Interferon-gamma is mainly produced by activated T helper cells and cytotoxic T lymphocytes and sustains the immune-defense against viral and bacterial infections. For a better understanding of IFN-gamma promoter regulation in T cells, different DNA-binding motivs were examined. Hereby, a new motiv (-196 to -183) was identified, that binds to the transcription factor AP-1 in T helper cells and Jurkat T cells. This factor acts as an essential activator protein. Further investigation demonstrated that IL-12 and IL-18 induce different regulatory pathways. Both AP-1 and STAT-4 bindings at their cognate DNA elements (-196 to -183 and -224 to -215) are required for the IL-12 dependent activation whereas IL-18 causes direct activation via AP-1.Moreover, the TH2 cytokine IL-4 represses significantly the IFN-gamma promoter activity in CD4+ T cells. IL-4 induces GATA-3, that interacts with two DNA-motivs (-111 to -87) at the IFN-gamma promoter.Furthermore, transgenic mice were generated, yielding a human IFN-gamma promoter construct (410 bp) under the control of a luciferase reporter gene. The data demonstrated a specific IFN-gamma promoter activation by antiCD3 plus antiCD28 in CD4+ and CD8+ T cells. The luciferase activty in CD4+ T cells was reinforced by addition of IL-12 and IL-18 and repressed by IL-4.

Interleukin-12 ist ein Schlüsselregulator zellvermittelter Immunantworten. In der vorliegenden Dissertation wurde die Regulation des IL-12 p40 Promotors in Monozyten und Makrophagen untersucht. Mit Hilfe von 'In Vivo Footprinting'-, Bandshift- und Transfektionsexperimenten konnte eine wichtige Funktion der Transkriptionsfaktoren NF-kappaB, C/EBP-beta und PU.1 bei der Aktivierung des Promotors gezeigt werden. 'In Vivo Footprinting'-Experimente führten auch zur Identifikation eines bisher nicht beschriebenen regulatorischen Elementes im IL-12 p40 Promotor. Dieses GA-12 genannte Motiv war in unstimulierten primären Monozyten protektiert, nicht jedoch in Zellen, die mit LPS oder LPS/ Interferon-gamma stimuliert wurden. Eine genauere Charakterisierung ergab, daß dieses Motiv eine repressorische Funktion auf den Promotor ausübt und in unstimulierten, nicht jedoch in stimulierten Monozyten und Makrophagen von einem spezifischen GATA-ähnlichen Komplex (GAP-12) erkannt wird. Zudem führte die Behandlung von Monozyten mit Interleukin-4 und Prostaglandin-E2, Inhibitoren der IL-12 Produktion, zu einer Verstärkung der Komplexbildung am GA-12 Motiv. Zur Analyse der Promotorregulation im nukleosomalen Umfeld wurden außerdem IL-12 p40 Promotor/ Luziferase transgene Mäuse hergestellt. Die Analyse dieser Mäuse zeigte, daß der proximale Promotor ausreichend Sequenzinformation beinhaltet, um in vivo die stimulations- und gewebespezifische Regulation des Gens zu gewährleisten. Anhand der erhobenen Daten konnte ein Modell der Regulation des IL-12 p40 Promotors in Monozyten und Makrophagen entworfen werden.

During vertebrate development, the lung inaugurates as an endodermal bud from the primitive foregut. Dichotomous subdivision of the bud results in arborizing airways that form the prospective gas exchanging chambers, where a thin blood-gas barrier (BGB) is established. In the mammalian lung, this proceeds through conversion of type II cells to type I cells, thinning, and elongation of the cells as well as extrusion of the lamellar bodies. Subsequent diminution of interstitial tissue and apposition of capillaries to the alveolar epithelium establish a thin BGB. In the noncompliant avian lung, attenuation proceeds through cell-cutting processes that result in remarkable thinning of the epithelial layer. A host of morphoregulatory molecules, including transcription factors such as Nkx2.1, GATA, HNF-3, and WNT5a; signaling molecules including FGF, BMP-4, Shh, and TFG- β and extracellular proteins and their receptors have been implicated. During normal physiological function, the BGB may be remodeled in response to alterations in transmural pressures in both blood capillaries and airspaces. Such changes are mitigated through rapid expression of the relevant genes for extracellular matrix proteins and growth factors. While an appreciable amount of information regarding molecular control has been documented in the mammalian lung, very little is available on the avian lung.

