Pharmaceutical residues contaminate aquatic ecosystems as a result of their widespread human and veterinary usage. Since continuously released and not efficiently removed, certain pharmaceuticals exhibit pseudo-persistence thus generating concerns for the health of aquatic wildlife. This work aimed at assessing on mussels Mytilus galloprovincialis, under laboratory conditions, the effects of three pharmaceuticals, carbamazepine (antiepileptic), propranolol (β-blocker) and oxytetracycline (antibiotic), to evaluate if the human-based mode of action of these molecules is conserved in invertebrates. Furthermore, in the framework of the European MEECE Programme, mussels were exposed to oxytetracycline and copper at increasing temperatures, simulating variations due to climate changes. The effects of these compounds were assessed evaluating a battery of biomarkers, the expression of HSP70 proteins and changes in cAMP-related parameters. A decrease in lysosomal membrane stability, induction of oxidative stress, alterations of cAMP-dependent pathway and the induction of defense mechanisms were observed indicating the development of a stress syndrome, and a worsening in mussels health status. Data obtained in MEECE Programme confirmed that the toxicity of substances can be enhanced following changes in temperature. The alterations observed were obtained after exposure to pharmaceuticals at concentrations sometimes lower than those detected in the aquatic environment. Hence, further research is advisable regarding subtle effects of pharmaceuticals on non-target organisms. Furthermore, results obtained during a research stay in the laboratories of Cádiz University (Spain) are presented. The project aimed at measuring possible effects of polluted sediments in Algeciras Bay (Spain) and in Cádiz Bay, by assessing different physiological parameters in caged crabs Carcinus maenas and clams Ruditapes decussatus exposed in situ for 28 days. The neutral red retention assay was adapted to these species and proved to be a sensitive screening tool for the assessment of sediment quality.

Negli ultimi decenni le nuove scoperte mediche e il miglioramento dello stile di vita nei paesi occidentali hanno determinato un aumento del consumo di sostanze terapeutiche, nonché della gamma di prodotti farmaceutici e parafarmaceutici comunemente utilizzati. Gli studi di cinetica dei farmaci dimostrano con certezza che essi, seppur metabolizzati, mantengono inalterato il loro potere d’azione, rimanendo biodisponibili anche una volta escreti dall’organismo. A causa della loro eccessiva polarità tali molecole sono inoltre difficilmente trattenute dai convenzionali impianti di depurazione dei reflui urbani, dai quali confluiscono inevitabilmente verso le acque fluviali e/o costiere le quali risultano. In ragione di ciò, la valutazione degli effetti che la loro presenza può provocare sulla qualità dei sistemi di approvigionamento idrico e sulla biologia delle specie tipiche degli ecosistemi acquatici, ha classificato tali composti come una nuova classe di inquinanti emergenti, il cui impatto ambientale non risulta ancora del tutto arginato attraverso adeguate contromisure legislative. I farmaci sono sostanze