The Saccharomyces cerevisiae RAD1 and human XPF genes encode a subunit of a nucleotide excision repair endonuclease that also is implicated in some forms of homologous recombination. An Arabidopsis thaliana gene (AtRAD1) encoding the orthologous plant protein has been identified recently. Here we report the isolation of three structurally distinct AtRAD1 cDNAs from A. thaliana leaf tissue RNA. One of the isolates (AtRAD1-1) corresponds to the cDNA previously shown to encode the full-length AtRad1 protein, whereas the other two (AtRAD1-2, AtRAD1-3) differ slightly in size due to variations at the 5′ end of exon 6 or the 3′ end of exon 7, respectively. The sequence differences argue that these cDNAs were probably templated by mRNAs generated via alternative splicing. Diagnostic polymerase chain reaction pointed to the presence of the AtRAD1-1 and AtRAD1-2 but not AtRAD1-3 transcripts in bud and root tissue, and to a fourth transcript (AtRAD1-4), having both alterations identified in AtRAD1-2 and AtRAD1-3, in root tissue. However, the low frequency of detection of AtRAD1-3 and AtRAD1-4 makes the significance of these tissue-specific patterns unclear. The predicted AtRad1-2, AtRad1-3 and AtRad1-4 proteins lack part of the region likely required for endonuclease complex formation. Expression of AtRAD1-2 and AtRAD1-3 in a yeast rad1 mutant did not complement the sensitivity to ultraviolet radiation or the recombination defect associated with the rad1 mutation. These results suggest that alternative splicing may modulate the levels of functional AtRad1 protein.

Buruli ulcer disease (BUD), a devastating tropical disease caused by Mycobacterium ulcerans, occurs in more than 80% of the administrative districts of Ghana. To elucidate community perceptions and
understanding of the aetiology of BUD, attitudes towards Buruli patients and treatment-seeking behaviours, we conducted a survey with 504 heads of households and seven focus group discussions in Ga West District, Ghana. Although 67% of participants regarded BUD as a health problem, 53% did not know its cause. Sixteen per cent attributed the cause to drinking non-potable water, 8.1% mentioned poor personal hygiene or dirty surroundings, and 5.5% identified swimming or wading in ponds as a risk factor. About 5.2% thought that witchcraft and curses cause BUD, and 71.8% indicated that BU sufferers first seek treatment from herbalists and only refer to the hospital as a last resort. The main
reasons were prospects of prolonged hospital stay, cost of transport, loss of earnings and opportunity associated with parents attending their children’s hospitalization over extended period, delays in being
attended by medical staff, and not knowing the cause of the disease or required treatment. The level of acceptance of BUD sufferers was high in adults but less so in children. The challenge facing health workers is to break the vicious cycle of poor medical outcomes leading to poor attitudes to hospital treatment in the community. Because herbalists are often the first people consulted by those who contract the disease, they need to be trained in early recognition of the pre-ulcerative stage of Buruli lesions.

In this paper we present our preliminary studies on the efficacy of eugenol for the recovery of erased characters on polymers. We have found that eugenol rapidly recovers erased characters when applied to the surface of polymer substrates. By applying eugenol to the surface to be treated using a cotton bud or paintbrush, we were able to revisualize erased characters from a range of polymers, inclluding acrylonitrile butadiene styrene, acrylic, high-impact polystrene, and polystyrene. Eugenol is safe, nonhazardour, and easily sourced reagent for this purpose.

A pollen chase experiment was performed upon three Costa Rican populations of Witheringia solanacea to examine the breakdown of genetically enforced self incompatibility (SI) and the extent of embryonic inbreeding depression. Self-pollen was applied in the bud, with outcross pollen applied one day later, and outcross pollinations at both intervals as a control. A variety of responses were found among the populations. BOHS readily accepted self pollen and suffered from very low inbreeding depression. Monteverde and Las Cruces both have lower fruit set with self-pollination precedence indicating that bud pollinations can overcome the self-incompatibility response and that embryonic death due to inbreeding depression causes fruit failure. The treatment:control fruit set is higher for the Las Cruces plants indicating stronger SI response Self-precedence seeds from the Las Cruces plants are likely to be outcrossed. Self-precedence seeds from Monteverde are likely selfed.

