Schizorchis caballeroi n. sp. has been described from the collared pika, Ochotona collaris (Nelson), from Alaska, and has been distinguished morphologically from its congeners, S. ochotonae Hansen, 1948, and S. altaica Gvozdev, 1951. Nine species of helminths have been described 10 dale from North American pikas, O. collaris and O. princeps (Richardson). These mammals are markedly allopatric and do not share any species of helminth. Nematodes of two genera, Eugenuris Shults and Labiostomum Akhtar, 1941, occur in O. collaris as weIl as in palearctic species of Ochotona. This, along with other pertinent information, is taken to suggest that O. collaris has invaded North American more recently than did O. princeps or its precursor.

In spite of the importance of the various species of shrews to the biotic community, several aspects of their ecology remain very incompletely known. The helminths parasitic in the more conspicuous small mammals in North America have been studied more or less completely, but those occurring in shrews have been largely disregarded. This situation is particularly inappropriate when one considers that shrews often constitute the most abundant mammalian group in a given area. We have attempted to secure an adequate amount of shrew helminth material for study, and it is the purpose of this paper to present observations resulting from this work.

Schistosoma mansoni is responsible for schistosomiasis, a parasitic disease that affects 200 million people worldwide. Molecular mechanisms of host-parasite interaction are complex and involve a crosstalk between host signals and parasite receptors. TGF-beta signaling pathway has been shown to play an important role in S. mansoni development and embryogenesis. In particular human (h) TGF-beta has been shown to bind to a S. mansoni receptor, transduce a signal that regulates the expression of a schistosome target gene. Here we describe 381 parasite genes whose expression levels are affected by in vitro treatment with hTGF-beta. Among these differentially expressed genes we highlight genes related to morphology, development and cell cycle that could be players of cytokine effects on the parasite. We confirm by qPCR the expression changes detected with microarrays for 5 out of 7 selected genes. We also highlight a set of non-coding RNAs transcribed from the same loci of protein-coding genes that are differentially expressed upon hTCF-beta treatment. These datasets offer potential targets to be explored in order to understand the molecular mechanisms behind the possible role of hTGF-beta effects on parasite biology. (C) 2012 Elsevier B.V. The purpose of this work is to analyze the parasitological risks of treated wastewater reuse from a stabilization pond in the city of Piracicaba, in the State of Sao Paulo (Brazil), and the level of treatment required to protect public health. Samples were taken from raw and treated wastewater in stabilization ponds and submitted to a parasitological, microbiological and physicochemical analysis. The study revealed on treated wastewater the presence of Ascaris sp. and Entamoeba coli with an average density of 1 cysts L-1 and 6 eggs L-1, respectively. For Ascaris, the annual risks of infection due to the accidental ingestion of wastewater irrigation were 7.5 x 10(-2) in 208 days and 8.7 x 10(-2) in 240 days. For Total Coliforms and Escherichia coli in treated wastewater, the average density was 1.0 x 10(5) MPN/100 ml and 2.7 x 10(4) MPN/100 ml respectively, representing 99% and 94% removal efficiency, respectively. For BOD, COD, TS and TSS removal efficiency was 69, 80, 50 and 71%, respectively. The removal efficiency for nitrogen; ammonia nitrogen and total phosphate was 24, 19 and 68%, respectively. The average density of helminths eggs in treated wastewater is higher compared to the density of the limit value of <= 1 egg L-1 and tolerable risk is above the level recommended by the World Health Organization. Multiple barriers are necessary for the reduction of organic matter, chemical contaminants and parasites from treated wastewater. Standards for the sanitary control of treated wastewater to be reused in agricultural irrigation areas should be compiled for developing countries in order to minimize public health risks.

