A total of 22 honey samples from the west of Tasmania were analysed. All of them were monofloral leatherwood honeys containing between 79.9% and 99.1% of Eucryphia sp. pollen. Furthermore, 27 different pollen types were identified, the most frequent of which were Eucalyptus, Leptospermum (Myrtaceae), Trifolium (Fabaceae), Rosaceae, Banksia (Proteaceae), Lotus (Fabaceae), Bursaria (Pittosporaceae) and Brassica (Brassicaceae), which appeared in a great number of honeys with different percentages. The pollen spectrum characteristics of Tasmania's monofloral leatherwood honeys differentiate them from Chilean honeys of the same floral origin.

This study explored the relationship between coping, alcohol expectancies and drinking refusal self-efficacy in predicting drinking behaviour in both community and clinical samples. These variables were found to have differential effects in their association with frequency and volume of alcohol consumption across the two samples. Generally, drinking refusal self-efficacy was a more salient factor in relation to frequency and volume of community drinking, while coping and expectancies were more strongly associated with frequency of drinking sessions by problem drinkers. The interaction between expectancies and drinking refusal self-efficacy was related to volume of consumption in both groups, while coping and expectancies interacted in their association with frequency in the clinical group. The findings are discussed with regard to the different patterns of cognitive variables governing the decision to drink and the amount consumed in each drinking session, which may differentiate community and problem drinkers.

Background: The aim of this study was to examine minor physical anomalies and quantitative measures of the head and face in patients with psychosis vs healthy controls. Methods: Based on a comprehensive prevalence study of psychosis, we recruited 310 individuals with psychosis and 303 controls. From this sample, we matched 180 case-control pairs for age and sex. Individual minor physical anomalies and quantitative measures related to head size and facial height and depth were compared within the matched pairs. Based on all subjects, we examined the specificity of the findings by comparing craniofacial summary scores in patients with nonaffective or affective psychosis and controls. Results: The odds of having a psychotic disorder were increased in those with wider skull bases (odds ratio [OR], 1.40; 95% confidence interval [CI], 1.02-1.17), smaller lower-facial heights (glabella to subnasal) (OR, 0.57; 95% CI, 0.44-0.75), protruding ears (OR, 1.72; 95% CI, 1.05-2.82), and shorter (OR, 2.29; 95% CI, 1.37-3.82) and wider (OR, 2.28; 95% CI, 1.43-3.65) palates. Compared with controls, those with psychotic disorder had skulls that were more brachycephalic. These differences were found to distinguish patients with nonaffective and affective psychoses from controls. Conclusions: Several of the features that differentiate patients from controls relate to the development of the neuro-basicranial complex and the adjacent temporal and frontal lobes. Future research should examine both the temporal lobe and the middle cranial fossa to reconcile our anthropomorphic findings and the literature showing smaller temporal lobes in patients with schizophrenia. Closer attention to the skull base may provide clues to the nature and timing of altered brain development in patients with psychosis.

The decision of the embryonic gonad to differentiate as either a testis or an ovary is a critical step in vertebrate development. The molecular basis of this decision has been the focus of much study, particularly over the past decade. Here we contrast the knowledge of early gonadal development and the switch to testis differentiation with the lack of molecular understanding of ovarian development at early stages. We review current knowledge regarding mechanisms of ovarian morphogenesis and propose a model for the hierarchical control of development of the fetal ovary, incorporating the few genes already known to be important and several signals or factors that are hypothesised to exist in the early ovary.

Bemisia tabaci (Hemiptera: Aleyrodidae) is a haplo-diploid species with a global distribution demonstrating strong geographical structure with eight recognizable genetic groups. Fifteen microsatellite loci (335 alleles, 6-44 alleles per locus) were derived from four of the eight groups and were then screened across 33 populations. These loci clearly differentiate the populations. The microsatellites amplified best in individuals from genetic groups representing the Mediterranean, Middle East, Asia (three groups) and Australasia/Oceania and amplified less well with populations from sub-Saharan Africa and the New World. This differential amplification pattern is a direct result of the relatedness to the microsatellite source material.

