Adenylate Kinase (AK) is a signal transducing protein that regulates cellular energy homeostasis balancing between different conformations. An alteration of its activity can lead to severe pathologies such as heart failure, cancer and neurodegenerative diseases. A comprehensive elucidation of the large-scale conformational motions that rule the functional mechanism of this enzyme is of great value to guide rationally the development of new medications. Here using a metadynamics-based computational protocol we elucidate the thermodynamics and structural properties underlying the AK functional transitions. The free energy estimation of the conformational motions of the enzyme allows characterizing the sequence of events that regulate its action. We reveal the atomistic details of the most relevant enzyme states, identifying residues such as Arg119 and Lys13, which play a key role during the conformational transitions and represent druggable spots to design enzyme inhibitors. Our study offers tools that open new areas of investigation on large-scale motion in proteins.

The condition of soft-textured flesh in commercially harvested sablefish, Anoplopoma fimbria, from southeastern Alaska was investigated by National Marine Fisheries Service (NMFS) scientists from the Alaska Fisheries Science Center’s Auke Bay Laboratories (ABL) in Alaska and the Northwest Fisheries Science Center in Seattle, Wash. Sablefish were sampled by longline, pot, and trawl at five sites around Chichagof Island at depths of 259–988 m in the summer of 1985 and at depths of 259–913 m in the winter of 1986. At the time of capture and data collection, sablefish were categorized as being “firm” or “soft” by visual and tactile examination, individually weighed, measured for length, and sexed. Subsamples of the fish were analyzed and linear regressions and analyses of variance were performed on both the summer (n = 242) and winter (n = 439) data for combinations of chemical and physical analyses, depth of capture, weight vs. length, flesh condition, gonad condition, and sex. We successfully identified and selected sablefish with firm- and soft-textured flesh by tactile and visual methods. Abundance of firm fish in catches varied by season: 67% in winter and 40% in summer. Winter catches may give a higher yield than summer catches. Abundance of firm fish catches also varied with depth. Firm fish were routinely found shallower than soft fish. The highest percentage of firm fish were found at depths less than 365 m in summer and at 365–730 m in winter, whereas soft fish were usually more abundant at depths greater than 731 m. Catches of firm fish declined with increasing depth. More than 80% of the fish caught during winter at depths between 365 and 730 m had firm flesh, but this declined to 48% at these depths in summer. Longlines and pots caught similar proportions of firm and soft fish with both gears catching more firm than soft fish. Trawls caught a higher proportion of soft fish compared to longlines and pots in winter. Chemical composition of “firm” and “soft” fish differed. On average “soft” fish had 14% less protein, 12% more lipid, and 3% less ash than firm fish. Cooked yields from sablefish with soft-textured flesh were 31% less than cooked yields from firm fish. Sablefish flesh quality (firmness) related significantly to the biochemistry of white muscle with respect to 11 variables. Summer fish of all flesh conditions averaged 6% heavier than winter fish. Regulating depth of fishing could increase the yield from catches, but the feasibility and benefits from this action will require further evaluation and study. Results of this study provide a basis for reducing the harvest of sablefish with soft flesh and may stimulate further research into the cause and effect relationship of the sablefish soft-flesh phenomenon.

O presente trabalho, plasmado em metodologia jurídica, reflete criticamente sobre o problema da motivação da sentença civil como elemento de organização e de funcionamento do Estado Constitucional Democrático de Direito. A motivação é condição essencial de jurisdicionalidade, no sentido de que sem motivação não há exercício legítimo da função jurisdicional. O trabalho faz uma abordagem da natureza da motivação como discurso justificativo, jurídico e racional, da validade dos critérios de escolha ou de valoração empregados pelo juiz em sua decisão. O raciocínio do juiz é apresentado sob dupla feição: raciocínio decisório interno (contexto de descoberta ou deliberação) e raciocínio justificativo externo (contexto de justificação ou de validação). O conjunto das funções técnico-instrumental (endoprocessual) e político-garantística (extraprocessual) é objeto de investigação. A motivação, nos planos teórico e prático, exerce também a função de garantia do garantismo processual. A tese da inexistência jurídica da sentença tem três eixos teóricos: omissão total da motivação gráfica; falta de motivação ideológica, equiparada à hipótese de ausência de motivação gráfica; incompatibilidade lógica radical entre as premissas ou entre as premissas e a conclusão final, que também equivale à ausência total de motivação. O trabalho retrata um modelo de injustiça atemporal vivificado pelo juiz Crono, oposto à motivação como inestimável fator de legitimação argumentativa da jurisdição. A obrigatoriedade de motivação pública é o traço característico da jurisdição de nossa contemporaneidade e representa a maior conquista civilizatória do processo équo e justo.

