Considering the importance of umbilical cord blood as a potential source of stem cell and, on the other hand, the use of the domestic swine (Sus scrofa) as a useful model for biomedical research in regenerative medicine and aiming to contribute about the quantification of lymphocyte subsets in umbilical cord blood and peripheral blood of newborn piglets, this study aimed to quantify CD4+, CD5+ and CD8+ cells from umbilical cord blood and peripheral blood from pigs at term blood samples. Were analyzed samples of the umbilical cord blood and peripheral of 48 piglets of Topigs lineage, from healthy mothers, artificially inseminated and natural birth. Blood samples were collected from the umbilical cord at birth, by the umbilical vein, and peripheral blood by venous sinus retro-ophthalmic. The immunological measurements of CD4+, CD5+ and CD8+ were obtained by flow cytometry. The relative average values for the CD4+, CD5+ e CD8+ counts in umbilical cord blood and peripheral blood of newborn piglets were inferior to those reported for peripheral blood in adult pigs, suggesting immunological immaturity. The ratio CD4+:CD8+ in umbilical cord blood (3.2±1.2%) and peripheral blood (3.2±1.7%) showed a predominance of TCD4+ over TCD8+. The percentage of CD4+ and CD8+ cells was 1.37±0.86% and 1.15±0.57%, respectively, in umbilical cord blood and peripheral blood.

O objetivo do trabalho é descrever uma estratégia para a obtenção de animais negativos para o PCV2 oriundos de uma granja positiva para este vírus. Dezesseis leitões foram obtidos de fêmeas que tiveram os títulos de IgG anti-PCV2 e o DNA viral testados durante a gestação. Esses leitões, aos sete e dez dias de idade, foram transferidos para a unidade de pesquisa. Durante o período de 7 e 10 aos 49 e 52 dias de idade, amostras de soro, suabes nasal e fecal foram coletadas, a cada sete dias. Após esse período, três animais permaneceram na unidade de pesquisa e foram acompanhados dos 49 aos 114 dias de idade, com coletas realizadas a cada 28 dias. Não houve diferença significativa (p = 0,317) de viremia entre marrãs (n = 6) e porcas (n = 10). Com relação aos níveis de IgG, observou-se diferença significativa (p = 0,0213) entre porcas e marrãs. Os leitões (n = 16), obtidos de duas fêmeas, foram transferidos para a unidade de pesquisa. Os animais entre 7 e 10 dias e aos 49 e 52 dias de idade apresentaram queda de IgG e ausência de IgM anti-PCV2; e as amostras de soro, suabe nasal e fecal foram negativos para o DNA de PCV2. Após os 49 dias, nos três animais mantidos isolados, a detecção de IgG, IgM e DNA para PCV2 permaneceu negativa. Concluindo, a estratégia de manejo utilizada permitiu obter suínos negativos para PCV2 oriundo de granjas positivas para o agente.

Escherichia coli é uma bactéria Gram-negativa, pertencente à família Enterobacteriaceae, que habita o intestino de animais de sangue quente, com 90% sendo comensais para o ser humano e 10% patogênicas. Quando presente em alimentos esse grupo é considerado um bom indicador higiênico-sanitário de alimentos. Em alimentos, a cepa patogênica mais estudada é E. coli produtora de toxina de Shiga (STEC) O157:H7. Entretanto, muitos casos vêm ocorrendo em todo o mundo devido a cepas patogênicas não O157, como O103, O111, O145 e O26. As STECs são responsáveis por desde uma simples diarréia até diarréia sanguinolenta que pode evoluir ainda para síndrome hemolítica urêmica e púrpura trombótica trombocitopênica, que podem ocasionar danos crônicos como falência renal. A transmissão destas cepas se deve a carnes mal cozidas, leites e derivados não pasteurizados, água e vegetais contaminados. Uma característica interessante de E. coli O157:H7 é o seu mecanismo de resistência ao estresse ácido – ácido tolerância – de modo que os alimentos ácidos não podem mais ser considerados seguros contra tais organismos. A fim de ampliar o conhecimento acerca das cepas não O157 e dos mecanismos de ácido resistência, este trabalho teve por objetivo verificar o comportamento de linhagens de STEC não O157:H7 submetidas às condições de estresse de baixo pH utilizando para tanto meios de cultura acidificados e polpas de frutas como matriz alimentar. Constatou-se que quando mantidas em condições-controle e as ácido-adaptadas mantêm populações da ordem de 2 a 3 log UFC/mL quando em polpas de frutas sabor cajá (pH 2,3) e tamarindo (pH 2,3) armazenadas a 4 C por até 30 dias