The expression pattern of angiotensin AT2 receptors with predominance during fetal life and upregulation under pathological conditions during tissue injury/repair process suggests that AT2 receptors may exert an important action in injury/repair adaptive mechanisms. Less is known about AT2 receptors in acute ischemia-induced cardiac injury. We aimed here to elucidate the role of AT2 receptors after acute myocardial infarction. Double immunofluorescence staining showed that cardiac AT2 receptors were mainly detected in clusters of small c-kit+ cells accumulating in peri-infarct zone and c-kit+AT2+ cells increased in response to acute cardiac injury. Further, we isolated cardiac c-kit+AT2+ cell population by modified magnetic activated cell sorting and fluorescence activated cell sorting. These cardiac c-kit+AT2+ cells, represented approximately 0.19% of total cardiac cells in infarcted heart, were characterized by upregulated transcription factors implicated in cardiogenic differentiation (Gata-4, Notch-2, Nkx-2.5) and genes required for self-renewal (Tbx-3, c-Myc, Akt). When adult cardiomyocytes and cardiac c-kit+AT2+ cells isolated from infarcted rat hearts were cocultured, AT2 receptor stimulation in vitro inhibited apoptosis of these cocultured cardiomyocytes. Moreover, in vivo AT2 receptor stimulation led to an increased c-kit+AT2+ cell population in the infarcted myocardium and reduced apoptosis of cardiomyocytes in rats with acute myocardial infarction. These data suggest that cardiac c-kit+AT2+ cell population exists and increases after acute ischemic injury. AT2 receptor activation supports performance of cardiomyocytes, thus contributing to cardioprotection via cardiac c-kit+AT2+ cell population.

Human steroid biosynthesis depends on a specifically regulated cascade of enzymes including 3β-hydroxysteroid dehydrogenases (HSD3Bs). Type 2 HSD3B catalyzes the conversion of pregnenolone, 17α-hydroxypregnenolone and dehydroepiandrosterone to progesterone, 17α-hydroxyprogesterone and androstenedione in the human adrenal cortex and the gonads but the exact regulation of this enzyme is unknown. Therefore, specific downregulation of HSD3B2 at adrenarche around age 6-8 years and characteristic upregulation of HSD3B2 in the ovaries of women suffering from the polycystic ovary syndrome remain unexplained prompting us to study the regulation of HSD3B2 in adrenal NCI-H295R cells. Our studies confirm that the HSD3B2 promoter is regulated by transcription factors GATA, Nur77 and SF1/LRH1 in concert and that the NBRE/Nur77 site is crucial for hormonal stimulation with cAMP. In fact, these three transcription factors together were able to transactivate the HSD3B2 promoter in placental JEG3 cells which normally do not express HSD3B2. By contrast, epigenetic mechanisms such as methylation and acetylation seem not involved in controlling HSD3B2 expression. Cyclic AMP was found to exert differential effects on HSD3B2 when comparing short (acute) versus long-term (chronic) stimulation. Short cAMP stimulation inhibited HSD3B2 activity directly possibly due to regulation at co-factor or substrate level or posttranslational modification of the protein. Long cAMP stimulation attenuated HSD3B2 inhibition and increased HSD3B2 expression through transcriptional regulation. Although PKA and MAPK pathways are obvious candidates for possibly transmitting the cAMP signal to HSD3B2, our studies using PKA and MEK1/2 inhibitors revealed no such downstream signaling of cAMP. However, both signaling pathways were clearly regulating HSD3B2 expression.