Tissues from Cook Inlet beluga whales, Delphinapterus leucas, that were collected as part of the Alaska Marine Mammal Tissue Archival Project were analyzed for polychlorinated biphenyls (PCB’s), chlorinated pesticides, and heavy metals and other elements. Concentrations of total PCB’s (ΣPCB’s), total DDT (ΣDDT), chlordane compounds, hexachlorobenzene (HCB), dieldrin, mirex, toxaphene, and hexachlorocyclohexane (HCH) measured in Cook Inlet beluga blubber were compared with those reported for belugas from two Arctic Alaska locations (Point Hope and Point Lay), Greenland, Arctic Canada, and the highly contaminated stock from the St. Lawrence estuary in eastern Canada. The Arctic and Cook Inlet belugas had much lower concentrations (ΣPCB’s and ΣDDT were an order of magnitude lower) than those found in animals from the St. Lawrence estuary. The Cook Inlet belugas had the lowest concentrations of all (ΣPCB’s aver-aged 1.49 ± 0.70 and 0.79 ± 0.56 mg/kg wet mass, and ΣDDT averaged 1.35 ± 0.73 and 0.59 ± 0.45 mg/kg in males and females, respectively). Concentrations in the blubber of the Cook Inlet males were significantly lower than those found in the males of the Arctic Alaska belugas (ΣPCB’s and ΣDDT were about half). The lower levels in the Cook Inlet animals might be due to differences in contaminant sources, food web differences, or different age distributions among the animals sampled. Cook Inlet males had higher mean and median concentrations than did females, a result attributable to the transfer of these compounds from mother to calf during pregnancy and during lactation. Liver concentrations of cadmium and mercury were lower in the Cook Inlet belugas (most cadmium values were <1 mg/kg and mercury values were 0.704–11.42 mg/kg wet mass), but copper levels were significantly higher in the Cook Inlet animals (3.97–123.8 mg/kg wet mass) than in Arctic Alaska animals and similar to those reported for belugas from Hudson Bay. Although total mercury levels were the lowest in the Cook Inlet population, methylmercury concentrations were similar among all three groups of the Alaska animals examined (0.34–2.11 mg/kg wet mass). Este estudo possui o interesse em ampliar as discussões, no campo da história da educação, sobre o uso de cartilhas como dispositivos por meio dos quais se procurou pôs em circulação, no espaço escolar, os ideais higiênicos, cuja aquisição permitiria a construção de uma vida vista como saudável e feliz. As cartilhas analisadas são: Cartilha de Higiene, de 1922, de Antônio de Almeida Júnior, médico e educador, conhecido por ter tido envolvimento no Movimento da Escola Nova; e Cartilha de Higiene, de 1936, de Renato Kehl, médico e defensor da eugenia no Brasil. As obras em questão foram produzidas em um cenário em que proliferaram intervenções oriundas do campo médico, entre outros campos científicos, com vistas à conformação da sociedade brasileira em bases modernas. Pretende-se analisar esses livros, valorizando sua dimensão de objeto cultural, explorando o caráter prescritivo de que se revestem, dirigido às crianças, em vários aspectos de sua vida escolar e doméstica, e ainda as representações produzidas sobre infância, família, escola e saúde.

