The aim of this study was to adapt the methodology of the accelerated aging and electrical conductivity tests for determination of physiological potential in crambe seeds. Six seed lots of crambe (cv. FMS Brilhante) were subjected to determination of moisture content, germination test, first count germination, emergence, and emergence speed index. For the accelerated aging test, the traditional methodology was used with water, and with a saturated potassium chloride and sodium chloride solution in three periods of exposure (24, 48, and 72 hours) at 41 degrees C; the electrical conductivity test was performed with four pre-soaking treatments (0, 2, 4, and 8 hours) and four soaking periods (4, 8, 16, and 24 hours) at 25 degrees C. The accelerated aging test with water for 72 hours and the electrical conductivity test with 2 hours of pre-soaking and assessment after 16 hours were effective for classification of the crambe seed lots in regard to physiological quality.

To evaluate the bond strength between two types of acrylic resin teeth and a microwave denture base resin after immersion in disinfectant solutions for 180 days. Eighty specimens made of acrylic resin teeth (Biotone and Biotone IPN) attached to a microwave polymerized denture base resin (Nature-Cryl MC) were divided into eight groups (n = 10) according to the treatment (distilled water-control, 2% chlorhexidine digluconate, 1% sodium hypochlorite and sodium perborate solution-Corega Tabs). The shear strength tests (MPa) were carried out using a universal testing machine with a 0.5 mm/min speed. Data analysis was performed using ANOVA and multiple comparison Student-Newman-Keuls post hoc test (α = 0.05). Biotone IPN showed similar results among the groups (distilled water, 8.25 ± 1.81 MPa; chlorhexidine, 7.81 ± 3.34 MPa; hypochlorite, 7.75 ± 3.72 MPa; and Corega Tabs, 7.58 ± 2.27 MPa, whereas Biotone showed significantly lower shear bond strength values for the groups immersed in Corega Tabs (5.25 ± 3.27 MPa) and chlorhexidine (6.08 ± 2.35 MPa). Soaking the dentures in 1% sodium hypochlorite could be recommended as a disinfectant solution for dentures fabricated with conventional acrylic resin denture teeth and microwave denture base resin. For dentures fabricated with IPN teeth and microwave denture base resin, all the soaking solutions evaluated in this study could be suggested to denture wearers.

Two experiments were conducted in order to study the effects of seed number, temperature and time of soaking sesame seeds on the results of the electrical conductivity test for physiological quality of seeds. In the experiment I evaluated the electrical conductivity varying the number of seeds (25, 50 and 100) and temperature (20, 25 and 30° C) in experiment II is different soaking periods (2, 4, 6, 8, 12, 16, 20 and 24 h). The tests were performed with three cultivars (cv. Trebol, cv. Cnpa G4 e cv. Comum). The treatments in factorial 3x3x3 and 3x8 in experiments I and II, respectively. The experimental design was completely randomized, with four replications. Means were compared by Tukey test at 5% probability. The characterization of cultivars held through the germination and vigor tests (physical, physiological and biochemical), which rated the Trebol cultivar seeds with better physiological then the Cnpa G4 and Comum. The electrical conductivity test was conducted with four sub-samples of 25 pure seeds soaked in 75 mL of deionized water at 25° C, to separate the lots from 2 hours of soaking, being feasible to evaluate the quality sesame seeds.

Purpose: Adequate denture hygiene can prevent and treat infection in edentulous patients, who are frequently elderly and have difficulty brushing their teeth. This study evaluated the efficacy of complete denture biofilm removal using a chlorhexidine solution in two concentrations: 0.12% and 2.0%. Materials and Methods: Sixty complete denture wearers participated in a trial for 21 days after receiving brushing instructions. They were distributed into three groups, according to the tested solution and regimen (n = 20): (G1) Control (daily overnight soaking in water); (G2) daily immersion at home in 0.12% chlorhexidine for 20 minutes after dinner; and (G3) a single immersion in 2.0% chlorhexidine for 5 minutes at the end of the experimental period, performed by a professional. Biofilm coverage area (%) was quantified on the internal surface of maxillary dentures at baseline and after 21 days. Afterward, the differences between initial and posttreatment results were compared by means of the Kruskal-Wallis test (a = 0.05). Results: Median values for biofilm coverage area after treatment were: (G1) 36.0%; (G2) 5.3%; and (G3) 1.4%. Differences were significant (KW = 35.25; p < 0.001), although G2 and G3 presented similar efficacy in terms of biofilm removal. Conclusions: Both chlorhexidine-based treatments had a similar ability to remove denture biofilm. Immersion in 0.12% or 2.0% chlorhexidine solutions can be used as an auxiliary method for cleaning complete dentures.

