Cholesterol is an essential component in the membranes of most eukaryotic cells, in which it mediates many functions including membrane fluidity, permeability and the formation of ordered membrane domains. In this work a fluorescent and a non-fluorescent cholesterol analog were characterized as tools to study cholesterol. Next, these analogs were used to study two specific cell biological processes that involve cholesterol, i.e. the structure and function of ordered membrane domains/rafts and intracellular cholesterol transport. The most common method for studying ordered membrane domains is by disrupting them by cholesterol depletion. Because cholesterol depletion affects many cellular functions besides those mediated by membrane domains, this procedure is highly unspecific. The cellular exchange of cholesterol by desmosterol as a tool to study ordered membrane domains was characterized. It turned out that the ability of desmosterol to form and stabilize membrane domains in vitro was weaker compared to cholesterol. This result was reinforced by atomistic scale simulations that indicated that desmosterol has a lower ordering effect on phospholipid acyl chains. Three procedures were established for exchanging cellular cholesterol by desmosterol. In cells in which desmosterol was the main sterol, insulin signaling was attenuated. The results suggest that this was caused by desmosterol destabilizing membrane rafts. Contrary to its effect on ordered membrane domains it was found that replacing cholesterol by desmosterol does not change cell growth/viability, subcellular sterol distribution, Golgi integrity, secretory pathway, phospholipid composition and membrane fluidity. Together these results suggest that exchanging cellular cholesterol by desmosterol provides a selective tool for perturbing rafts. Next, the importance of cholesterol for the structure and function of caveolae was analyzed by exchanging the cellular cholesterol by desmosterol. The sterol exchange reduced the stability of caveolae as determined by detergent resistance of caveolin-1 and heat resistance of caveolin-1 oligomers. Also the sterol exchange led to aberrations in the caveolar structure; the morphology of caveolae was altered and there was a larger variation in the amount of caveolin-1 molecules per caveola. These results demonstrate that cholesterol is important for caveolar stability and structural homogeneity. In the second part of this work a fluorescent cholesterol analog was characterized as a tool to study cholesterol transport. Tight control of the intracellular cholesterol distribution is essential for many cellular processes. An important mechanism by which cells regulate their membrane cholesterol content is by cholesterol traffic, mostly from the plasma membrane to lipid droplets. The fluorescent sterol probe BODIPY-cholesterol was characterized as a tool to analyze cholesterol transport between the plasma membrane, the endoplasmic reticulum (ER) and lipid droplets. The behavior of BODIPY-cholesterol was compared to that of natural sterols, using both biochemical and live-cell microcopy assays. The results show that the transport kinetics of BODIPY-cholesterol between the plasma membrane, the ER and lipid droplets is similar to that of unesterified cholesterol. Next, BODIPY-cholesterol was utilized to analyze the importance of oxysterol binding protein related proteins (ORPs) for cholesterol transport between the plasma membrane, the ER, and lipid droplets in mammalian cells. By overexpressing all human ORPs it turned out that especially ORP1S and ORP2 enhanced sterol transport from the plasma membrane to lipid droplets. Our results suggest that the increased sterol transport takes place between the plasma membrane and ER and not between the ER and lipid droplets. Simultaneous knockdown of ORP1S and ORP2 resulted in a moderate but significant inhibition of sterol traffic from the plasma membrane to ER and lipid droplets, suggesting a physiological role for these ORPs in this process. The two phenylalanines in an acidic tract (FFAT) motif in ORPs, which mediates interaction with vesicle associated membrane protein associated proteins (VAPs) in the ER, was not necessary for mediating sterol transport. However, VAP silencing slowed down sterol transport, most likely by destabilizing ORPs containing a FFAT motif.

