An in vitro model, the aggregating brain cell culture of fetal rat telencephalon, has been used to study the maturation-dependent sensitivity of brain cells to two organophosphorus pesticides (OPs), chlorpyrifos and parathion, and to their oxon derivatives. Immature (DIV 5-15) or differentiated (DIV 25-35) brain cells were treated continuously for 10 days. Acetylcholinesterase (AChE) inhibitory potency for the OPs was compared to that of eserine (physostigmine), a reversible AChE inhibitor. Oxon derivatives were more potent AChE inhibitors than the parent compounds, and parathion was more potent than chlorpyrifos. No maturation-dependent differences for AChE inhibition were found for chlorpyrifos and eserine, whereas for parathion and paraoxon there was a tendency to be more effective in immature cultures, while the opposite was true for chlorpyrifos-oxon. Toxic effects, assessed by measuring protein content as an index of general cytotoxicity, and various enzyme activities as cell-type-specific neuronal and glial markers (ChAT and GAD, for cholinergic and GABAergic neurons, respectively, and GS and CNP, for astrocytes and oligodendrocytes, respectively) were only found at more than 70% of AChE inhibition. Immature compared to differentiated cholinergic neurons appeared to be more sensitive to OP treatments. The oxon derivates were found to be more toxic on neurons than the parent compounds, and chlorpyrifos was more toxic than parathion. Eserine was not neurotoxic. These results indicate that inhibition of AChE remains the most sensitive macromolecular target of OP exposure, since toxic effects were found at concentrations in which AChE was inhibited. Furthermore, the compound-specific reactions, the differential pattern of toxicity of OPs compared to eserine, and the higher sensitivity of immature brain cells suggest that the toxic effects and inhibition of AChE are unrelated.

RESUMO Objetivo Analisar o processo de construção da identidade docente na perspectiva do passado, presente e futuro na formação de professores de enfermagem por meio da integração de tecnologias digitais. Método Pesquisa descritivo-exploratória, vertente qualitativa, consolidada na aula presencial “Construção da Identidade Docente” em 2012. Das atividades síncronas e assíncronas, quatro foram analisadas por meio da análise de conteúdo. Resultados No Chat identificaram-se marcas positivas e negativas e a reflexão da construção da própria identidade (seis categorias); no “Túnel do tempo” a projeção da futura identidade (quatro categorias); no “portfólio” a percepção da própria identidade (quatro categorias) e na avaliação da aula, a percepção das experiências vivenciadas na aula (duas categorias). Conclusão A integração das tecnologias digitais favoreceu a discussão e compreensão dos licenciandos, de maneira colaborativa, acerca de como sua identidade é construída e a projeção de sua futura identidade alinhada ao perfil e ao papel do professor exigidos na contemporaneidade.

Hyperinsulinemia increases lactate release by various organs and tissues. Whereas it has been shown that aerobic glycolysis is linked to Na+-K+-ATPase activity, we hypothesized that stimulation by insulin of skeletal muscle Na+-K+-ATPase is responsible for increased muscle lactate production. To test this hypothesis, we assessed muscle lactate release in healthy volunteers from the [13C]lactate concentration in the effluent dialysates of microdialysis probes inserted into the tibialis anterior muscles on both sides and infused with solutions containing 5 mmol/l [U-13C]glucose. On one side, the microdialysis probe was intermittently infused with the same solution additioned with 2.10(-5) M ouabain. In the basal state, [13C]lactate concentration in the dialysate was not affected by ouabain. During a euglycemic-hyperinsulinemic clamp, [13C]lactate concentration increased by 135% in the dialysate without ouabain, and this stimulation was nearly entirely reversed by ouabain (56% inhibition compared with values in the dialysate collected from the contralateral probe). These data indicate that insulin stimulates muscle lactate release by activating Na+-K+-ATPase in healthy humans.

