We demonstrate that during inflammatory responses the nuclear factor kappa B (NF-kappa B) induces the synthesis of melatonin by macrophages and that macrophage-synthesized melatonin modulates the function of these professional phagocytes in an autocrine manner. Expression of a DsRed2 fluorescent reporter driven by regions of the aa-nat promoter, that encodes the key enzyme involved in melatonin synthesis (arylalkylamine-N-acetyltransferase), containing one or two upstream kappa B binding sites in RAW 264.7 macrophage cell lines was repressed when NF-kappa B activity was inhibited by blocking its nuclear translocation or its DNA binding activity or by silencing the transcription of the RelA or c-Rel NF-kappa B subunits. Therefore, transcription of aa-nat driven by NF-kappa B dimers containing RelA or c-Rel subunits mediates pathogen-associated molecular patterns (PAMPs) or pro-inflammatory cytokine-induced melatonin synthesis in macrophages. Furthermore, melatonin acts in an autocrine manner to potentiate macrophage phagocytic activity, whereas luzindole, a competitive antagonist of melatonin receptors, decreases macrophage phagocytic activity. The opposing functions of NF-kappa B in the modulation of AA-NAT expression in pinealocytes and macrophages may represent the key mechanism for the switch in the source of melatonin from the pineal gland to immune-competent cells during the development of an inflammatory response.

Background: Most investigations regarding the First Americans have primarily focused on four themes: when the New World was settled by humans; where they came from; how many migrations or colonization pulses from elsewhere were involved in the process; and what kinds of subsistence patterns and material culture they developed during the first millennia of colonization. Little is known, however, about the symbolic world of the first humans who settled the New World, because artistic manifestations either as rock-art, ornaments, and portable art objects dated to the Pleistocene/Holocene transition are exceedingly rare in the Americas. Methodology/Principal Findings: Here we report a pecked anthropomorphic figure engraved in the bedrock of Lapa do Santo, an archaeological site located in Central Brazil. The horizontal projection of the radiocarbon ages obtained at the north profile suggests a minimum age of 9,370640 BP, (cal BP 10,700 to 10,500) for the petroglyph that is further supported by optically stimulated luminescence (OSL) dates from sediment in the same stratigraphic unit, located between two ages from 11.7 +/- 0.8 ka BP to 9.9 +/- 0.7 ka BP. Conclusions: These data allow us to suggest that the anthropomorphic figure is the oldest reliably dated figurative petroglyph ever found in the New World, indicating that cultural variability during the Pleistocene/Holocene boundary in South America was not restricted to stone tools and subsistence, but also encompassed the symbolic dimension.

A two-stage bioreactor was operated for a period of 140 days in order to develop a post-treatment process based on anaerobic bioxidation of sulfite. This process was designed for simultaneously treating the effluent and biogas of a full-scale UASB reactor, containing significant concentrations of NH4 and H2S, respectively. The system comprised of two horizontal-flow bed-packed reactors operated with different oxygen concentrations. Ammonium present in the effluent was transformed into nitrates in the first aerobic stage. The second anaerobic stage combined the treatment of nitrates in the liquor with the hydrogen sulfide present in the UASB-reactor biogas. Nitrates were consumed with a significant production of sulfate, resulting in a nitrate removal rate of 0.43 kg N m(3) day(-1) and a parts per thousand yen92 % efficiency. Such a removal rate is comparable to those achieved by heterotrophic denitrifying systems. Polymeric forms of sulfur were not detected (elementary sulfur); sulfate was the main product of the sulfide-based denitrifying process. S-sulfate was produced at a rate of about 0.35 kg m(3) day(-1). Sulfur inputs as S-H2S were estimated at about 0.75 kg m(3) day(-1) and Chemical Oxygen Demand (COD) removal rates did not vary significantly during the process. DGGE profiling and 16S rRNA identified Halothiobacillus-like species as the key microorganism supporting this process; such a strain has not yet been previously associated with such bioengineered systems.

