This study was conducted to identify the concentration dependence of the operating wavelengths and the relative intensities in which a dye mixture doped polymer optical fibre can operate. A comparative study of the radiative and Forster type energy transfer processes in Coumarin 540:Rhodamine 6G, Coumarin 540:Rhodamine B and Rhodamine 6G:Rhodamine B in methyl methacrylate (MMA) and poly(methyl methacrylate) (PMMA) was done by fabricating a series of dye mixture doped polymer rods which have two emission peaks with varying relative intensities. These rods can be used as preforms for the fabrication of polymer optical fibre amplifiers operating in the multi-wavelength regime. The 445 nm line from an Nd:YAG pumped optical parametric oscillator (OPO) was used as the excitation source for the first two dye pairs and a frequency doubled Nd:YAG laser emitting at 532 nm was used to excite the Rh 6G:Rh B pair. The fluorescence lifetimes of the donor molecule in pure form as well as in the mixtures were experimentally measured in both monomer and polymer matrices by time-correlated single photon counting technique. The energy transfer rate constants and transfer efficiencies were calculated and their dependence on the acceptor concentration was analysed. It was found that radiative energy transfer mechanisms are more efficient in all the three dye pairs in liquid and solid matrices.

Dependence of energy transfer parameters on excitation wavelength has been investigated in poly (methyl methacrylate) (PMMA) optical fibre preforms doped with C 540:Rh B dye mixture by studying the fluorescence intensity and the lifetime variations. A fluorescence spectrophotometer was used to record the excitation spectra of the samples for the emission wavelengths 495 and 580 nm. The fluorescence emission from the polymer rods was studied at four specific excitation wavelengths viz; 445, 465, 488 and 532 nm. The fluorescence lifetime of the donor molecule was experimentally measured in polymer matrix by time correlated single photon counting technique. The energy transfer rate constants and transfer efficiencies were calculated and their dependence on the acceptor concentration was analysed for three excitation wavelengths. It was found that any change in the excitation wavelength leads to significant variations in the quenching characteristics, which in turn affect the calculated energy transfer parameters.

In der vorliegenden Arbeit wird ein Unterrichtskonzept für die gymnasiale Oberstufe beschrieben, das anhand der Polarisationseigenschaft des Lichts von der Beobachtung ausgehend einen Zugang zur Quantenphysik ermöglicht. Die Unterrichtsinhalte bauen so aufeinander auf, dass ein "harter Bruch" zwischen der klassischen und der quantenphysikalischen Beschreibung von Licht vermieden wird. Das methodische Vorgehen des Unterrichtskonzeptes führt vom Phänomen ausgehend zu quantitativen Experimenten hin zu einer Einführung in quantenphysikalische Begriffe und Prinzipien. Dabei bildet der elektrische Feldvektor die Verknüpfung zwischen der klassischen und der quantenphysi-kalischen Beschreibung der Polarisationsexperimente, in dem er zunächst die Polarisationsexperimente beschreibt und im weiteren Verlauf des Unterrichtsganges als Wahrscheinlichkeitsamplitude gedeutet wird. Die Polarisation von Licht wird zu Beginn des Unterrichtsganges im Rahmen eines fächerübergreifenden Kontextes eingeführt, wobei die Navigation der Insekten nach dem polarisierten Himmelslicht als Einstieg dient. Die Erzeugung und die Eigen-schaften von polarisiertem Licht werden anhand von einfachen qualitativen Schüler- und Demonstrationsexperimenten mit Polarisationsfolien erarbeitet. Das Polarisationsphänomen der Haidinger-Büschel, das bei der Beobachtung von polarisiertem Licht wahrgenommen werden kann, ermöglicht eine Anbindung an das eigene Erleben der Schülerinnen und Schüler. Zur Erklärung dieser Experimente auf der Modellebene wird der elektrische Feldvektor und dessen Komponentenzerlegung benutzt. Im weiteren Verlauf des Unterrichtsganges wird die Komponentenzerlegung des elektrischen Feldvektors für eine quantitative Beschreibung der Polarisationsexperimente wieder aufgegriffen. In Experimenten mit Polarisationsfiltern wird durch Intensitätsmessungen das Malussche Gesetz und der quadratische Zusammenhang zwischen Intensität und elektrischem Feldvektor erarbeitet. Als Abschluss der klassischen Polarisationsexperimente wird das Verhalten von polarisiertem Licht bei Überlagerung in einem Michelson-Interferometer untersucht. Das in Abhängigkeit der Polarisationsrichtungen entstehende Interferenzmuster wird wiederum mit Hilfe der Komponentenzerlegung des elektrischen Feldvektors beschrieben und führt zum Superpositionsprinzip der elektrischen Feldvektoren. Beim Übergang zur Quantenphysik werden die bereits durchgeführten Polarisationsexperimente als Gedankenexperimente in der Photonenvorstellung gedeutet. Zur Beschreibung der Polarisation von Photonen wird der Begriff des Zustandes eingeführt, der durch die Wechselwirkung der Photonen mit dem Polarisationsfilter erzeugt wird. Das Malussche Gesetz wird in der Teilchenvorstellung wieder aufgegriffen und führt mit Hilfe der statistischen Deutung zum Begriff der Wahrscheinlichkeit. Bei der Beschreibung von Interferenzexperimenten mit einzelnen Photonen wird die Notwendigkeit eines Analogons zum elektrischen Feldvektor deutlich. Diese Betrachtungen führen zum Begriff der Wahrscheinlichkeitsamplitude und zum Superpositionsprinzip der Wahrscheinlichkeitsamplituden. Zum Abschluss des Unterrichtsganges wird anhand des Lokalisationsproblems einzelner Photonen das Fundamentalprinzip der Quantenphysik erarbeitet.

