Symphonia globulifera Linn. f., popularmente comumente conhecida como anani, citada em relatos de utilização popular em diferentes países, como Camarões, e em várias regiões do Brasil, principalmente na Amazônia, no tratamento de inflamações na pele, tônico geral e laxante para mulheres grávidas, além de sua seiva/látex ser empregada contra reumatismo e tumores. Embora alguns trabalhos tenham descrito o perfil fitoquímico de S. globulifera, não existem estudos, até o presente, avaliando as propriedades botânicas, químicas (análise térmica) e farmacêutica (spray dryer) desta planta. Assim, foram utilizadas metodologias clássicas contidas em farmacopéias e métodos analíticos mais modernos. Os resultados obtidos para a caracterização botânica são: as células epidérmicas de ambas as faces da folha são heterodimensionais. O complexo estomático é do tipo anomocítico encontrado apenas na face abaxial. O pecíolo apresenta-se plano-convexo revestido por células quadrangulares e espessa cutinização. A caracterização do pó são: a análise granulométrica classifica-o como sendo grosso. Na perda por dessecação foi de 9%; no teor de cinzas totais foi de 4,3%; com a análise térmica foi possível verificar em TG entre 31 a 100°C a primeira perda de massa de 7,8%, em DTA a 64°C um evento endotérmico e em DSC a 84°C um evento endotérmico de 169 J/g; o perfil espectroscópico na região do IV sugere a presença de compostos fenólicos. Enquanto os resultados para a tintura e extrato liofilizado são: densidade aparente de 0,97%g/mL; pH de 6; percentual de resíduo seco de 9%; na prospecção química verificou-se a presença de saponinas, açúcares redutores e fenóis e taninos para o extrato e algumas frações; na análise térmica em TG foi encontrado uma perda de massa de 6,55% entre 28 a 100°C, em DSC somente um evento exotérmico a 169°C; o perfil espectroscópico na região do IV apresentou bandas de absorção forte características de alcoóis e fenóis. No processo de secagem, os excipientes que apresentaram os melhores resultados foram aerosil 40% e celulose microcristalina mais aerosil na proporção de 35:5. O extrato apresentou características antioxidantes. A matéria-prima vegetal também foi ii testada quanto a sua atividade antimicrobiana e antifúngica, fornecendo resultados positivos para algumas cepas testadas e apresentado atividade bactericida para frações acetato de etila e hexânica. O extrato e a fração hexânica apresentaram atividade fungistática,Todos esses resultados contribuem para futuros estudos com essa planta, aumentando assim seu valor agregado.

As opalas de Pedro II e Buriti dos Montes, no estado do Piauí, constituem as mais importantes ocorrências brasileiras dessa gema, tanto em termos de volume quanto pela qualidade gemológica, que é comparável à das famosas opalas australianas. No entanto, a informalidade na extração e comercialização destas opalas, assim como a falta de informações quanto à gênese destes depósitos não permitem a prospecção por novas jazidas e o estabelecimento de um certificado de procedência para as opalas do Piauí que permitisse sua inserção formal no mercado gemológico internacional. Alguns autores têm se dedicado ao estudo dessas opalas, revelando fortes evidências de sua origem hidrotermal, mas até então, nenhum trabalho abordou as características físico-químicas dos fluidos que teriam originado esses depósitos de opalas. Diante disso, o principal objetivo deste trabalho foi entender o sistema hidrotermal responsável pela gênese das opalas do Piauí, ou seja, caracterizar os fluidos que originaram a mineralização e mostrar sua relação com o contexto geológico da região. Os municípios de Pedro II e Buriti dos Montes se localizam na porção nordeste do estado do Piauí, a aproximadamente 230 km a leste da capital Teresina, e as ocorrências de opala se encontram na porção basal da Bacia do Parnaíba, constituindo veios e vênulas nos arenitos dos grupos Serra Grande (Buriti dos Montes) e Canindé (Pedro II), os quais são seccionados por soleiras e diques de diabásio da Formação Sardinha. Elas também ocorrem cimentando brechas e como depósitos coluvionares e de paleocanal. Associados às opalas, localmente encontram-se veios de quartzo, calcedônia, barita e hematita (ou goethita). De maneira geral, as opalas de Pedro II apresentam jogo de cores, são predominantemente brancas ou azuladas com aspecto leitoso, semitranslúcidas a opacas e com inclusões sólidas pouco aparentes. Em contrapartida, as opalas de Buriti dos Montes não apresentam jogo de cores, a cor varia entre amarelo claro e vermelho amarronzado, são semitransparentes a translúcidas e contêm grande variedade de inclusões sólidas. Os dados obtidos revelam que as opalas de Pedro II são tipicamente do tipo amorfo (opala-A), enquanto as opalas de Buriti dos Montes variam entre amorfas e cristobalita-tridimita (opala-CT). Na opala preciosa, o típico jogo de cores é causado pelo arranjo regular das esferas de sílica que as constituem. A ausência de cimento opalino entre as esferas reforça a beleza desse efeito. Em contrapartida, as opalas laranja não apresentam jogo de cores, mas têm maior transparência devido ao diminuto tamanho das esferas. As inclusões sólidas também produzem belos efeitos nas opalas estudadas, principalmente na variedade laranja, que é mais transparente. Além disso, o conjunto de inclusões sólidas revela características intrínsecas aos processos hidrotermais que originaram as opalas estudadas. Agregados botrioidais, dendríticos e nodulares são exemplos de inclusões formadas por fragmentos dos arenitos hospedeiros carreados pelos fluidos hidrotermais que geraram as opalas. As inclusões sólidas também têm relação direta com a cor das opalas. Nas opalas de Buriti dos Montes, os tons de vermelho, laranja e amarelo são produzidos pela dissolução parcial das inclusões constituídas por oxihidróxidos de Fe. De maneira semelhante, a cor verde nas opalas preciosas está relacionada aos microcristais de Co-pentlandita inclusos nas mesmas. O conjunto de minerais associados às opalas conduz a uma assinatura mineralógicogeoquímica marcada pelos elevados teores de Fe e Al nas opalas com inclusões de hematita/goethita e caulinita, e assim também com aumento considerável dos teores de elementos terras raras nas opalas em que se concentram as inclusões de caulinita e apatita. Entre os elementos-traço, Ba é o mais abundante, e provavelmente foi incorporado pelo fluido hidrotermal, tendo em vista que veios de barita são encontrados com frequência nessa região da Bacia do Parnaíba. Várias feições como estruturas de fluxo nas opalas, corrosão e dissolução parcial dos cristais de quartzo hialino e de inclusões mineralógicas, vênulas de quartzo hidrotermal sobrecrescidas aos grãos detríticos, e zoneamento dos cristais de quartzo confirmam que essas opalas têm origem hidrotermal. A ruptura do Gondwana teria provocado um vasto magmatismo básico fissural, que por sua vez foi responsável pelo aporte de calor que gerou as primeiras células convectivas de fluidos quentes. A água contida nos arenitos certamente alimentou o sistema e se enriqueceu em sílica através da dissolução parcial ou total dos próprios grãos de quartzo dos arenitos. Este fluido hidrotermal foi posteriormente aprisionado em sistemas de fraturas e nelas se resfriou, precipitando a opala e minerais associados.

The socioeconomic importance of sugar cane in Brazil is unquestionable because it is the raw material for the production of ethanol and sugar. The accurate spatial intervention in the management of the crop, resulting zones of soil management, increases productivity as well as its agricultural yields. The spatial and Person's correlations between sugarcane attributes and physico-chemical attributes of a Typic Tropustalf were studied in the growing season of 2009, in Suzanápolis, State of São Paulo, Brazil (20°28'10'' S lat.; 50°49'20'' W long.), in order to obtain the one that best correlates with agricultural productivity. Thus, the geostatistical grid with 120 sampling points was installed to soil and data collection in a plot of 14.6 ha with second crop sugarcane. Due to their substantial and excellent linear and spatial correlations with the productivity of the sugarcane, the population of plants and the organic matter content of the soil, by evidencing substantial correlations, linear and spatial, with the productivity of sugarcane, were indicators of management zones strongly attached to such productivity.