Seventy-seven males of Hypsiboas prasinus from 2 Atlantic forest fragments in the municipalities of Botucatu and Jundiai, sao Paulo State, Brazil, were examined for endoparasites. The frogs were captured in summer (January until March) and winter (July/August) of 2008 and 2009. Thirty-three males (75%) from Botucatu were infected with Rhabdias cf. fuelleborni, cosmocerciid nematodes, and Cylindrotaenia americana. Twenty-five tree frogs (78.5%) from Jundiai were infected by Rhabdias cf. fuelleborni, Physaloptera sp., and cosmocerciid nematodes. Only cosmocerciid nematodes presented a statistically significance difference in prevalence (z = 4.345; P < 0.001) and mean abundance (t = 562.0; P < 0.001) between Botucatu and Jundiai during the winter. Also, the cosmocerciids exhibited higher mean abundance (t = 196.0; P = 0.034) in winter when compared with summer at the Jundiai site. Moreover, to our knowledge, this is the first report of C. americana in the Brazilian Hylidae. This study presents 4 new records of nematodes in H. prasinus.

Tropiduridae (Squamata: Iguania) is a lizard taxon widely distributed in the neotropics. Among its representatives, some species are classified as generalists regarding habitat usage. Others exhibit a very restricted and probably relict distribution, and are strongly associated with predominantly sandy and dry habitats. Within this rather ecologically similar than phylogenetically closely related group we examined specimens of Eurolophosaurus amathites, E. divaricatus, Tropidurus hygomi, T. psammonastes for endoparasites. In all four species examined we recorded parasitic nematodes (Nemathelminthes: Nematoda). At least three nematode species were recovered: Parapharyngodon sp., Physaloptera lutzi and Strongyluris oscari, with Ph. lutzi being the most abundant parasite encountered in all lizard species examined. In spite of the hosts' habitat specialization, these parasites are also found frequently in non-psammophilous tropidurid species as well as in other squamates. Individual species richness per lizard was low, with usually just one species parasitizing at a time. These are the first parasites registered for these tropidurids and constitute a total of six new host records.

Le attività di ricerca della presente tesi di dottorato si sono focalizzata principalmente sulla parassitofauna dei pesci marini allevati in Grecia ed in Italia con particolare attenzione allo studio degli ectoparassiti di maggior rilievo sanitario in maricoltura ed alla ricerca di endoparassiti di potenziale interesse zoonosico, in particolare larve di nematodi Anisakidae del genere Anisakis. Nel corso del triennio sono stati sottoposti ad esami parassitologici 916 spigole (Dicentrarchus labrax) e 462 orate (Sparus aurata) prelevate presso diverse tipologie di allevamenti greci ed italiani. Per quanto concerne le spigole, la presenza di ectoparassiti è stata riscontrata nel 29,2% e nel 61,9% dei soggetti provenienti rispettivamente da impianti siti in Grecia ed in Italia, mentre le orate hanno presentato percentuali di positività rispettivamente del 87,5% e del 26,7%. Gli ectoparassiti dominanti sono risultati essere il monogeneo Diplectanum aequans nelle spigole ed il ciliato Cryptocaryon irritans e il monogeneo Furnestinia echeneis nelle orate, sebbene sia stato possibile studiare anche il coinvolgimento di altri ectoparassiti, quali il monogeneo Sparicotyle chrysophrii ed il dinoflagellato Amyloodinium ocellatum, nel determinismo di alcuni episodi morbosi. Le osservazioni istopatologiche hanno permesso di caratterizzare le lesioni causate dagli ectoparassiti a diverse intensità d’infestazione. Per quanto concerne la ricerca di parassiti zoonosici, con particolare riferimento agli stadi larvali di nematodi Anisakidae del genere Anisakis, si sono condotti esami parassitologici a livello di cavità viscerale e di muscolo laterale in tutti i soggetti provenienti da allevamenti in gabbia (626 soggetti, di cui 441 spigole e 185 orate). Tutti i soggetti esaminati sono risultati negativi, indicando come il rischio di infestazione da larve di nematodi anisakidi possa essere considerato trascurabile in spigole ed orate allevate in gabbia, come già dimostrato per il salmone atlantico (EFSA, 2010).

