Plate counts at R T and 8 C on the skin with muscle and the gut contents of absolutely fresh sardines (Sardinella longiceps) caught off Cochin showed a seasonal variation when sampling was done over a period of 12 months. The counts of the gut contents ran parallel with those of the skin with muscle, but at a higher level of magnitude. Qualitatively, the analysis of 360 strains of bacteria isolated from the skin with muscle and 100 strains from the guts during a year's study revealed a very high preponderance of Gram negative rods, mainly of Achromobacter, Vibrio, and Pseudomonas groups. The percentage of Gram positive organism was very low or nil at times in the ocean fresh sardines.

An elaborate survey was carried out to ascertain the common types of micro-organisms responsible for spoilage in canned prawns. Among nearly 1.500 strains isolated from bacteriologically defective cans, 60% were Gram positive spore formers of the Bacillus type. Other types isolated belonged to Gram positive cocci, Gram negative rods, Gram positive non-spore forming rods, Gram negative cocci and coccoids. No anaerobe could be isolated. The predominant Gram positive spore formers were identified as Bacillus pantothenticus, B. firmus, B. brevis and B. Pumilus species.

The ink of the Indian squid Loligo duvauceli (d'Orbigny) was tested for antibacterial activity. The antibacterial effect of bacteria present in the ink gland was also tested. Only one type of bacteria was found to be present in the ink gland of squid and was identified as Photobacterium leiognathi. Among the various forms of ink extracts, the precipitated and freeze-dried ink showed more pronounced antibacterial effect against Gram-negative bacteria, Salmonella, spp. Escherichia coli, Vibrio cholerae, V. parahaemolyticus and Pseudoinonas spp., and a less pronounced effect against Gram-positive bacteria, Staphylococcus spp. and Micrococcus spp., P. leiognathi did not inhibit any of the above bacteria. The antibacterial activity was associated with the compounds of the ink.

Bacteriological examination of the gastrointestinal microflora of 2 freshwater cichlid fish species (Sarotherodon mossambicus and Tilapia nilotica ) was performed, resulting in the bacteria enumeration of total viable counts of 1.06x10⁷/g and 7.75x10⁷/g of gastro-bacteria intestinal tract plus contents (wet weight) respectively, by aerobic incubation at 30+1°C. The majority (78%) of the total gut isolates from both fish species was Gram positive mesophilic which is characteristic of the higher ambient temperature in the tropics. These isolates were fastidious in their nutritional requirements and together with the rest are isogenous to bacteria autochthonous to soil and water. The occurrence of such organisms is attributed to the feeding habits of these fish. The gastrointestinal bacteria isolated in this study are transient residents but not "indigenous" in these cichlid fish.

Bacterial flora associated with tail rot/fin rot of Carassius auratus, Xiphophorus helleri and hemorrhagic ulcers of Clarias spp were studied. Sensitivity pattern of 33 isolates comprising Aeromonas spp, Pseudomonas spp and Gram-positive rods from diseased C. auratus, X. helleri and Clarias spp were screened against six broad-spectrum antibiotics viz. ciprofloxacin, chloramphenicol, co-trimoxazole, gentamycin, nitro-furantoin and oxytetracycline. Ciprofloxacin was the most effective in inhibiting bacteria at 0.05-0.10 µg/ml level. About 44% of Pseudomonas spp. was resistant to nitrofurantoin. Resistance to oxytetracycline was seen in 27% of Aeromonas spp Gram-positive rods were comparatively more resistant to antibiotics. The multiple antibiotic resistances were seen in 21% of the bacterial isolates of diseased fish.

