A standardised nested-PCR method that amplifies a region of the glycoprotein E gene of avian infectious laryngotracheitis virus (ILTV) has been developed for the diagnosis of infection by Gallid herpesvirus 1. The two sets of primers employed produced the expected ampIification products of 524bp(externa I primers) and 219bp (internal primers) in the presence of ILTV DNA, whereas no Such amplicons were obtained with other avian respiratory pathogens or with DNA extracted from the cells of uninfected chickens. The identity of the 219bp amplified product was con firmed by DNA sequencing. The standardised nested-PCR method detected ILTV DNA from trachea, lung, conjunctiva and trigeminal ganglia samples from flocks of birds with and without clinical signs. and showed hi.-h sensitivity (95.4%) and specificity (93.1%) when compared with the reference test involving virus isolation in specific-pathogen-free chicken embryos. The standardised nested-PCR method described may be used to detect clinical and latent ILTV infections, and will be of significant value for both diagnostic and epidemiological Studies. A person working for the Centre for Indigenous Health, Education and Research provides an insight into the personal journey of an Indigenous professional embarking on a career in health and research, specifying the difficulties and problems within the course of development. He suggests that to increase the number and level of involvement of Indigenous researchers in research field the need for providing the opportunity for Indigenous people should be considered.

In a previous study (Jones and Smith, 1999) we established that much the same core pattern of national identity characterizes many developed countries. Using the national identity module from the 1995 International Social Survey Programme, we identified two dimensions of national identity: an ascriptive dimension resembling the concept of ethnic identity described in the historical and theoretical literature, and a voluntarist dimension closer to the notion of civic identity. Some writers view these dimensions in terms of a historical sequence but we find that both constructs coexist in the minds of individual respondents in the nations we examine (we exclude Bulgaria and the Philippines from the present but not the earlier analysis). The dataset used for the multilevel analyses reported here consists of 28 589 respondents in the remaining 21 countries included in the national identity database for the 1995 round of surveys. The macrosociological literature on national identity does not offer well-defined predictions about what precise patterns of national identification we might expect to find among the masses of the developed countries. There are, however, recurring themes from which one can construct plausible hypotheses about how countries might differ according to their level of development, broadly conceived. Thus, we hypothesize that forces such as post-industrialism and globalization tend to favour the more open voluntaristic form of national identity over the more restrictive ascribed form. We develop different multi-level models in order to evaluate specific hypotheses pertaining to such issues, by simultaneously relating individual and societal characteristics to the relative strength of individual commitment to these different types of national identity.

Frizzled genes encode a family of Wnt ligand receptors, which have a conserved cysteine-rich Wnt binding domain and include both transmembrane and secreted forms. Work by others has shown that experimental perturbation of Wnt signaling results in aberrant hair formation, hair growth, and hair structure. To date, however, there is no information on the contribution of individual Frizzled proteins to hair development. We now report that Frizzled-3 expression in skin is restricted to the epidermis and to the developing hair follicle. Northern analysis on total mouse skin mRNA revealed a single Frizzled-3 transcript of 3.7 kb. Reverse transcription-polymerase chain reaction and in situ hybridization analysis revealed Frizzled-3 expression in epidermal and hair follicle keratinocytes. Frizzled-3 transcripts are first detected in discrete foci in the developing epidermis of 13 d embryos and later in the hair follicle placodes of 15 d embryos, suggesting a role for this Frizzled isoform in follicle development. In 17 d embryos and id old newborn mice Frizzled-3 expression is limited to suprabasal keratinocytes and is not seen in pelage follicles until 3 d postpartum. In 7 d old neonatal skin, Frizzled-3 is expressed throughout the epidermis and in the outer cell layers of hair follicles. We have also identified the mRNA encoding human Frizzled-3 in epidermal keratinocytes and in the HaCaT keratinocyte cell line. Human Frizzled-3 mRNA encodes a 666 amino acid protein with 97.8% identity to the mouse protein. The human Frizzled-3 gene was mapped using a radiation-hybrid cell line panel to the short arm of chromosome 8 between the markers WI-1172 and WI-8496 near the loci for the Hypotrichosis of Marie Unna and Hairless genes.

Sorghum ergot was first discovered in Australia in 1996. It affects seed production and grain usage in stock feed due to concerns of animal toxicity. Three species of Claviceps are known to cause ergot of sorghum with different epidemiological, animal toxicity, and management implications. Claviceps africana was identified as the causal agent but morphological differences between isolates raised the possibility of more than one species being involved. The major aim of this study was to identify the Claviceps species causing sorghum ergot and to determine the genetic diversity among isolates of the ergot pathogen from Australia and overseas. Symptom development, sequencing of the ITS1 region, and radiolabelled DNA amplification fingerprints (RAF) were used to confirm that ergot of sorghum in Australia is caused by C. africana. The morphology of sphacelia, microconidia, macroconidia, and secondary conidia of all 36 Australian isolates studied matched the description for C. africana and the DNA sequence of the ITS1 region of 2 selected Australian isolates was identical to that of C. africana. This report sets out the findings and recommendations of a review of the Public Management Career (EPPGG) undertaken between 5 and 9 January 2009. The purpose of the review was to clarify EPPGG???s role, functions and profile, and to develop proposals for improving its effectiveness and management, taking account of the context within which such improvements would have to be implemented. Specific objectives were to examine a number of human resources management (HRM) aspects of the career in the light of international practice, including recruitment; initial education; professional development; performance evaluation; deployment; mobility and networking.

