This thesis examines the transition of employees into entrepreneurship, with particular emphasis on the role of workplace characteristics in influencing this movement. The first main chapter examines whether the determinants of becoming an intrapreneur differ from those that support transitions into independent entrepreneurship. The results show that intrapreneurs resemble employees rather than entrepreneurs, contrary to what the entrepreneurship theory would suggest. Yet it shows that those intrapreneurs that expect to acquire an ownership stake in the business, unlike the rest of intrapreneurs, possess traditional entrepreneurial traits. Chapter 3 investigates how workers’ degree of specialisation determines their decision to found a firm. It shows that entrepreneurs emerging from small firms, i.e. generalists, transfer knowledge from more diverse aspects of the business and create firms more related to the main activity of their last employer. Workers in large firms, however, benefit from higher returns to human capital that increase their opportunity costs to switch to entrepreneurship. Since becoming an entrepreneur would make part of their specialised skills unutilised, the minimum quality of the idea at which they would be willing to leave will be higher and, therefore, entrepreneurs emerging from large firms will be of highest quality. Chapter 4 analyses whether the reason to terminate an employment contract is associated with the fact that the majority of entrepreneurs appear to set up their business after having worked for a small firm. Moreover, it studies how this pattern varies as the labour market conditions worsen. The effect of layoffs turns out to be a key driver in the entry to entrepreneurship and it is found to exert a greater effect the smaller the firm workers are dismissed from. This has been reflected in an overall larger flow of employees from small firms moving into entrepreneurship over the recession.

Community structure of sediment bacteria in the Everglades freshwater marsh, fringing mangrove forest, and Florida Bay seagrass meadows were described based on polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) patterns of 16S rRNA gene fragments and by sequencing analysis of DGGE bands. The DGGE patterns were correlated with the environmental variables by means of canonical correspondence analysis. There was no significant trend in the Shannon–Weiner index among the sediment samples along the salinity gradient. However, cluster analysis based on DGGE patterns revealed that the bacterial community structure differed according to sites. Not only were these salinity/vegetation regions distinct but the sediment bacteria communities were consistently different along the gradient from freshwater marsh, mangrove forest, eastern-central Florida Bay, and western Florida Bay. Actinobacteria- and Bacteroidetes/Chlorobi-like DNA sequences were amplified throughout all sampling sites. More Chloroflexi and members of candidate division WS3 were found in freshwater marsh and mangrove forest sites than in seagrass sites. The appearance of candidate division OP8-like DNA sequences in mangrove sites distinguished these communities from those of freshwater marsh. The seagrass sites were characterized by reduced presence of bands belonging to Chloroflexi with increased presence of those bands related to Cyanobacteria, γ-Proteobacteria, Spirochetes, and Planctomycetes. This included the sulfate-reducing bacteria, which are prevalent in marine environments. Clearly, bacterial communities in the sediment were different along the gradient, which can be explained mainly by the differences in salinity and total phosphorus.

1. Apex predators are often assumed to be dietary generalists and, by feeding on prey from multiple basal nutrient sources, serve to couple discrete food webs. But there is increasing evidence that individual level dietary specialization may be common in many species, and this has not been investigated for many marine apex predators. 2. Because of their position at or near the top of many marine food webs, and the possibility that they can affect populations of their prey and induce trophic cascades, it is important to understand patterns of dietary specialization in shark populations. 3. Stable isotope values from body tissues with different turnover rates were used to quantify patterns of individual specialization in two species of ‘generalist’ sharks (bull sharks, Carcharhinus leucas, and tiger sharks, Galeocerdo cuvier). 4. Despite wide population-level isotopic niche breadths in both species, isotopic values of individual tiger sharks varied across tissues with different turnover rates. The population niche breadth was explained mostly by variation within individuals suggesting tiger sharks are true generalists. In contrast, isotope values of individual bull sharks were stable through time and their wide population level niche breadth was explained by variation among specialist individuals. 5. Relative resource abundance and spatial variation in food-predation risk tradeoffs may explain the differences in patterns of specialization between shark species. 6. The differences in individual dietary specialization between tiger sharks and bull sharks results in different functional roles in coupling or compartmentalizing distinct food webs. 7. Individual specialization may be an important feature of trophic dynamics of highly mobile marine top predators and should be explicitly considered in studies of marine food webs and the ecological role of top predators.