While the lungfish dentition is partially understood as far as morphology and light microscopic structure is concerned, the ultrastructure is not. Each tooth plate is associated with a dental lamina that develops from the inner layer of endodermal cells that form the oral epithelium. Dentines, bone and cartilage of the jaws differentiate from mesenchyme cells aggregating beneath the oral endothelium. Enamel, in the developing and in the mature form, has similarities to that of other early vertebrates, but unusual characters appear as development proceeds. Ameloblasts are capable of secreting enamel, and, with mononuclear osteoclasts, of remodelling the bone below the tooth plate. The forms of dentine, all based largely on an extracellular matrix of collagen and mineralised with biological apatite, differ from each other and from the underlying bone in the ultrastructure of associated cells and in the mineralised extracellular matrices produced. Cell processes emerging from the odontoblasts and from the osteoblasts vary in length, degree of branching and of anastomoses between the processes, although all of the cell types have large amounts of rough endoplasmic reticulum. Mineralisation of the extracellular matrices varies among the enamel, dentines and bone in the tooth plate. In addition, the development of the hard tissues of the tooth plates indicates that many of the similarities in fine structure of the dentition in lungfish, to tissues in other fish and amphibia, apparent early in development, disappear as the dentition matures. Background Wide testing of the aldosterone: renin ratio among hypertensive individuals has revealed primary aldosteronism to be common, with most patients normokalaemic. Some investigators, however, have reported aldosterone-producing adenoma to be rare among patients so detected. Objective To test the hypothesis that differences among reported studies in the rate of detection of aldosterone-producing adenoma (as opposed to bilateral adrenal hyperplasia) reflect differences in the procedures used for diagnosis of primary aldosteronism, and the methods used to identify aldosterone-producing adenomas. Methods In the newly established Princess Alexandra Hospital Hypertension Unit (PAHHU), we used procedures developed by Greenslopes Hospital Hypertension Unit (which reports that more than 30% of patients with primary aldosteronism have aldosterone-producing adenomas) to diagnose primary aldosteronism and determine the subtype. All patients with an increased aldosterone: renin ratio (measured after correction for hypokalaemia and while the patient was not receiving interfering medications) underwent fludrocortisone suppression testing to confirm or exclude primary aldosteronism; if they were positive, they underwent genetic testing to exclude glucocorticoid-remediable aldosteronism before adrenal venous sampling was used to differentiate lateralizing from bilateral primary aldosteronism. Results This approach allowed PAHHU to diagnose, within 2 years, 54 patients [only seven (13%) hypokalaemic] with primary aldosteronism. All tested negative for glucocorticoid-remediable aldosteronism. Aldosterone production was lateralized to one adrenal in 15 patients (31%; only six hypokalaemic) and was bilateral in 34 (69%; all normokalaemic) of 49 patients who underwent adrenal venous sampling. Among patients with lateralizing adrenal hyperplasia, computed tomography revealed an ipsilateral mass in only six and a contralateral lesion in one. Fourteen patients underwent unilateral adrenalectomy, which cured the hypertension in seven and improved it in the remainder. In patients with bilateral primary aldlosteronism, hypertension responded to spironolactone (112.5-50 mg/ day) or amiloride (2.5-10 mg/day). Conclusion When performed with careful regard to confounding factors, measurement of the aldosterone: renin ratio in all hypertensive individuals, followed by fludrocortisone suppression testing to confirm or exclude primary aldosteronism and adrenal venous sampling to determine the subtype, can result in the detection of significant numbers of patients with specifically treatable or potentially curable hypertension. Proteolysis of UHT milk during storage at room temperature is a major factor limiting its shelf-life through changes in its flavour and texture. The latter is characterised by increases in viscosity leading in some cases to gel formation. The enzymes responsible for the proteolysis are the native milk alkaline proteinase, plasmin, and heat-stable, extracellular bacterial proteinases produced by psychrotrophic bacterial contaminants in the milk prior to heat processing. These proteinases react differently with the milk proteins and produce different peptides in the UHT milk. In order to differentiate these peptide products, reversed-phase HPLC and the fluorescamine method were used to analyse the peptides soluble in 12% trichloroacetic acid (TCA) and those soluble at pH 4.6. The TCA filtrate showed substantial peptide peaks only if the milk was contaminated by bacterial proteinase, while the pH 4.6 filtrate showed peptide peaks when either or both bacterial and native milk proteinases caused the proteolysis. Results from the fluorescamine test were in accordance with the HPLC results whereby the TCA filtrate exhibited significant proteolysis values only when bacterial proteinases were present, but the pH 4.6 filtrates showed significant values when the milk contained either or both types of proteinase. A procedure based on these analyses is proposed as a diagnostic test for determining which type of proteinase-milk plasmin, bacterial proteinase, or both-is responsible for proteolysis in UHT milk. Recent advances in molecular biology have made it possible to use the trace amounts of DNA in faeces to non-invasively sample endangered species for genetic studies. Here we use faeces as a source of DNA and mtDNA sequence data to elucidate the relationship among Spanish and Moroccan populations of great bustards. 834 bp of combined control region and cytochrome-b mtDNA fragments revealed four variable sites that defined seven closely related haplotypes in 54 individuals. Morocco was fixed for a single mtDNA haplotype that occurs at moderate frequency (28%) in Spain. We could not differentiate among the sampled Spanish populations of Caceres and Andalucia but these combined populations were differentiated from the Moroccan population. Estimates of gene flow (Nm = 0.82) are consistent with extensive observations on the southern Iberian peninsular indicating that few individuals fly across the Strait of Gibraltar. We demonstrate that both this sea barrier and mountain barriers in Spain limit dispersal among adjacent great bustard populations to a similar extent. The Moroccan population is of high ornithological significance as it holds the only population of great bustards in Africa. This population is critically small and genetic and observational data indicate that it is unlikely to be recolonised via immigration from Spain should it be extirpated. In light of the evidence presented here it deserves the maximum level of protection.