T helper (Th) 9 cells are an important subpopulation of the CD4+ T helper cells. Due to their ability to secrete Interleukin-(IL-)9, Th9 cells essentially contribute to the expulsion of parasitic helminths from the intestinal tract but they play also an immunopathological role in the course of asthma. Recently, a beneficial function of Th9 cells in anti-tumor immune responses was published. In a murine melanoma tumor model Th9 cells were shown to enhance the anti-melanoma immune response via the recruitment of CD8+ T cells, dendritic cells and mast cells. In contrast to Th9 effector cells regulatory T cells (Tregs) are able to control an immune response with the aid of different suppressive mechanisms. Based on their ability to suppress an immune response Tregs are believed to be beneficial in asthma by diminishing excessive allergic reactions. However, concerning cancer they can have a detrimental function because Tregs inhibit an effective anti-tumor immune reaction. Thus, the analysis of Th9 suppression by Tregs is of central importance concerning the development of therapeutic strategies for the treatment of cancer and allergic diseases and was therefore the main objective of this PhD thesis.rnIn general it could be demonstrated that the development of Th9 cells can be inhibited by Tregs in vitro. The production of the lineage-specific cytokine IL-9 by developing Th9 cells was completely suppressed at a Treg/Th9 ratio of 1:1 on the transcriptional (qRT-PCR) as well as on the translational level (ELISA). In contrast, the expression of IRF4 that was found to strongly promote Th9 development was not reduced in the presence of Tregs, suggesting that IRF4 requires additional transcription factors to induce the differentiation of Th9 cells. In order to identify such factors, which regulate Th9 development and therefore represent potential targets for Treg-mediated suppressive mechanisms, a transcriptome analysis using “next-generation sequencing” was performed. The expression of some genes which were found to be up- or downregulated in Th9 cells in the presence of Tregs was validated with qRT-PCR. Time limitations prevented a detailed functional analysis of these candidate genes. Nevertheless, the analysis of the suppressive mechanisms revealed that Tregs probably suppress Th9 cells via the increase of the intracellular cAMP concentration. In contrast, IL-9 production by differentiated Th9 cells was only marginally affected by Tregs in vitro and in vivo analysis (asthma, melanoma model). Hence, Tregs represent very effective inhibitors of Th9 development whereas they have only a minimal suppressive influence on differentiated Th9 cells.rn