Hantaviruses have a tri-segmented negative-stranded RNA genome. The S segment encodes the nucleocapsid protein (N), M segment two glycoproteins, Gn and Gc, and the L segment the RNA polymerase. Gn and Gc are co-translationally cleaved from a precursor and targeted to the cis-Golgi compartment. The Gn glycoprotein consists of an external domain, a transmembrane domain and a C-terminal cytoplasmic domain. In addition, the S segment of some hantaviruses, including Tula and Puumala virus, have an open reading frame (ORF) encoding a nonstructural potein NSs that can function as a weak interferon antagonist. The mechanisms of hantavirus-induced pathogenesis are not fully understood but it is known that both hemorrhagic fever with renal syndrome (HFRS) and hantavirus (cardio) pulmonary syndrome (HCPS) share various features such as increased capillary permeability, thrombocytopenia and upregulation of TNF-. Several hantaviruses have been reported to induce programmed cell death (apoptosis), such as TULV-infected Vero E6 cells which is known to be defective in interferon signaling. Recently reports describing properties of the hantavirus Gn cytoplasmic tail (Gn-CT) have appeared. The Gn-CT of hantaviruses contains animmunoreceptor tyrosine-based activation motif (ITAM) which directs receptor signaling in immune and endothelial cells; and contain highly conserved classical zinc finger domains which may have a role in the interaction with N protein. More functions of Gn protein have been discovered, but much still remains unknown. Our aim was to study the functions of Gn protein from several aspects: synthesis, degradation and interaction with N protein. Gn protein was reported to inhibit interferon induction and amplication. For this reason, we also carried out projects studying the mechanisms of IFN induction and evasion by hantavirus. We first showed degradation and aggresome formation of the Gn-CT of the apathogenic TULV. It was reported earlier that the degradation of Gn-CT is related to the pathogenicity of hantavirus. We found that the Gn-CT of the apathogenic hantaviruses (TULV, Prospect Hill virus) was degraded through the ubiquitin-proteasome pathway, and TULV Gn-CT formed aggresomes upon treatment with proteasomal inhibitor. Thus the results suggest that degradation and aggregation of the Gn-CT may be a general property of most hantaviruses, unrelated to pathogenicity. Second, we investigated the interaction of TULV N protein and the TULV Gn-CT. The Gn protein is located on the Golgi membrane and its interaction with N protein has been thought to determine the cargo of the hantaviral ribonucleoprotein which is an important step in virus assembly, but direct evidence has not been reported. We found that TULV Gn-CT fused with GST tag expressed in bacteria can pull-down the N protein expressed in mammalian cells; a mutagenesis assay was carried out, in which we found that the zinc finger motif in Gn-CT and RNA-binding motif in N protein are indispensable for the interaction. For the study of mechanisms of IFN induction and evasion by Old World hantavirus, we found that Old World hantaviruses do not produce detectable amounts of dsRNA in infected cells and the 5 -termini of their genomic RNAs are monophosphorylated. DsRNA and tri-phosphorylated RNA are considered to be critical activators of innate immnity response by interacting with PRRs (pattern recognition receptors). We examined systematically the 5´-termini of hantavirus genomic RNAs and the dsRNA production by different species of hantaviruses. We found that no detectable dsRNA was produced in cells infected by the two groups of the old world hantaviruses: Seoul, Dobrava, Saaremaa, Puumala and Tula. We also found that the genomic RNAs of these Old World hantaviruses carry 5´-monophosphate and are unable to trigger interferon induction. The antiviral response is mainly mediated by alpha/beta interferon. Recently the glycoproteins of the pathogenic hantaviruses Sin Nombre and New York-1 viruses were reported to regulate cellular interferon. We found that Gn-CT can inhibit the induction of IFN activation through Toll-like receptor (TLR) and retinoic acid-inducible gene I-like RNA helicases (RLH) pathway and that the inhibition target lies at the level of TANK-binding kinase 1 (TBK-1)/ IKK epislon complex and myeloid differentiation primary response gene (88) (MyD88) / interferon regulatory factor 7 (IRF-7) complex.