Microorganisms inhabit very different soil habitats in the ice-free areas of Antarctica, playing a major role in nutrient cycling in cold environments. We studied the soil characteristics and the dominant bacterial composition from nine different soil profiles located on Livingston Island (maritime Antarctica). The total carbon (TC) and total nitrogen (TN) values were high for the vegetated soils, decreasing with depth, whereas the values for the mineral soils were generally low. Soil pH was more acidic for moss-covered soils and neutral to alkaline for mineral soils. Numbers of culturable heterotrophic bacteria were higher at vegetated sites, but significant numbers were also detectable in carbon-depleted soils. Patterns of denaturing gradient gel electrophoresis (DGGE) revealed a highly heterogeneous picture throughout the soil profiles. Subsequent sequencing of DGGE bands revealed in total 252 sequences that could be assigned to 114 operational taxonomic units, showing the dominance of members of the Bacteroidetes and Acidobacteria. The results of phospholipid fatty acid analysis showed a lack of unsaturated fatty acids for most of the samples. Samples with a prevalence of unsaturated over saturated fatty acids were restricted to several surface samples. Statistical analysis showed that the dominant soil bacterial community composition is most affected by TC and TN contents and soil physical factors such as grain size and moisture, but not pH. Antarctic glacier forefields are extreme environments and pioneer sites for ecological succession. Increasing temperatures due to global warming lead to enhanced deglaciation processes in cold-affected habitats, and new terrain is becoming exposed to soil formation and microbial colonization. However, only little is known about the impact of environmental changes on microbial communities and how they develop in connection to shifting habitat characteristics. In this study, using a combination of molecular and geochemical analysis, we determine the structure and development of bacterial communities depending on soil parameters in two different glacier forefields on Larsemann Hills, East Antarctica. Our results demonstrate that deglaciation-dependent habitat formation, resulting in a gradient in soil moisture, pH and conductivity, leads to an orderly bacterial succession for some groups, for example Cyanobacteria, Bacteroidetes and Deltaproteobacteria in a transect representing 'classical' glacier forefields. A variable bacterial distribution and different composed communities were revealed according to soil heterogeneity in a slightly 'matured' glacier forefield transect, where Gemmatimonadetes, Flavobacteria, Gamma- and Deltaproteobacteria occur depending on water availability and soil depth. Actinobacteria are dominant in both sites with dominance connected to certain trace elements in the glacier forefields.

The anaerobic process was efficient in organic matter removal. During the process, an interesting compound as quercetin was produced inside of reactor. Phylogenetic analysis showed the presence of phylotypes affiliated with gamma-Proteobacteria, Choroflexi, and Bacteroidetes. Archaea were represented by phylotypes belonging to the genus Methanosarcina and Methanosaeta.

Sea urchins are benthic macroinvertebrates that inhabit shallow coastal waters in tropical and temperate zones. Urchins are usually classified as generalists or omnivores as they can adjust their diet according to the food resources available in the environment. Due to the strong grazing pressure they may exert, urchins have an important role in marine ecosystems, occupying different trophic levels and stimulating the intensification of the dynamics of communities where they occur. In 2004, a monitoring program focused on the population dynamics of the white sea urchin, Tripneustes ventricosus, has been initiated in the Fernando de Noronha Archipelago. At the same time, a surprisingly lack of information on the species biology has been noted, despite their wide geographical distribution and economic importance in many parts of its range. Hence, this work was developed to provide information on the feeding habits of T. ventricosus in the archipelago. Ten specimens were collected between December 2006 and July 2007 at two sites of the archipelago, Air France and Sueste Bay for biometrics and analysis of gut contents. Test diameters ranged from 9.19 cm (± 1.1) to 10.08 cm (± 0.58). Calculated stomach repletion index (IRE) was higher (p <0.05) in the Air France site and also during January and July. The IRE was not correlated to the gonad index. Fifteen different species of algae were detected in a total of 120 stomachs examined: 4 Chlorophytas, 4 Phaeophytas and 6 Rhodophytas. Food diversity (p <0.05) was higher in December 2006 and January 2007. Although several items had a high frequency of occurrence, they were low represented in terms of weight, and consequently, had a low level of relative importance. The brown algae Dictyopteris spp and Dictyota spp, followed by the green algae Caulerpa verticillata accounted for the greatest importance in T. ventricosus diet, comprising about 90% of the consumed items

Sir Wilfred Grenfell Colleges Fine Arts Division offers two BFA programs: visual arts and theatre. They are the only BFA degree programs available in the province. The bachoelor of Fine arts(Visual Arts)program is a professional program designed to educate and train students in the history, theory and practice of the visual arts. The curriculum has been devised to produce well-rounded generalists with solid grounding in all aspects of visual arts. The Curriculum illustrates the philosophy that artistic freedom and creative expression require technical skill, intellectual awareness and personal vision, acquired through a disciplined application of effort and critical understanding of artistic issues, past and present. Academic electives are tended to provide a broad exposure to the liberal arts.