Propósito de la revisión: la enfermedad de Parkinson (EP) es un trastorno degenerativo caracterizado clínicamente por presentar temblor en reposo, rigidez y bradicinesia. El propósito es determinar la utilidad de la molécula TRODAT-1 en el diagnóstico de la EP. Metodología: se realizó una búsqueda en las bases de datos: PUBMED, COCHRANE, MEDLINE, LILACS y SCIELO en un período de 10 años desde enero de 1998 a enero de 2008. Se obtuvieron 26 artículos, estos se analizaron y se seleccionaron 10 artículos, de los cuales sólo 6 respondían a las necesidades del estudio, de acuerdo a los criterios de inclusión. De los 6 artículos analizados, 4 fueron clasificados como evidencia grado (+) y los 2 restantes evidencia grado (-) de acuerdo con las guías NICE. Todos los artículos revisados reportan una disminución importante en la captación del TRODAT-1 a nivel estriatal, su utilidad en el diagnóstico de EP en estadios tempranos, bajo costo y seguridad. Sólo tres reportan valores de sensibilidad y especificidad, pero su nivel de calidad no permite hacer una comparación de los mismos. Conclusiones: se propone realizar estudios de prueba diagnóstica comparados con el diagnóstico clínico de la enfermedad, que tengan un acuerdo en la forma de plantear las mediciones semicuantitativas de las unidades de captación utilizando las mismas fórmulas para hacerlos comparables.

In positron emission tomography and single photon emission computed tomography studies using D2 dopamine (DA) receptor radiotracers, a decrease in radiotracer binding potential (BP) is usually interpreted in terms of increased competition with synaptic DA. However, some data suggest that this signal may also reflect agonist (DA)-induced increases in D2 receptor (D2R) internalization, a process which would presumably also decrease the population of receptors available for binding to hydrophilic radioligands. To advance interpretation of alterations in D2 radiotracer BP, direct methods of assessment of D2R internalization are required. Here, we describe a confocal microscopy-based approach for the quantification of agonist-dependent receptor internalization. The method relies upon double-labeling of the receptors with antibodies directed against intracellular as well as extracellular epitopes. Following agonist stimulation, DA D2R internalization was quantified by differentiating, in optical cell sections, the signal due to the staining of the extracellular from intracellular epitopes of D2Rs. Receptor internalization was increased in the presence of the D2 agonists DA and bromocriptine, but not the D1 agonist SKF38393. Pretreatment with either the D2 antagonist sulpiride, or inhibitors of internalization (phenylarsine oxide and high molarity sucrose), blocked D2-agonist induced receptor internalization, thus validating this method in vitro. This approach therefore provides a direct and streamlined methodology for investigating the pharmacological and mechanistic aspects of D2R internalization, and should inform the interpretation of results from in vivo receptor imaging studies.

The chemical specificity of terahertz spectroscopy, when combined with techniques for sub-wavelength sensing, is giving new understanding of processes occurring at the nanometre scale in biological systems and offers the potential for single molecule detection of chemical and biological agents and explosives. In addition, terahertz techniques are enabling the exploration of the fundamental behaviour of light when it interacts with nanoscale optical structures, and are being used to measure ultrafast carrier dynamics, transport and localisation in nanostructures. This chapter will explain how terahertz scale modelling can be used to explore the fundamental physics of nano-optics, it will discuss the terahertz spectroscopy of nanomaterials, terahertz near-field microscopy and other sub-wavelength techniques, and summarise recent developments in the terahertz spectroscopy and imaging of biological systems at the nanoscale. The potential of using these techniques for security applications will be considered.