A intensa atividade humana devasta grandes extensões de florestas nativas, seja para expansão da agricultura seja para suprir a crescente demanda do mercado por madeira, de uma forma ou de outra os pequenos fragmentos florestais remanescentes sofrem constantes pressões antrópicas, contudo, o uso de espécies arbóreas adaptadas pode contribuir para a proteção desses recursos naturais. Buscou-se neste estudo avaliar os padrões fenológicos de 109 espécies florestais arbóreas pertencentes a 37 famílias e 82 gêneros, dentre nativas e exóticas, com nove anos de idade, para diferenciar as mais adaptadas e com potencial de serem utilizadas em programas de reflorestamento. O estudo foi realizado na Fazenda Experimental do Incaper, em Jucuruaba, município de Viana-ES, (UTM E-345524, N- 7741039). Foram realizadas análises químicas do solo na área plantada e os dados climatológicos obtidos na estação meteorológica de Viana. O estudo baseou-se na observação do número de plantas sobreviventes de cada espécie e da avaliação do seu crescimento. Foram realizadas observações das fenofases de brotação, senescência de folhas, floração e frutificação. As avaliações fenológicas foram realizadas em intervalos mensais, no período de novembro de 2012 a outubro de 2013. Realizou-se a medição da altura das árvores, diâmetro à altura do peito (DAP), índice de enfolhamento, taxa de sobrevivência e cálculo do ICC (Índice Combinado de Crescimento), bem como a determinação das espécies mais adaptadas. Das 109 espécies estudadas, 64,22% apresentaram adaptação funcional e estrutural às condições de solo e clima da região experimental, 42,22 % floresceram e frutificaram e 90% apresentaram senescência e brotação acompanhando a sazonalidade climática. Vinte e nove espécies apresentaram ICC maior do que o ICC médio. A maioria das espécies destacou-se como alternativa para recuperação da cobertura vegetal local, com destaque para Inga uruguensis e Schizolobium amazonicum. As análises de fluorescência da clorofila revelaram que o aparato fotossintético da Schizolobium amazonicum foi capaz de protegê-la da fotoinibição e promover boa conversão da energia luminosa em fotoquímica.