Gewebe, Zellen und speziell Zellkompartimente unterscheiden sich in ihrer Sauerstoffkonzentration, Stoffwechselrate und in der Konzentration an gebildeten reaktiven Sauerstoffspezies. Um eine mögliche Änderung in der Aminosäurennutzung durch den Einfluss von Sauerstoff und seinen reaktiven Spezies untersuchen zu können wurden, Bereiche bzw. Kompartimente der menschlichen Zelle definiert, die einen Referenzrahmen bildeten und bekannt dafür sind, einen relativ hohen Grad an reaktiven Sauerstoffspezies aufzuweisen. Aus dem Vergleich wurde deutlich, dass vor allem die beiden redox-aktiven und schwefeltragenden Aminosäuren Cystein und Methionin durch eine besondere Verteilung und Nutzung charakterisiert sind. Cystein ist hierbei diejenige Aminosäure mit den deutlichsten Änderungen in den fünf untersuchten Modellen der oxidativen Belastung. In all diesen Modellen war die Nutzung von Cystein deutlich reduziert, wohingegen Methionin in Proteinen des Mitochondriums und der Elektronentransportkette angereichert war. Dieser auf den ersten Blick paradoxe Unterschied zwischen Cystein und Methionin wurde näher untersucht, indem die differenzierte Methioninnutzung in verschiedenen Zellkompartimenten von Homo sapiens charakterisiert wurde.rnDie sehr leicht zu oxidierende Aminosäure Methionin zeigt ein ungewöhnliches Verteilungsmuster in ihrer Nutzungshäufigkeit. Entgegen mancher Erwartung wird Methionin in zellulären Bereichen hoher oxidativer Belastung und starker Radikalproduktion intensiv verwendet. Dieses Verteilungsmuster findet man sowohl im intrazellulären Vergleich, als auch im Vergleich verschiedener Spezies untereinander, was daraufhin deutet, dass es einen lokalen Bedarf an redox-aktiven Aminosäuren gibt, der einen sehr starken Effekt auf die Nutzungshäufigkeit von Methionin ausübt. Eine hohe Stoffwechselrate, die im Allgemeinen mit einer erhöhten Produktion von Oxidantien assoziiert wird, scheint ein maßgeblicher Faktor der Akkumulation von Methionin in Proteinen der Atmungskette zu sein. Die Notwendigkeit, oxidiertes Antioxidans wieder zu reduzieren, findet auch bei Methionin Anwendung, denn zu Methioninsulfoxid oxidiertes Methionin wird durch die Methioninsulfoxidreduktase wieder zu Methionin reduziert. Daher kann die spezifische Akkumulation von Methionin in Proteinen, die verstärkt reaktiven Sauerstoffspezies ausgesetzt sind, als eine systematische Strategie angesehen werden, um andere labile Strukturen vor ungewollter Oxidation zu schützen. rnDa Cystein in allen untersuchten Modellen der oxidativen Belastung und im Besonderen in Membranproteinen der inneren Mitochondrienmembran lebensspannenabhängig depletiert war, wurde dieses Merkmal näher untersucht. Deshalb wurde die Hypothese getestet, ob ein besonderer Redox-Mechanismus der Thiolfunktion für diese selektive Depletion einer im Allgemeinen als harmlos oder antioxidativ geltenden Aminosäure verantwortlich ist. Um den Effekt von Cysteinresten in Membranen nachzustellen, wurden primäre humane Lungenfibroblasten (IMR90) mit diversen Modellsubstanzen behandelt. Geringe Konzentrationen der lipophilen Substanz Dodecanthiol verursachten eine signifikante Toxizität in IMR90-Zellen, die von einer schnellen Zunahme an polyubiquitinierten Proteinen und anderen Indikatoren des proteotoxischen Stresses, wie Sequestosom 1 (P62), HSP70 und HSP90 begleitet wurde. Dieser Effekt konnte spezifisch der Chemie der Thiolfunktion in Membranen zugeordnet werden, da Dodecanol (DOH), Dodecylmethylsulfid (DMS), Butanthiol oder wasserlösliche Thiole weder eine cytotoxische Wirkung noch eine Polyubiquitinierung von Proteinen verursachten. Die Ergebnisse stimmen mit der Hypothese überein, dass Thiole innerhalb von biologischen Membranen als radikalische Kettentransferagentien wirken. Diese Eigenschaft wird in der Polymerchemie durch Nutzung von lipophilen Thiolen in hydrophoben Milieus technisch für die Produktion von Polymeren benutzt. Da die Thiylradikal-spezifische Reaktion von cis-Fettsäuren zu trans-Fettsäuren in 12SH behandelten Zellen verstärkt ablief, kann gefolgert werden, dass 12SH zellulär radikalisiert wurde. In lebenden Organismen kann demnach die Oxidation von Cystein die Schädigung von Membranen beschleunigen und damit Einfallstore für die laterale Radikalisierung von integralen Membranproteinen schaffen, welche möglicherweise der Langlebigkeit abträglich ist, zumindest, wenn sie in der inneren Mitochondrienmembran auftritt.