Em solos recentemente fertilizados, as plantas absorvem os nutrientes que estão principalmente nas adjacências dos grânulos, cuja composição química é pouco conhecida. Este trabalho teve como objetivo avaliar o gradiente de concentração de K e a concentração eletrolítica a partir da região fertilizada com KCl em cinco solos, que tinham quantidades variáveis de argila (95 a 569 g kg-1) e de matéria orgânica (13 a 54 g kg-1). Os tratamentos consistiram de doses de KCl equivalentes a 0, 50, 100 e 200 kg ha-1 de K2O. Para obter as quantidades a serem aplicadas, considerou-se que essas doses seriam adicionadas sobre a superfície do solo, em linhas distanciadas de 0,70 m, e atingiria uma faixa de 2,5 cm, ou seja, entrariam em contato com 357 m² ha-1 de solo. Cada unidade experimental foi formada pela união de dois tubos horizontais de PVC, cada um com 7,0 cm de diâmetro e 9,0 cm de comprimento, preenchida com solo. Os grânulos de KCl foram distribuídos uniformemente entre duas folhas circulares de papel-filtro (7,0 cm de diâmetro), que foram colocadas na superfície de junção dos dois tubos. O gradiente de concentração de K nos solos foi quantificado por meio da concentração do K trocável (Mehlich-1) em diferentes distâncias a partir do local de aplicação dos grânulos. A movimentação de K variou com o tipo de solo, aumentou com a dose de KCl e foi menor no solo que tinha mais areia e menos matéria orgânica. Após 7 dias, com a maior dose de KCl aplicada (200 mg kg-1 K2O), a movimentação de K foi maior no Cambissolo Húmico léptico CHat (8,4 cm) e menor no Latossolo Vermelho distrófico LVd (6,4 cm). A amplitude de concentração de K no centímetro mais próximo do adubo variou de 9.800 a 16.475 mg kg-1; na camada distante de 4,0 a 5,0 cm dos grânulos, o K variou de 244 a 2.254 mg kg-1. A condutividade elétrica não atingiu valores prejudiciais ao processo germinativo de espécies com média tolerância a sais, mesmo no centímetro mais próximo do fertilizante. O movimento do K nos solos aconteceu em poucos dias, porém a movimentação máxima em cada dose de KCl não teve correlação com nenhum atributo de solo.

A limited number of foods explain the majority of food allergies. These allergies can be due to a weak allergenicity (garlic, onion, potato), or a weak (or increasing) exposure to emergent food allergens which can be imported (exotic fruits), or recently introduced (lupin, buckwheat, sesame, inulin) or modified by the industry (lysats, lecithins, traces of antibiotics, caseinates, molds, dust mite). Others are in relation with rarer cross-reactivity food allergy syndrome (Apiaceae-Compositae-mugwort syndrome, egg-bird syndrome, cat epithelium-pork meat syndrome). Others are rarely identified, because the food is masked (pepper, basilic). We illustrate rare cases of food allergy and discuss the diagnostic management which is based on a meticulous patient history. Un nombre restreint d'aliments explique la majorité des allergies alimentaires. Les allergies alimentaires rares sont dues à une faible allergénicité (ail, oignon, pomme de terre) ou à une exposition faible ou croissante à des aliments émergents, importés (fruits exotiques), introduits (lupin, sarrasin, sésame, inuline), ou modifiés par l'industrie (lysats, lécithines, traces d'antibiotiques, caséinates, moisissures, acariens). D'autres sont en relation avec des croisements d'allergènes rares (syndrome croisé ombellifères-composées-armoise, syndrome oeuf-oiseau, syndrome épithélium de chat-viande de porc). D'autres enfin sont rarement identifiées, car l'allergène est masqué (poivre, basilic). Nous décrivons des cas rares illustratifs et rappelons la démarche diagnostique qui s'appuie sur une anamnèse minutieuse.

