que tanto as linhagens de L. edodes como as partes dos basidiomas analisados influenciaram nas respostas nutricionais das amostras. O estípete apresentou as maiores médias de fibra bruta e as menores médias de proteína bruta. O píleo, por sua vez, apresentou as maiores médias de cinzas. Nas avaliações de basidiomas inteiros, as maiores médias, quanto ao teor de proteína bruta, foram obtidas nas linhagens LE-95/07, LE-96/17 e LE-96/18. Já a maior média de fibra bruta foi obtida pela linhagem LE-96/13 de L. edodes. O conteúdo de lipídios do píleo e do estípete não variaram entre as linhagens de L. edodes.

O valor nutricional do Lentinula edodes (Berk.) Pegler varia em função da linhagem cultivada, do processamento após colheita, do estágio de desenvolvimento do basidioma e do tipo de substrato utilizado. Este trabalho teve como objetivo avaliar nutricionalmente basidiomas de L. edodes em função da linhagem e do tipo de eucalipto cultivado. O delineamento experimental foi inteiramente casualizado, em esquema fatorial 2 x 10 (linhagens de L. edodes x tipo de eucalipto), totalizando 20 tratamentos com 2 repetições, sendo que cada repetição correspondeu a uma amostra de basidiomas desidratados e moídos. de acordo com os resultados obtidos, verificou-se que as propriedades nutricionais do L. edodes (proteína bruta, extrato etéreo, cinzas e fibra bruta) demonstraram sofrer influência da interação eucalipto x fungo. Assim, os melhores resultados foram: Proteína bruta: Linhagem LE-96/18 cultivada em toras de E. urophylla, cuja média foi de 24,3%; Extrato etéreo: Linhagem LE-96/18 cultivada em toras do clone 23, cuja média foi de 3,0%; Cinzas: Linhagem LE-96/18 cultivada em toras de E. paniculata e E. camaldulensis e Linhagem LE-95/01 cultivada em toras de E. citriodora, cujas médias foram de 5,0%; Fibra bruta: Linhagem LE-95/01 cultivada em toras de E. paniculata, cuja média foi de 20,5%.

The experiment was conducted to investigate the viability of the production of the edible fungus Pleurotus sajor-caju (Fr.) Singer in substrates in which main source of carbon was the cotton textille mill waste. Two substrates compositions were tested: C1 (cotton textille mill waste, wheat bran, plaster and limestone) and C2 (cotton textille mill waste, wheat bran, bean straw, plaster and limestone). A DIC experimental design was used, with nine repetitions for treatment and the production data and biological efficiency were analyzed being used the procedure ANOVAG of the statistical package SAEG. The cotton textille mill waste improvement, as the main ingredient of the substrate, was shown efficient for the production of the mushroom P. sajor-caju, presenting satisfactory values of productivity (0,56 and 0,5 kg/kg substratum) and biological efficiency (55,76 and 55,39%), respectively for C1 and C2. These results showed us that cotton textille mill waste, could be recommended as less onerous alternative of commercial substrate for the species Pleurotus sajor-caju.

The mushroom Agaricus blazei has been extensively investigated because of evidence of its antimutagenic, antitumor, and anticarcinogenic activities. This study investigated the clastogenic and/or anticlastogenic activity of aqueous extract of Agaricus blazei (10% w/v) in drug-metabolizing rat hepatoma tissue cells (HTCs), with continuous treatment and treatment during different phases of the cell cycle. DNA damage was induced utilizing two directacting agents-methyl methane sulfonate and ethyl methane sulfonate-and two indirect-acting agents-2-aminoanthracene and cyclophosphamide. The aqueous extract of A. blazei with either continuous treatment or treatment during different phases of the cell cycle showed clastogenic activity. The results with continuous treatment showed that A. blazei does not protect against DNA damage-inducing agents that are direct acting. Meanwhile, when combined with indirect-acting agents, a protective effect was demonstrated. A protective effect was also found during different phases of the cell cycle when cells were treated with indirect-acting agents. The protective effects against indirect-acting agents (continuous treatment and during the different phases of the cell cycle) suggest that A. blazei may provide some health benefits to the public when used as a functional food.

