O trabalho visa estudar a ocorrência e os aspectos epidemiológicos do parasitismo intestinal na aldeia Paranatinga da tribo indígena Parakanã, Amazônia Oriental Brasileira. Foram realizados dois inquéritos coproparasitológicos, em abril de 1992 e em fevereiro de 1995. Os métodos utilizados na identificação dos agentes parasitários foram os de Hoffman e exame direto, dois métodos simples, facilmente exeqüíveis em aldeias indígenas. Da amostra de 126 índios em abril de 1992 (população de 215 índios), 101 (80,2%) encontravam-se parasitados por pelo menos um enteroparasita. Ancilostomídeos foram encontrados em 33,3%, A. lumbricoides, em 42,8%, T. trichiura, em 0,8%, e S. stercoralis, em 5,6%. Em relação aos protozoários, E. histolytica foi encontrada em 65,0% e G. lamblia, em 46,8%. No inquérito de fevereiro de 1995, apesar do aumento da prevalência total em comparação com o de abril de 1992 (p = 0,04), houve diminuição das prevalências de ancilostomídeos, E. histolytica, G. lamblia, e ausência de S. stercoralis (p<0,05). A despeito das ações de saúde e saneamento empreendidas nas aldeias Parakanã, a prevalência de enteroparasitas ainda se encontra elevada na aldeia Paranatinga, sugerindo que as medidas de atenção primária e secundária devem ser imediatamente incrementadas.

As cisteína-proteases estão entre os alvos mais promissores para o desenvolvimento de novos agentes terapêuticos, visto que participam de eventos fundamentais do ciclo de vida de muitos microorganismos, inclusive Giardia. Como a atividade das proteases pode ser controlada por inibidores específicos, essas substâncias têm sido avaliadas quanto ao potencial antiparasitário. Diante disso, o presente estudo teve por objetivo avaliar o efeito in vitro do inibidor de cisteína-proteases E-64 sobre o crescimento, a aderência e a viabilidade de trofozoítos de cepa de Giardia isolada em Botucatu. Nos ensaios de crescimento e aderência, o número de trofozoítos foi estimado microscopicamente em hemocitômetro, enquanto que a viabilidade celular foi avaliada pelo método do MTT. No presente estudo, embora o metronidazol tenha se apresentado bastante efetivo, o E-64 mostrou ser capaz de inibir o crescimento, a aderência e a viabilidade em taxas superiores a 50%, especialmente nos cultivos expostos à concentração de 100 µM. A despeito de preliminares, esses resultados demonstram que o inibidor E-64 pode interferir em processos primordiais para a sobrevivência do parasita, além do que, abrem novas perspectivas para investigações futuras a fim de se avaliar o real potencial giardicida dos inibidores de proteases.

The scope of this article is to analyze the prevalence and factors associated with the development of infectious diseases that affect children in daycare centers, namely respiratory infections, diarrheal disease and parasitic infections. Bibliographic research was conducted in the MEDLINE, LILACS and SciELO databases, and observational studies were included. 129 studies were identified, of which 21 were considered relevant to this study, namely two longitudinal and 19 cross-sectional studies. The systematization of the reviewed studies highlighted: i) the presence of intestinal parasites was the main outcome analyzed, followed by respiratory infections; ii) only one study investigated the occurrence of diarrheal disease; iii) the Giardia lamblia was the most prevalent parasitosis; iv) the variables that were most often associated with the development of intestinal parasitosis were child age, family income and maternal education; v) the attendance at daycare centers was a risk factor for intestinal parasites and respiratory infections. Respiratory and parasitic infections are major problems in institutionalized children in daycare centers. The reduction of such diseases involves a complex web of socio-economic, sanitation and daycare center infrastructure aspects.

There is little research on the practices of sanitary inspection in the chain of production of vegetables to the consumer, especially those eaten raw, they are liable to serve microorganism such as bacteria, fungi and parasites, contributing to possible health hazards. The aim of this study was to assess qualitatively contamination by parasites and / or commensals of medical interest in lettuce leaves (Lactuca sativa) fresh market in the municipality of Quata Sao Paulo. A total of 15 random samples were analyzed every other day of the three different places that sell vegetables a grocery store, a supermarket and a vegetable garden during the month of May 2011. The parasites and / or commensals found in lettuce were Entamoeba coli (67%), Entamoeba histolytica (20%), Giardia sp (13%) and Ascaris lumbricoides (7%). The analysis showed the presence of parasites and / or commensals in all samples, except in the cultivated garden which showed poor sanitary conditions, probably due to contamination in the shipping and handling by third parties in supermarket and grocery store. The parasite monitoring sanitary conditions of vegetables sold in urban environments becomes relevant for preventive measures to avoid the continued parasitic cycle and possible future health complications.