The identity [r]evolution is happening. Who are you, who am I in the information society? In recent years, the convergence of several factors - technological, political, economic - has accelerated a fundamental change in our networked world. On a technological level, information becomes easier to gather, to store, to exchange and to process. The belief that more information brings more security has been a strong political driver to promote information gathering since September 11. Profiling intends to transform information into knowledge in order to anticipate one's behaviour, or needs, or preferences. It can lead to categorizations according to some specific risk criteria, for example, or to direct and personalized marketing. As a consequence, new forms of identities appear. They are not necessarily related to our names anymore. They are based on information, on traces that we leave when we act or interact, when we go somewhere or just stay in one place, or even sometimes when we make a choice. They are related to the SIM cards of our mobile phones, to our credit card numbers, to the pseudonyms that we use on the Internet, to our email addresses, to the IP addresses of our computers, to our profiles... Like traditional identities, these new forms of identities can allow us to distinguish an individual within a group of people, or describe this person as belonging to a community or a category. How far have we moved through this process? The identity [r]evolution is already becoming part of our daily lives. People are eager to share information with their "friends" in social networks like Facebook, in chat rooms, or in Second Life. Customers take advantage of the numerous bonus cards that are made available. Video surveillance is becoming the rule. In several countries, traditional ID documents are being replaced by biometric passports with RFID technologies. This raises several privacy issues and might actually even result in changing the perception of the concept of privacy itself, in particular by the younger generation. In the information society, our (partial) identities become the illusory masks that we choose -or that we are assigned- to interplay and communicate with each other. Rights, obligations, responsibilities, even reputation are increasingly associated with these masks. On the one hand, these masks become the key to access restricted information and to use services. On the other hand, in case of a fraud or negative reputation, the owner of such a mask can be penalized: doors remain closed, access to services is denied. Hence the current preoccupying growth of impersonation, identity-theft and other identity-related crimes. Where is the path of the identity [r]evolution leading us? The booklet is giving a glance on possible scenarios in the field of identity.

Se presenta el paradigma temporal desde la perspectiva actual de la sociedad de la información y el conocimiento. Se analiza el tiempo virtual en el ámbito de las TIC. Se estudia el tiempo en los medios de comunicación y la oferta de "tiempo real", lo inmediato, en la sociedad global y su relación con las demandas de información de los usuarios y ciudadanos. Se propone un esfuerzo ético y de transparencia en plena era global y en un mundo marcado por las tecnologías y el tiempo de la información.

Extensive programmed laboratory tests involving some 400 asphalt emulsion slurry seals (AESS) were conducted. Thirteen aggregates including nine Iowa sources, a quartzite, a synthetic aggregate (Haydite), a limestone stone from Nebraska, and a Chat aggregate from Kansas were tested in combination with four emulsions and two mineral fillers, resulting in a total of 40 material combinations. A number of meetings were held with the Iowa DOT engineers and 12 state highway departments that have had successful slurry seal experiences and records, and several slurry seal contractors and material and equipment suppliers were contacted. Asphalt emulsion slurry seal development, uses, characteristics, tests, and design methods were thoroughly reviewed in conjunction with Iowa's experiences through these meetings and discussions and through a literature search (covering some 140 articles and 12 state highway department specifications). It was found that, while asphalt emulsion slurry seals (when properly designed and constructed) can economically improve the quality and extend the life of existing pavement surface, experiences with them had been mixed due to the many material, slurry, and construction variables that affect their design, construction, and performance. The report discusses those variables identified during the course of the project and makes recommendations concerning design procedures, design criteria, specifications and the means of evaluating them.