A retinocoroidite é a manifestação mais comum causada pela infecção congênita por Toxoplasma gondii. Devido a gravidade das lesões oculares que podem até levar à perda completa da visão, a detecção precoce da toxoplasmose congênita e da lesão ocular são essenciais para o tratamento. Este trabalho possuiu o objetivo de avaliar a aplicabilidade da pesquisa de anticorpos IgG e das subclasses IgG1, IgG2, IgG3 e IgG4 anti-T. gondii por citometria de fluxo como marcador laboratorial das diferentes formas de lesões retinocoroidais na toxoplasmose congênita. Foram analisadas 88 amostras de soro de recém-nascidos com toxoplasmose congênita, sendo 25 sem lesão ocular (SL), 10 com lesão ocular ativa (RA), 26 com lesão ocular ativa e cicatricial (RAC) e 27 com lesão ocular cicatricial (RC). Foram também utilizadas 19 amostras de soro de recém-nascidos não infectados que apresentaram IgG positivo após o nascimento (NI). Essas amostras foram obtidas a partir de soros de recémnascidos participantes de um programa de triagem neonatal realizado em Minas Gerais realizado nos anos de 2006 e 2007. Os resultados demonstraram que os recém-nascidos com toxoplasmose congênita apresentam maior reatividade de anticorpos IgG total e subclasses IgG1, IgG2 e IgG3 do que indivíduos não infectados. No grupo não infectado, o único anticorpo com mais de 50% de indivíduos com alta reatividade de anticorpos foi IgG4. Ao comparar os grupos de indivíduos com toxoplasmose congênita, foi observado que o grupo RAC, seguido de RC, apresentou maior reatividade principalmente para os anticorpos IgG1 e IgG3 comparado aos recém-nascidos dos grupos RA e SL, enquanto que pacientes do grupo RA apresentaram maior reatividade para IgG4 do que indivíduos dos outros grupos. IgG1 foi a única subclasse capaz de diferenciar os grupos NI, SL dos grupos RAC e RC. Também foi avaliado o índice de avidez de IgG total, que não permitiu estabelecer nenhum critério de diferenciação das formas de lesão ocular causadas pelo T. gondii. Portanto, a citometria de fluxo demonstrou que pode ser um método laboratorial complementar para ser utilizado como indicador das diferentes lesões oculares causadas pela toxoplasmose congênita.

O estresse pode afetar qualquer pessoa, independente de idade, sexo ou etnia. O organismo humano o utiliza como uma resposta adaptativa frente a situações diversas, as quais requeiram alguma adaptação do organismo para que possa enfrentar tal situação. Dependendo do estímulo estressor, pode ser gerado no indivíduo desgastes físico, mental ou emocional, no entanto, o estresse não representa necessariamente algo ruim ou patológico; este é um mecanismo de adaptação vital para a sobrevivência da espécie humana. Porém, o número de pessoas que são afetadas de forma negativa pelo estresse tem crescido imensamente nas últimas décadas. Pesquisas destacam que nos Estados Unidos cerca de 60% a 90% dos atendimentos médicos estão relacionados de alguma maneira com o estresse, enquanto que no Brasil aproximadamente 80% da população sofre de estresse, sendo que desses, 30% encontram-se na fase mais crítica, a chamada fase de exaustão. Tendo em vista que a principal forma de identificação de estresse ainda é realizada através do uso de questionário de autorrelato. O presente estudo apresenta como contribuição uma metodologia de análise do nível de estresse baseada na variação da condutância galvânica da pele e de sinais de eletroencefalografia, sendo utilizados como parâmetros a assimetria do ritmo alfa, assim como a razão entre os ritmos beta e alfa no córtex frontal e pré-frontal. Para a gravação dos sinais de EEG foi utilizado um dispositivo portátil, com eletrodos especificamente situados nas posições aF3, F3, F4 e aF4, de acordo com o Sistema Internacional 10/20 de posicionamento de eletrodos. Os participantes deste estudo são Bombeiros Militares da 1ª Cia de Vitória-ES. Foram utilizadas três classes de estímulos emocionais positivos, calmos e negativos, através da utilização de imagens pertencentes ao banco de dados IAPS (International Affective Picture System). Os resultados de acurácia obtidos através de um classificador SVM (Support Vector Machine) chegam a 88,24% para classe de estímulos positivos, 84,09% para classe calma e de 92,86% para os estímulos negativos. Deste modo, esta pesquisa apresenta uma combinação de parâmetros que podem ser aferidos com equipamentos de baixo custo, e fornecem condições de diferenciar estímulos estressantes, podendo assim, ser utilizada para auxiliar no treinamento de profissionais da área de urgência e emergência.