Abstract Background Assuming a higher risk of latent tuberculosis (TB) infection in the population of Rio de Janeiro, Brazil, in October of 1998 the TB Control Program of Clementino Fraga Filho Hospital (CFFH) routinely started to recommend a two-step tuberculin skin test (TST) in contacts of pulmonary TB cases in order to distinguish a boosting reaction due to a recall of delayed hypersensitivity previously established by infection with Mycobacterium tuberculosis (M.tb) or BCG vaccination from a tuberculin conversion. The aim of this study was to assess the prevalence of boosted tuberculin skin tests among contacts of individuals with active pulmonary tuberculosis (TB). Methods Retrospective cohort of TB contacts ≥ 12 years old who were evaluated between October 1st, 1998 and October 31st 2001. Contacts with an initial TST ≤ 4 mm were considered negative and had a second TST applied after 7–14 days. Boosting reaction was defined as a second TST ≥ 10 mm with an increase in induration ≥ 6 mm related to the first TST. All contacts with either a positive initial or repeat TST had a chest x-ray to rule out active TB disease, and initially positive contacts were offered isoniazid preventive therapy. Contacts that boosted did not receive treatment for latent TB infection and were followed for 24 months to monitor the development of TB. Statistical analysis of dichotomous variables was performed using Chi-square test. Differences were considered significant at a p < 0.05. Results Fifty four percent (572/1060) of contacts had an initial negative TST and 79% of them (455/572) had a second TST. Boosting was identified in 6% (28/455). The mean age of contacts with a boosting reaction was 42.3 ± 21.1 and with no boosting was 28.7 ± 21.7 (p = 0.01). Fifty percent (14/28) of individuals whose test boosted met criteria for TST conversion on the second TST (increase in induration ≥ 10 mm). None of the 28 contacts whose reaction boosted developed TB disease within two years following the TST. Conclusion The low number of contacts with boosting and the difficulty in distinguishing boosting from TST conversion in the second TST suggests that the strategy of two-step TST testing among contacts of active TB cases may not be useful. However, this conclusion must be taken with caution because of the small number of subjects followed.

Este trabalho teve como objetivo avaliar diferenças genéticas entre três grupos de matrizes de Cedrela fissilis a partir de variáveis quantitativas juvenis em teste de progênies, para delinear zonas de coleta e uso de sementes da espécie na região de estudo, bem como avaliar a variabilidade genética do material amostrado. Instalou-se um teste de progênies, em viveiro, a partir de sementes de 48 matrizes amostradas nos Municípios de Rio Negrinho, Mafra e São Bento do Sul, no Estado de Santa Catarina; e em Lapa, Rio Negro, Campo do Tenente e Antonio Olinto, no Estado do Paraná. Das matrizes coletadas, 33 encontravam-se distribuídas em três grupos espaciais e 15 dispersas na região. O delineamento em blocos casualizados com oito repetições e 20 plantas por parcela foi empregado. Os dados avaliados incluíram: índice de velocidade de emergência, diâmetro do colo e altura das mudas (aos 61, 102 e 145 dias após a semeadura); sobrevivência, número de folhas por muda, massa seca da parte aérea e da raiz e área foliar da terceira folha totalmente expandida a contar do ápice. A metodologia de máxima verossimilhança restrita foi utilizada para a análise estatística, com o auxílio do software SELEGEN. Verificou-se que os caracteres juvenis apresentam elevado controle genético, podendo ser utilizados para avaliação da variabilidade genética de amostras de populações da espécie. Os três grupos de matrizes delimitados espacialmente não apresentam diferenças genéticas significativas, sendo possível inferir que as três áreas pertencem a uma mesma zona de coleta e uso de sementes

Este artigo apresenta algumas reflexões em torno das formas e volumes das louças em faiança fina produzidas, e consumidas, na cidade de São Paulo, durante o período de 1913 e 1937, relacionando-as a alguns hábitos levados a cabo na Paulicéia, como o crescente costume do "cafezinho". Para tal, parte-se das análises do acervo gerado pelo resgate do sítio arqueológico Petybon, localizado na zona metropolitana da cidade, no bairro da Lapa, região da Água Branca/Vila Romana. Aponta-se que a diversidade de formas das louças dialoga com os projetos de modernidade pensados para São Paulo e as demandas dos consumidores cujas diversas práticas culturais influenciaram na produção das faianças finas pela Fábrica de Louças Santa Catharina e Indústrias Reunidas Fábricas Matarazzo.

