A new conglomerate family sample of 194 dwellings with 996 resident persons were studied in the town of Barcelos, State of Amazonas, in order to re-evaluate the risk of Chagas disease. During the survey the persons were interviewed and in this occasion we showed to them a collection of Panstrongylus, Rhodnius and Triatoma, asking if they recognized and eventually have been bitten by this kind of bugs. At this time we collected 500 ul of blood in microtainer® tubes from 886 interviewed persons who gave permission after informed consent. A screening test for T. cruzi antibodies based on agglutination of colored polymer particles, sensitized with three different synthetic peptides of T. cruzi (ID-PaGIA Chagas Test)®, showed 13.2% of sera positivity, but only 6.8% were confirmed by indirect immunofluorescence, and ELISA with purified T. cruzi antigens. Two hundred and six interviewed persons (20.7%) recognized the triatomines, as "piaçavas' lice" and 62 (30%) confirmed that have been bitten by the bugs, 25.8% of them had a positive serology for T. cruzi infection. Electrocardiographic alterations were shown in 9.3% of the seropositives and in 11.9% of the seronegative cases. This was considered not statistically significant.

O Estágio Pedagógico (EP) é um dos momentos com mais significado para a formação inicial dos professores. O objetivo deste relatório foi descrever e refletir as estratégias pedagógicas utilizadas nas quatro áreas de intervenção das linhas programáticas do EP: a prática letiva, as atividades de integração no meio, as atividades de intervenção na comunidade escolar e as atividades de natureza científico-pedagógica. O nosso EP foi realizado na Escola Básica dos 2º e 3º Ciclos Dr. Eduardo Brazão de Castro durante o ano letivo de 2014/2015. O núcleo de estágio foi constituído por duas estagiárias. A prática letiva foi realizada em duas turmas da escola, uma do 9º ano do ensino regular e uma do 12º ano do ensino profissional (turma partilhada). Inclui-se ainda neste ponto a conceptualização de um instrumento de observação e a assistência a aulas. As atividades de integração no meio foram compostas pelas atividades de apoio à direção de turma: (1) caraterização da turma, e (2) estudo caso. Um conhecimento aprofundado da turma permite aos professores delinear de uma forma mais adequadas as atividades pedagógicas ao longo do ano letivo. Estas atividades culminaram com a ação de extensão curricular, onde se promoveu a aproximação entre a escola e os encarregados de educação. Relativamente à atividade de intervenção na comunidade escolar, dirigida para toda a comunidade educativa, teve como objetivo a promoção de atividade física na escola. Finalmente, as atividades de natureza científico-pedagógica tiveram como propósito refletir sobre o papel e a importância do Professor de Educação Física na promoção da atividade física utilizando o exemplo do Voleibol e do Atletismo (ação coletiva), e consciencializar os professores sobre as potencialidades e contributos do Voleibol num plano formativo/educativo, facultando ferramentas metodológicas (ação individual). O Estágio Pedagógico veio contribuir para a aquisição de uma nova visão sobre a prática profissional docente em Educação Física e um desenvolvimento da capacidade autocrítica sobre a prática profissional.

Chromoblastomycosis is a chronic human melanized fungi infection of the subcutaneous tissue caused by traumatic inoculation of a specific group of dematiaceous fungi through the skin, often found in barefooted agricultural workers, in tropical and subtropical climate countries. We report the case of a male patient presenting a slow-growing pruriginous lesion on the limbs for 20 years, mistreated over that time, which was diagnosed and successfully treated as chromoblastomycosis. Besides the prevalence of this disease, treatment is still a clinical challenge.

The present study aimed at standardizing a real-time quantitative polymerase chain reaction assay to evaluate the presence of GBV-C/HGV RNA. A "TaqMan" assay using primers and probe derived from the 5¢ NCR region was developed and validated. Two hundred and fifty-three plasma samples from HIV-infected women were tested for GBV-C viremia and antibody against the envelope protein 2. GBV-C RNA was detected in 22.5% of the patients whereas the antibody was identified in 25.3% of the cohort. Detection of viral RNA and of antibodies was mutually exclusive. Viral loads showed a mean of 1,777 arbitrary units / mL, being 1.1 and 13,625 arbitrary units / mL respectively the lowest and highest values measured. We conclude that the real-time quantitative polymerase chain reaction method developed is appropriate for the investigation of GBV-C RNA since it was shown to be highly specific and sensitive, as well as requiring few steps, preventing contamination and providing additional information as to the relative viremia of carriers, a parameter that must be included in studies evaluating the co-factors influencing the clinical outcome of HIV/AIDS.