The LIM domain-binding protein Ldb1 is an essential cofactor of LIM-homeodomain (LIM-HD) and LIM-only (LMO) proteins in development. The stoichiometry of Ldb1, LIM-HD, and LMO proteins is tightly controlled in the cell and is likely a critical determinant of their biological actions. Single-stranded DNA-binding proteins (SSBPs) were recently shown to interact with Ldb1 and are also important in developmental programs. We establish here that two mammalian SSBPs, SSBP2 and SSBP3, contribute to an erythroid DNA-binding complex that contains the transcription factors Tal1 and GATA-1, the LIM domain protein Lmo2, and Ldb1 and binds a bipartite E-box-GATA DNA sequence motif. In addition, SSBP2 was found to augment transcription of the Protein 4.2 (P4.2) gene, a direct target of the E-box-GATA-binding complex, in an Ldb1-dependent manner and to increase endogenous Ldb1 and Lmo2 protein levels, E-box-GATA DNA-binding activity, and P4.2 and beta-globin expression in erythroid progenitors. Finally, SSBP2 was demonstrated to inhibit Ldb1 and Lmo2 interaction with the E3 ubiquitin ligase RLIM, prevent RLIM-mediated Ldb1 ubiquitination, and protect Ldb1 and Lmo2 from proteasomal degradation. These results define a novel biochemical function for SSBPs in regulating the abundance of LIM domain and LIM domain-binding proteins.

Tumor necrosis factor receptor p75/80 ((TNF-R p75/80) is a 75 kDa type 1 transmembrane protein expressed predominately on cells of hematopoietic lineage. TNF-R p75/80 belongs to the TNF receptor superfamily characterized by cysteine-rich extracellular regions composed of three to six disulfide-linked domains. In the present report, we have characterized, for the first time, the complete gene structure for human TNF-R p75/80 which spans approximately 43 kbp. The gene consists of 10 exons (ranging from 34 bp to 2.5 kbp) and 9 introns (343 bp to 19 kbp). Consensus elements for transcription factors involved in T cell development and activation were noted in the 5$\sp\prime$ flanking region including TCF-1, Ikaros, AP-1, CK-2, IL-6RE, ISRE, GAS, NF-$\kappa$B and SP1, as well as an unusually high GC content and CpG frequency that appears characteristic of some TNF-R family members. The unusual (GATA)$\sb{\rm n}$ and (GAA)(GGA) repeats found within intron 1 may prove useful for further genome analysis within the 1p36 chromosomal locus. The human TNF-R p75/80 gene structure will permit further assessment of its involvement in normal hematopoietic cell development and function, autoimmune disease, and non-random translocations in hematopoietic malignancies. The region 1.8 kb 5$\sp\prime$ of the ATG was able to drive luciferase expression when transfected into cell lines expressing TNF-R p75/80. Further characterization of the 5$\sp\prime$-regulatory region will aid in determining factors and signal transduction pathways involved in regulating TNF-R p75/80 expression. ^

The Wilms' tumor 1 gene (WT1) encodes a zinc-finger transcription factor and is expressed in urogenital, hematopoietic and other tissues. It is expressed in a temporal and spatial manner in both embryonic and adult stages. To obtain a better understanding of the biological function of WT1, we studied two aspects of WT1 regulation: one is the identification of tissue-specific cis-regulatory elements that regulate its expression, the other is the downstream genes which are modulated by WT1.^ My studies indicate that in addition to the promoter, other regulatory elements are required for the tissue specific expression of this gene. A 259-bp hematopoietic specific enhancer in intron 3 of the WT1 gene increased the transcriptional activity of the WT1 promoter by 8- to 10-fold in K562 and HL60 cells. Sequence analysis revealed both GATA and c-Myb motifs in the enhancer fragment. Mutation of the GATA motif decreased the enhancer activity by 60% in K562 cells. Electrophoretic mobility shift assays showed that both GATA-1 and GATA-2 proteins in K562 nuclear extracts bind to this motif. Cotransfection of the enhancer containing reporter construct with a GATA-1 or GATA-2 expression vector showed that both GATA-1 and GATA-2 transactivated this enhancer, increasing the CAT reporter activity 10-15 fold and 5-fold respectively. Similar analysis of the c-Myb motif by cotransfection with the enhancer CAT reporter construct and a c-Myb expression vector showed that c-Myb transactivated the enhancer by 5-fold. A DNase I-hypersensitive site has been identified in the 258 bp enhancer region. These data suggest that GATA-1 and c-Myb are responsible for the activity of this enhancer in hematopoietic cells and may bind to the enhancer in vivo. In the process of searching for cis-regulatory elements in transgenic mice, we have identified a 1.0 kb fragment that is 50 kb downstream from the promoter and is required for the central nervous system expression of WT1.^ In the search for downstream target genes of WT1, we noted that the proto-oncogene N-myc is coexpressed with the tumor suppressor gene WT1 in the developing kidney and is overexpressed in many Wilms' tumors. Sequence analysis revealed eleven consensus WT1 binding sites located in the 1 kb mouse N-myc promoter. We further showed that the N-myc promoter was down-regulated by WT1 in transient transfection assays. Electrophoretic mobility shift assays showed that oligonucleotides containing the WT1 motifs could bind WT1 protein. Furthermore, a Denys-Drash syndrome mutant of WT1, R394W, that has a mutation in the DNA binding domain, failed to repress the N-myc promoter. This suggests that the repression of the N-myc promoter is mediated by DNA binding of WT1. This finding helps to elucidate the relationship of WT1 and N-myc in tumorigenesis and renal development. ^