The high perishability of the onion has limited its conservation period and caused great post-harvest losses. The need of consuming fresh products has led the market of minimally processed products to considerably rise. Cut operations during the minimal processing may not only cause important biochemical reactions in the onion tissues, but also affect its sensorial quality, resulting in a drastic reduction in the post-harvest life of the product. In this study the influence of different cut types was assessed on the quality of minimally processed onions. 'Baia Periforme'onions were transported to the laboratory, where they were selected, cleaned, classified, peeled, minimally processed into slices, grated and chopped, and subsequently sanitized for 10 minutes in sodium hypochlorite at 100 mg L-1. The product was kept on polystyrene trays covered with plastic film (PVC) as 80 g portions and conserved in a cold chamber at 5 +/- 1 degrees C and 85 +/- 5% RH, during 6 days. Physical, chemical and physico-chemical assays were carried out. Statistical design was completely randomized with 3 treatments and 3 replications (plot group) and 5 replicationss for the control group. For sensorial analysis (aspect and smell), 10 replications were used. To compare means, Tukey test at 5% significance was adopted. During the refrigerated conservation, sliced onions kept more stable levels of soluble solids, titratable acidity, total sugars, reducing and non-reducing sugars. The grated onion was superior considering the sensorial characteristics, indicating that the cut types interfered in these features and the grated onion was more accepted by evaluators.

The search for bioactive molecules to be employed as recognition elements in biosensors has stimulated researchers to pore over the rich Brazilian biodiversity. In this sense, we introduce the use of natural cashew gum (Anacardium occidentale L) as an active biomaterial to be used in the form of layer-by-layer films, in conjunction with phthalocyanines, which were tested as electrochemical sensors for dopamine detection. We investigated the effects of chemical composition of cashew gum from two different regions of Brazil (Piaui and Ceara states) on the physico-chemical characteristics of these nanostructures. The morphology of the nanostructures containing cashew gum was studied by atomic force microscopy which indicates that smooth films punctuated by globular features were formed that showed low roughness values. The results indicate that, independent of the origin, cashew gum stands out as an excellent film forming material with potential application in nanobiomedical devices as electrochemical sensors. In this thesis we focussed on the characterization of the reaction center (RC) protein purified from the photosynthetic bacterium Rhodobacter sphaeroides. In particular, we discussed the effects of native and artificial environment on the light-induced electron transfer processes. The native environment consist of the inner antenna LH1 complex that copurifies with the RC forming the so called core complex, and the lipid phase tightly associated with it. In parallel, we analyzed the role of saccharidic glassy matrices on the interplay between electron transfer processes and internal protein dynamics. As a different artificial matrix, we incorporated the RC protein in a layer-by-layer structure with a twofold aim: to check the behaviour of the protein in such an unusual environment and to test the response of the system to herbicides. By examining the RC in its native environment, we found that the light-induced charge separated state P+QB - is markedly stabilized (by about 40 meV) in the core complex as compared to the RC-only system over a physiological pH range. We also verified that, as compared to the average composition of the membrane, the core complex copurifies with a tightly bound lipid complement of about 90 phospholipid molecules per RC, which is strongly enriched in cardiolipin. In parallel, a large ubiquinone pool was found in association with the core complex, giving rise to a quinone concentration about ten times larger than the average one in the membrane. Moreover, this quinone pool is fully functional, i.e. it is promptly available at the QB site during multiple turnover excitation of the RC. The latter two observations suggest important heterogeneities and anisotropies in the native membranes which can in principle account for the stabilization of the charge separated state in the core complex. The thermodynamic and kinetic parameters obtained in the RC-LH1 complex are very close to those measured in intact membranes, indicating that the electron transfer properties of the RC in vivo are essentially determined by its local environment. The studies performed by incorporating the RC into saccharidic matrices evidenced the relevance of solvent-protein interactions and dynamical coupling in determining the kinetics of electron transfer processes. The usual