bioattive progettate per avere effetti specifici a bassissime concentrazioni negli organismi target attraverso specifici meccanismi d’azione. Nel caso in cui i bersagli cellulari su cui agiscono siano evolutivamente conservati negli organismi non target, essi possono esercitare le proprie funzioni attraverso i medesimi meccanismi di regolazione fisiologica attivati nelle specie target, dando origine a effetti specifici, o anche aspecifici, nel caso in cui tali bersagli siano deputati alla regolazioni di funzioni differenti. Pertanto lo scopo del presente lavoro di tesi è stato quello di analizzare le possibili alterazioni di carattere fisiologico in individui di mitilo mediterraneo (Mytilus galloprovincialis) esposti a concentrazioni ambientali di fluoxetina, farmaco antidepressivo appartenente alla classe degli inibitori selettivi del riassorbimento presinaptico della serotonina (SSRI). Più nel dettaglio, a seguito di un’esposizione per 7 giorni a dosaggi compresi tra 0.03 e 300 ng/L di FX, sono stati analizzati i livelli intracellulari di AMPc e l’attività dell’enzima PKA nei diversi trattamenti sperimentali effettuati. Inoltre sono stati valutati i livelli di espressione genica del recettore serotoninergico 5HTmyt1 e della la P-glicoproteina (Pgp, gene ABCB1), trasportatore di membrana responsabile del sistema di detossificazione noto come Multi-xenobiotic resistance (MXR). Nella ghiandola digestiva, la FX causa una diminuzione statisticamente significativa dei livelli di AMPc, dell’attività della PKA e dell’espressione del gene ABCB1 rispetto al controllo. Al contrario nel mantello il farmaco non produce alterazioni dei livelli intracellulari di AMPc e dell’attività della PKA mentre si apprezza una sottoespressione del gene ABCB1 nei trattamenti a dosaggi intermedi. In entrambi i tessuti si nota un aumento dell’espressione genica di 5HTmyt1 alle minori concentrazioni di FX, mentre ai dosaggi più alti non si registrano alterazioni significative rispetto al controllo. Nel loro insieme i risultati indicano che nei mitili, concentrazioni ambientali di FX producono significative alterazioni di diversi parametri fisiologici attraverso una modulazione specifica dei medesimi bersagli molecolari coinvolti nella terapia umana. La riduzione dei livelli di AMPc/PKA apprezzata nella ghiandola digestiva risulta in linea con la funzione inibitoria svolta dal recettore 5HTmyt1 su tale via di trasduzione, mentre l’assenza di variazioni significative registrata nel mantello supporta l’ipotesi di un’interazione tra il sistema serotoninergico e catecolaminergico nella regolazione dei processi legati al ciclo riproduttivo che si verificano in tale tessuto. In conclusione, i dati dimostrano che l’espressione del gene codificante la proteina Pgp è regolata dalla FX attraverso uno specifico meccanismo d'azione AMPc-dipendente modulato dalla serotonina; tuttavia, è ipotizzabile anche un effetto non specifico indotto dalla FX stessa, per esempio attraverso l’induzione di stress ossidativo. Inoltre essi evidenziano la presenza di un meccanismo di regolazione retroattivo sulla espressione dei recettori 5HTmyt1 in funzione delle concentrazioni extracellulari di serotonina modulate dall’azione della FX.