This study examined the susceptibility of peritoneal macrophage (PM) from the Neotropical primates: Callithrix jacchus, Callithrix penicillata, Saimiri sciureus, Aotus azarae infulatus and Callimico goeldii to ex vivo Leishmania (L.) infantum chagasi-infection, the etiological agent of American visceral leishmaniasis (AVL), as a screening assay for evaluating the potential of these non-human primates as experimental models for studying AVL. The PM-susceptibility to infection was accessed by the PM-infection index (PMI) at 24, 72 h and by the mean of these rates (FPMI), as well as by the TNF-α, IL-12 (Capture ELISA) and Nitric oxide (NO) responses (Griess method). At 24h, the PMI of A. azarae infulatus (128) was higher than those of C. penicillata (83), C. goeldii (78), S. sciureus (77) and C. jacchus (55). At 72h, there was a significant PMI decrease in four monkeys: A. azarae infulatus (128/37), C. penicillata (83/38), S. sciureus (77/38) and C. jacchus (55/12), with exception of C. goeldii (78/54). The FPMI of A. azarae infulatus (82.5) and C. goeldii (66) were higher than C. jacchus (33.5), but not higher than those of C. penicillata (60.5) and S. sciureus (57.5). The TNF-a response was more regular in those four primates which decreased their PMI at 24/72 h: C. jacchus (145/122 pg/mL), C. penicillata (154/130 pg/mL), S. sciureus (164/104 pg/mL) and A. azarae infulatus (154/104 pg/mL), with exception of C. goeldii (38/83 pg/mL). The IL-12 response was mainly prominent in A. infulatus and C. goeldii which presented the highest FPMI and, the NO response was higher in C. goeldii, mainly at 72 h. These findings strongly suggest that these New World primates have developed a resistant innate immune response mechanism capable of controlling the macrophage intracellular growth of L. (L.) i. chagasi-infection, which do not encourage their use as animal model for studying AVL.

Bacteria of the genus Bartonella are emerging pathogens detected in lymph node biopsies and aspirates probably caused by increased concentration of bacteria. Twenty-three samples of 18 patients with clinical, laboratory and/or epidemiological data suggesting bartonellosis were subjected to three nested amplifications targeting a fragment of the 60-kDa heat shock protein (HSP), the internal transcribed spacer 16S-23S rRNA (ITS) and the cell division (FtsZ) of Bartonella henselae, in order to improve detection in clinical samples. In the first amplification 01, 04 and 05 samples, were positive by HSP (4.3%), FtsZ (17.4%) and ITS (21.7%), respectively. After the second round six positive samples were identified by nested-HSP (26%), eight by nested-ITS (34.8%) and 18 by nested-FtsZ (78.2%), corresponding to 10 peripheral blood samples, five lymph node biopsies, two skin biopsies and one lymph node aspirate. The nested-FtsZ was more sensitive than nested-HSP and nested-ITS (p < 0.0001), enabling the detection of Bartonella henselae DNA in 15 of 18 patients (83.3%). In this study, three nested-PCR that should be specific for Bartonella henselae amplification were developed, but only the nested-FtsZ did not amplify DNA from Bartonella quintana. We conclude that nested amplifications increased detection of B. henselae DNA, and that the nested-FtsZ was the most sensitive and the only specific to B. henselae in different biological samples. As all samples detected by nested-HSP and nested-ITS, were also by nested-FtsZ, we infer that in our series infections were caused by Bartonella henselae. The high number of positive blood samples draws attention to the use of this biological material in the investigation of bartonellosis, regardless of the immune status of patients. This fact is important in the case of critically ill patients and young children to avoid more invasive procedures such as lymph nodes biopsies and aspirates.

O autor estuda os dados obtidos pela infecção de moluscos de Campinas e de Belo Horizonte por miracídios de S. mansoni. Chega à conclusão de que a população de Belo Horizonte mostrou-se mais susceptível à infecção pelo helminto do que a população de moluscos Campinas.

Foi avaliado, comparativamente, o desempenho de fêmeas e machos de Triatoma infestans, em 5º estádio de desenvolvimento, em xenodiagnósticospara a tripanossomiase americana. Estes exames foram realizados em 40 pacientes, em fase crônica da doença de Chagas. Em cada exame utilizou-se 20 ninfas fêmeas e 20 ninfas machos, que foram pesadas imediatamente antes e após o repasto. O exame individual do conteúdo intestinal dos triatomíneos foi efetuado aos 20, 30, 60 e 90 dias após a aplicação dos xenodiagnósticos. As ninfas fêmeas e machos, ingeriram, em média, 230 e210 mg de sangue, respectivamente. Os resultados revelaram 15 (37,5%) xenodiagnósticos positivos; todavia, em nenhum destes exames foi verificada positivação, para T. cruzi, de todas as ninfas utilizadas. As ninfas fêmeas ingeriram significativamente mais sangue que as ninfas machos, isto vem de encontro com as observações de maior positividade, para T. cruzi entre as fêmeas, sugerindo, serem estas ninfas capazes de possibilitar maior incremento à sensibilidade do xenodiagnóstico na fase crônica da doença de Chagas, sendo necessários novos estudos para confirmação desta hipótese.