RESUME Il a longtemps été admis que le glucose était le principal, sinon le seul substrat du métabolisme énergétique cérébral. Néanmoins, des études récentes indiquent que dans des situations particulières, d'autres substrats peuvent être employés. C'est le cas des monocarboxylates (lactate et pyruvate principalement). Bien que la barrière hématoencéphalique soit peu perméable à ces molécules, elles deviennent néanmoins des substrats possibles si elles sont produites localement. Les deux systèmes enzymatiques pivots des voies glycolytiques et oxydatives sont la lactate déshydrogénase (LDH, EC 1.1.1.27) qui catalyse l'interconversion du pyruvate et du lactate et le complexe pyruvate déshydrogénase qui catalyse la conversion irréversible du pyruvate en acétyl-CoA qui entre dans la respiration mitochondriale. Nous avons étudié la localisation, tant régionale que cellulaire, des isoformes LDH-1, LDH-5 et PDHEla dans le cerveau du chat et dé l'homme au moyen de diverses techniques histologiques. Dans un premier temps, des investigations par hybridation in situ au moyen d'oligosondes marquées au 33P sur de coupes de cerveau de chat ont permis de montrer une différence de l'expression des enzymes à vocation oxydative (LDH-1 et PDHA1, le gène codant pour la protéine PDHEIa) par rapport à LDH-5, isoforme qui catalyse préférentiellement la formation de lactate. LDH-1 et PDHA 1 ont des distributions similaires et sont enrichies dans de nombreuses structures cérébrales, comme l'hippocampe, de nombreux noyaux thalamiques et des structures pontiques. Le cortex cérébral exhibe également une expression importante de LDH-1 et PDH. LDH-5 a par contre une expression largement plus diffuse à travers le cerveau, bien que l'on trouve néanmoins un enrichissement plus important dans l'hippocampe. Ces résultats sont en accord avec les observations que nous avons précédemment publiées chez le rongeur pour LDH-1 et LDH-5 (Laughton et collaborateurs, 2000). Des analyses par PCR en temps réel ont confirmé que dans certaines régions, LDH-1 est exprimée de façon nettement plus importante que LDH-5. Dans un deuxième temps, nous avons appliqué sur des coupes histologiques d'hippocampe et de cortex occipital humain post-mortem des anticorps monoclonaux spécifiques de l'isoforme LDH-5 et la sous-unité PDHela du complexe pyruvate déshydrogénase. Là aussi, les immunoréactions révèlent une ségrégation régionale mais aussi cellulaire des deux enzymes. Dans les deux régions étudiées, LDH-5 est localisée exclusivement dans les astrocytes. Dans le cortex occipital, la matière blanche et également la couche I corticale sont immunopositives pour LDH-5. Dans l'hippocampe, le CA4 et l'alveus exhibe l'immunomarquage le plus intense pour LDH-5. Seuls des neurones (à de rares exceptions quelques astrocytes) sont immunopositifs à l'anticorps monoclonal dirigé contre PDHela. La couche IV du cortex occipital présente la plus forte immunoréaction. Dans l'hippocampe, une immunoréactivité est observée dans le stratum granulosum et à travers la région CA1 jusqu'à la région CA3. L'ensemble de ces résultats montre une hétérogénéité métabolique dans le cerveau et étaye l'hypothèse "astrocyte-neurone lactate shuttle" (ANL5) (Bittar et collaborateurs, 1996; Magistretti et Pellerin, 1999) qui propose que les astrocytes fournissent aux neurones activés du lactate comme substrat alternatif de leur métabolisme énergétique. ABSTRACT For a long time now, glucose has been thought to be the main, if not the sole substrate for brain energy metabolism. Recent data nevertheless suggest that other molecules, such as monocarboxylates (lactate and pyruvate mainly) could be suitable substrates. Although monocarboxylates poorly cross the blood brain barrier (BBB), such substrates could replace glucose if produced locally. The two key enzymatic systems required for the use and production of these substats are lactate dehydrogenase (LDH; EC 1.1.1.27) that catalyses the interconversion of lactate and pyruvate and the pyruvate dehydrogenase complex that irreversibly funnels pyruvate towards the mitochondrial TCA cycle and oxydative phosphorylation. Our study consisted in localizing these different systems with various histochemical procedures in the cat brain and two regions, i.e. hippocampus and primary visual cortex, of the human brain. First, by means of in situ hybridization with 33P labeled oligoprobes, we have demonstrated that the more oxidative enzymes (LDH-1 and PDHA1, the gene coding for PDHEla) are highly expressed in a variety of feline brain structures. These structures include the hippocampus, various thalamic nuclei and the pons. The cerebral cortex exhibits also a high LDH-1 and PDHAl expression. On the other hand, LDH-5 expression is poorer and more diffuse, although the hippocampus does seem to have a higher expression. These fmdings are consistent with our previous observation of the expression of LDH1 and LDH-5 in the rodent brain (Laughton et al, 2000). Real-time PCR (TagMan tm) revealed that, in various regions, LDH-1 is effectively more highly expressed than LDH-5. In a second set of experiments, monoclonal antibodies to LDH-5 and PDHeIa were applied to cryostat sections of post-mortem human hippocampus and occipital cortex. These procedures revealed not only that the two enzymes have different regional distributions, but also distinct cellular localisation. LDH-5 immunoreactivity is solely observed in astrocytes. In the occipital cortex, the white matter and layer I are immunopositive. In the hippocampus, the alveus and CA4 show LDH-5 immunoréactivity. PDHeIa has been detected, with few exceptions, only in neurons. Layer IV of the occipital cortex was most immmunoreactive. In the hippocampus, PDHela immunoreactivity is noticed in the stratum granulosum and through CA 1 to CA3 areas. The overall observations made in this study show that there is a metabolic heterogeneity in the brain and our findings support the hypothesis of an astrocyte-neuron lactate shuttle (ANLS)(Bittar et al., 1996; Magistretti & Pellerin, 1999) where astrocytes export to active neurons lactate to fuel their energy demands.