Herbicides are becoming emergent contaminants in Italian surface, coastal and ground waters, due to their intensive use in agriculture. In marine environments herbicides have adverse effects on non-target organisms, as primary producers, resulting in oxygen depletion and decreased primary productivity. Alterations of species composition in algal communities can also occur due to the different sensitivity among the species. In the present thesis the effects of herbicides, widely used in the Northern Adriatic Sea, on different algal species were studied. The main goal of this work was to study the influence of temperature on algal growth in the presence of the triazinic herbicide terbuthylazine (TBA), and the cellular responses adopted to counteract the toxic effects of the pollutant (Chapter 1 and 2). The development of simulation models to be applied in environmental management are needed to organize and track information in a way that would not be possible otherwise and simulate an ecological prospective. The data collected from laboratory experiments were used to simulate algal responses to the TBA exposure at increasing temperature conditions (Chapter 3). Part of the thesis was conducted in foreign countries. The work presented in Chapter 4 was focused on the effect of high light on growth, toxicity and mixotrophy of the ichtyotoxic species Prymnesium parvum. In addition, a mesocosm experiment was conducted in order to study the synergic effect of the pollutant emamectin benzoate with other anthropogenic stressors, such as oil pollution and induced phytoplankton blooms (Chapter 5).

L’elevata presenza dei residui dei farmaci nell’ambiente acquatico desta preoccupazione per la salute della fauna acquatica e per un eventuale rischio per l’uomo. Dopo l’assunzione, i farmaci vengono escreti come tali o come metaboliti attivi, risultando spesso resistenti ai processi di trattamento delle acque. Per questo motivo alcuni di essi sono pseudo-persistenti raggiungendo concentrazioni di ng-µg/L. I farmaci sono molecole disegnate per essere biologicamente attive a basse concentrazioni su bersagli specifici, per questo motivo possono indurre effetti anche su specie non target con bersagli molecolari simili all’uomo a concentrazioni ambientali. A tal proposito il presente lavoro intende investigare gli effetti della caffeina, ampiamente usata come costituente di bevande e come farmaco, presente nelle acque superficiali a concentrazioni di ng-µg/L. Come organismo di studio è stato scelto il Mytilus galloprovincialis, sfruttando le conoscenze disponibili in letteratura circa le sue risposte ai contaminanti ambientali. I mitili sono stati esposti in acquario a caffeina (5, 50 e 500 ng/L) per 7 giorni e poi analizzati attraverso una batteria di otto biomarker, alterazioni fisiologiche o biochimiche che forniscono informazioni circa lo stato di salute degli animali. I metodi utilizzati sono stati diversi a seconda dei biomarker (analisi citochimiche e saggi enzimatici). Le concentrazioni sono state scelte nel range ambientale. L’esposizione ha prodotto alterazioni della stabilità della membrana lisosomiale negli emociti e l’instaurarsi di processi di detossificazione nella ghiandola digestiva, evidenziati dall’aumento dell’attività della glutatione S-transferasi. Gli altri biomarker non mettono in evidenza che la caffeina, in queste condizioni sperimentali, possa indurre alterazioni della funzionalità lisosomiale, effetti ossidativi o neurotossici. I dati ottenuti sui mitili, quindi, suggeriscono che la caffeina, anche nel range di concentrazioni ambientali più elevato, possa essere considerata un contaminante che desta bassa preoccupazione.

This thesis aimed at addressing some of the issues that, at the state of the art, avoid the P300-based brain computer interface (BCI) systems to move from research laboratories to end users’ home. An innovative asynchronous classifier has been defined and validated. It relies on the introduction of a set of thresholds in the classifier, and such thresholds have been assessed considering the distributions of score values relating to target, non-target stimuli and epochs of voluntary no-control. With the asynchronous classifier, a P300-based BCI system can adapt its speed to the current state of the user and can automatically suspend the control when the user diverts his attention from the stimulation interface. Since EEG signals are non-stationary and show inherent variability, in order to make long-term use of BCI possible, it is important to track changes in ongoing EEG activity and to adapt BCI model parameters accordingly. To this aim, the asynchronous classifier has been subsequently improved by introducing a self-calibration algorithm for the continuous and unsupervised recalibration of the subjective control parameters. Finally an index for the online monitoring of the EEG quality has been defined and validated in order to detect potential problems and system failures. This thesis ends with the description of a translational work involving end users (people with amyotrophic lateral sclerosis-ALS). Focusing on the concepts of the user centered design approach, the phases relating to the design, the development and the validation of an innovative assistive device have been described. The proposed assistive technology (AT) has been specifically designed to meet the needs of people with ALS during the different phases of the disease (i.e. the degree of motor abilities impairment). Indeed, the AT can be accessed with several input devices either conventional (mouse, touchscreen) or alterative (switches, headtracker) up to a P300-based BCI.