Coenzyme Q (ubiquinone), a fully substituted benzoquinone with polyprenyl side chain, participates in many cellular redox activities. Paradoxically it was discovered only in 1957, albeit being ubiquitous. It required a person, F. L. Crane, a place, Enzyme Institute, Madison, USA, and a time when D. E. Green was directing vigorous research on mitochondria. Located at the transition of 2-electron flavoproteins and 1-electron cytochrome carriers, it facilitates electron transfer through the elegant Q-cycle in mitochondria to reduce O-2 to H2O, and to H2O2, now a significant signal-transducing agent, as a minor activity in shunt pathway (animals) and alternative oxidase (plants). The ability to form Q-radical by losing an electron and a proton was ingeniously used by Mitchell to explain the formation of the proton gradient, considered the core of energy transduction, and also in acidification in vacuoles. Known to be a mobile membrane constituent (microsomes, plasma membrane and Golgi apparatus), allowing it to reach multiple sites, coenzyme Q is expected to have other activities. Coenzyme Q protects circulating lipoproteins being a better lipid antioxidant than even vitamin E. Binding to proteins such as QPS, QPN, QPC and uncoupling protein in mitochondria, QA and QB in the reaction centre in R. sphaeroides, and disulfide bond-forming protein in E. coli (possibly also in Golgi), coenzyme Q acquires selective functions. A characteristic of orally dosed coenzyme Q is its exclusive absorption into the liver, but not the other tissues. This enrichment of Q is accompanied by significant decrease of blood pressure and of serum cholesterol. Inhibition of formation of mevalonate, the common precursor in the branched isoprene pathway, by the minor product, coenzyme Q, decreases the major product, cholesterol. Relaxation of contracted arterial smooth muscle by a side-chain truncated product of coenzyme Q explains its effect of decreasing blood pressure. Extensive clinical studies carried out on oral supplements of coenzyine Q, initially by K. Folkers and Y. Yamamura and followed many others, revealed a large number of beneficial effects, significantly in cardiovascular diseases. Such a variety of effects by this lipid quinone cannot depend on redox activity alone. The fat-soluble vitamins (A, D, E and K) that bear structural relationship with coenzyme Q are known to be active in their polar forms. A vignette of modified forms of coenzyme Q taking active role in its multiple effects is emerging.

Boswellia papyrifera and Boswellia carterii, known as Arabian incense, diffuses smoke, contaminating the air, which adversely affects human health. Therefore, this study was designed to ascertain the effect of these plants on histopathological and ultrastructure changes in cauda epididymis of Albino rats. Animals were exposed to 4 g/kg body weight of B. papyrifera and B. carterii daily for 120 days along with suitable controls. Our study indicates a significant reduction in epithelial heights. Cells showed signs of degeneration. The ultrastructural study revealed that the cauda epididymis was affected, including its cell types. Furthermore, a decrease in the size of mitochondria, Golgi complex, and both ERs was observed. In all treated groups, plasma fructose decreased considerably, indicating the sign of reduced energy, vital for motility and other sperm functions. The results of this study suggest that these plants systematically affect cauda epididymal cell types and its lumen through its potential toxicity. Hermansky-Pudlak syndrome (HPS) is a group of disorders characterized by the malformation of lysosome-related organelles, such as pigment cell melanosomes. Three of nine characterized HPS subtypes result from mutations in subunits of BLOC-2, a protein complex with no known molecular function. In this paper, we exploit melanocytes from mouse HPS models to place BLOC-2 within a cargo transport pathway from recycling endosomal domains to maturing melanosomes. In BLOC-2-deficient melanocytes, the melanosomal protein TYRP1 was largely depleted from pigment granules and underwent accelerated recycling from endosomes to the plasma membrane and to the Golgi. By live-cell imaging, recycling endosomal tubules of wild-type melanocytes made frequent and prolonged contacts with maturing melanosomes; in contrast, tubules from BLOC-2-deficient cells were shorter in length and made fewer, more transient contacts with melanosomes. These results support a model in which BLOC-2 functions to direct recycling endosomal tubular transport intermediates to maturing melanosomes and thereby promote cargo delivery and optimal pigmentation.

The coupling of endocytosis and exocytosis underlies fundamental biological processes ranging from fertilization to neuronal activity and cellular polarity. However, the mechanisms governing the spatial organization of endocytosis and exocytosis require clarification. Using a quantitative imaging-based screen in budding yeast, we identified 89 mutants displaying defects in the localization of either one or both pathways. High-resolution single-vesicle tracking revealed that the endocytic and exocytic mutants she4 Delta and bud6 Delta alter post-Golgi vesicle dynamics in opposite ways. The endocytic and exocytic pathways display strong interdependence during polarity establishment while being more independent during polarity maintenance. Systems analysis identified the exocyst complex as a key network hub, rich in genetic interactions with endocytic and exocytic components. Exocyst mutants displayed altered endocytic and post-Golgi vesicle dynamics and interspersed endocytic and exocytic domains compared with control cells. These data are consistent with an important role for the exocyst in coordinating endocytosis and exocytosis.