Esquistossomose e desnutrição são graves problemas de saúde pública nos países em desenvolvimento. A esquistossomose provoca uma série de morbidades, que é influenciada, em grande maioria, pela natureza do estímulo à resposta imunológica e ao estado nutricional do hospedeiro. Durante a infecção esquistossomótica, a resposta imune produz citocinas que são liberadas e estimulam a produção de óxido nítrico. Numerosos estudos demonstraram que o estado nutricional e a resposta imune afetam as características fenotípicas dos vermes adultos. No entanto, se o óxido nítrico desempenha um papel neste fenômeno é desconhecido. Neste estudo, os camundongos do tipo selvagem (grupo controle) e camundongos knockout com deficiência na produção de óxido nítrico foram alimentados com uma dieta comercial (UNILAB) ou uma dieta básica regional, dieta com baixa concentração de proteína (7,87%). Por meio de um sistema digital de análise de imagem (Image Pro Plus, USA) e microscopia confocal, estudou-se a morfologia da ventosa oral, tegumento e o sistema reprodutor de vermes machos e fêmeas. Vermes machos e fêmeas recuperados de camundongos desnutridos com deficiência de oxido nítrico mostraram descamação do tegumento. A superfície dorsal de vermes desnutridos machos apresentaram tubérculos irregulares, distribuídos de forma desigual, e escassos. O sistema reprodutivo de vermes fêmeas desnutridas não demonstrou alterações morfológicas. No entanto, os machos deste grupo exibiram menor número de células no processo de diferenciação dentro dos lobos testiculares. Em conclusão, nossos resultados sugerem que a deficiência de produção de óxido nítrico não induz alterações morfológicas em nível do tegumento e do sistema reprodutor de vermes machos. Em contraste, a desnutrição é responsável por tais alterações morfológicas, principalmente no sistema reprodutivo e na ventosa oral.

Este estudo refere-se às evidências de necessidades de autocuidado de militares com hipertensão sistêmica da Marinha do Brasil. A hipertensão arterial é uma das doenças crônicas mais frequentes, de maior prevalência no mundo e a sua evolução leva a complicações que comprometem todos os sistemas orgânicos. Neste sentido o problema apresentado é: como se caracterizam as necessidades de autocuidado de militares hipertensos da Marinha do Brasil. Para tanto, o objetivo geral da pesquisa foi: Discutir as necessidades de cuidado no processo saúde/adoecimento do militar da Marinha do Brasil e os objetivos específicos: Descrever as características sociodemográficas e de autocuidado dos militares atendidos no ambulatório de cardiologia do HNMD; Identificar através dos depoimentos dos militares, as necessidades de autocuidado e; Propor estratégias de autocuidado em saúde aos militares. O referencial teórico utilizado foi o Modelo de Promoção de Saúde de Nola J Pender. Trata-se de um estudo misto de caráter exploratório e descritivo, realizado no ambulatório de Cardiologia do Hospital Naval Marcilio Dias, no município do Rio de Janeiro, no período de julho a agosto de 2014. Os sujeitos do estudo foram 20 militares hipertensos da Marinha do Brasil com idade acima dos 25 anos. Os dados foram coletados por meio de entrevista semiestruturada, em seguida, submetidos à análise de conteúdo de Bardin, emergindo três categorias e subcategorias assim denominadas: Primeira: Evidências de necessidades de autocuidado. Segunda: Identificação situacional da saúde dos militares com a subcategoria: A experiência do autocuidado para o militar com HAS. Terceira: Interesse pelo autocuidado com duas subcategorias: Interesse pelo aprendizado e Estratégias para o aprendizado do autocuidado. O perfil dos participantes caracterizou-se da seguinte maneira: Dos 20 militares, 19 são homens e 1 mulher, sendo 3 da ativa e 17 da reserva, a maioria são casados, idosos, de etnia parda e crença religiosa cristã, possuem ensino médio completo, com remuneração acima de 4 salários mínimos, residindo em local com saneamento básico. Quanto ao autocuidado referido pelos participantes: a maioria controla o uso de sal, ingesta hídrica adequada, utilizam alimentação balanceada, praticam algum tipo de exercício diário, a maioria referiu uso regular de anti-hipertensivos, e procura pelos serviços de saúde, quando necessário. Conclui-se que apesar do autocuidado referido, estes estão aquém às necessidades de saúde e não atendem às demandas de autocuidado necessária à manutenção da vida. Apesar de diversos fatores contribuírem para desmotivar os militares em relação ao autocuidado, eles apresentam interesse em aprender novas tecnologias de cuidar para adequação de sua saúde atual e melhora da qualidade de vida. Para atender às necessidades de autocuidado identificadas, sugere-se planejamento das ações estratégicas de promoção à saúde conjunta, com a equipe multiprofissional, instituindo a consulta de enfermagem, avaliação do autocuidado, prevenção dos fatores de risco e acompanhamento regular para análise das mudanças efetivas dos hábitos de vida saudáveis.