In Medicina Veterinaria l'avvelenamento da rodenticidi anticoagulanti è conosciuto e studiato ormai da anni, essendo una delle intossicazioni più comunemente riscontrate nelle specie non target. In letteratura si rinvengono numerose pubblicazioni ma alcuni aspetti sono rimasti ancora inesplorati.Questo studio si propone di valutare il processo infiammatorio, mediante le proteine di fase acuta (APPs), in corso di fenomeni emorragici, prendendo come modello reale un gruppo di soggetti accidentalmente avvelenati da rodenticidi anticoagulanti. I 102 soggetti avvelenati presentano un valore più elevato di proteina C reattiva (CRP)con una mediana di 4.77 mg/dl statisticamente significativo rispetto alla mediana delle due popolazioni di controllo di pari entità numerica create con cross match di sesso, razza ed età; rispettivamente 0.02 mg/dl dei soggetti sani e 0.37 mg/dl dei soggetti malati di altre patologie. Inoltre all'interno del gruppo dei soggetti avvelenati un valore di CRP elevato all'ammissione può predisporre al decesso. La proteina C reattiva assume quindi un ruolo diagnostico e prognostico in questo avvelenamento. Un'altra finalità, di non inferiore importanza, è quella di definire una linea guida terapeutica con l'ausilio di biomarker coagulativi e di valutare la sicurezza della vitamina K per via endovenosa: in 73 cani, non in terapia con vitamina k, intossicati da rodenticidi anticoagulanti, i tempi della coagulazione (PT ed aPTT) ritornano nel range di normalità dopo 4 ore dalla prima somministrazione di 5 mg/kg di vitamina k per via endovenosa e nessun soggetto durante e dopo il trattamento ha manifestato reazioni anafilattiche, nessuno dei pazienti ha necessitato trasfusione ematica e tutti sono sopravvissuti. Infine si è valutata l'epidemiologia dell'ingestione dei prodotti rodenticidi nella specie oggetto di studio e la determinazione dei principi attivi mediante cromatografia liquida abbinata a spettrofotometria di massa (UPLC-MS/MS).

Eisbohrkerne stellen wertvolle Klimaarchive dar, da sie atmosphärisches Aerosol konservieren. Die Analyse chemischer Verbindungen als Bestandteil atmosphärischer Aerosole in Eisbohrkernen liefert wichtige Informationen über Umweltbedingungen und Klima der Vergangenheit. Zur Untersuchung der α-Dicarbonyle Glyoxal und Methylglyoxal in Eis- und Schneeproben wurde eine neue, sensitive Methode entwickelt, die die Stir Bar Sorptive Extraction (SBSE) mit der Hochleistungsflüssigchromatographie-Massenspektrometrie (HPLC-MS) kombiniert. Zur Analyse von Dicarbonsäuren in Eisbohrkernen wurde eine weitere Methode entwickelt, bei der die Festphasenextraktion mit starkem Anionenaustauscher zum Einsatz kommt. Die Methode erlaubt die Quantifizierung aliphatischer Dicarbonsäuren (≥ C6), einschließlich Pinsäure, sowie aromatischer Carbonsäuren (wie Phthalsäure und Vanillinsäure), wodurch die Bestimmung wichtiger Markerverbindungen für biogene und anthropogene Quellen ermöglicht wurde. Mit Hilfe der entwickelten Methoden wurde ein Eisbohrkern aus den Schweizer Alpen analysiert. Die ermittelten Konzentrationsverläufe der Analyten umfassen die Zeitspanne von 1942 bis 1993. Mittels einer Korrelations- und Hauptkomponentenanalyse konnte gezeigt werden, dass die organischen Verbindungen im Eis hauptsächlich durch Waldbrände und durch vom Menschen verursachte Schadstoffemissionen beeinflusst werden. Im Gegensatz dazu sind die Konzentrationsverläufe einiger Analyten auf den Mineralstaubtransport auf den Gletscher zurückzuführen. Zusätzlich wurde ein Screening der Eisbohrkernproben mittels ultrahochauflösender Massenspektrometrie durchgeführt. Zum ersten Mal wurden in diesem Rahmen auch Organosulfate und Nitrooxyorganosulfate in einem Eisbohrkern identifiziert.