El presente trabajo se realizó en el periodo comprendido entre Noviembre y Diciembre de 1989, con el objetivo de investigar la infestación de parásitos gastrointestinales y pulmonares de mayor importancia económica en los bovinos del complejo 1 y 2 de la empresa genética Agenor Gómez. Boaco, Nicaragua. Se muestrearon 242 animales, entre bovinos de la raza Aberdeen Angus y Brahman, distribuidos en las categorías siguientes: C1 (0-1 año), C2 (1-3 años) y C3 (>3 años). Las muestras fueron extraídas directamente del recto animal, luego refrigeradas a 5°C y fueron analizadas en laboratorio de parasitología del MIDINRA, localizado en municipio de Camoapa V región. La cantidad de huevos y larvas se registró para cada género empleando las técnicas de sedimentación en agua, flotación con solución salina saturada y el método de larvoscopia e identificación con fotografías. Al realizar el ANDEVA por género de parásitos reportados en el análisis coprológico para la variable N° de huevos y larvas/gr de heces, se encontró que existen diferencias significativas con P>0.05 entre los promedios observados de los diferentes hatos. Los niveles de infestación promedio en los distintos hatos oscilaron entre 0u y 2lu; donde u= número de huevos y larvas/gramos de heces. De estos 48 promedios (8 hatos por 6 géneros de parásitos), 6 alcanzaron valores >9; 1 hato C1 de la upe Las Brisas con media de 2lu del género Haemonchus, 1 hato e de la upe San Miguel con media de 20.87u del género Strongyloides, 1 hato C2 de la upe San Jorge con media de 17.69u del género Strongyloides, 2 hatos C1 de las upe las Brisas y San Miguel con medias de 10.67u y medias de 10.5u respectivamente, ambos del género Coccidia y 1 hato e2 de la upe San Jorge con media de 9.6lu del género Coccidia. En los promedios restantes se obtuvieron valores <6u perteneciendo a la calificación leve. Posteriormente se realizó un análisis de correlación para determinar el grado de asociación existente entre cada uno de los géneros de parásitos económicamente importantes, encontrándose que el género Coccidia presentó el mayor grado de correlación con respecto a los demás géneros encontrados, se encontró que las correlaciones >= 0.25 corresponden a los géneros Coccidia, Moniezia, Neoascaris, Strongyloides y Haemonchus con r de 0.25, 0.25, 0.48, 0.30 respectivamente, similar para Haemonchus y Moniezia con r de 0.289.

As one of the most abundant polysaccharides on Earth, xylan will provide more than a third of the sugars for lignocellulosic biofuel production when using grass or hardwood feedstocks. Xylan is characterized by a linear β(1,4)-linked backbone of xylosyl residues substituted by glucuronic acid, 4-O-methylglucuronic acid or arabinose, depending on plant species and cell types. The biological role of these decorations is unclear, but they have a major influence on the properties of the polysaccharide. Despite the recent isolation of several mutants with reduced backbone, the mechanisms of xylan synthesis and substitution are unclear. We identified two Golgi-localized putative glycosyltransferases, GlucUronic acid substitution of Xylan (GUX)-1 and GUX2 that are required for the addition of both glucuronic acid and 4-O-methylglucuronic acid branches to xylan in Arabidopsis stem cell walls. The gux1 gux2 double mutants show loss of xylan glucuronyltransferase activity and lack almost all detectable xylan substitution. Unexpectedly, they show no change in xylan backbone quantity, indicating that backbone synthesis and substitution can be uncoupled. Although the stems are weakened, the xylem vessels are not collapsed, and the plants grow to normal size. The xylan in these plants shows improved extractability from the cell wall, is composed of a single monosaccharide, and requires fewer enzymes for complete hydrolysis. These findings have implications for our understanding of the synthesis and function of xylan in plants. The results also demonstrate the potential for manipulating and simplifying the structure of xylan to improve the properties of lignocellulose for bioenergy and other uses.

