The stem cell niche organization and dynamics provide valuable cues for the development of mimetic environments that could have potential to stimulate the regenerative process. We propose the use of biodegradable biomaterials to produce closed miniaturised structures able to encapsulate different cell types or bioactive molecules. In particular, capsules are fabricated using the so-called layer-by-layer technology, where the consecutive (nano-sized) layers are well stabilized by electrostatic interactions or other weak forces. Using alginate-based spherical templates containing cells or other elements (e.g. proteins, magnetic nanoparticles, microparticles) it is possible to produce liquefied capsules that may entrap the entire cargo under mild conditions. The inclusion of liquefied micropcapsules may be used to produce hierarchical compartmentalised systems for the delivery of bioactive agents. The presence of solid microparticles inside such capsules offers adequate surface area for adherent cell attachment increasing the biological performance of these hierarchical systems, while maintain both permeability and injectability. We demonstrated that the encapsulation of distinct cell types (including mesenchymal stem cells and endothelial cells) enhances the osteogenic capability of this system, that could be useful in bone tissue engineering applications.
Superhydrophobic surfaces as a tool for the fabrication of hierarchical spherical polymeric carriers

Hierarchical polymeric carriers with high encapsulation efficiencies are fabricated via a biocompatible strategy developed using superhydrophobic (SH) surfaces. The carries are obtained by the incorporation of cell/BSA-loaded dextran-methacrylate (DEXT-MA) microparticles into alginate (ALG) macroscopic beads. Engineered devices like these are expected to boost the development of innovative and customizable systems for biomedical and biotechnological purposes.

During blood banking, erythrocytes undergo storage lesions, altering or degrading their metabolism, rheological properties, and protein content. Carbonylation is a hallmark of protein oxidative lesions, thus of red blood cell oxidative stress. In order to improve global erythrocyte protein carbonylation assessment, subcellular fractionation has been established, allowing us to work on four different protein populations, namely soluble hemoglobin, hemoglobin-depleted soluble fraction, integral membrane and cytoskeleton membrane protein fractions. Carbonylation in erythrocyte-derived microparticles has also been investigated. Carbonylated proteins were derivatized with 2,4-dinitrophenylhydrazine (2,4-DNPH) and quantified by western blot analyses. In particular, carbonylation in the cytoskeletal membrane fraction increased remarkably between day 29 and day 43 (P<0.01). Moreover, protein carbonylation within microparticles released during storage showed a two-fold increase along the storage period (P<0.01). As a result, carbonylation of cytoplasmic and membrane protein fractions differs along storage, and the present study allows explaining two distinct steps in global erythrocyte protein carbonylation evolution during blood banking. We investigated the use of in situ implant formation that incorporates superparamagnetic iron oxide nanoparticles (SPIONs) as a form of minimally invasive treatment of cancer lesions by magnetically induced local hyperthermia. We developed injectable formulations that form gels entrapping magnetic particles into a tumor. We used SPIONs embedded in silica microparticles to favor syringeability and incorporated the highest proportion possible to allow large heating capacities. Hydrogel, single-solvent organogel and cosolvent (low-toxicity hydrophilic solvent) organogel formulations were injected into human cancer tumors xenografted in mice. The thermoreversible hydrogels (poloxamer, chitosan), which accommodated 20% w/v of the magnetic microparticles, proved to be inadequate. Alginate hydrogels, however, incorporated 10% w/v of the magnetic microparticles, and the external gelation led to strong implants localizing to the tumor periphery, whereas internal gelation failed in situ. The organogel formulations, which consisted of precipitating polymers dissolved in single organic solvents, displayed various microstructures. A 8% poly(ethylene-vinyl alcohol) in DMSO containing 40% w/v of magnetic microparticles formed the most suitable implants in terms of tumor casting and heat delivery. Importantly, it is of great clinical interest to develop cosolvent formulations with up to 20% w/v of magnetic microparticles that show reduced toxicity and centered tumor implantation.

