We report on the nonlinear (NL) optical properties of glassy TeO2-GeO2-K2O-Bi2O3 at lambda = 800 nm and lambda = 1,064 nm. Using the Kerr gate technique with a laser delivering 150 fs pulses at 800 nm, we demonstrated the fast NL response of the samples. The modulus of the NL refractive index, n (2), at 800 nm was similar to 10(-15) cm(2)/W. The Z-scan technique was used to determine n (2) a parts per thousand +10(-15) cm(2)/W, at 1,064 nm with pulses of 17 ps. The two-photon absorption coefficient, alpha (2), was smaller than the minimum that we can measure (< 0.003 cm/GW). The figure of merit n (2)/alpha (2) lambda was calculated and indicates that this glass composition has large potential to be used for all-optical switching.

The relentless pursuit for materials containing rare earth ions with photoluminescent properties has led to several studies with applications in the development of new technologies. The main focus of this work is the preparation of Er3+-doped polycrystalline Y2O3 with photoluminescent properties using PEG as an organic precursor and heat-treated at different temperatures. The methodology used in this synthesis is highly attractive due to its high feasibility for improved technology and low cost for preparing materials. The behavior of the viscous resin has been evaluated and the final compounds exhibited the formation of a cubic polycrystalline phase, which is able to support variations in Er3+ doping concentrations up to 10 mol%, without significant changes in the polycrystalline parameters. The values of the nanocrystallite size calculated by Scherrer's equation showed direct dependence on the heat-treatment temperature as well as the Er3+ concentration. Intense emission in the visible region under excitation at 980 nm was attributed to an upconversion phenomenon assigned to the intraconfigurational f-f transitions of Er3+ ions. The upconversion mechanism was investigated and it was demonstrated that the higher intense emission in the red region in comparison to the emission in the green region is related to the crystallite size. The studies about the intensity showed the dependence of upconversion emission of power source, indicating that two-photon are responsible for the green and red photoluminescence. These polycrystalline materials exhibit properties that make them promising for use in solar energy systems, C-telecom band or solid-state laser devices. (C) 2014 Elsevier B.V. All rights reserved.

Functionalization with surfactants and with active molecules of deoxyribonucleic acid (DNA), thin film processing as well as their nonlinear optical and electrical properties are reviewed and discussed. On the basis of a quantum three level model, we show that the anomalous concentration variation of cubic susceptibility chi((3))(-3 omega; omega, omega, omega) in thin films of DNA-CTMA complexes doped with Disperse Red 1 chromophore can be explained by the concentration variation of two-photon resonance contribution. We show also that the DNA complexes, plasticized with glycerol and adequately doped can be processed into self standing conducting membranes with a high electrical conductivity. The measured ionic conductivity at room temperature, depending on dopant used and its concentration, is in the range of 3.5 x 10(-4)-10(-5) S/cm and increases linearly as a function of temperature, reaching 10(-3) S/cm at 358 K for the most conducting sample, obeying predominantly the Arrhenius law. The use of laser light to modify the material's surface or bulk as well as to induce changes in the volume through a chemical reaction has received great attention in the last few years, due to the possibility of tailoring the material's properties aiming at technological applications. Here, we report on recent progress of microstructuring and microfabrication in polymeric materials by using femtosecond lasers. In the first part, we describe how polymeric materials' micromachining, either on the surface or bulk, can be employed to change their optical and chemical properties promising for fabricating waveguides, resonators, and self-cleaning surfaces. In the second part, we discuss how two-photon absorption polymerization can be used to fabricate active microstructures by doping the basic resin with molecules presenting biological and optical properties of interest. Such microstructures can be used to fabricate devices with applications in optics, such as microLED, waveguides, and also in medicine, such as scaffolds for tissue growth.