Interaction is a basic element in any educational process, and it is something that needs to be reconsidered in the light of technology. In order to examine the methodological changes that ICTs bring to teaching from an interaction perspective, a study was carried out at the University of Lleida to observe interaction processes in various face-to-face, blended learning and e-learning subjects. The methodological design was based on three data collection techniques: documentary analysis of subject curricula, lecturer and student questionnaires, and lecturer interviews. The data showed that, as the online component of subjects increased, the lecturers and students used more technological tools to communicate (e-mail, forums, chats, social networks, etc.). Furthermore, we found that the lecturers and students basically communicated for academic purposes. While they hardly ever communicated for personal reasons (guidance, support, etc.), they claimed that closer contact with a non-academic focus would be preferable. We also observed that the students’ work was more individual in e-learning subjects. Although there is still a considerable way to go in ICT-mediated lecturer-student interaction, both the lecturers and students recognise the potential of such technologies, even though they still do not use them as they feel they should.

La interacción, un elemento básico en cualquier proceso formativo, debe replantearse con la irrupción de la tecnología. Con la intención de abordar los cambios metodológicos que las TIC implican en la docencia desde la perspectiva de la interacción que generan, se planteó un estudio en la Universidad de Lérida para observar los procesos que interactúan en diferentes asignaturas en función de si éstas se desarrollaban bajo la modalidad presencial, semipresencial o no presencial. El diseño metodológico se articuló alrededor de tres técnicas de recogida de datos: análisis documental de programas de asignaturas, cuestionarios a profesores y estudiantes y entrevistas a profesores. Los datos mostraron que, conforme aumenta la no presencialidad de las asignaturas, profesorado y estudiantado utilizaban más herramientas tecnológicas (correo electrónico, foro, chat, redes sociales...) para comunicarse. Además, el tipo de comunicación imperante tenía finalidades académicas, mientras que se producía una escasa interacción para aspectos más personales (de orientación, apoyo...); en este sentido, tanto profesorado como estudiantado preferirían un contacto más cercano no tan centrado en los elementos académicos del proceso. También observamos que en las asignaturas en línea se desarrolla un trabajo más individual por parte del estudiantado. Aunque todavía queda camino por recorrer en la interacción docente-discente a través de las TIC, los agentes implicados en el proceso reconocen sus potencialidades, pero aún no las utilizan como consideran que convendría hacerse.

El projecte es basarà en realitzar una aplicació web amb l'entorn .NET per implementar una aplicació de xat i/o videoconferència en temps real. Aquesta aplicació necessitarà únicament un navegador per funcionar, i utilitzarà els frameworks WebRTC o SignalR, els quals ambdós són frameworks Open Source especialitzats en Real Time Comunications existents a la xarxa.