The last decades of the 20th century defined the genetic engineering advent, climaxing in the development of techniques, such as PCR and Sanger sequencing. This, permitted the appearance of new techniques to sequencing whole genomes, identified as next-generation sequencing. One of the many applications of these techniques is the in silico search for new secondary metabolites, synthesized by microorganisms exhibiting antimicrobial properties. The peptide antibiotics compounds can be classified in two classes, according to their biosynthesis, in ribosomal or nonribosomal peptides. Lanthipeptides are the most studied ribosomal peptides and are characterized by the presence of lanthionine and methylanthionine that result from posttranslational modifications. Lanthipeptides are divided in four classes, depending on their biosynthetic machinery. In class I, a LanB enzyme dehydrate serine and threonine residues in the C-terminus precursor peptide. Then, these residues undergo a cyclization step performed by a LanC enzyme, forming the lanthionine rings. The cleavage and the transport of the peptide is achieved by the LanP and LanT enzymes, respectively. Although, in class II only one enzyme, LanM, is responsible for the dehydration and cyclization steps and also only one enzyme performs the cleavage and transport, LanT. Pedobacter sp. NL19 is a Gram-negative bacterium, isolated from sludge of an abandon uranium mine, in Viseu (Portugal). Antibacterial activity in vitro was detected against several Gram-positive and Gram-negative bacteria. Sequencing and in silico analysis of NL19 genome revealed the presence of 21 biosynthetic clusters for secondary metabolites, including nonribosomal and ribosomal peptides biosynthetic clusters. Four lanthipeptides clusters were predicted, comprising the precursor peptides, the modifying enzymes (LanB and LanC), and also a bifunctional LanT. This result revealed the hybrid nature of the clusters, comprising characteristics from two distinct classes, which are poorly described in literature. The phylogenetic analysis of their enzymes showed that they clustered within the bacteroidetes clade. Furthermore, hybrid gene clusters were also found in other species of this phylum, revealing that it is a common characteristic in this group. Finally, the analysis of NL19 colonies by MALDI-TOF MS allowed the identification of a 3180 Da mass that corresponds to the predicted mass of a lanthipeptide encoded in one of the clusters. However, this result is not fully conclusive and further experiments are needed to understand the full potential of the compounds encoded in this type of clusters. In conclusion, it was determined that NL19 strain has the potential to produce diverse secondary metabolites, including lanthipeptides that were not functionally characterized so far.

A fauna de mamíferos do estado de São Paulo consta de 231 táxons, sendo este número uma estimativa da real diversidade presente na região, dado a falta de amostragem em grandes extensões do estado, e também de revisões taxonômicas para determinados grupos. Ainda assim, nosso conhecimento aumentou em 20% desde a última estimativa em 1998, principalmente em relação aos quirópteros e roedores. Estes dados são provenientes de inventários faunísticos, e também do estudo de espécimes depositados em coleções científicas oriundos de revisões taxonômicas. Também temos um maior volume de dados a respeito da distribuição dos mamíferos em relação às diferentes paisagens presentes no estado, o que nos permite dividir a mastofauna em três componentes distintos: o mais importante desses é o das espécies generalistas, que ocorrem em todas as principais paisagens do estado; o segundo grupo concentra espécies das formações abertas, e o terceiro grupo inclui as espécies essencialmente florestais. Além disso, o número de estudos que tem se preocupado com o efeito da fragmentação de hábitats sobre as comunidades de mamíferos, bem como a respeito da permeabilidade das espécies em áreas alteradas, também aumentaram. Dados a respeito da ocorrência, abundância e vulnerabilidade das espécies foram essenciais para traçar estratégias em relação à escolha de áreas e à indicação de ações prioritárias para a conservação dos mamíferos no estado, assim como classificar as espécies nas diferentes categorias de ameaças propostas, culminando na Lista das Espécies Ameaçadas do Estado de São Paulo. Entretanto, ainda existem inúmeras lacunas de conhecimento, que vão desde o número limitado de amostras zoológicas, até a falta de informações acerca da ecologia e história natural de várias espécies. É imprescindível que aumentemos as amostras de mamíferos em coleções zoológicas, principalmente em áreas de Floresta Ombrófila Densa, nos fragmentos de Cerrado, bem como em áreas do centro e oeste do Estado, que permanecem ainda pouco estudadas, com o objetivo de produzir um maior número de revisões taxonômicas em diversos grupos de mamíferos, e de estudos com abordagens filogeográficas e de genética de populações, para diagnosticarmos de forma efetiva a riqueza de mamíferos no estado, bem como os mecanismos evolutivos responsáveis por esta diversificação. Aliados a esses estudos serão necessárias abordagens ecológicas para gerarmos conhecimento, que em conjunto, nos permitirá avaliarmos o estado de conservação dos mamíferos de São Paulo e tomarmos decisões sobre as melhores estratégias para manejarmos e preservarmos estas espécies.