Abstract Background Delignification pretreatments of biomass and methods to assess their efficacy are crucial for biomass-to-biofuels research and technology. Here, we applied confocal and fluorescence lifetime imaging microscopy (FLIM) using one- and two-photon excitation to map the lignin distribution within bagasse fibers pretreated with acid and alkali. The evaluated spectra and decay times are correlated with previously calculated lignin fractions. We have also investigated the influence of the pretreatment on the lignin distribution in the cell wall by analyzing the changes in the fluorescence characteristics using two-photon excitation. Eucalyptus fibers were also analyzed for comparison. Results Fluorescence spectra and variations of the decay time correlate well with the delignification yield and the lignin distribution. The decay dependences are considered two-exponential, one with a rapid (τ1) and the other with a slow (τ2) decay time. The fastest decay is associated to concentrated lignin in the bagasse and has a low sensitivity to the treatment. The fluorescence decay time became longer with the increase of the alkali concentration used in the treatment, which corresponds to lignin emission in a less concentrated environment. In addition, the two-photon fluorescence spectrum is very sensitive to lignin content and accumulation in the cell wall, broadening with the acid pretreatment and narrowing with the alkali one. Heterogeneity of the pretreated cell wall was observed. Conclusions Our results reveal lignin domains with different concentration levels. The acid pretreatment caused a disorder in the arrangement of lignin and its accumulation in the external border of the cell wall. The alkali pretreatment efficiently removed lignin from the middle of the bagasse fibers, but was less effective in its removal from their surfaces. Our results evidenced a strong correlation between the decay times of the lignin fluorescence and its distribution within the cell wall. A new variety of lignin fluorescence states were accessed by two-photon excitation, which allowed an even broader, but complementary, optical characterization of lignocellulosic materials. These results suggest that the lignin arrangement in untreated bagasse fiber is based on a well-organized nanoenvironment that favors a very low level of interaction between the molecules.

The nonlinear index of refraction (n(2)) and the two-photon absorption coefficient (beta) of water-based ferrofluids made of magnetite nanocrystals of different sizes and with different coatings have been measured through the Z-scan technique, with ultrashort (femtoseconds) laser pulses. Their third-order susceptibility is calculated from the values of n(2) and beta. The influence of different particles' coatings and sizes on these nonlinear optical properties are investigated. The values of n(2) and beta depend more significantly on the nanoparticles' size than on the particular coating. We observe a decrease of beta as the nanoparticles' diameters decrease, although the optical gap is found to be the same for all samples. The results are interpreted considering modifications in the electronic orbital shape due to the particles' nanosize effect.

Intravital imaging techniques are the best approach to investigate in situ cellular behavior under physiological conditions. Many techniques have emerged during these last few years for this purpose. We recently described an intravital imaging technique that allows for the observation of placenta physiological responses at the labyrinth layer of this tissue. This technique will be very useful to study many placental opportunistic infections and in this article we reinforce its usefulness by analyzing placental physiological entrapment of beads and parasites. In particular, our results show that small beads (1.0 μm) or Plasmodium chabaudi-GFP-infected-Red Blood Cells (Pc-GFP-iRBCs) cannot get trapped inside small or large blood vessels of popliteal lymph nodes (PLNs). Inside the placenta, clusters of beads could only be found inside the maternal blood vessels. However, Pc-GFP-iRBCs were found inside and outside the maternal blood vessels. We observed that trophoblasts can ingest infected-Red Blood Cells (iRBCs) in vitro and immunofluorescence of placenta revealed Pc-GFP-iRBCs inside and outside the maternal blood vessels. Taken together, we conclude that fast deposition of particles inside blood vessels seems to be an intrinsic characteristic of placenta blood flow, but iRBCs could be internalized by trophoblast cells. Thus these results represent one of the many possible uses of our intravital imaging technique to address important questions inside the parasitological field.

This work reports on the two-photon absorption (2PA) cross-section and first hyperpolarizability of dibenzoylmethane solutions using femtosecond Z-scan and hyper-Rayleigh scattering techniques. The 2PA spectrum, spanning the wavelength range from 460 to 740 nm, presents a band centered at 510 nm, with a cross-section value estimated as 37 GM at this wavelength. Owing to the molecular symmetry, this band is not observed in the linear absorption spectrum. The sum-over-state approach was adopted to evaluate various spectroscopic parameters. Experimental and theoretical values of the first hyperpolarizability values were estimated in ethanol and DMSO solutions.

Tailoring properties of materials by femtosecond laser processing has been proposed in the last decade as a powerful approach for technological applications, ranging from optics to biology. Although most of the research output in this field is related to femtosecond laser processing of single either organic or inorganic materials, more recently a similar approach has been proposed to develop advanced hybrid nanomaterials. Here, we report results on the use of femtosecond lasers to process hybrid nanomaterials, composed of polymeric and glassy matrices containing metal or semiconductor nanostructures. We present results on the use of femtosecond pulses to induce Cu and Ag nanoparticles in the bulk of borate and borosilicate glasses, which can be applied for a new generation of waveguides. We also report on 3D polymeric structures, fabricated by two-photon polymerization, containing Au and ZnO nanostructures, with intense two-photon fluorescent properties. The approach based on femtosecond laser processing to fabricate hybrid materials containing metal or semiconductor nanostructures is promising to be exploited for optical sensors and photonics devices.