Anemophilous plants described as catapulting pollen explosively into the air have rarely attracted detailed examination. We investigated floral anthesis in a male mulberry tree with high-speed video and a force probe. The stamen was inflexed within the floral bud. Exposure to dry air initially resulted in a gradual movement of the stamen. This caused fine threads to tear at the stomium, ensuring dehiscence of the anther, and subsequently enabled the anther to slip off a restraining pistillode. The sudden release of stored elastic energy in the spring-like filament drove the stamen to straighten in less than 25 μs, and reflex the petals to velocities in excess of half the speed of sound. This is the fastest motion yet observed in biology, and approaches the theoretical physical limits for movements in plants.

Yeast cells begin to bud and enter S phase when growth conditions are favourable during G1 phase. When subjected to some oxidative stresses, cells delay entry at G1 allowing repair of cellular damage. Hence, oxidative stress sensing is coordinated with the regulation of cell cycle. We identified a novel function of the cell-cycle regulator of Saccharomyces cerevisiae, Swi6p, as a redox sensor through its cysteine residue at position 404. When alanine was substituted at this position, the resultant mutant, C404A, was sensitive to several reactive oxygen species and oxidants including linoleic acid hydroperoxide, the superoxide anion and diamide. This mutant lost the ability to arrest in G1 phase upon treatment with lipid hydroperoxide. The Cys404 residue of Swi6p in wild-type cells was oxidised to a sulfenic acid when cells were subjected to linoleic acid hydroperoxide. Mutation of Cys404 to Ala abolished the down-regulation of expression of the G1 cyclin genes CLN1, CLN2, PCL1 and PCL2 that occurred when cells of the wild type were exposed to the lipid hydroperoxide. In conclusion, oxidative stress signaling for cell-cycle regulation occurs through oxidation of the G1/S-speicific transcription factor Swi6p and consequently leads to suppression of the expression of G1-cyclins and delay in cells entering the cell cycle.

Taste is the chemical sense responsible for the detection of non-volatile chemicals in potential foods. For fat to be considered as one of the taste primaries in humans, certain criteria must be met including: class of affective stimuli; receptors specific for the class of stimuli on taste bud cells (TBC); afferent fibers from TBC to taste processing regions of the brain; perception independent of other taste qualities; and downstream physiological effects. The breakdown products of the macronutrients carbohydrates (sugars) and protein (amino acids) are responsible for activation of sweet and umami tastes respectively. Following the same logic the breakdown products of fat being fatty acids are the likely class of stimuli for fat taste. Indeed, psychophysical studies have confirmed fatty acids of varying chain length and saturation are orally detectable by humans. The most likely fatty acid receptor candidates located on TBC are CD36, G protein-coupled receptor 120. Once the receptors are activated by fatty acids a series of transduction events occurs causing the release of neurotransmitters towards afferent fibers signalling the brain. Whether fatty acids elicit any direct perception independent of other taste qualities is still open to debate with only poorly defined perceptions for fatty acids reported. Others suggest that the fatty acid taste component is at detection threshold only and any perceptions are associated with either aroma or chemesthesis. It has also been established that oral exposure to fat via sham feeding stimulates increases blood triacylglycerol concentrations in humans. Therefore, overall, with the exception of an independent perception, there is consistent emerging evidence that fat is the sixth taste primary. The implications of fatty acid taste go further into health and obesity research with the gustatory detection of fats and their contributions to energy and fat intake receiving increasing attention. There appears to be a coordinated bodily response to fatty acids throughout the alimentary canal; those who are insensitive orally are also insensitive in the gastrointestinal tract and overconsume fatty food and energy. The likely mechanism linking fatty acid taste insensitivity with overweight and obesity is development of satiety after consumption of fatty foods.