This review deals with surface-enhancved Raman scattering (SERS) employing Langmuir-Blodgett (LB) films, which serve as model systems for developing theoretical and experimental studies to elucidate the SERS effect. In addition, LB films have be used as integral parts of molecular architectures for SERS-active substrates. On the other hand, SERS and surface-enhaced resonant Raman scattering (SERRS) have allowed various properties of LB films to be investigated, especially those associated with molecular-level interactions. In the paper, emphasis is placed on single molecule detection (SMD), where the target molecule is diluted on an LB matrix of spectral silent material (low Raman cross section). The perspectives and challenges for combining SERS and LB films are also discussed.

Cooperative spontaneous emission of a single photon from a cloud of N atoms modifies substantially the radiation pressure exerted by a far-detuned laser beam exciting the atoms. On one hand, the force induced by photon absorption depends on the collective decay rate of the excited atomic state. On the other hand, directional spontaneous emission counteracts the recoil induced by the absorption. We derive an analytical expression for the radiation pressure in steady-state. For a smooth extended atomic distribution we show that the radiation pressure depends on the atom number via cooperative scattering and that, for certain atom numbers, it can be suppressed or enhanced. Cooperative scattering of light by extended atomic clouds can become important in the presence of quasi-resonant light and could be addressed in many cold atoms experiments.

The excited-state dynamics of two polyfluorene copolymers, one fully conjugated containing phenylene vinylene units alternated with 9,9`-dihexylfluorenyl groups and the other segmented by -(CH2)(8)- spacer, were studied in dilute solution of different solvents using a picosecond single-photon timing technique. The excited-state dynamics of the segmented copolymer follows the Forster resonant energy-transfer model which describes intrachain energy-transfer kinetics among random oriented chromophores. Energy transfer is confirmed by analysis of fluorescence anisotropy relaxation with the measurement of a short decay component of about 60 ps. The fluorescence decay surface of the fully conjugated copolymer is biexponential with decay times of about 470 and 900 ps, ascribed to deactivation of chain moieties containing trans and cis isomers already in a photostationary condition. Thus, energy transfer is very fast due to the conjugated nature and rigid-rod-like structure of this copolymer chain.

Objective: Since blood viscosity (BV) is one of the most important factors determining blood flow, this study aimed to investigate the possible correlation between increased blood viscosity and reduction of regional cerebral blood flow (rCBF) in healthy ageing.Methods: Male subjects were distributed in two groups: "young", aged 20-30 (27 volunteers), or "elderly", aged 60-70 (50 volunteers). Whole blood viscosity was obtained with a Wells-Brookfield Cone/Plate Viscometer. Cerebral blood flow was analysed by means of single photon emission computed tomography (SPECT).Results: The mean BV values were 3.28 +/- 0.43 mPa in the group of young volunteers and 4.33 +/- 0.73 mPa in the group of elderly volunteers (t = -6.9, p < 0.0001). The elderly had a lower blood flow than the young in the following regions: bilateral parietal; temporal-parietal and temporal of the left hemisphere. Pearson's correlation between BV and rCBF showed a good inverse correlation when the BV was above 3.95 +/- 0.83 mPa.Conclusions: Our results point to a close relationship between the two parameters analysed, BV and rCBF. The impairment in rCBF observed in the elderly volunteers might be due to an increase in BV, among other factors.Significance: These findings suggest interesting possibilities for the treatment/prevention of brain ageing. Purpose of reviewTo critically review data on the use of neuroimaging tools in the clinical diagnostic investigation of dementias.Recent findingsFor many years, the use of neuroimaging tools in the evaluation of dementias has been restricted to excluding neurosurgical lesions that may account for the cognitive decline. However, modern neuroimaging extends beyond this traditional role of excluding other conditions and has a key role in the clinical investigation of Alzheimer's disease and of other degenerative cortical dementias. MRI, PET with fluorodeoxyglucose, and single-photon emission computed tomography are topographic markers of neural damage and enable the identification of specific lesional patterns that characterize Alzheimer's disease and other cortical dementias. More recently, PET amyloid markers have enabled the in-vivo assessment of amyloid load, a key feature in the physiopathology of Alzheimer's disease.SummaryThe combined use of neuroimaging examinations with clinical, neuropsychological, and cerebrospinal fluid markers can improve the specificity of the diagnosis of Alzheimer's disease, even at early stages of the disease. In the following years, progress in research will provide standardized and validated imaging markers of Alzheimer's disease and other dementias, which may increase their application in clinical settings.