Lim domain only 2 (LMO2) is a transcriptional co-factor required for angiogenesis and the specification of haematopoietic cells during development. LMO2 is widely expressed within haematopoiesis with the exception of T-cells. Failure to downregulate LMO2 during T-cell maturation leads to leukaemia, thus underlining the critical nature of context-dependent regulation of LMO2 expression. We previously identified a distal regulatory element of LMO2 (element -25) that cooperates with the proximal promoter in directing haematopoietic expression. Here we dissected the functional activity of element -25 and showed it to consist of two modules that conferred independent and cell-type specific activities: a 3' myeloid enhancer and a 5' T-cell repressor. The myeloid enhancer was bound by GATA2 in progenitors and its activity depended on a highly conserved GATA motif, whereas the T-cell repressor moiety of element -25 was bound by the Core Binding Factor in T-cells and its repressive activity depended on a highly conserved RUNT motif. Since the myeloid enhancer and nearby downstream region is recurrently involved in oncogenic translocations, our data suggest that the -25 enhancer region provides an open chromatin environment prone to translocations, which in turn cause aberrant LMO2 expression in T-cells due to the removal of the adjacent T-cell repressor.

Molecular mechanisms that underlie preleukemic myelodysplasia (MDS) and acute myelogenous leukemia (AML) are poorly understood. In MDS or AML with a refractory clinical course, more than 30% of patients have acquired interstitial or complete deletions of chromosome 5. The 5q13.3 chromosomal segment is commonly lost as the result of 5q deletion. Reciprocal and unbalanced translocations of 5q13.3 can also occur as sole anomalies associated with refractory AML or MDS. This study addresses the hypothesis that a critical gene at 5q13.3 functions either as a classical tumor suppressor or as a chromosomal translocation partner and contributes to leukemogenesis. ^ Previous studies from our laboratory delineated a critical region of loss to a 2.5–3.0Mb interval at 5q13.3 between microsatellite markers D5S672 and GATA-P18104. The critical region of loss was later resolved to an interval of approximately 2Mb between the markers D5S672 and D5S2029. I, then generated a long range physical map of yeast artificial chromosomes (YACs) and developed novel sequence tagged sites (STS). To enhance the resolution of this map, bacterial artificial chromosomes (BACs) were used to construct a triply linked contig across a 1 Mb interval. These BACs were used as probes for fluorescent in situ hybridization (FISH) on an AML cell line to define the 5q13.3 critical region. A 200kb BAC, 484a9, spans the translocation breakpoint in this cell line. A novel gene, SSDP2 (single stranded DNA binding protein), is disrupted at the breakpoint because its first four exons are encoded within 140kb of BAC 484a9. This finding suggests that SSDP2 is the critical gene at 5q13.3. ^ In addition, I made an observation that deletions of chromosome 5q13 co-segregate with loss of the chromosome 17p. In some cases the deletions result from unbalanced translocations between 5q13 and 17p13. It was confirmed that the TP53 gene is deleted in patients with 17p loss, and the remaining allele harbors somatic mutation. Thus, the genetic basis for the aggressive clinical course in AML and MDS may be caused by functional cooperation between deletion or disruption of the 5q13.3 critical gene and inactivation of TP53. ^