OBJECTIVE: To compare the classical neurosurgical technique with a new simplified technique for prenatal repair of a myelomeningocelelike defect in sheep. METHODS: A myelomeningocele-like defect (laminectomy and dural excision) was created in the lumbar region on day 90 of gestation in 9 pregnant sheep. Correction technique was randomized. In Group 1 the defect was corrected using the classic neurosurgical technique of three-layer suture (dura mater, muscle and skin closure) performed by a neurosurgeon. In Group 2, a fetal medicine specialist used a biosynthetic cellulose patch to protect the spinal cord and only the skin was sutured above it. Near term (day 132 of gestation) fetuses were sacrificed for pathological analysis. RESULTS: There were two miscarriages and one maternal death. In total, six cases were available for pathological analysis, three in each group. In Group 1, there were adherence of the spinal cord to the scar (meningo-neural adhesion) and spinal cord architecture loss with posterior funiculus destruction and no visualization of grey matter. In Group 2, we observed in all cases formation of a neo-dura mater, separating the nervous tissue from adjacent muscles, and preserving the posterior funiculus and grey matter. CONCLUSION: The new simplified technique was better than the classic neurosurgical technique. It preserved the nervous tissue and prevented the adherence of the spinal cord to the scar. This suggests the current technique used for the correction of spina bifida in humans may need to be reassessed.

The umbilical cord is a structure that provides vascular flow between the fetus and the placenta. It contains two arteries and one vein, which are surrounded and supported by gelatinous tissue known as Wharton’s jelly. There are many umbilical cord abnormalities that are related to the prognosis of fetus survival and birth weight. The authors report a case of umbilical cord constriction due to the localized absence of Wharton’s jelly, which was undiagnosed antenatally and had a fatal outcome. A review of the association between the absence of Wharton’s jelly and an unfavorable pregnancy outcome was undertaken.

Abstract.- This study assessed the effects of human influences over the abundance and size patterns of five species of gastropods (top-shell snails and limpets) commonly collected in the Canarian Archipelago: ‘burgado hembra’ (Osilinus atrata), ‘burgado macho’ (Osilinus sauciatus), ‘lapa blanca’ (Patella aspera), ‘lapa negra’ (Patella candei crenata) and ‘lapa de sol’ (Patella rustica). We studied patterns of abundance and size of these species across three islands (Lanzarote, La Graciosa and Alegranza) corresponding to three levels of human influence: high, medium and low, respectively; which were quantified through three indicators: human pressure (inhabitants km-1 d-1), accessibility to the coast (km of sealed and unsealed roads), and volumes of capture (kg). The abundances of O. atrata and P. aspera were statistically higher at Alegranza than at La Graciosa and Lanzarote, whereas the density was 10 to 15 times larger at Alegranza for four of the five studied species. Significant differences in the size structure of the species among islands (= levels of human influence) were also observed. Not only all large-sized individuals disappeared at Lanzarote and La Graciosa, yet there was a decrease in numbers for the majority of size ranges. Thought populations of top-shell snails and limpets were affected by natural variability, human activities turned out to be the major driver for the observed differences among islands. These results seriously question the effectiveness of the current shellfish regulations contained in the Regional Fish Law. Resumen.- Este estudio determinó los efectos de la influencia humana sobre la abundancia y la estructura de talla de cinco especies de moluscos gasterópodos comúnmente recolectados en el Archipiélago Canario: el ‘burgado hembra’ (Osilinus atrata), el ‘burgado macho’ (Osilinus sauciatus), la ‘lapa blanca’ (Patella aspera), la ‘lapa negra’ (Patella candei crenata) y la ‘lapa de sol’ (Patella rustica). Se analizaron tres islas (Lanzarote, La Graciosa y Alegranza) correspondientes a tres categorías de influencia humana: alta, media y baja, respectivamente; cuantificadas a través de tres indicadores: la presión humana (hab km-1 d-1), la accesibilidad a la costa (km de red viaria) y los volúmenes de extracción de los recursos (kg). La abundancia de O. atrata y P. aspera fue estadísticamente mayor en Alegranza que en La Graciosa y Lanzarote, mientras que los valores de densidad fueron de 10 a 15 veces superiores en Alegranza para cuatro de las cinco especies estudiadas. Para la estructura de talla, se detectaron notables diferencias entre islas (= niveles de influencia humana). En Lanzarote y La Graciosa no sólo desaparecieron los ejemplares de mayor talla, sino que disminuyeron los ejemplares para la mayoría de rangos de talla. A pesar que la variabilidad natural de las poblaciones de burgados y lapas es alta, la actividad humana parece ser la causa principal de las diferencias observadas entre islas. Estos resultados cuestionan seriamente la efectividad de las normas sobre marisqueo de la actual Ley de Pesca de Canarias.