Speläotheme und Tropfwasser spielen eine wichtige Rolle bei der Erforschung von Naturerscheinungen. Während sich bisherige Studien größtenteils auf anorganische Proxies konzentrieren, wächst das Interesse an organischen Biomarkern, vor allem Lipidbiomarkern.rnAufgrund dessen wurde eine neue Methode entwickelt, um Fettsäuren mit einer geradzahligen Kettenlänge C12-C20 in Speläothem- und Tropfwasserproben zu bestimmen. Dabei kam eine Festphasenextraktion mit anschließender HPLC-MS Messung zum Einsatz. Die Methode wurde auf mehrere Proben aus der Herbstlabyrinth-Adventhöhle angewendet. Die benötigte Probenmenge wurde im Vergleich zu früheren Studien von etwa 4 L auf 60-100 mL bei Tropfwasser und von 10-100 g auf ca. 0,5-3,5 g bei Stalagmitproben reduziert. Es konnte eine interne Variation der Analyten festgestellt werden. Korrelationen der Fettsäuren ließen vermuten, dass C12-C18 von der gleichen Quelle stammen, während C20 teilweise andere Quellen besitzt. Korrelationen mit δ13C verdeutlichten, dass es einen Zusammenhang zwischen der Vegetationsdichte und dem Vorkommen der Fett-säuren in dem Probenmaterial gibt. Vergleiche mit Mg Konzentrationen zeigten, dass die Niederschlagsmenge zwar den Transport der Fettsäuren mit dem Tropfwasser beeinflusst, allerdings nicht ihre ermittelte Konzentration in Stalagmitproben. Die ermittelten Ergebnisse ließen darauf schließen, dass eindeutigere Vegetations-Proxies als die Fettsäuren gefunden werden müssen. Ein erstmaliges non-target screening verdeutlichte, dass Lignine und Tannine als charakteristische Biomarker eingesetzt werden können.rn

The cultivation of genetically modified (GM) plants has raised several environmental concerns. One of these concerns regards non-target soil fauna organisms, which play an important role in the decomposition of organic matter and hence are largely exposed to GM plant residues. Soil fauna may be directly affected by transgene products or indirectly by pleiotropic effects such as a modified plant metabolism. Thus, ecosystem services and functioning might be affected negatively. In a litterbag experiment in the field we analysed the decomposition process and the soil fauna community involved. Therefore, we used four experimental GM wheat varieties, two with a race-specific antifungal resistance against powdery mildew (Pm3b) and two with an unspecific antifungal resistance based on the expression of chitinase and glucanase. We compared them with two non-GM isolines and six conventional cereal varieties. To elucidate the mechanisms that cause differences in plant decomposition, structural plant components (i.e. C:N ratio, lignin, cellulose, hemicellulose) were examined and soil properties, temperature and precipitation were monitored. The most frequent taxa extracted from decaying plant material were mites (Cryptostigmata, Gamasina and Uropodina), springtails (Isotomidae), annelids (Enchytraeidae) and Diptera (Cecidomyiidae larvae). Despite a single significant transgenic/month interaction for Cecidomyiidae larvae, which is probably random, we detected no impact of the GM wheat on the soil fauna community. However, soil fauna differences among conventional cereal varieties were more pronounced than between GM and non-GM wheat. While leaf residue decomposition in GM and non-GM wheat was similar, differences among conventional cereals were evident. Furthermore, sampling date and location were found to greatly influence soil fauna community and decomposition processes. The results give no indication of ecologically relevant adverse effects of antifungal GM wheat on the composition and the activity of the soil fauna community.

PURPOSE: The aim of this study was to evaluate [(99m)Tc]Demotate 2 ([(99m)Tc-N(4) (0-1),Asp(0),Tyr(3)]octreotate) as a candidate for in vivo imaging of sst(2)-positive tumours and to compare it with [(111)In]DOTA-tate ([(111)In-DOTA(0),Tyr(3)]octreotate). METHODS: Labelling of Demotate 2 with (99m)Tc was performed at room temperature using SnCl(2) as reductant in the presence of citrate at alkaline pH. Radiochemical analysis involved ITLC and HPLC methods. Peptide conjugate affinities for sst(2) were determined by receptor autoradiography on rat brain cortex sections using [DOTA(0),(125)I-Tyr(3)]octreotate as the radioligand. The affinity profile of Demotate 2 for human sst(1)-sst(5) was studied by receptor autoradiography in cell preparations using the universal somatostatin radioligand [(125)I][Leu(8),(D: )Trp(22),Tyr(25)]somatostatin-28. The internalisation rates of [(99m)Tc]Demotate 2 and [(111)In]DOTA-tate were compared in sst(2)-positive and -negative control cell lines. Biodistribution of radiopeptides was studied in male Lewis rats bearing CA20948 tumours. RESULTS: Peptide conjugates showed selectivity and a high affinity binding for sst(2) (Demotate 2 IC(50)=3.2 nM and DOTA-tate IC(50)=5.4 nM). [(99m)Tc]Demotate 2, like [(111)In]DOTA-tate, internalised rapidly in all sst(2)-positive cells tested, but not in sst(2)-negative control cells. After injection in CA20948 tumour-bearing rats both radiopeptides showed high and specific uptake in the sst(2)-positive organs and in the implanted tumour and rapid excretion from non-target tissues via the kidneys. CONCLUSION: [(99m)Tc]Demotate 2, similarly to the known sst(2)-targeting agent [(111)In]DOTA-tate, showed promising biological qualities for application in the scintigraphy of sst(2)-positive tumours.

