Response to mineral fertilization and inoculation with rhizobia and/or arbuscular mycorrhiza fungi (AMF) of the Anadenanthera colubrina, Mimosa bimucronata and Parapiptadenia rigida (Leguminosae-Mimosoideae) native trees from Brazilian riparian forests, were studied in nursery conditions. Each species was submitted to seven treatments, varying nitrogen and phosphorous fertilization and inoculation with rhizobia (r), mycorrhiza (m) or both (rm): NP, P, P + r, P + rm, N, N + m and N + rm. Results showed that AMF inoculations did not enhance the mycorrhizal colonization, and P uptake was not sufficient to sustain good growth of plants. The level of P mineral added affected negatively the AMF colonization in A. colubrina and M. bimucronata, but not in P. rigida. Native fungi infected the three legume hosts. The absence of mineral N limited growth of A. colubrina and P. rigida, but in M. bimucronata the lack of N was corrected by biological nitrogen fixation. N mineral added inhibited the nodulation, although spontaneous nodulation had occurred in A. colubrina and M. bimucronata. Rhizobia inoculation enhanced the number of nodules, nitrogenase activity and leghemoglobin content of these two species. Thus, the extent of rhizobial and mycorrhizal symbiosis in these species under nursery conditions can affect growth and consequently the post-planting success. (C) 2004 Elsevier B.V. Lotus tetragonolobus lectin (LTA) is a fucose-specific legume lectin. Although several studies report a diverse combination of biological activities for LTA, little is known about the mechanisms involved in L-fucosyl oligosaccharide recognition. The crystal structure of LTA at 2.0 angstrom resolution reveals a different legume lectin tetramer. Its structure consists of a homotetramer composed of two back-to-back GS4-like dimers arranged in a new mode, resulting in a novel tetramer. The LTA N-linked carbohydrate at Asn4 and the unusual LTA dimer-dimer interaction are related to its particular mode of tetramerization. In addition, we used small angle X-ray scattering to investigate the quaternary structure of LTA in solution and to compare it to the crystalline structure. Although the crystal structure of LTA has revealed a conserved metal-binding site, its L-fucose-binding site presents some punctual differences. Our investigation of the new tetramer of LTA and its fucose-binding site is essential for further studies related to cross-linking between LTA and complex divalent L-fucosyl carbohydrates. O experimento foi conduzido em um solo Latossolo Vermelho Escuro álico, textura média, na Fazenda Experimental da UNESP - Campus de Ilha Solteira, SP. O trabalho teve como objetivo estudar o comportamento, da Galactia striata(Jacq.) Urb., quanto ao aspecto do valor nutritivo, em duas épocas de semeadura (28/09/79 e 25/03/80) e épocas de coleta (de 28 em 28 dias após emergência das plantas). O delineamento experimental foi em blocos casualizados com parcelas subdivididas, considerando as épocas de semeadura, as parcelas,e as épocas de coleta , as subparcelas. A aplicação de calcário se processou 30 dias antes de cada época de semeadura e a adubação fundamental na semeadura consistiu na aplicação de 20 kg/ha de nitrogênio na forma de sulfato de amônio(21% N), 120 kg de P2O5 na forma de cloreto de potássio (49,8% K). As semeaduras foram realizadas em linhas espaçadas de 0,30 m, com dez linhas de 5 m por subparcela, a uma profundidade de 2,5 cm, sendo deixada após o desbaste 10-15 plantas por metro linear. No material coletado separaram-se as folhas de hastes e efetuou-se a análise de digestibilidade in vitro da matéria seca. Conclui-se que: em função do decréscimo da digestibilidade in vitro da matéria seca por ser lento com o desenvolvimento vegetativo, a época de corte da Galactia striata pode ser determinada em função da produção de matéria seca. A Galactia striata é capaz de prover forragem de alto valor nutritivo, tanto no período de verão corno no de inverno.