Os microrganismos do solo são componentes essenciais na manutenção do equilíbrio físico-químico e biológico do mesmo e exercem importante função que inclui a degradação de resíduos de plantas e animais e a liberação de nutrientes na cadeia alimentar. Este trabalho teve como objetivo comparar a microbiota de um solo com cobertura de mata (SMS) e outro cultivado com hortaliças (SHC), supressivos ou não a Rhizoctonia solani. Foram feitas extrações do DNA total dos solos e a partir dos mesmos, amplificação por PCR dos genes 16S rDNA, clonagem dos fragmentos e seqüenciamento dos genes do RNA ribossomal. A análise dos resultados demonstrou que essa metodologia foi eficiente para avaliação de bactérias. No solo supressivo de mata os filos mais encontrados pertencem aos das Acidobactérias, Verrucomicrobia e Actinobactérias e no solo conducente cultivado com hortaliças a maioria pertence aos filos das Proteobactérias, Firmicutes e Bacteroidetes.

Globally, peatlands occupy a small portion of terrestrial land area but contain up to one-third of all soil organic carbon. This carbon pool is vulnerable to increased decomposition under projected climate change scenarios but little is known about how plant functional groups will influence microbial communities responsible for regulating carbon cycling processes. Here we examined initial shifts in microbial community structure within two sampling depths under plant functional group manipulations in mesocosms of an oligotrophic bog. Microbial community composition for bacteria and archaea was characterized using targeted 16S rRNA Illumina gene sequencing. We found statistically distinct spatial patterns between the more shallow 10-20 cm sampling depth and the deeper 30-40 cm depth. Significant effects by plant functional groups were found only within the 10-20 cm depth, indicating plant-mediated microbial community shifts respond more quickly near the peat surface. Specifically, the relative abundance of Acidobacteria decreased under ericaceous shrub treatments in the 10-20 cm depth and was replaced by increased abundance of Gammaproteobacteria and Bacteroidetes. In contrast, the sedge rhizosphere continued to be dominated by Acidobacteria but also promoted an increase in the relative recovery of Alphaproteobacteria and Verrucomicrobia. These initial results suggest microbial communities under ericaceous shrubs may be limited by anaerobic soil conditions accompanying high water table conditions, while sedge aerenchyma may be promoting aerobic taxa in the upper peat rhizosphere regardless of ambient soil oxygen limitations.

Objectifs: L’objectif principal de ce mémoire consiste à comprendre les caractéristiques des carrières criminelles d’individus connus de la police pour avoir perpétré une infraction de leurre d’enfants sur Internet. Aussi, par une analyse typologique à l’aide des antécédents criminels, il sera possible d’établir une typologie d’individus ayant leurré des enfants sur Internet. Également, il sera question de vérifier s’il y a un lien entre les caractéristiques des antécédents criminels de ces individus sur la perpétration de l’agression sexuelle hors ligne. Méthodologie: Provenant de données officielles de la communauté policière du Québec, l’échantillon comprend les parcours de criminels ayant perpétré une infraction de leurre d’enfants sur Internet. Des analyses descriptives en lien avec les différents paramètres de la carrière criminelle seront effectuées. Ensuite, des tests de moyenne et une analyse de régression Cox permettront de vérifier la présence ou non d’un lien statistique entre les caractéristiques des antécédents criminels des individus connus de la police pour leurre d’enfants sur Internet et le passage à l’acte physique. Résultats: Les analyses ont montré que la majorité des sujets n’avaient aucun antécédent judiciaire. Pour la plupart, le leurre d’enfants est le crime le plus grave perpétré au cours de leur carrière criminelle. Trois catégories d’individus ont été décelées : les amateurs, les spécialistes et les généralistes. Ce sont les individus polymorphes ayant une carrière criminelle plus grave et plus longue qui sont portés à agresser sexuellement avant le leurre. Cependant, ce sont des individus spécialisés ayant une importante proportion de délits sexuels dans leurs antécédents criminels qui ont plus de chance d’agresser sexuellement suite à l’exploitation sexuelle sur Internet.