PURPOSE: We aimed at designing and developing a novel bombesin analogue, DOTA-PEG(4)-BN(7-14) (DOTA-PESIN), with the goal of labelling it with (67/68)Ga and (177)Lu for diagnosis and radionuclide therapy of prostate and other human cancers overexpressing bombesin receptors. METHODS: The 8-amino acid peptide bombesin (7-14) was coupled to the macrocyclic chelator DOTA via the spacer 15-amino-4,7,10,13-tetraoxapentadecanoic acid (PEG(4)). The conjugate was complexed with Ga(III) and Lu(III) salts. The GRP receptor affinity and the bombesin receptor subtype profile were determined in human tumour specimens expressing the three bombesin receptor subtypes. Internalisation and efflux studies were performed with the human GRP receptor cell line PC-3. Xenografted nude mice were used for biodistribution. RESULTS: [Ga(III)/Lu(III)]-DOTA-PESIN showed good affinity to GRP and neuromedin B receptors but no affinity to BB3. [(67)Ga/(177)Lu]-DOTA-PESIN internalised rapidly into PC-3 cells whereas the efflux from PC-3 cells was relatively slow. In vivo experiments showed a high and specific tumour uptake and good retention of [(67)Ga/(177)Lu]-DOTA-PESIN. [(67)Ga/(177)Lu]-DOTA-PESIN highly accumulated in GRP receptor-expressing mouse pancreas. The uptake specificity was demonstrated by blocking tumour uptake and pancreas uptake. Fast clearance was found from blood and all non-target organs except the kidneys. High tumour-to-normal tissue ratios were achieved, which increased with time. PET imaging with [(68)Ga]-DOTA-PESIN was successful in visualising the tumour at 1 h post injection. Planar scintigraphic imaging showed that the (177)Lu-labelled peptide remained in the tumour even 3 days post injection. CONCLUSION: The newly designed ligands have high potential with regard to PET and SPECT imaging with (68/67)Ga and targeted radionuclide therapy with (177)Lu.

Ecological disturbances may be caused by a range of biotic and abiotic factors. Among these are disturbances that result from human activities such as the introduction of exotic plants and land management activities. This dissertation addresses both of these types of disturbance in ecosystems in the Upper Peninsula of Michigan. Invasive plants are a significant cause of disturbance at Pictured Rocks Natural Lakeshore. Management of invasive plants is dependent on understanding what areas are at risk of being invaded, what the consequences of an invasion are on native plant communities and how effective different tools are for managing the invasive species. A series of risk models are described that predict three stages of invasion (introduction, establishment and spread) for eight invasive plant species at Pictured Rocks National Lakeshore. These models are specific to this location and include species for which models have not previously been produced. The models were tested by collecting point data throughout the park to demonstrate their effectiveness for future detection of invasive plants in the park. Work to describe the impacts and management of invasive plants focused on spotted knapweed in the sensitive Grand Sable Dunes area of Pictured Rocks National Lakeshore. Impacts of spotted knapweed were assessed by comparing vegetation communities in areas with varying amounts of spotted knapweed. This work showed significant increases in species diversity in areas invaded by knapweed, apparently as a result of the presence of a number of non-dune species that have become established in spotted knapweed invaded areas. An experiment was carried out to compare annual spot application of two herbicides, Milestone® and Transline® to target spotted knapweed. This included an assessment of impacts of this type of treatment on non-target species. There was no difference in the effectiveness of the two herbicides, and both significantly reduced the density of spotted knapweed during the course of the study. Areas treated with herbicide developed a higher percent cover of grasses during the study, and suffered limited negative impacts on some sensitive dune species such as beach pea and dune stitchwort, and on some other non-dune species such as hawkweed. The use of these herbicides to reduce the density of spotted knapweed appears to be feasible over large scales.