approach when studying the interplay between internal motions and protein function consists in freezing the degrees of freedom of the protein at cryogenic temperature. We proved that the “trehalose approach” offers distinct advantages with respect to this traditional methodology. We showed, in fact, that the RC conformational dynamics, coupled to specific electron transfer processes, can be modulated by varying the hydration level of the trehalose matrix at room temperature, thus allowing to disentangle solvent from temperature effects. The comparison between different saccharidic matrices has revealed that the structural and dynamical protein-matrix coupling depends strongly upon the sugar. The analyses performed in RCs embedded in polyelectrolyte multilayers (PEM) structures have shown that the electron transfer from QA - to QB, a conformationally gated process extremely sensitive to the RC environment, can be strongly modulated by the hydration level of the matrix, confirming analogous results obtained for this electron transfer reaction in sugar matrices. We found that PEM-RCs are a very stable system, particularly suitable to study the thermodynamics and kinetics of herbicide binding to the QB site. These features make PEM-RC structures quite promising in the development of herbicide biosensors. The studies discussed in the present thesis have shown that, although the effects on electron transfer induced by the native and artificial environments tested are markedly different, they can be described on the basis of a common kinetic model which takes into account the static conformational heterogeneity of the RC and the interconversion between conformational substates. Interestingly, the same distribution of rate constants (i.e. a Gamma distribution function) can describe charge recombination processes in solutions of purified RC, in RC-LH1 complexes, in wet and dry RC-PEM structures and in glassy saccharidic matrices over a wide range of hydration levels. In conclusion, the results obtained for RCs in different physico-chemical environments emphasize the relevance of the structure/dynamics solvent/protein coupling in determining the energetics and the kinetics of electron transfer processes in a membrane protein complex.

The main objective of this thesis was the chemical characterization of synthetic secondary organic aerosol (SOA) produced from atmospherically relevant anthropogenic and biogenic VOCs during reaction chamber experiments. In parallel, the resulting chemical features of these laboratory-SOA were used to interpret the composition of ambient samples of atmospheric fine particulate matter collected at several sites in Europe, in order to determine the fraction of ambient aerosol organic mass accounted for by biogenic and anthropogenic SOA.

Transmissible spongiform encephalopathies (TSE) are neurodegenerative diseases caused by the conversion of the host-encoded cellular protein (PrPC) to a disease-associated isoform (PrPSc). The agent responsible for prion diseases may exist as different strains with specific biological and biochemical properties. According to the protein-only hypothesis, prion strain diversity is enciphered in PrPSc conformation. Molecular strain typing methods are based on the electrophoretic mobility of protease resistant core of PrPSc, on the susceptibility to protease digestion, on the glycosylation profile of PrPres and on the conformational stability of PrPSc. In this study a new conformational stability assay was developed based on the differential solubility of PrPC and PrPSc: CSSA (conformational stability and solubility assay). The conformational stability assay was performed by measuring PrPSc solubility in homogenates treated with increasing concentrations of GdnHCl, in the absence of proteinase K. Indeed, dose-response curves allowed estimation of the concentration of GdnHCl able to solubilise 50% of PrPSc. The results showed that this method is valuable for the biochemical typing of strains in bank voles and it is also a promising tool for molecular analysis of natural prion isolates. CSSA also revealed strain-specific PrPSc conformational stabilities of ovine natural isolates so that this feature, combined with the N-terminal PrPSc cleavage, allowed differentiation of classical scrapie, including CH1641-like, from natural goat BSE and experimental sheep BSE. In view of the implications concerning strain similarity between animal and human TSEs, the physico-chemical properties of the Nor98 with two human prion diseases (VPSPr and GSS) were compared in order to investigate the extent of the similarity between animal and human prion strains. The results showed an unexpected heterogeneity of the molecular features among human and sheep TSEs associated with internal PrPres fragments with the possible exception of Nor98 and a case of GSS P102L. These similarities and differences need further investigation by N- and C-terminal sequencing and biological characterization.