Em março de 1907, o Decreto no 6.640 enunciou uma reforma policial na cidade do Rio de Janeiro que, entre outras modificações, fez emergir o periódico Boletim Policial. Este trabalho analisa como essa revista, como enunciado do discurso policial, foi um arquivo construtor da polícia da Capital Federal como polícia científica e moderna entre os anos 1907 e 1918. Esse período é marcado pela reorganização dos parâmetros científicos acerca do crime e do criminoso, em várias partes do mundo, que serviram de base para a polícia científica. É o momento em que a aplicação da antropologia criminal ainda estava em voga, do uso de novos métodos de identificação de criminosos, de inovações na análise do local do crime, entre outras transformações. Abordo, também, a aplicabilidade do discurso policial modernizador em diálogo com os pressupostos da Escola Positiva e com novas formas de tecnologias de identificação dos criminosos nos ditos sobre os portugueses na esfera criminal.

Ecosystem-based management is one of many indispensable components of objective, holistic management of human impacts on nonhuman systems. By itself, however, ecosystem-based management carries the same risks we face with other forms of current management; holism requires more. Combining single-species and ecosystem approaches represents progress. However, it is now recognized that management also needs to be evosystem-based. In other words, management needs to account for all coevolutionary and evolutionary interactions among all species; otherwise we fall far short of holism. Fully holistic practices are quite distinct from the approaches to the management of fisheries that are applied today. In this paper, we show how macroecological patterns can guide management consistently, objectively, and holistically. We present one particular macroecological pattern with two applications. The first application is a case study of fisheries from the Baltic Sea involving historical data for two species; the second involves a sample of 44 species of primarily marine fish worldwide. In both cases we evaluate historical fishing rates and determine holistic/systemic sustainable single-species fishing rates to illustrate that conventional fisheries management leads to much more extensive and pervasive overfishing than currently realized; harvests are, on average, over twenty-fold too large to be fully sustainable. In general, our approach involves not only the sustainability of fisheries and related resources but also the sustainability of the ecosystems and evosystems in which they occur. Using macroecological patterns accomplishes four important goals: 1) Macroecology becomes one of the interdisciplinary components of management. 2) Sustainability becomes an option for harvests from populations of individual species, species groups, ecosystems, and the entire marine environment. 3) Policies and goals are reality-based, holistic, or fully systemic; they account for ecological as well as evolutionary factors and dynamics (including management itself). 4) Numerous management questions can be addressed.

Autonomous underwater vehicles (AUV’s) are increasingly used to collect physical, chemical, and biological information in the marine environment. Recent efforts include merging AUV technology with acoustic telemetry to provide information on the distribution and movements of marine fish. We compared surface vessel and AUV tracking capabilities under rigorous conditions in coastal waters near Juneau, Alaska. Tracking surveys were conducted with a REMUS 100 AUV equipped with an integrated acoustic receiver and hydrophone. The AUV was programmed to navigate along predetermined routes to detect both reference transmitters at 20–500 m depths and tagged fish and crabs in situ. Comparable boat surveys were also conducted. Transmitter depth had a major impact on tracking performance. The AUV was equally effective or better than the boat at detecting reference transmitters in shallow water, and significantly better for transmitters at deeper depths. Similar results were observed for tagged animals. Red king crab, Paralithodes camtschaticus, at moderate depths were recorded by both tracking methods, while only the AUV detected Sablefish, Anoplopoma fimbria, at depths exceeding 500 m. Strong currents and deep depths caused problems with AUV navigation, position estimation, and operational performance, but reflect problems encountered by other AUV applications that will likely diminish with future advances, enhanced methods, and increased use.