A study was conducted to evaluate the s of dietary inorganic and organic trace minerals in two levels of supplementation regarding performance, diarrhea occurrence, hematological parameters, fecal mineral excretion and mineral retention in metacarpals and liver of weanling pigs. Seventy piglets weaned at 21 days of age with an average initial body weight of 6.70 ± 0.38 kg were allotted in five treatments: control diet (no added trace mineral premix); 50% ITMP (control diet with inorganic trace mineral premix supplying only 50% of trace mineral requirements); 50% OTMP (control diet with organic trace mineral premix supplying only 50% of trace mineral requirements); 100% ITMP (control diet with inorganic trace mineral premix supplying 100% of trace mineral requirements); and 100% OTMP (control diet with organic trace mineral premix supplying 100% of trace mineral requirements). Feed intake and daily weight gain were not affected by treatments, however, piglets supplemented by trace minerals presented better gain:feed ratio. No differences were observed at calcium, phosphorus, potassium, magnesium, sodium and sulfur excreted in feces per kilogram of feed intake. Treatments did not affect calcium, phosphorus, magnesium, sulfur and iron content in metacarpals. Trace mineral supplementation, regardless of level and source, improved the performance of piglets.

Some authors consider minerals from organic sources more bioavailable for pig nutrition in comparison with inorganic sources. To evaluate the relative iron bioavailability from the organic source iron carbo-amino-phospho-chelate (ICAPC) to weanling piglets, it was conducted an experiment with 126 commercial piglets, using iron sulfate monohydrate (S) as standard. The experiment had a randomized block design with seven treatments (diet without adding specific source of iron, diet with 50, 100 and 150 ppm iron from S and diet with 50, 100 and 150 ppm iron from ICAPC), six replications and three animals per experimental unit. Performance parameters (average daily gain - ADG, feed: gain ratio - F:G) and blood variables (hemoglobin - Hb, hematocrit - Ht, transferrin - TR, latent iron-binding capacity - LIBC, total iron-binding capacity - TIBC, serum iron - Fe and transferrin saturation index - TSI) were evaluated. At the end of the experiment a piglet from each experimental unit was slaughtered and its liver and spleen removed for assessment of iron concentration by flame atomic absorption spectrometry (FAAS). The evaluated sources of iron yielded similar results for the variables of interest, but the increase in iron intake was followed by a linear increase in ADG, Hb, Ht, Fe and TSI as well as a linear decrease in the values of F:G, TR, LIBC and TIBC. Iron bioavailabilities from both ICAPC and S sources are similar for weanling piglets.

The weaning period of piglets is characterized by physiological alterations, such as decreased weight gain, increased reactive oxygen species (ROS) and increased serum cortisol levels with possible effects on the immune response. The effect of parenteral administration of vitamins A, D and E on production performance, oxidative metabolism, and the function of polymorphonuclear leukocytes (PMNLs) was assessed in piglets during the weaning period. The sample was comprised of 20 male piglets that were given an injectable ADE vitamin combination (135,000 IU vitamin A, 40,000 IU vitamin D and 40mg vitamin E/animal) at 20 and 40 days of age. Weight gain, concentration of reduced glutathione (GSH), malondialdehyde (MDA), superoxide dismutase (SOD) and the microbicidal and phagocytic activity of PMNLs were assessed. No difference was observed in the average piglet weight during the study; however, a greater percentage of weight gain was observed after weaning in the treated group. The concentrations of GSH and SOD did not differ between groups, although lipid peroxidation was greater in the control group at 60 days of age. The investigated variables of oxidative metabolism were correlated as follows: -0.41 for GSH and MDA, -0.54 for GSH and SOD and 0.34 for MDA and SOD. The intensity of intracellular ROS production, the percentage of ROS-producing PMNLs and the intensity of phagocytosis by PMNLs did not differ between treatment groups. Administration of the injectable ADE combination improved the percentage of weight gain between 20 and 40 days of age, decreased oxidative stress at 60 days of age and did not influence the function of PMNLs in piglets.