The formation of the placenta is one of the first and most important developmental events that occur in early mammalian embryogenesis. Even given this importance of the placenta, the academic community has largely ignored studying gene regulation during the development and maturation of the placenta. For this reason, an in-depth study of gene regulation in the trophoblast layer of the placenta using murine Adenosine Deaminase (Ada) as a model system has been undertaken. It has been determined that Ada is highly expressed in the placenta and is critical for embryo development. Dr. Kellems' laboratory has previously described a 1.8 kb fragment of the Ada 5 ′ flanking region that is capable of directing trophoblast specific expression in a transgenic model system. Preliminary studies have demonstrated several critical portions of this fragment that are necessary for the correct tissue specific expression in the placenta. My first specific aim was to elucidate the trans factor binding to one of these sequences, the FP3. Through electromobility shift assays (EMSA), the 30 bp FP3 was narrowed to a 5 bp sequence which computer databases predicted bound to Acute Myeloid Leukemia 1 (AML-1). This was confirmed by supershift analysis. The functional importance of this binding was demonstrated by a transgenic approach. A significant difference in expression of the reporter in the placenta was seen when the 5 bp sequence was mutated. This finding is a novel use for the AML-1 transcription factor which is the DNA binding portion of the heterodimer Core Binding Protein (CBP). The 5′ 240 bp region has also been demonstrated to contain functionally significant sequence. Through EMSA assays and computer predictions, the area has been narrowed to two pertinent regions that are predicted to contain GATA binding motifs. ^

Cardiovascular disease (CVD) is the leading cause of death in the United States. One manifestation of CVD known to increase mortality is an enlarged, or hypertrophic heart. Hypertrophic cardiomyocytes adapt to increased contractile demand at the genetic level with a re-emergence of the fetal gene program and a downregulation of fatty acid oxidation genes with concomitant increased reliance on glucose-based metabolism. To understand the transcriptional regulatory pathways that implement hypertrophic directives we analyzed the upstream promoter region of the muscle specific isoform of the nuclear-encoded mitochondrial gene, carnitine palmitoyltransferase-1β (CPT-1β) in cultured rat neonatal cardiac myocytes. This enzyme catalyzes the rate-limiting step of fatty acid entry into β-oxidation and is downregulated in cardiac hypertrophy and failure, making it an attractive model for the study of hypertrophic gene regulation and metabolic adaptations. We demonstrate that the muscle-enriched transcription factors GATA-4 and SRF synergistically activate CPT-1β; moreover, DNA binding to cognate sites and intact protein structure are required. This mechanism coordinates upregulation of energy generating processes with activation of the energy consuming contractile promoter for cardiac α-actin. We hypothesized that fatty acid or glucose responsive transcription factors may also regulate CPT-1β. Oleate weakly stimulates CPT-1β activity; in contrast, the glucose responsive Upstream Stimulatory Factors (USF) dramatically depresses the CPT-1β reporter. USF regulates CPT-1β through a novel physical interaction with the cofactor PGC-1 and abrogation of MEF2A/PGC-1 synergistic stimulation. In this way, USF can inversely regulate metabolic gene programs and may play a role in the shift of metabolic substrate preference seen in hypertrophy. Failing hearts have elevated expression of the nuclear hormone receptor COUP-TF. We report that COUP-TF significantly suppresses reporter transcription independent of DNA binding and specific interactions with GATA-4, Nkx2.5 or USF. In summary, CPT-1β transcriptional regulation integrates mitochondrial gene expression with two essential cardiac functions: contraction and metabolic substrate oxidation. ^