Insect bite hypersensitivity (IBH) is an IgE-mediated dermatitis of horses caused by bites of Culicoides spp. IBH does not occur in Iceland where Culicoides are absent. However, following importation into continental Europe where Culicoides are present, >or=50% of Icelandic horses (1st generation) develop IBH but helminths and allergens induce Th2 responses, we investigated whether horses domiciled in Iceland are Th2-biased, and whether this is determined by helminth infection. We compared the parasite burden and T cell responses between Icelandic horses living in Iceland or Switzerland. Horses in Iceland have higher faecal egg counts, higher tapeworm-specific IgG(T) levels and higher total serum IgE levels than horses in Switzerland. Nevertheless, horses in Iceland displayed a low proportion of IL-4-producing cells in PBMC cultures after polyclonal or parasite extracts stimulation. No IL-4-producing cells were found in PBMC from horses after stimulation by Culicoides extract. Addition of anti-IL-10 and anti-TGF-beta1 to PBMC cultures of horses in Iceland increased the proportion of IL-4-producing cells after polyclonal or parasite antigens stimulation but not stimulation with Culicoides extract. This paralleled the high levels of IL-10 and TGF-beta1 found in supernatants from PBMC cultures of horses in Iceland. Collectively, horses living in Iceland have a high parasite burden but low IL-4 production. This supports the hypothesis that heavy helminth infections have a suppressive effect on IL-4 production mediated by IL-10 and TGF-beta1.

Parasites are linked with their host in a trophic interaction with implications for both hosts and parasites. Interaction stretches from the host's immune response to the structuring of communities and the evolution of biodiversity. As in many species sex determines life history strategy, response to parasites may be sex-specific. Males of vertebrate species tend to exhibit higher rates of parasites than females. Sex-associated hormones may influence immunocompetence and are hypothesised to lead to this bias. In a field study, we tested the prediction of male biased parasitism (MBP) in free ranging chamois (Rupicapra rupicapra rupicapra), which are infested intensely by gastrointestinal and lung helminths. We further investigated sex differences in faecal androgen (testosterone and epiandrosterone), cortisol and oestrogen metabolites using enzyme immunoassays (EIA) to evaluate the impact of these hormones on sex dependent parasite susceptibility. Non-invasive methods were used and the study was conducted throughout a year to detect seasonal patterns. Hormone levels and parasite counts varied significantly throughout the year. Male chamois had a higher output of gastrointestinal eggs and lungworm larvae when compared to females. The hypothesis of MBP originating in sex related hormone levels was confirmed for the elevated output of lungworm larvae, but not for the gastrointestinal nematodes. The faecal output of lungworm larvae was significantly correlated with androgen and cortisol metabolite levels. Our study shows that sex differences in steroid levels play an important role to explain MBP, although they alone cannot fully explain the phenomenon.

Nitazoxanide (2-acetolyloxy-N-(5-nitro 2-thiazolyl) benzamide; NTZ) represents the parent compound of a novel class of broad-spectrum anti-parasitic compounds named thiazolides. NTZ is active against a wide variety of intestinal and tissue-dwelling helminths, protozoa, enteric bacteria and a number of viruses infecting animals and humans. While potent, this poses a problem in practice, since this obvious non-selectivity can lead to undesired side effects in both humans and animals. In this study, we used real time PCR to determine the in vitro activities of 29 different thiazolides (NTZ-derivatives), which carry distinct modifications on both the thiazole- and the benzene moieties, against the tachyzoite stage of the intracellular protozoan Neospora caninum. The goal was to identify a highly active compound lacking the undesirable nitro group, which would have a more specific applicability, such as in food animals. By applying self-organizing molecular field analysis (SOMFA), these data were used to develop a predictive model for future drug design. SOMFA performs self-alignment of the molecules, and takes into account the steric and electrostatic properties, in order to determine 3D-quantitative structure activity relationship models. The best model was obtained by overlay of the thiazole moieties. Plotting of predicted versus experimentally determined activity produced an r2 value of 0.8052 and cross-validation using the "leave one out" methodology resulted in a q2 value of 0.7987. A master grid map showed that large steric groups at the R2 position, the nitrogen of the amide bond and position Y could greatly reduce activity, and the presence of large steric groups placed at positions X, R4 and surrounding the oxygen atom of the amide bond, may increase the activity of thiazolides against Neospora caninum tachyzoites. The model obtained here will be an important predictive tool for future development of this important class of drugs.