Background: Infectious diarrhea can be caused by bacteria, viruses, or protozoan organisms, or a combination of these. The identification of co-infections in dogs is important to determine the prognosis and to plan strategies for their treatment and prophylaxis. Although many pathogens have been individually detected with real-time polymerase chain reaction (PCR), a comprehensive panel of agents that cause diarrhea in privately owned dogs has not yet been established. The objective of this study was to use a real-time PCR diarrhea panel to survey the frequencies of pathogens and co-infections in owned dogs attended in a veterinary hospital with and without diarrhea, as well the frequency in different countries. Feces samples were tested for canine distemper virus, canine coronavirus, canine parvovirus type 2 (CPV-2), Clostridium perfringens alpha toxin (CPA), Cryptosporidium spp., Giardia spp., and Salmonella spp. using molecular techniques.Results: In total, 104 diarrheic and 43 control dogs that were presented consecutively at a major private veterinary hospital were included in the study. Overall, 71/104 (68.3%) dogs with diarrhea were positive for at least one pathogen: a single infection in 39/71 dogs (54.9%) and co-infections in 32/71 dogs (45.1%), including 21/32 dogs (65.6%) with dual, 5/32 (15.6%) with triple, and 6/32 (18.8%) with quadruple infections. In the control group, 13/43 (30.2%) dogs were positive, all with single infections only. The most prevalent pathogens in the diarrheic dogs were CPA (40/104 dogs, 38.5%), CPV-2 (36/104 dogs, 34.6%), and Giardia spp. (14/104 dogs, 13.5%). CPV-2 was the most prevalent pathogen in the dual co-infections, associated with CPA, Cryptosporidium spp., or Giardia spp. No statistical difference (P = 0.8374) was observed in the duration of diarrhea or the number of deaths (P = 0.5722) in the presence or absence of single or co-infections.Conclusions: Diarrheic dogs showed a higher prevalence of pathogen infections than the controls. Whereas the healthy dogs had only single infections, about half the diarrheic dogs had co-infections. Therefore, multiple pathogens should be investigated in dogs presenting with diarrhea. The effects of multiple pathogens on the disease outcomes remain unclear because the rate of death and the duration of diarrhea did not seem to be affected by these factors.

Introduction: The intestinal parasites continue to represent a significant public health problem, due to the large number of infected people and the various organic changes that can cause. The intestinal parasites become more prevalent in villages with high population density absence or inadequacy of sanitation and adequate, and personal hygiene practices and inadequate housing. Objective: To determine the prevalence of intestinal parasites Ascaris lumbricoides, Trichuris trichiura and Giardia duodenalis in children of school age (7-14 years) living in the city of Salvador, Bahia, which were part of the Environmental Sanitation Program (Bahia Azul) covering eight different sewage basins in the years 1997/1998 and 2003/2004. Methods: Data were collected pre-coded questionnaire administered to the parents of students, collecting information socioeconomic, environmental and housing conditions. Stool samples were analyzed for each school in the Laboratory of Parasitology of the ISC, UFBA. The database was built and analyzed by Epi Info program (version 3.5.1/2008) were built where overall prevalence rate and Basin depletion of parasites. We compared the overall prevalence rates (at least one parasite) before and after the intervention health, as well as species of parasite. Results: The overall prevalence before the intervention was 56,2% (N = 729) and 42,5% after the intervention (N = 890), down -24,5%. The prevalence observed for each species A. lumbricoides, T. trichiura and G. duodenalis before intervention was respectively 31,4%, 35,8% and 9,9% respectively after the intervention and 24,4%, 28,8% and 6,0%. In relation to variations in prevalence were found to decrease A. lumbricoides, T. Trichiura and G. duodenalis respectively -22,4%, -19,7% and -39,7%. The objective of this study was to verify the prevalence of intestinal parasites and/or commensals in the neoplastic patients undergoing chemotherapy. Stool samples were analyzed by the method of Lutz (1919) and Rugai (1954), in triplicate. This work was composed of three groups, the first one (GI) formed by neoplastic patients that are not undergoing chemotherapy, the second (GII) comprised patients who were undergoing chemotherapy, and the third group (GIII) consisting of patients who completed chemotherapy. A total of 30 patients (GI-5, GII-18 and GIII-7) were screened at the Assis Regional Hospital of the Unified Health System of Assis, São Paulo.Additional information on antiparasitic treatment and tumor type were obtained by questionnaire. The positivity was 66.7% (20 cases) for intestinal parasites and/or commensals. The helminths were Ascaris lumbricoides (36.7%), Hookworms (20%) and Hymenolepis diminuta (3.3%). Among the highlights are protozoan Giardia lamblia (46.7%), Entamoeba coli (6.7%), E. histolytica /dispar (3.3%), Endolimax nana (3.3%) and Iodameba butschlii (3.3%). The high frequency of intestinal parasites and/or commensals in the neoplastic patients can be attributed to poor personal hygiene and lack of immunity to reinfection and poor knowledge of the prophylaxis of infection by protozoa and helminths. The results indicate the necessity of adopting a new criterion for neoplastic patients undergoing chemotherapy, primarily performing parasitological diagnosis, treatment and monitoring of cure of intestinal parasitic infections in this risk group.