In dieser Arbeit wurden Untersuchungen mit verschiedenenMethoden derMikrospektroskopie unter Verwendung einesPhotoemissions-Elektronenmikroskops(PEEM) durchgeführt. Es wurde ein Energiefilterfür den Einsatzmit dem PEEM aufgebaut und getestet. Dieses Instrument wurdezusammen miteinem Multilayer-Monochromator am Synchrotron eingesetzt.Der Energiefilterwurde unter anderem für Photoelektronenspektroskopie anAuxCs1-xund Pb/Si-Schichten, sowie zur Charakterisierung desMonochromators eingesetzt.
Die Transmissionsfunktion eines abbildendenGegenfeldanalysators fürdas PEEM wurde berechnet und mit Messungen verglichen. InÜbereinstimmungmit der Berechnung zeigt sich ein charakteristischesasymmetrisches Profilder mit dem Analysator gemessenen Photoelektronenpeaks. Eswurde eine Verbesserungder Ortsauflösung durch energieselektive Abbildungnachgewiesen.
Mit dem PEEM wurde erstmals Mehr-Photonen Photoemissionzur Abbildungausgenutzt. Es wurde ein Verfahren gezeigt, mit dem sich dieOrdnung dernichtlinearen Photoemission aus zwei mit unterschiedlicherLaserleistungaufgenommenen Bildern lateral abbilden läßt. DiePolarisationsabhängigkeitder Photoemission von glatten Probenstellen läßtsich mit einercos4-Funktion anpassen, wie es fürZwei-Photonen Photoemissionzu erwarten ist. Alle Proben wiesen Zentren erhöhterPhotoemissionsintensitätauf, welche durch Anregung lokalisierter Plasmonen auf derOberflächean kleinen Partikeln entstehen. An diesen erstmalig sichtbargemachten'Hot Spots' zeigt sich eine charakteristischePolarisationsabhängigkeitder Photoemission mit bevorzugter Anregung vonPhotoelektronen bei s-polarisiertemLicht.

Zusammenfassung Kooperativität bei Atmungsproteinen bedeutet eine Änderung der O2-Affinität während der Beladung mit O2 und läßt sich durch die Wechselbeziehung zwischen O2-Beladung und Konformation beschreiben. In dieser Arbeit wurde das 24-mere Hämocyanin der Vogelspinne Eurypelma californicum bzgl. Beladung und Konformationen sowohl auf der Ensemble- wie auch auf der Einzel-Molekül-Ebene charakterisiert. EnsembleDie Bindung von O2 an Hämocyanine ist mit einer drastischen Abnahme der Fluoreszenz-Quantenausbeute verbunden. Durch Vergleich von theoretisch und experimentell bestimmten Quantenausbeuten konnte gezeigt werden, daß die Löschung auf Förster-Transfer zurückzuführen ist, und daß kein Einfluß der Oligomerisierung, Protein-Konformation, Beweglichkeit oder Tierart besteht. Die Konformation von Hämocyaninen konnte mit Crosslinkern im oxy- und deoxy-Zustand fixiert werden. Die Charakterisierung der Produkte führte zu einer neuen Vorstellung, wie unterschiedliche Affinitäten realisiert sein können. Hierbei kommt der Dynamik der Protein-Matrix eine entscheidende Rolle zu. Einzel-MoleküleMittels Zwei-Photonen-Anregung konnten erstmalig einzelne Proteine über ihre intrinsische Tryptophan-Fluoreszenz nachgewiesen werden. Zum einen gelang es, Modellsysteme mit nur 340 Trp abzubilden, andererseits konnte die Diffusion einzelner Hämocyanine (148 Trp) mit Hilfe der Fluoreszenz-Korrelationsspektroskopie nachgewiesen werden.Einzelne Hämocyanine konnten durch Adsorption und mildes Eintrocknen an verschiedenen Oberflächen immobilisiert werden. Mittels Atomarer Kraft-Mikroskopie (AFM) ließen sich individuelle Hämocyanine abgebilden, wobei Details der Quarärstruktur aufgelöst werden konnten.