Por otra parte, en la segunda parte del libro, le interesa arribar, después de presentar el choque entre culturas, -la letrada representada por los españoles y la oral representada por los timbúes- a una síntesis de opuestos que rescate lo mejor de unos y otros; y en ese intento deja traslucir el proyecto ideológico al que adscribe. El juego de apropiaciones, proyecciones y entrecruzamientos que vinculan a Lucía Miranda con el romancero, las escenas de lectura y el Cantar del Mío Cid, permiten dar cuenta de una elección que no escapa a condicionamientos personales, familiares y de clase que tienen como marco la historia argentina y el horizonte de expectativas de la época

Two sheep and two goats, fitted with a ruminal cannula, received two diets composed of 30% concentrate and 70% of either alfalfa hay (AL) or grass hay (GR) as forage in a two-period crossover design. Solid and liquid phases of the rumen were sampled from each animal immediately before feeding and 4 h post-feeding. Pellets containing solid associated bacteria (SAB) and liquid associated bacteria (LAB) were isolated from the corresponding ruminal phase and composited by time to obtain 2 pellets per animal (one SAB and one LAB) before DNA extraction. Denaturing gradient gel electrophoresis (DGGE) analysis of 16S ribosomal DNA was used to analyze bacterial diversity. A total of 78 and 77 bands were detected in the DGGE gel from sheep and goats samples, respectively. There were 18 bands only found in the pellets from sheep fed AL-fed sheep and 7 found exclusively in samples from sheep fed the GR diet. In goats, 21 bands were found only in animals fed the AL diet and 17 were found exclusively in GR-fed ones. In all animals, feeding AL diet tended (P < 0.10) to promote greater NB and SI in LAB and SAB pellets compared with the GR diet. The dendrogram generated by the cluster analysis showed that in both animal species all samples can be included in two major clusters. The four SAB pellets within each animal species clustered together and the four LAB pellets grouped in a different cluster. Moreover, SAB and LAB clusters contained two clear subclusters according to forage type. Results show that in all animals bacterial diversity was more markedly affected by the ruminal phase (solid vs. liquid) than by the type of forage in the diet.

Los sistemas in vitro de simulación de la fermentación ruminal constituyen herramientas muy útiles para el estudio de la función ruminal. El mantenimiento de poblaciones microbianas representativas de las existentes en el rumen de los animales es uno de los requisitos que deberían cumplir los sistemas in vitro, pero la disminución del número de protozoos en los fermentadores es un hecho bien constatado (Moumen et al., 2009; Martínez et al., 2011). Dado el importante papel que los protozoos desempeñan en la función ruminal, el diseño de modificaciones técnicas que permitan mantener estas poblaciones en fermentadores supondría un importante avance para su utilización en el estudio del ecosistema ruminal. El objetivo del presente trabajo fue analizar dos modificaciones técnicas para mejorar la retención de los protozoos en fermentadores de flujo continuo.