Besides the risk of filling material extrusion throughout the apex, a satisfactory apical seal can be difficult to achieve in canals with open apices or iatrogenic enlargements of the apical constriction. These situations pose a challenge to root canal filling. This paper describes the root canal filling of a maxillary right canine with an overinstrumented apex, complete loss of the apical stop, extensive canal transportation and apical periodontitis. A 5 mm calcium hydroxide apical plug was placed before root canal filling. The plug was made by soaking paper points with saline, dipping the points in calcium hydroxide powder and then applying it to the apex several times, until a consistent apical plug was obtained. The canal was then irrigated with saline in order to remove any residual calcium hydroxide from the root canal walls, dried with paper points and obturated with an inverted #80 gutta-percha cone and zinc oxide-eugenol based sealer by the lateral condensation technique. An 8-year radiographic follow-up showed formation of mineralized tissue sealing the apical foramen, apical remodeling and no signs of apical periodontitis.

Kristallisation der Arbutin-Synthase und der Strictosidin Glukosidase - zwei Enzyme aus dem sekundären Glykosidstoffwechsel von Rauvolfia serpentina Die vorliegende Arbeit befasst sich mit der Kristallisation und der strukturellen Auswertung der Arbutin-Synthase (AS) und der Strictosidin Glukosidase (SG). Beide Enzyme stammen aus der Medizinalpflanze Rauvolfia serpentina. Für die Kristallisation der Arbutin-Synthase wurden ca. 2500 verschiedene Beding-ungen experimentell untersucht. Für einige dieser Experimente wurde das Enzym molekularbiologisch und chemisch verändert. Trotzdem konnten keine Kristalle erhalten werden. Die bei diesen Veränderungen erhaltenen Ergebnisse wurden anhand von Vergleichen mit Strukturen anderer Glykosyltransferasen der gleichen Familie analysiert. Bei der Reinigung der AS konnte mit verschiedenen Trennsystemen nie eine homogene Lösung produziert werden. Der wahrscheinliche Grund für diese schlechte Isolierbarkeit, und damit der wahrscheinliche Grund für die schwierige Kris-tallisation, liegt in der überdurchschnittlich hohen Anzahl an Cysteinen in der Proteinsequenz. Mit den Aminosäuren Cys171, Cys253 und Cys461 wurden drei Cysteine gefunden, die einem Strukturvergleich nach an der Proteinoberfläche liegen und möglicherweise durch Quervernetzungen mit anderen Proteinmolekülen ein heterogenes Gemisch bilden, das nicht geordnet kristallisieren kann. Durch gezielte Mutationen dieser drei Aminosäuren könnte die Kristallisation zukünftig ermöglicht werden. Für die SG waren bereits Bedingungen bekannt bei denen nicht vermessbare Enzymkristalle (Nadeln) wuchsen. In weit gefächerten Versuchen konnten diese Kristalle jedoch nicht zu 3D-Wachstum angeregt werden. Es wurden mit einem HTS-Screening neue Bedingungen zur Kristallisation gefunden. Anschließend konnten die native Struktur und der Strictosidin/Enzym-Komplex vermessen und aufgeklärt werden. Die SG gehört zur Familie 1 der Glukosidasen (GH-1) und besitzt die in dieser Familie konservierte (beta/alpha)8-Barrel-Faltung. Im Vergleich mit 16 bekannten Glykosidasen der Familie GH-1 wurde die Substratbindung untersucht. Dabei wurde die in der Familie konservierte Zuckerbindung vorgefunden, jedoch große Unterschiede in der Aglykonbindung entdeckt. Es wurden Bedingungen für die Konformationsänderung des Trp388 erkannt. Diese Konformationsänderung dirigiert den Aglykonteil des Substrates auf verschiedene Seiten der Substratbindungstasche und teilt so die Familie GH-1 in zwei Gruppen.

Numerous bacterial pathogens subvert cellular functions of eukaryotic host cells by the injection of effector proteins via dedicated secretion systems. The type IV secretion system (T4SS) effector protein BepA from Bartonella henselae is composed of an N-terminal Fic domain and a C-terminal Bartonella intracellular delivery domain, the latter being responsible for T4SS-mediated translocation into host cells. A proteolysis resistant fragment (residues 10-302) that includes the Fic domain shows autoadenylylation activity and adenylyl transfer onto Hela cell extract proteins as demonstrated by autoradiography on incubation with α-[(32)P]-ATP. Its crystal structure, determined to 2.9-Å resolution by the SeMet-SAD method, exhibits the canonical Fic fold including the HPFxxGNGRxxR signature motif with several elaborations in loop regions and an additional β-rich domain at the C-terminus. On crystal soaking with ATP/Mg(2+), additional electron density indicated the presence of a PP(i) /Mg(2+) moiety, the side product of the adenylylation reaction, in the anion binding nest of the signature motif. On the basis of this information and that of the recent structure of IbpA(Fic2) in complex with the eukaryotic target protein Cdc42, we present a detailed model for the ternary complex of Fic with the two substrates, ATP/Mg(2+) and target tyrosine. The model is consistent with an in-line nucleophilic attack of the deprotonated side-chain hydroxyl group onto the α-phosphorus of the nucleotide to accomplish AMP transfer. Furthermore, a general, sequence-independent mechanism of target positioning through antiparallel β-strand interactions between enzyme and target is suggested.