The influence of the immediate prestimulus EEG microstate (sub-second epoch of stable topography/map landscape) on the map landscape of visually evoked 47-channel event-related potential (ERP) microstates was examined using the frequent, non-target stimuli of a cognitive paradigm (12 volunteers). For the two most frequent prestimulus microstate classes (oriented left anterior-right posterior and right anterior-left posterior), ERP map series were selectively averaged. The post-stimulus ERP grand average map series was segmented into microstates; 10 were found. The centroid locations of positive and negative map areas were extracted as landscape descriptors. Significant differences (MANOVAs and t-tests) between the two prestimulus classes were found in four of the ten ERP microstates. The relative orientation of the two ERP microstate classes was the same as prestimulus in some ERP microstates, but reversed in others. — Thus, brain electric microstates at stimulus arrival influence the landscapes of the post-stimulus ERP maps and therefore, information processing; prestimulus microstate effects differed for different post-stimulus ERP microstates.

Conventional risk assessments for crop protection chemicals compare the potential for causing toxicity (hazard identification) to anticipated exposure. New regulatory approaches have been proposed that would exclude exposure assessment and just focus on hazard identification based on endocrine disruption. This review comprises a critical analysis of hazard, focusing on the relative sensitivity of endocrine and non-endocrine endpoints, using a class of crop protection chemicals, the azole fungicides. These were selected because they are widely used on important crops (e.g. grains) and thereby can contact target and non-target plants and enter the food chain of humans and wildlife. Inhibition of lanosterol 14α-demethylase (CYP51) mediates the antifungal effect. Inhibition of other CYPs, such as aromatase (CYP19), can lead to numerous toxicological effects, which are also evident from high dose human exposures to therapeutic azoles. Because of its widespread use and substantial database, epoxiconazole was selected as a representative azole fungicide. Our critical analysis concluded that anticipated human exposure to epoxiconazole would yield a margin of safety of at least three orders of magnitude for reproductive effects observed in laboratory rodent studies that are postulated to be endocrine-driven (i.e. fetal resorptions). The most sensitive ecological species is the aquatic plant Lemna (duckweed), for which the margin of safety is less protective than for human health. For humans and wildlife, endocrine disruption is not the most sensitive endpoint. It is concluded that conventional risk assessment, considering anticipated exposure levels, will be protective of both human and ecological health. Although the toxic mechanisms of other azole compounds may be similar, large differences in potency will require a case-by-case risk assessment.

During November 2010–February 2011, we used camera traps to estimate the population density of Eurasian lynx Lynx lynx in Ciglikara Nature Reserve, Turkey, an isolated population in southwest Asia. Lynx density was calculated through spatial capture—recapture models. In a sampling eff ort of 1093 camera trap days, we identifi ed 15 independent individuals and estimated a density of 4.20 independent lynx per 100 km2, an unreported high density for this species. Camera trap results also indicated that the lynx is likely to be preying on brown hare Lepus europaeus, which accounted for 63% of the non-target species pictured. As lagomorph populations tend to fl uctuate, the high lynx density recorded in Ciglikara may be temporary and may decline with prey fl uctuation. Therefore we recommend to survey other protected areas in southwestern Turkey where lynx is known or assumed to exist, and continuously monitor the lynx populations with reliable methods in order to understand the populations structure and dynamics, defi ne sensible measures and management plans to conserve this important species.