The objective of this study was to detect possible carriers of parasites and/or commensals between the horticulturists of Fair of the Producer from Upper Paranaiba, Minas Gerais. A total of 30 horticulturists were instructed to collect three stool samples on alternate days during the months of August and September 2007. Horticulturists were positive 40% for one or more parasite and / or dinner, and found: Giardia lamblia (3.5%), Entamoeba histolytica / E. dispar (7%), Entamoeba coli (13%), Endolimax nana (13%) and Ascaris lumbricoides (3.5%). Stool of gardeners is of importance in monitoring parasite sanitary conditions of vegetables sold in urban environments.

Florianopolis, a city located in the Santa Catarina State in southern Brazil, is the national leading producer of bivalve mollusks. The quality of bivalve mollusks is closely related to the sanitary conditions of surrounding waters where they are cultivated. Presently, cultivation areas receive large amounts of effluents derived mainly from treated and non-treated domestic, rural, and urban sewage. This contributes to the contamination of mollusks with trace metals, pesticides, other organic compounds, and human pathogens such as viruses, bacteria, and protozoan. The aim of this study was to perform a thorough diagnosis of the shellfish growing areas in Florianopolis, on the coast of Santa Catarina. The contamination levels of seawater, sediments, and oysters were evaluated for their microbiological, biochemical, and chemical parameters at five sea sites in Florianopolis, namely three regular oyster cultivation areas (Sites 1, 2, and oyster supplier), a polluted site (Site 3), and a heavily polluted site (Site 4). Samples were evaluated at day zero and after 14 days. Seawater and sediment samples were collected just once, at the end of the experiment. Antioxidant defenses, which may occur in contaminated environments in response to the increased production of reactive oxygen species (ROS) by organisms, were analyzed in oysters, as well as organic compounds (in oysters and sediment samples) and microbiological contamination (in oysters and seawater samples). The results showed the presence of the following contaminants: fecal coliforms in seawater samples (four sites), human adenovirus (all sites), human noroviruses GI and GII (two sites), Hepatitis A viruses (one site), JC Polyomavirus in an oyster sample from the oyster supplier, Giardia duodenalis cysts, and Cryptosporidium sp oocysts (one site). Among organochlorine pesticides, only DDT (dichlorodiphenyltrichloroethane) and HCH (hexachlorocyclohexane) were detected in some sediment and oysters samples in very low levels; site 4 had the highest concentrations of total aliphatic hydrocarbons. PAHs, and linear alkylbenzenes (LABs) found either in oysters or in sediment samples. The major concentration of fecal sterol coprostanol was found at site 4, followed by site 3. After 14 days of allocation in the four selected sites, there was a significant difference in the enzymes analyzed at the monitored spots. The detection of different contaminants in oysters, seawater, and sediment samples in the present study shows the impact untreated or inadequately treated effluents have on coastal areas. These results highlight the need for public investment in adequate wastewater treatment and adequate treatment of oysters, ensuring safe areas for shellfish production as well as healthier bivalve mollusks for consumption.

Cryptosporidium parvum infection is very important with respect to public health, owing to foodborne and waterborne outbreaks and gastrointestinal illness in immunocompetent and immunocompromised persons. In cattle, infection with this species manifests either as a subclinical disease or with diarrheal illness, which occurs more often in the presence of other infectious agents than when alone. The aim of this study was to develop a real-time polymerase chain reaction (PCR) assay for the detection of C. parvum in calf fecal samples and to compare the results of this assay with those of the method routinely used for the diagnosis of Cryptosporidium spp., nested PCR targeting the 18S rRNA gene. Two hundred and nine fecal samples from calves ranging in age from 1 day to 6 months were examined using real-time PCR specific for the actin gene of C. parvum and by a nested PCR targeting the 18S rRNA gene of Cryptosporidium spp. Using real-time PCR detection, 73.2% (153 out of 209) of the samples were positive for C. parvum, while 56.5% (118 out of 209) of the samples were positive for Cryptosporidium spp. when the nested PCR amplification method was used for the detection. The analytical sensitivity of the real-time PCR was approximately one C. parvum oocyst. There was no significant nonspecific DNA amplification of any of the following species and genotype: Cryptosporidium andersoni, Cryptosporidium baileyi, Cryptosporidium bovis, Cryptosporidium canis, Cryptosporidium galli, Cryptosporidium ryanae, Cryptosporidium serpentis, or avian genotype II. Thus, we conclude that real-time PCR targeting the actin gene is a sensitive and specific method for the detection of C. parvum in calf fecal samples.