Con el objetivo de determinar como influyen los niveles de tecnologia empleado por los productores en el manejo agronomico del cultivo de guineo cuadrado y por ende en su rentabilidad. Se desarrolló en el período correspondiente del 15 de Septiembre al 15 de Diciembre de 1992 el estudio "DIAGNOSTICO AGROSOCIOECONOM1CO DEL CULTIVO DE GUINEO CUADRADO", Dicho estudio abarco principalmente el Departamento de Rivas por ser éste el que mayor áreas de siembra destina para la produccion de éste Rubro en la IV Región de pais. Para la realización del estudio, se tomaron datos de campo, muestras de suelo para el análisis fisico - quimico, así como la aplicación de encuestas a los productores, Se hizo análisis del manejo agronomico del cultivo y análisis económico a cada productor en estudio de acuerdo al nivel de tecnologia estudiado. Como resultado del estudio se observó que la rentabilidad del cultivo, considerando los productores en estudio tiene un 60% de rentabilidad, presentándose los mayores indices de rentabilidad en aquellos productores que utilizan el nivel de tecnologia media. Cabe mencionar que el 40% restante de los productores obtuvieron malos resultados, obteniendo algunos rentabilidades negativas. La rentabilidad se vio afectada directamente por los bajos rendimientos debidos principalmente a un mal manejo agronomico, que se refleja en la falta de adopción de técnicas mejoradas, para el manejo del cultivo, sumado a ello la falta de interés institucional y los problemas de comercialización. Todos estos factores en su conjunto influyen negativamente en la rentabilidad del cultivo de guineo cuadrado en la Cuarta Región del país.

El presente trabajo se desarrolló con el objetivo de generar información agronómica a través de la caracterización y evaluación preliminar de 24 líneas de frijol común (Phaseolus vulgaris L.) en sus diferentes estados fenológicos. El experimento fue establecido en la Estación Experimental “La Compañía”, ubicada en el Departamento de Carazo. El diseño experimental ut ilizado fue bloques completos al azar (BCA) con 24 tratamientos, un testigo (variedad INTA ROJO) y 3 réplicas. Para la caracterización se tomaron 28 caracteres cualitativos y 9 caracteres cuantitativos. Las variables evaluadas fueron días a la floración, días a la madurez fisiológica, días a cosecha, número de vainas por planta, número de semillas por vaina, número de plantas cosechadas, peso de 100 semillas, rendimiento (kg ha-1) y reacción a enfermedades. El tamaño de la muestra fue de 15 plantas dentro de la parcela útil, a excepción de las variables fenológicas, rendimiento (kg ha-1), reacción a enfermedad y peso de 100 semillas. El análisis estadístico hecho para las variables fue el ANDEVA, separación de medias por Tukey al 95% de confianza y análisis descriptivo para los 9 caracteres cuantitativos de la caracterización. Las variables fenológicas resultaron con una alta significancia, demostrando un comportamiento de precoz a intermedio, cumpliendo su ciclo en un rango de 65 a 72 días; los componentes de rendimiento fueron de significativo a altamente significativo para el número de semillas por vaina y peso de 100 semillas, respectivamente; no así para el número de vainas por planta y número plantas cosechadas, que resultaron no significativos. El rendimiento (kg ha-1) resultó significativo, con promedios que fueron de 1307 a 2264 kg ha-1. La separación de medias por Tukey demostró que ninguno de los materiales supera estadísticamente al testigo en casi todas las variables, a excepción del peso de 100 semillas. En la evaluación de la enfermedad Mustia hilachosa (Thanatephorus cucumeris); de las líneas evaluadas resultaron 3 resistentes, 20 intermedias y 1 susceptible.

Resumen: En los últimos cuatro años la Argentina creció 39% aproximadamente. Varios factores se conjugaron para permitir este crecimiento, pero sin duda, uno clave fue la existencia de una importante capacidad productiva ociosa, algo que se está diluyendo. En este contexto es importante conocer cuánto margen hay para crecer en el corto plazo, o sea, cuánto es el output gap o brecha del producto, y a qué ritmo se puede crecer en el largo plazo, o sea, la tasa de crecimiento del PBI potencial. Para ello en el presente artículo se calcula el PBI potencial en base al “método de la función de producción”. La principal conclusión es que para crecer a 4% en el largo plazo, una tasa mucho menor a la registrada recientemente, se requiere un esfuerzo inversor mayor al actual, tanto en cantidad como en calidad.