A mucuna-preta é uma leguminosa empregada para adubação verde e como forrageira, cujas sementes apresentam dormência causada pela impermeabilidade do tegumento à água. O objetivo deste trabalho foi estudar a intensidade de dormência das sementes de mucuna-preta em diferentes estádios de maturação quando submetidas à secagem no interior das vagens. Rácemos foram colhidos, semanalmente, a partir de 35 dias do início de florescimento (35 DAF) até o estádio de vagens secas (98 DAF). As sementes de cada colheita foram secadas no interior das vagens, em condições ambientais não controladas de laboratório; quando secas foram extraídas, avaliadas quando a massa de 100 sementes, espessura das sementes e submetidas ao teste de germinação. Foram analisadas as porcentagens de sementes duras, de germinação, de embebição e o índice de velocidade de embebição. Pode-se concluir que o estádio de maturação em que ocorre a secagem afeta a intensidade de dormência das sementes de mucuna-preta.

Plant lectins, especially those purified from species of the Legummosae family, represent the best studied group of carbohydrate-binding proteins. The legume lectins from Diocleinae subtribe are highly similar proteins that present significant differences in the potency/ efficacy of their biological activities. The structural studies of the interactions between lectins and sugars may clarify the origin of the distinct biological activities observed in this high similar class of proteins. In this way, this work presents a crystallographic study of the ConM and CGL (agglutinins from Canavalia maritima and Canavalia gladiata, respectively) in the following complexes: ConM/ CGL:Man(alpha 1-2)Man(alpha 1-0)Me, ConM/CGL:Man(alpha 1-O)Man(alpha 1-O)Me and ConM/CGL:Man(alpha 1-4)Man(alpha 1-O)Me, which crystallized in different conditions and space group from the native proteins.The structures were solved by molecular replacement, presenting satisfactory values for R-factor and R-factor. Comparisons between ConM, CGL and ConA (Canavalia ensiformis lectin) binding mode with the dimannosides in subject, presented different interactions patterns, which may account for a structural explanation of the distincts biological properties observed in the lectins of Diocleinae subtribe. The present study aimed at determining the influence of condensed tannins present in the Brazilian legume species Mimosa hostilis, Mimosa caesalpinifolia and Bauhinia cheilantha on ruminal degradability, microbial colonization and enzymatic activity. Polyethylene glycol (PEG) was used to reduce the astringency and concentration of soluble condensed tannins. Four ruminally-cannulated Saanen goats (60 +/- 8 kg BW) were fed, in two experimental periods, with a hay diet based on the studied legumes treated or non-treated with PEG. Voluntary intake, microbial colonization, DM, CP, NDF, and ruminal degradability of PEG treated and non-treated forage leaves, as well as pH, ammonia and 1,4 P-endoglucanase activity of the rumen content were evaluated. Astringency and soluble tannin concentration of the studied legumes were reduced by approximately 70% and 50%, respectively, with PEG treatment. Average DM intake was higher for the treated diet (16.76 g DM/kg BW/day against 13.06 g DM/kg BW/day). Percentile values for degradation parameters and for potential and effective degradabilities of DM, CP and NDF were also affected by the tannins, but at different intensities. Electron microscopic observations of ruminally-incubated legume leaves showed a more effective microbial colonization of PEG-treated leaves for all legume species. A decrease in pH and an increase in ammonia concentration and in endoglucanase activity in the ruminal content was also observed for PEG-treated diets at all sampling periods. Condensed tannins of the studied legume species have influenced the adhesion conditions, colonization and enzymatic activity of the microbial ecosystem, and consequently the ruminal degradation of the different dietary fractions. For this reason, the reduction in condensed tannin would be of great importance to improve the nutrition of ruminant feeding of these species. (c) 2005 Elsevier B.V. All rights reserved.