To date few studies have been undertaken in Portugal dealing with the attitudes, motivations, and profile of tourists who visit World Heritage Sites. Also, few studies have dealt with destination image (e.g., Agapito, Mendes & Valle, 2010; Lopes, 2011). As far as it is known, none have approached the issue of gender differences in the choice of a Portuguese heritage destination. Since cultural tourism destinations need to differentiate themselves from each other, appropriate market segmentation must be based on a deep understanding of the customers’ motivations and preferences. Keeping in mind results from empirical literature (e.g., Silberberg, 1995; Beerli & Martin, 2004; Richards, 2004; Pérez, 2009; Sheng, Shen, & Chen, 2008), gender seems to be a possible approach to market segmentation, whether for Guimarães or for other cultural tourism destinations around the world. Located in the north-western region of Portugal, Guimarães is a city of strong symbolic and cultural significance, and the nomination of its historical centre as a World Heritage Site in 2001 enhanced its tourism potential. This study analyses the possible relation between gender and attitudes and motivations towards a World Heritage Site, such as Guimarães. Additionally, the empirical approach used in the study tries to capture differences in the perceived attributes of the city. Commonalities and distinctions within and between groups of tourists, by focusing on the specific characteristic of gender, were analysed. The study addressed two main questions: first, whether males and females have similar or different preferences in choosing the city as their destination; and, second, whether there are gender differences in the perception of the attributes of Guimarães. A better understanding of the gendered nature of the destination is a valuable cue for shaping products and services according to visitors’ preferences.

Discovering the ways through which firms develop and maintain competitive advantage is a central research stream in management theory. The objective of this paper is to present a contribution to the discussion of the knowledge of the firm as a source of competitive advantage. The paper states that a firm's success is a consequence of its ability in the continuous development of core competencies that will sustain its competitiveness over time. Core competencies are understood as the sets of knowledge that differentiate a company strategically. The firm must discover, develop, share and update the knowledge that sustains the present and future core competencies. Knowledge management, through processes of knowledge creation and integration, is one way of doing this.

RESUMO: Hoje em dia o software tornou-se num elemento útil na vida das pessoas e das empresas. Existe cada vez mais a necessidade de utilização de aplicações de qualidade, com o objectivo das empresas se diferenciarem no mercado. As empresas produtoras de software procuram aumentar a qualidade nos seus processos de desenvolvimento, com o objectivo de garantir a qualidade do produto final. A dimensão e complexidade do software aumentam a probabilidade do aparecimento de não-conformidades nestes produtos, resultando daí o interesse pela actividade de testes de software ao longo de todo o seu processo de concepção, desenvolvimento e manutenção. Muitos projectos de desenvolvimento de software são entregues com atraso por se verificar que na data prevista para a sua conclusão não têm um desempenho satisfatório ou por não serem confiáveis, ou ainda por serem difíceis de manter. Um bom planeamento das actividades de produção de software significa usualmente um aumento da eficiência de todo o processo produtivo, pois poderá diminuir a quantidade de defeitos e os custos que decorrem da sua correcção, aumentando a confiança na utilização do software e a facilidade da sua operação e manutenção. Assim se reconhece a importância da adopção de boas práticas no desenvolvimento do software. Para isso deve-se utilizar uma abordagem sistemática e organizada com o intuito de produzir software de qualidade. Esta tese descreve os principais modelos de desenvolvimento de software, a importância da engenharia dos requisitos, os processos de testes e principais validações da qualidade de software e como algumas empresas utilizam estes princípios no seu dia-a-dia, com o intuito de produzir um produto final mais fiável. Descreve ainda alguns exemplos como complemento ao contexto da tese. ABSTRACT: Nowadays the software has become a useful element in people's lives and it is increasingly a need for the use of quality applications from companies in order to differentiate in the market. The producers of software increase quality in their development processes, in order to ensuring final product quality. The complexity and size of software, increases the probability of the emergence of non-conformities in these products, this reason increases of interest in the business of testing software throughout the process design, development and maintenance. Many software development projects are postpone because in the date for delivered it’s has not performed satisfactorily, not to be trusted, or because it’s harder to maintain. A good planning of software production activities, usually means an increase in the efficiency of all production process, because it can decrease the number of defects and the costs of it’s correction, increasing the reliability of software in use, and make it easy to operate and maintenance. In this manner, it’s recognized the importance of adopting best practices in software development. To produce quality software, a systematic and organized approach must be used. This thesis describes the main models of software development, the importance of requirements engineering, testing processes and key validation of software quality and how some companies use these principles daily, in order to produce a final product more reliable. It also describes some examples in addition to the context of this thesis.