The proposed plan for enrichment of the Sulu Sea, Philippines, a region of rich marine biodiversity, with thousands of tonnes of urea in order to stimulate algal blooms and sequester carbon is flawed for multiple reasons. Urea is preferentially used as a nitrogen source by some cyanobacteria and dinoflagellates, many of which are neutrally or positively buoyant. Biological pumps to the deep sea are classically leaky, and the inefficient burial of new biomass makes the estimation of a net loss of carbon from the atmosphere questionable at best. The potential for growth of toxic dinoflagellates is also high, as many grow well on urea and some even increase their toxicity when grown on urea. Many toxic dinoflagellates form cysts which can settle to the sediment and germinate in subsequent years, forming new blooms even without further fertilization. If large-scale blooms do occur, it is likely that they will contribute to hypoxia in the bottom waters upon decomposition. Lastly, urea production requires fossil fuel usage, further limiting the potential for net carbon sequestration. Skin flap procedures are commonly used in plastic surgery. Failures can follow, leading to the necrosis of the flap. Therefore, many studies use LLLT to improve flap viability. Currently, the LED has been introduced as an alternative to LLLT. the objective of this study was to evaluate the effect of LLLT and LED on the viability of random skin flaps in rats. Forty-eight rats were divided into four groups, and a random skin flap (10 x 4 cm) was performed in all animals. Group 1 was the sham group; group 2 was submitted to LLLT 660 nm, 0.14 J; group 3 with LED 630 nm, 2.49 J, and group 4 with LLLT 660 nm, with 2.49 J. Irradiation was applied after surgery and repeated on the four subsequent days. On the 7th postoperative day, the percentage of flap necrosis was calculated and skin samples were collected from the viable area and from the transition line of the flap to evaluate blood vessels and mast cells. the percentage of necrosis was significantly lower in groups 3 and 4 compared to groups 1 and 2. Concerning blood vessels and mast cell numbers, only the animals in group 3 showed significant increase compared to group 1 in the skin sample of the transition line. LED and LLLT with the same total energies were effective in increasing viability of random skin flaps. LED was more effective in increasing the number of mast cells and blood vessels in the transition line of random skin flaps.

The phototherapy effects in the skin are related to biomodulation, usually to accelerate wound healing. However, there is no direct proof of the interrelation between the effects of low-level laser therapy (LLLT) and light-emitting diode (LED) in neuropeptide secretion, these substances being prematurely involved in the neurogenic inflammation phase of wound healing. This study therefore focused on investigating LLLT and LED in Calcitonin gene-related peptide (CGRP) and substance P (SP) secretion in healthy rat skin. Forty rats were randomly distributed into five groups with eight rats each: Control Group, Blue LED Group (470 nm, 350 mW power), Red LED Group (660 nm, 350 mW power), Red Laser Group (660 nm, 100 mW power), and Infrared Laser Group (808 nm, 100 mW power) (DMCA (R) Equipamentos Ltda., So Carlos, So Paulo, Brazil). the skin of the animals in the experimental groups was irradiated using the punctual contact technique, with a total energy of 40 J, single dose, standardized at one point in the dorsal region. After 14 min of irradiation, the skin samples were collected for CGRP and SP quantification using western blot analysis. SP was released in Infrared Laser Group (p = 0.01); there was no difference in the CGRP secretion among groups. Infrared (808 nm) LLLT enhances neuropeptide SP secretion in healthy rat skin.