Este trabalho teve como objetivo estudar a manutenção da viabilidade das borbulhas de laranjeira ‘Valência’ e tangerineira ‘Montenegrina’, oriundas de ambiente protegido e pomar, sob diferentes processos de desinfestação e períodos de armazenamento em câmara fria, assim como avaliar o comportamento das substâncias de reserva contidas nos ramos porta-borbulhas nos diferentes períodos de armazenamento. Com estes objetivos foram conduzidos dois experimentos em câmara fria com temperatura em torno de 5ºC. No experimento 1, testou-se borbulhas de duas cultivares de citros oriundas de ambiente protegido, três tratamentos químicos (testemunha, 1x tratado com Captan e 2x tratado com Captan na dose de 10g/L) e três períodos de armazenamento (0, 90, e 180 dias), com quatro repetições. No experimento 2, testou-se borbulhas de duas cultivares de citros oriundas do pomar de citros, três tratamentos químicos (testemunha, 1x tratado com Captan e 2x tratado com Captan na dose de 10g/L) e cinco períodos de armazenamento (0, 45, 90, 135 e 180 dias), com quatro repetições. As borbulhas da cultivar Montenegrina, mantiveram-se viáveis por 90 dias sem tratamento com fungicida e por, no mínimo, 180 dias se submetidas a 1 tratamento com fungicida no momento de seu armazenamento. As borbulhas da cultivar Valência, coletadas de ambiente protegido, mantiveram-se viáveis por, no mínimo, 180 dias sem a necessidade de tratamento com fungicida. As substâncias de reserva dos ramos porta-borbulhas sofreram redução ao longo do armazenamento.

A radioproteção dos pacientes submetidos a exames radiográficos está diretamente ligada à qualidade e à repetição das radiografias realizadas. Esta dissertação é apresentada sob a forma de três artigos. O artigo I avaliou a qualidade de 300 radiografias panorâmicas enviadas a clínicas de ortodontia. As radiografias foram classificadas como excelentes (não se observam erros), aceitáveis para diagnóstico (observam-se erros, contudo os mesmos não impedem o diagnóstico) e inaceitáveis (imagem sem valor diagnóstico). Um total de 16,33% das radiografias foi considerado excelente, 78,66% aceitáveis para o diagnóstico e 5% inaceitáveis. Encontrou-se uma média de 1.54 erros por radiografia, sendo os mais freqüentemente encontrados a falta de contato da língua com o palato (21%), aparecimento de imagens fantasma (19,66%), mento inclinado para cima (15,66%), paciente à frente do plano de foco (13,33%), cabeça girada (13,33%), imagens com alta densidade (10,33%) e com baixa densidade (8,66%). Concluiu-se que os padrões de qualidade da amostra estão de acordo com o preconizado em Guidelines on Radiology Standards for Primary Dental Care, segundo os quais se admite até 10% de imagens inaceitáveis. Contudo, cabe salientar que não se conhece o índice de repetições nas clínicas onde as radiografias da amostra foram obtidas e, tendo passado por um controle de qualidade prévio, todas as imagens deveriam ser classificadas como excelentes ou aceitáveis. O artigo II avaliou a freqüência dos erros que levaram à repetição de radiografias panorâmicas realizadas no Serviço de Radiologia da FO-UFRS. O livro de registros do Serviço mostrou um total de 3815 radiografias panorâmicas realizadas no período de junho de 2002 a junho de 2005. No mesmo período o Serviço apresentou 330 radiografias panorâmicas repetidas, resultando em índice de repetição de 8,65% dos exames. Os erros mais freqüentemente encontrados foram: paciente posicionado à frente do plano de foco (25,15%); cabeça girada para direita ou esquerda (24,84%); cabeça inclinada para frente (21,21%); paciente posicionado atrás do plano de foco (20,30%); imagem com a alta densidade (19,69%); imagem com baixa densidade (17,27%); imagem com baixo contraste (16,96%); imagem com alto contraste (12,72%); cabeça inclinada para direita ou esquerda (12,42%); corte do côndilo na radiografia (11,21); corte do mento na radiografia (8,48%); ausência de contato da língua com o palato (7,27%); paciente se moveu durante a exposição (4,94%); cabeça inclinada para trás (2,72%) e aparecimento de imagem fantasma (2,12%). Encontrou-se uma média de 2,07 erros por radiografia. Conclui-se que os erros mais freqüentemente cometidos foram classificados como erros de posicionamento do paciente e que o Serviço de Radiologia da FO-UFRGS apresenta um índice de repetição de radiografias panorâmicas satisfatório, estando de acordo com os padrões de qualidade estabelecidos pelo Guidelines on Radiology Standards for Primary Dental Care. O artigo III teve por objetivo verificar o efeito da radiação emitida em radiografias panorâmicas sobre as células da borda lateral direita da língua, através da avaliação das alterações nucleares, antes e depois da exposição aos raios X. A amostra foi constituída de 42 indivíduos adultos jovens do gênero masculino, sendo 22 deles pertencentes ao grupo que realizou uma radiografia panorâmica (grupo I) e os outros 20 pacientes pertencentes ao grupo II, que realizou duas radiografias panorâmicas. O exame citopatológico das células esfoliadas da mucosa da língua foi realizado antes da incidência radiográfica e 10 dias após. As células foram obtidas através da raspagem e as lâminas foram coradas pela técnica de Feulgen. Para observação das alterações citopatológicas foram analisadas 2.000 células em cada lâmina e quantificados os micronúcleos, buds, broken eggs, cariorrexes e células binucleadas. As lâminas foram analisadas por um único observador. Constatou-se que existe diferença significativa (p=0,01) para as variáveis broken egg, bud, cariorrexe e célula binucleada antes e depois da exposição à radiação ionizante. Na comparação entre os grupos, verificou-se que as variáveis cariorrexe e célula binucleada apresentaram diferença significativa (p=0,01), ambas com valores superiores para o grupo II.