A survey was conducted to generate holistic information on the production and utilization of local white lupin in two lupin growing districts, namely, Mecha and Sekela, representing mid and high altitude areas, respectively in North-western Ethiopia. During the survey, two types of participatory rural appraisal (PRA) techniques, namely, individual farmer interview (61 farmers from Mecha and 51 from Sekela) and group discussion (with 20 farmers from each district) were employed. There are significant differences (P<0.05) between the two study districts for the variables like total land holding, frequency of ploughing during lupin planting, days to maturity, lupin productivity, and number of days of soaking lupin in running water. However, there are no significant differences (P>0.05) between the two study districts for the variables like land allocated for lupin cultivation, lupin seed rate, lupin soaking at home, lupin consumption per family per week and proportion of lupin used for household consumption. The use of the crop as livestock feed is negligible due to its high alkaloid content. It is concluded that the local white lupin in Ethiopia is a valuable multipurpose crop which is being cultivated in the midst of very serious shortage of cropland. Its ability to maintain soil fertility and serve as a source of food in seasons of food scarcity makes it an important crop. However, its bitter taste due to its high alkaloid content remains to be a big challenge and any lupin improvement strategy has to focus on minimizing the alkaloid content of the crop.

En semillas de diferentes cultivares de maíz provenientes de 12 lotes adquiridos en distintos años se estudió la relación entre la conductividad eléctrica y el pH del agua de remojo y la germinación y crecimiento de plántulas en invernadero, utilizando semillas sin tratar y semillas sometidas a la cámara de envejecimiento acelerado (72 h a 45 ± 1°C y 100% de humedad relativa). Se empleó un diseño de bloques completos al azar con cuatro repeticiones, se realizó el análisis de varianza y las diferencias entre cultivares se detectaron mediante la prueba de la mínima diferencia significativa. Los lotes mostraron a los 16 días de evaluación altos porcentajes de germinación en semillas no tratadas y se evidenció el efecto desfavorable sobre la germinación previo envejecimiento de las semillas. La altura de la planta, el peso seco del vástago y de las raíces mostraron diferencias significativas entre los lotes estudiados. La conductividad eléctrica se relacionó con los porcentajes de germinación de semillas sometidas a envejecimiento. Los lotes de semillas con altos valores de altura de planta y peso seco de vástago y de raíces presentaron bajos valores de conductividad eléctrica, por lo tanto, esta prueba podría ser indicativa del comportamiento de semillas de maíz en condiciones de invernadero. La prueba de pH no arrojó relación directa con la emergencia y el vigor de las semillas.

La conservación de alimentos por tecnología de barreras es una alternativa de procesamiento, en la cual se aplican las técnicas de conservación tradicionales pero de una manera menos intensa, mediante una combinación de obstáculos a fin de mantener las cualidades organolépticas en el producto final. El objetivo de este trabajo fue desarrollar un método de conservación de porotos con la aplicación de tecnología de bajo costo y de aceptabilidad comparable al método convencional (Método Appert). La metodología que se empleó en la elaboración de conservas de porotos por tecnologías de barrera fue la siguiente: selección, lavado, remojado durante 24 horas, escurrido, cocinado a presión en salmuera al 0,5% durante cinco minutos a 121°C. Posteriormente se envasaron en recipientes esterilizados, se agregó la solución de relleno en ebullición (vinagre 7,50%, ácido láctico 0,30%, ácido ascórbico 0,35%, ácido cítrico 0,35%, sal 1,00% y azúcar 0,50%. No se esterilizó y se dejó enfriar hasta alcanzar la temperatura ambiente. La determinación de la calidad de las conservas elaboradas se realizó a los seis meses de almacenadas realizando una evaluación microbiológica, físicoquímica y sensorial. La conserva obtenida es aceptable, inocua y puede ser preservada a temperatura ambiente.

En este trabajo se exponen los resultados obtenidos en pruebas de germinación realizadas durante los años 1947, 1948 y 1949 de semillas de Gymnocladus dioica L. producidas en esta región, previamente tratadas con ácido sulfúrico concentrado (d. 1,84) . Estos ensayos fueron efectuados con el objeto de lograr una buena germinación, ya que la aplicación del tratamiento empleado por Wiesehugel (1935) resultó ineficaz. Los porcentajes más altos de germinación, 72% y 75%, se obtuvieron con inmerciones de las semillas en ácido, durante 16 y 18 horas.