Amphibian skin is a rich resource of antimicrobial peptides like maximins and maximins H from toad Bombina maxima. A novel cDNA clone encoding a precursor protein that comprises maximin 3 and a novel peptide. named maximin H5. was isolated from a skin cDNA library of B. maxima. The predicted primary structure of maximin H5 is ILGPVLGLVSDTLDDVLGIL-NH2,. Containing three aspartate residues and no basic amino acid residues. maximin H5 is characterized by an anionic property. Different from cationic maximin H peptides. only Gram-positive strain Staphylococcus aureus was sensitive to maximin H5. while the other bacteria] and fungal strains tested ere resistant to it. The presence of metal ions. like Zn2+ and Mg2+, did not increase its antimicrobial potency. Maximin H5 represents the first example of potential anionic antimicrobial peptides from amphibians, The results provide the first evidence that. together kith cationic antimicrobial peptides. anionic antimicrobial peptides may also exist naturally as part of the innate defense system. A Gram-positive bacterium, designated strain CW 7(T), was isolated from forest soil in Anhui Province, south-east China. Cells were strictly aerobic, motile with peritrichous flagella and rod-shaped. The strain grew optimally at 30-37 degrees C and pH 7.0-8.0. The major fatty acids of strain CW 7(T) were anteiso-C-15:0, iso-C-15:0 and anteiso-C-17:0. The predominant menaquinone was MK-7. The cell-wall peptidoglycan contained meso-diaminopimelic acid. The G + C content of the genomic DNA was 42.3 mol%. Phylogenetic analysis indicated that strain CW 7(T) belonged to a monophyletic cluster within the genus Bacillus and showed 16S rRNA gene sequence similarities of less than 96.5% to recognized species of the genus Bacillus. The results of the polyphasic taxonomic study, including phenotypic, chemotaxonomic and phylogenetic analyses, showed that strain CW 7(T) represents a novel species of the genus Bacillus, for which the name Bacillus pallidus sp. nov. is proposed. In the present study, EA-CATH1 and EA-CATH2 were identified from a constructed lung cDNA library of donkey (Equus asinus) as members of cathelicidin-derived antimicrobial peptides, using a nested PCR-based cloning strategy. Composed of 25 and 26 residues, respectively, EA-CATH1 and EA-CATH2 are smaller than most other cathelicidins and have no sequence homology to other cathelicidins identified to date. Chemically synthesized EA-CATH1 exerted potent antimicrobial activity against most of the 32 strains of bacteria and fungi tested, especially the clinically isolated drug-resistant strains, and minimal inhibitory concentration values against Gram-positive bacteria were mostly in the range of 0.3-2.4 mu g center dot mL-1. EA-CATH1 showed an extraordinary serum stability and no haemolytic activity against human erythrocytes in a dose up to 20 mu g center dot mL-1. CD spectra showed that EA-CATH1 mainly adopts an alpha-helical conformation in a 50% trifluoroethanol/water solution, but a random coil in aqueous solution. Scanning electron microscope observations of Staphylococcus aureus (ATCC2592) treated with EA-CATH1 demonstrated that EA-CATH could cause rapid disruption of the bacterial membrane, and in turn lead to cell lysis. This might explain the much faster killing kinetics of EA-CATH1 than conventional antibiotics revealed by killing kinetics data. In the presence of CaCl2, EA-CATH1 exerted haemagglutination activity, which might potentiate an inhibition against the bacterial polyprotein interaction with the host erythrocyte surface, thereby possibly restricting bacterial colonization and spread.

Crude mucus and its partially purified fractions from two marine catfish from Mumbai, Arius dussumieri and Osteogeneiosus militaris were assayed for their crinotoxicity through assays for hemolysis and haemagglutination of chicken erythrocytes, formation of paw edema in mice, and antibacterial activity against one gram-positive and four gram-negative bacteria. Assays were also done to block the edema using Phineramine maleate, Piroxicam, and Atropine sulfate. Crude toxin as well as their fractions from both the fishes exhibited haemolytic and haemagglutinating activities on chicken blood, besides edematous activity in mice models. The edematous activity was blocked by Phineramine maleate and Piroxicam but enhanced by Atropine sulfate; however, all these activities, either blocking or enhancing, were statistically insignificant. Antibacterial activity was absent in all the extracts tested.

Spoilage characteristics of different types of bacteria isolated from bacteriologically defective cans and processing factory environs were studied by inoculating pure cultures into sterile prawn meat. The pattern of spoilage, namely, production of off odour, bulging of the cans and disintegration of meat were observed. Data on spoilage under aerobic as well as anaerobic conditions are presented. Most of the cultures produced some kind of spoilage, though differences were observed in the extent of spoilage produced by different types of bacteria. Gram positive spore formers were found to be the major spoilers and the extent of spoilage was more with mixed cultures.

The sensitivity of 61 cultures of bacteria isolated from fish towards chlortetracycline (CTC) at 5 ppm and 20 ppm levels has been determined on two solid media: sea water agar (SWA) and a distilled water based medium consisting of peptone, beef extract, glucose and NaCI (PBGA). The cultures employed consisted of (i) gram-negative rods of marine origin (Achromobacter, Pseudomonas, Vibrio and Flavobacterium) and (ii) gram positive organisms (Micrococci and Corynebacterium). A detailed bacteriological survey of the prawn canneries of Cochin area was carried out to study the nature and type of micro-organisms present in the factory environs and their role in causing contamination of the canned products. About 26% of the total of 1030 strains isolated was found to be gram positive spore-formers of the Bacillus type, the cooling water being their major source. Similar types of organisms formed the major group often met with in defective canned prawn samples picked up from the factories for examination, thus establishing a correlation between bacterial characteristics and load of cooling water and can contamination.