The external morphology of seeds and post-germination developmental stages of Angelonia salicariifolia Bonpl. (Scrophulariaceae) were investigated using scanning electron microscopy. Some structural features of the seed exotesta and seedling in Angelonia are presented for the first time and are of potential taxonomic value for this neotropical genus. The seeds are very small (0.9-1.7 mm long and 0.5-0.9 mm wide), ovate, with a reticulate-crested exotesta, reticules arranged uniformly in longitudinal rows, with a high density of microcilia-like projections on the cell wall of the reticule base and on the edge of the crests. The hilum is located beside the micropyle at the narrow end of the seed. Germination is epigeal. During germination the radicle develops, followed by elongation of the hypocotyl and primary root. At this stage dense root hairs develop on the lower part of the hypocotyl. The apical bud-located between the cotyledons-begins to develop after the cotyledons have unfolded. The cotyledons are equal in size, sessile and ovate. The seedlings have two types of trichomes, one characteristic of the cotyledons and first pair of leaves (glandular, sessile, four-celled head with quadrangular shape) and the other characteristic of the hypocotyl and epicotyl (stalked, erect, elongate and three-celled with dome-shaped unicellular head). Vulvovaginal candidiasis (VVC) is one of the most common causes of vaginitis and affects about 75% of women of reproductive age. The majority of cases (80 to 90%) are due to C. albicans, the most virulent species of the genus Candida. Virulence attributes are scarcely investigated and the source of infection remains uncertain. Objective: This study aimed to evaluate the virulence factors and genotypes of clinical isolates of C. albicans sequentially obtained from the anus and vagina of patients with sporadic and recurrent VVC. Materials and methods: We analyzed 62 clinical isolates of C. albicans (36 vaginal and 26 anal strains). Direct examination of vaginal and anal samples and colony forming units (CFU) counts were performed. Yeasts were identified using the chromogenic media CHROMagar Candida® and by classical methodology, and phenotypically characterized regarding to virulence factors, including the ability to adhere to epithelial cells, proteinase activity, morphogenesis and biofilm formation. The genotypes of the strains were investigated with ABC genotyping, microsatellite genotyping with primer M13 and RAPD. Results: We found 100% agreement between direct examination and culture of vaginal samples. Filamentous forms were present in most of the samples of vaginal secretion, which presented CFU counts significantly higher than the samples of anal secretion. There was no statistically significant difference between virulence factors of infecting vaginal isolates and those presented by colonizing anal isolates; as well as for the comparison of the vaginal isolates from patients with different clinical conditions (sporadic or recurrent VVC). There was a decrease in the ability to adhere to HBEC, morphogenesis and biofilm formation of the vaginal isolates during the progress of infection. There was an association between the ability to express different virulence factors and the clinical manifestations presented by the patients. Genotype A was the most prevalent (93.6%), followed by genotype C (6.4%). We found maintenance of the same ABC genotype and greater prevalence of microevolution for the vaginal strains of C. albicans sequentially obtained. Vaginal and anal isolates of C. albicans obtained simultaneously from the same patient presented the same ABC genotype and high genetic relatedness. Conclusion: It is noteworthy that the proliferation of yeast and bud-to-hypha transition are important for the establishment of CVV. The expression of virulence factors is important for the pathogenesis of VVC, although it does not seem to be determinant in the transition from colonization to infection or to the installation of recurrent condition. Genotype A seems to be dominant over the others in both vaginal and anal isolates of patients with VVC. The most common scenario was microevolution of the strains of C. albicans in the vaginal environment. It is suggested that the anal reservoir constituted a possible source of vaginal infection, in most cases assessed