Shiitake (Lentinula edodes (Berkeley) Pegler) is one of the most consumed mushrooms, for both therapeutic purposes and as food, therefore, the study of its biological properties is of great interest for producers and consumers. Aqueous extracts of the shiitake mushroom (L. edodes (Berkeley) Pegler) were evaluated by the micronucleus test (MN) in HEp-2 cells in vitro, to analyze their possible mutagenic and antimutagenic activities. None of the three extract concentrations tested (0.5, 1.0 and 1.5 mg/mL) presented mutagenicity at any of the preparation temperatures (4 degrees C, 22 +/- 2 degrees C and 60 degrees C). In the antimutagenicity evaluation, all extract concentrations at all preparation temperatures presented a strong protective activity for the HEp-2 cells in response to the alkylating agent methyl methanesulfonate (MMS) in the different treatment protocols: pre-treatment, simultaneous treatment and post-treatment. The extracts prepared at 22 +/- 2 degrees C presented the lowest frequencies of MN in the evaluations of mutagenicity and antimutagenicity, indicating these as the best option for potential therapeutic use. In a culture of a Pleurotus ostreatus (oyster mushroom) strain, macro and micronutrients of the raw material and the initial and spent substrates were evaluated. Substrates were formulated with sawdust from Simarouba amara Aubl. and Ochroma piramidale Cav. ex. Lam., crushed Bactris gasipaes Kunth and crushed Saccharum officinarum (sugar cane). Samples were solubilized by means of acid digestion (nitric-peridrol). Ca, Mg, Fe, Cu, Zn and Mn were determined by atomic absorption spectrophotometry, Na and K by atomic emission, and P by colorimetry. The mineral composition of the fruiting body varied with the substrates, which made possible the production of a fruiting body rich in K, P Mg and Fe. Potassium was the mineral with the highest content in the fruiting body in all substrates tested (36.83-42.18g.kg(-1)). There was an increase of protein and mineral content in the spent substrate in relation to the initial one.

The viability of the utilization of wood and agroindustrial residues available in the Amazon region in the formulation of alternative substrates for the cultivation of Pleurotus ostreatus was tested. Thus, two wood residues: marupa sawdust (SIAMP) and pau-de-balsa sawdust (SAPB), and two substrates derived from agroindustrial residues: sugar-cane bagasse (SIACN) and pupunheira stem (SIAPP), were used. These were supplemented with a mixture of rice bran, wheat and corn as protein source, with addition of 2-3% of CaCO(3) for pH correction (6.5). The substrates were placed in polyethylene (HDPE) bags, sterilized at 121 degrees C for 1h and inoculated in a laminar flow chamber. The cultivation was carried out in an axenic way, in a modified atmosphere. The productivity of the substrates was evaluated in relation to the biological efficiency, with the following mean results: 125.60, 99.80%, 94.00 and 64.60% for SIAPP, SIACN, SIAMP and SIAPB, respectively. The high biological efficiency of the substrates and the cultivation process clearly showed the viability of the utilization of the residues, suggesting the commercial cultivation of this mushroom, which may contribute for improving the social and economical conditions and sustainability of the regional biodiversity resources of Amazonia.

The effects of crude extracts of the mushroom Agaricus blazei Murrill (Agaricaceae) on both DNA damage and placental form glutathione S-transferase (GST-P)-positive liver foci induced by diethylnitrosamine (DEN) were investigated. Six groups of adult male Wistar rats were used. For two weeks, animals of groups 3 to 6 were treated with three aqueous solutions of A. blazei (mean dry weight of solids being 1.2, 5.6, 11.5 and 11.5 mg/ml, respectively). After this period, groups 2 to 5 were given a single ip injection 200 mg/kg DEN and groups 1 and 6 were treated with 0.9% NaCl. All animals were subjected to 70% partial hepatectomy at week five and sacrificed 4, 24 and 48 h or 8 weeks after DEN or 0.9% NaCl treatments (10th week after the beginning of the experiment). The alkaline comet assay and GST-P-positive liver foci development were used to evaluate the influence of the mushroom extracts on liver cell DNA damage and on the initiation of liver carcinogenesis, respectively. Previous treatment with the highest concentration of A. blazei (11.5 mg/ml) significantly reduced DNA damage, indicating a protective effect against DEN-induced liver cytotoxicity/genotoxicity. However, the same dose of mushroom extract significantly increased the number of GST-P-positive liver foci.