Fundamentos de la Tesis La presente Tesis investiga acerca de las primeras arquitecturas que surgieron a lo largo de La Costa del Sol con motivo de la apertura al exterior de la España turística de los años cincuenta, y cómo su implantación iba a suponer, a lo largo de la segunda mitad del siglo, la paulatina aparición de modos de vida y desarrollos urbanos que pueden ser considerados hoy día como contemporáneos. El Pacto Americano de 1953, pese a su origen militar, supuso en España el surgimiento a su vez de contraprestaciones económicas y de relación que permitirían salir al país de la autarquía y el inicio de un proceso de homologación con el entorno occidental de consecuencias imprevisibles. La Costa del Sol nace de esta manera como fenómeno de explotación turística de un territorio de paisaje amable y de buen clima. La investigación profundiza en el hecho turístico y en sus derivaciones antropológicas, sociales, económicas y culturales como resultado del intercambio de poblaciones autóctonas y lejanas en un lugar donde se relacionan diferentes modos de vida. En este sentido, la visita de Dalí a Torremolinos en 1930 permite introducir en la investigación conceptos relacionados con el azar como integrador de errores, deformaciones o transformaciones no previstas. Estas primeras arquitecturas de llamativo corte moderno aparecieron durante la década entre 1959 y 1969, en el entorno virgen preexistente, comprendido entre la playa y la montaña, proporcionando densidad y singularidad con sus diseños, y alentando un carácter de multiplicidad desde sus propios modelos, lo que favorecería un fenómeno interactivo entre las distintas soluciones. Visto desde la capital del Estado se reconocería un singular territorio supra-provincial entre el Cabo de Gata y la Punta de Tarifa. En dicho territorio, una serie de pequeñas poblaciones se enlazarían a través del cordón litoral gracias a la carretera nacional 340. Esta primera arquitectura comercial privada y de calidad surgió también gracias a la conexión del aeropuerto de Málaga con el turismo internacional. Gracias a esta conexión se exhibirán rasgos pertenecientes a la cultura arquitectónica de la época, a lo moderno, como reclamo, fundamentalmente edifi cios exóticos que mostrarán rasgos de progreso en una periferia empobrecida y rural. Metodología empleada • Búsqueda de fuentes primarias como documentación original de los diferentes proyectos y entrevistas con arquitectos protagonistas de esa época. • Búsqueda, catalogación y ordenación de los diferentes planes de ordenación de la Costa del Sol que se hicieron durante la década de estudio así como un análisis de sus propuestas y consecuencias. • Estudio de casos relativos a los dos modelos de actuaciones arquitectónicas predominantes en el periodo de estudio: los hoteles y los complejos de apartamentos. • Establecimiento de un planteamiento gradual por escalas, trabajando como “múltiplos” y permitiendo generar lazos de autosimilitud. • Análisis de la relación de estos proyectos con el paisaje circundante, con lo urbano, existiese o no, y con el interior de las células de apartamentos. Aportaciones de la Tesis El fenómeno estudiado ha permitido atisbar otra escala más, que atiende a la aparición de un nuevo ente urbano. Para ello las teorías sobre la ciudad vertidas durante los años 30 por F. Ll. Wright, junto con la visión reveladora de Reyner Banham sobre Los Ángeles a principios de los 70 o de Rem Koolhaas sobre la ciudad genérica como apoteosis del concepto de elección múltiple, permiten establecer parámetros que enmarcan el estudio del nuevo ente urbano de difícil legibilidad que se funda en la Costa del Sol durante la década de los 60. La investigación concluye acerca del papel de estas primeras arquitecturas del turismo dentro del proceso dinámico, imparable y crecedero que se inició durante la época de estudio. Un fenómeno que ha dado lugar a un desarrollo suburbial y a-jerárquico, sin centro concreto, donde se dan lugar multitud de relaciones en continua adaptación que permiten defi nirlo como una nueva ciudad. En este proceso estas primeras arquitecturas surgieron como objetos exóticos por su modernidad desafi ante. Esta cualidad convirtió a estos edifi cios en verdaderos atractores de actividad, embajadores de nuevos modos de vida contemporáneos. Por lo que en sus entornos próximos a lo largo de este territorio se provocó la cristalización paulatina de un hecho urbano. Su evolución posterior ha derivado en un progresivo relleno de huecos y vacíos. De manera no planifi cada, azarosa e incluso inesperada, las primeras arquitecturas, que servían a la vida a medias que supone el hecho turístico, han derivado en una paulatina ciudad completa, exenta de jerarquías. Conclusión final La arquitectura se hace más real que nunca como algo tangible en el fl uir de la ciudad. El proceso imparable que se inició en la Costa del Sol sólo puede comprenderse desde sus arquitecturas, que acogen momentos y lugares. En ellas se entremezclan signos, cultura, modos de vida, estilo, construcción, economía y vida contemporánea que han ido colonizando poco a poco este territorio en un proceso irreversible como en tantos otros lugares del planeta. Este proceso de desvelamiento ha constituido el motivo principal de esta Tesis