Este artigo busca revelar o processo de desenvolvimento da categoria dos auditores fiscais da Receita Federal do Brasil (AFRFBs), a partir das mudanças no contexto social e profissional, e a forma como essas mutações concorreram para a construção de uma identidade profissional própria desses servidores. O que se pretende é entender como o contexto político-econômico vem alterando as percepções que esses profissionais têm de si próprios e como as reformas transformaram o modo de exercerem suas funções. A pesquisa, que se dá a partir de uma proposta de transação "quanti-quali", permitiu explorar, na trilha teórica de Dubar, aspectos relevantes do contínuo processo de construção das identidades profissionais ou de perfis identitários desses servidores. Os conhecimentos sobre o trabalho e as formas de identificação profissional na categoria dos auditores fiscais da Receita Federal do Brasil podem contribuir para a reflexão sobre relações de trabalho e processos de gestão pública no Brasil.

The idiomatic expression “In Rome be a Roman” can be applied to leadership training and development as well. Leaders who can act as role models inspire other future leaders in their behaviour, attitudes and ways of thinking. Based on two examples of current leaders in the fields of Politics and Public Administration, I support the idea that exposure to role models during their training was decisive for their career paths and current activities as prominent characters in their profession. Issues such as how students should be prepared for community or national leadership as well as cross-cultural engagement are raised here. The hypothesis of transculturalism and cross-cultural commitment as a factor of leadership is presented. Based on current literature on Leadership as well as the presented case studies, I expect to raise a debate focusing on strategies for improving leaders’ training in their cross-cultural awareness.

A problemática da inserção profissional dos novos professores tem sido largamente estudada sob várias perspetivas das quais destacamos, as dificuldades enfrentadas no início do seu desempenho como docentes (Alen y Sardi, 2009) Krichesky y Merodo (2009), o vinculo entre inserção e identidade profissional (Martineau y Portelance, 2005), o desenvolvimento da competência ética nos novos professores (St. Vincent 2011) e as trajetórias de emprego, entre outras.Os resultados destas pesquisas têm revelado o tipo de problemas enfrentados pelos novos professores à entrada na carreira docente e confirmado que nem todos os percursos se iniciam de modo tranquilo e sereno. É esta a problemática que serve de base ao objeto de estudo desta dissertação, pretendendo-se mais precisamente, centrar a nossa pesquisa no percurso profissional de seis professores diplomados pela Escola Superior de Educação de Lisboa há dois anos. O objetivo geral do estudo é, assim, descrever e compreender o desenvolvimento profissional desses professores, identificar as dimensões mais relevantes, os fatores que o condicionam e os contextos que o limitam ou facilitam. Tendo em conta o objeto de estudo identificaram-se as seguintes questões: 1) Quais as implicações da formação inicial no desempenho da profissão? 2) Quais as principais dificuldades / problemas com que se defrontaram os novos professores na inserção na profissão?; 3) Como atuam os professores principiantes?; O que mudam nas suas práticas, o que perdura?; 4) Como se incrementa o sentimento de pertença ao novo grupo profissional?; 5) Quais as particularidades dos contextos de trabalho em que estes professores se encontram integrados que se revelam facilitadoras e/ou constrangedoras da sua inserção profissional? Para o efeito recorremos a uma metodologia qualitativa, e privilegiámos como técnica de recolha de dados a entrevista semiestruturada. Para o tratamento dos dados sustentámo-nos na análise de conteúdo seguindo os passos recomendados para a análise de conteúdo (Bardin, 2009). Os resultados do estudo permitiram identificar os constrangimentos experimentados por estes professores à entrada da profissão, assim como os modos como os ultrapassaram. Destes, destacam-se dificuldades relacionadas com a falta de autonomia, com aspetos de natureza metodológica, ao nível da organização e desenvolvimento curricular e de relacionamento com os elementos da direção das instituições nos contextos de trabalho.Abstract The issue of employability of new teachers has been widely studied from various perspectives. Research has revealed the sort of problems faced by new teachers at the beginning of their career. It confirms that frequently it is not a peaceful and serene start. We highlight the following difficulties: the link between integration and professional identity (Alen y Sardi, 2009; Krichesky y Merodo, 2009), the development of ethical competence in new teachers (y Portelance Martineau, 2005), and the professional path (St. Vincent 2011). This is the hypothesis explored in this thesis. Specifically, we focus our research on the career of six graduate teachers by the Lisbon School of Education. The main objective is to describe and understand the professional development of these teachers. For that we identify the most relevant dimensions, the factors that constrain and contexts that limit or facilitate their development. A qualitative methodology was used throughout the study. Semi structured interviews were the main data collection process. Data treatment followed the recommended framework for content analysis (Bardin, 2009). The following research questions were put forward: 1) What are the impacts of base level of knowledge in the professional performance? 2) What are the main difficulties / problems that new teachers face? 3) How do novice teachers perform? What changes in their methods, what endures? 4) What increases the feeling of belonging to the new professional group? 5) What are the peculiarities of work contexts that act as facilitators and/or barriers to professional acceptance? The results allowed identifying the constraints experienced by teachers at the beginning of their careers, and the ways to surpass them. We highlight the difficulties related to: lack of autonomy; methodologies; curricular organization and development; and the relationship with the directors at their workplace.