Casing layer is one of the most important components of Agaricus spp. production and it directly affects mushroom productivity, size and mass. The aim of this study was to evaluate potential raw materials as a casing layer and their effect on Agaricus brasiliensis productivity. Raw materials from Brazil with potential use were selected and characterized, and the most promising ones were tested as casing layers for mushroom yield. Evaluated raw materials included lime schist, vermiculite, eucalyptus sawdust, sand, São Paulo peat, Santa Catarina peat, subsoil and charcoal. Particle size, porosity and water absorption in relation to mushroom yield for casing layers were determined. Lime schist, an alternate casing layer to peat, is presented and the effects of the casing layer on the mushroom yield are discussed. © 2009 Elsevier Ltd. All rights reserved.

Agaricus brasiliensis is a Brazilian basidiomycete which has been cultivated and consumed around the world as a therapeutic food. Casing layer is one of the most important steps on A. brasiliensis cultivation and European peat is the most used casing layer on Agaricus bisporus cultivation. Besides the importance of it on mushroom cultivation the peat import could be a risk of exotic organism introduction. Alternative as Brazilian peat is barely used for mushroom growers in Brazil. Thus, the objective of this work was to evaluate Brazilian peat with and without pasteurization as casing layer on A. brasiliensis cultivation. The fungus was previously grown on wheat grains and transferred to a substratum prepared by composted traditional method. After mycelium colonization of the substratum a pasteurized or non pasteurized Brazilian peat (casing layer) was added. It was concluded that pasteurization of the casing layer increases in 30% yield after 65 days of cultivation. There is no difference of yield for pasteurized and non pasteurized casing layer until 30 days of cultivation. An increase of flies is observed in non pasteurized casing layer. The production flush is easily perceived with pasteurized casing layer but not with non pasteurized casing layer.

The development of various types of cancer results from the interaction among endogenous, environmental and hormonal factors, where the most notable of these factors is diet. The aim of the present study was to determine the antigenotoxic, anticarcinogenic, phagocytic and immunomodulatory activities of Agaricus blazei. The test antigenotoxicity (Comet Assay) and anticarcinogenic (Test of Aberrant Crypt Foci) assess changes in DNA and/or intestinal mucosa that correlate to cancer development. Tests of phagocytosis in the spleen and differential count in blood cells allow the inference of modulation of the immune system as well as to propose a way of eliminating cells with DNA damage. Supplementation with the mushroom was carried out under pre-treatment, simultaneous treatment, post-treatment and pre-treatment. +. continuous conditions. Statistical analysis demonstrated that the mushroom did not have genotoxic activity but showed antigenotoxic activity. Supplementation caused an increase in the number of monocytes and in phagocytic activity, suggesting that supplementation increases a proliferation of monocytes, consequently increasing phagocytic capacity especially in the groups pre-treatment, simultaneous and pre-treatment. +. continuous. The data suggest that A. blazei could act as a functional food capable of promoting immunomodulation which can account for the destruction of cells with DNA alterations that correlate with the development of cancer, since this mushroom was demonstrated to have a preventive effect against pre-neoplastic colorectal lesions evaluated by the aberrant crypt foci assay. According to these results and the literature, it is believed that supplementation with A. blazei can be an efficient method for the prevention of cancer as well as possibly being an important coadjuvant treatment in chemotherapy. © 2011 Elsevier Inc.