This work was conducted to evaluate food intake and digestive efficiency of temperate wool and tropic semi-arid hair lambs, according to different concentrate: forage ratios in diet. Twenty-four lambs, averaging 90 +/- 1.8 days old and a mean body weight of 20 +/- 0.69 kg, 12 of them wool lambs, F, from Ideal x Ile de France crossing, and 12 others pure Santa Ines hair lambs, were distributed into a four replication 3 x 2 factorial arrangement consisting of three diets and two genotypes. Experimental diets consisted of: D1 = 60% concentrate mix (C) and 40% Cynodon sp. cv. Tifton-85 hay (F), D2 = 40% C and 60% F, and D3 = 20% C and 80% F. D1 was formulated for a daily gain of 300g per animal. Increasing forage levels in diets resulted in linear reductions (P < 0.01) in DM, OM, CP, TCH and metabolizable energy (ME) intake, and a linear increase (P < 0.01) in NDF ingestion. Tropic semi-arid hair lambs had higher DM, OM, NDF, CP, and TCH intake than temperate wool lambs. Although there were no genotype effects in OM and GE coefficient of digestibility, hair lambs showed more efficient (P < 0.05) digestibility of DM, CP, NDF and TCH. Increases in forage levels of diets corresponded to a negative linear effect (P < 0.01) in the apparent digestibility of DM, OM, CP, TCH and GE, while apparent digestibility of NDF increased linearly (P < 0.01). Total endogenous nitrogen (fecal plus urinary N) for F(1) Ideal x Ilede France wool and Santa Ines hair lambs were, respectively, 182 and 312 mg/kg(0.75) per day. Thus, Santa Ines tropic semi-arid hair lambs showed to be more responsive than F(1) Ideal x Ile de France temperate wool lambs to low quality fibrous diets. (C) 2004 Elsevier B.V. Using simulated ceramic refuse chambers, field decomposition studies were performed on the spent fungal refuse of the lead-cutting ant Atta sexdens rubropilosa. Refuse half life was estimated at 40 days, with complete decomposition at 100 days. These results suggest that the conversion-factor method used to estimate forage input into leaf-cutting ant colonies must be corrected for decomposition, or serious estimation errors will occur.

Forage plants, particularly the Brachiaria genus, are the main source of nutrients for cattle and are at times the only feed offered. The concentration of elements in the plant is related to the soil, fertilization, climate, season, variety, and cultural practices. An experiment on dystrophic Red-Yellow Latosol soil in Aracatuba, São Paulo was performed to evaluate the effects of the doses and sources of nitrogen fertilizers on the chemical properties of the soil and the dry matter yield of the grass Brachiaria brizantha cv. Xaraes. A randomized block design was employed involving three replicates in a 3 x 3 factorial, with three doses (100, 200 and 400 kg ha(-1) year(-1)) and three sources (Ajifer (R) L40, ammonium sulfate and urea) of nitrogen and a control treatment without nitrogen (zero). The greatest effects on the chemical properties of the soil as a function of nitrogen fertilization in the Xaraes grass were observed in the topsoil. The use of Ajifer (R) L40 and ammonium sulfate as sources of nitrogen had similar effects, with an increase in the sulfur content and a reduction in the soil pH at the superficial layer. The use of the fertilizers Ajifer (R) L40, ammonium sulfate and urea did not affect the micronutrient contents, except for Fe and Mn, and did not alter the sodium concentration or electrical conductivity of the soil. The dry matter yield of Xaraes grass was similar for all three nitrogen sources.

Arachis pintoi is an alternative to forage production in the tropics. Its germplasm comprises more than 150 accessions, that could be used to improve it. Our objective was the isolation and characterization of microsatellite loci in A. pintoi to be used to molecular evaluation of this germplasm and of A. repens (section Caulorrhizae). Seven loci were analyzed using five accessions of A. repens and 20 accessions of A. pintoi. The high variation found makes clear the high potential of this marker in genetic studies in these species. The developed markers showed total transferability to A. repens.

This review present a discussion abouth the tannins. The tannins are compounds that have the ability to bind other macromolecules, reducing their availability to animal metabolism. This characteristic can be benefic or deleterious, depending on the tannin concentration degree in the plant and its structure. Positive effects are mainly related with a better use of the dietary protein and an increased efficiency of microbial protein synthesis in the rumen. The use of tannins to reduce ruminal methane emission has been subject of current researches, firmly indicating a decrease of ruminal methanogenesis. However, many of these studies, carried out to evaluate the effects of tannins on forage fermentation and animal metabolism, are based on legume plants, so their results can not be surpassed to studies concerning tanniniferous grasses. By improving the characterization of the profile and reactivity of tannins in tanniniferous forages, such as some sorghum hybrids, as well as conducting experiments to demonstrate the effects of tannins on animal metabolism and performance, the real potential of forages with tannin will be enlightened.