Although exposure to secondhand smoke (SHS) is reportedly high in prison, few studies have measured this in the prison environment, and none have done so in Europe. We measured two indicators of SHS exposure (particulate matter PM10 and nicotine) in fixed locations before (2009) and after (2010) introduction of a partial smoking ban in a Swiss prison. Access to smoking cessation support was available to detainees throughout the study. Objectives To measure SHS before and after the introduction of a partial smoking ban. Methods Assessment of particulate matter PM10 (suspended microparticles of 10 μm) and nicotine in ambient air, collected by real-time aerosol monitor and nicotine monitoring devices. Results The authors observed a significant improvement of nicotine concentrations in the air after the introduction of the smoking ban (before: 7.0 μg/m(3), after: 2.1 μg/m(3), difference 4.9 μg/m(3), 95% CI for difference: 0.52 to 9.8, p=0.03) but not in particulate matter PM10 (before: 0.11 mg/m(3), after: 0.06 mg/m(3), difference 0.06 mg/m(3), 95% CI for difference of means: -0.07 to 0.19, p=0.30). Conclusions The partial smoking ban was followed by a decrease in nicotine concentrations in ambient air. These improvements can be attributed to the introduction of the smoking ban since no other policy change occurred during this period. Although this shows that concentrations of SHS decreased significantly, protection was still incomplete and further action is necessary to improve indoor air quality.

An increased plasma concentration of von Willebrand factor (vWF) is detected in individuals with many infectious diseases and is accepted as a marker of endothelium activation and prothrombotic condition. To determine whether ExoU, a Pseudomonas aeruginosa cytotoxin with proinflammatory activity, enhances the release of vWF, microvascular endothelial cells were infected with the ExoU-producing PA103 P. aeruginosa strain or an exoU-deficient mutant. Significantly increased vWF concentrations were detected in conditioned medium and subendothelial extracellular matrix from cultures infected with the wild-type bacteria, as determined by enzyme-linked immunoassays. PA103-infected cells also released higher concentrations of procoagulant microparticles containing increased amounts of membrane-associated vWF, as determined by flow cytometric analyses of cell culture supernatants. Both flow cytometry and confocal microscopy showed that increased amounts of vWF were associated with cytoplasmic membranes from cells infected with the ExoU-producing bacteria. PA103-infected cultures exposed to platelet suspensions exhibited increased percentages of cells with platelet adhesion. Because no modulation of the vWF mRNA levels was detected by reverse transcription-polymerase chain reaction assays in PA103-infected cells, ExoU is likely to have induced the release of vWF from cytoplasmic stores rather than vWF gene transcription. Such release is likely to modify the thromboresistance of microvascular endothelial cells.

Cette thèse a pour but de caractériser les microparticules isolées à partir des concentrés érythrocytaires et plus précisément de déterminer la présence d'antigènes de groupes sanguins à leur surface. Elle est divisée en trois parties, sous forme d'articles publiés à partir d'un travail de recherche mené au Centre de Transfusion Sanguine d'Epalinges. Dans l'article « Microparticles in stored red blood cells : an approach using flow cytometry and proteomic tools » publié dans Vox Sanguinis en 2008, il est question de la lésion de stockage des globules rouges. Grâce à des techniques alliant la cytométrie de flux et la protéomique, il a été montré que la génération de microparticules augmente au cours du stockage des concentrés érythrocytaires et que leur composition se modifie au cours du temps. L'article de revue « Analysis and clinical relevance of microparticles from red blood cells » publié dans Current Opinion in Hematology en 2010, explique les mécanismes de formation et d'élimination des microparticules de globules rouges. Il fait une revue des implications cliniques liées à la génération de microparticules et discute leur conséquences potentielles dans le domaine de la médecine transfusionnelle. L'article « Red blood cell microparticles and blood group antigens : an analysis by flow cytometry » publié dans Blood Transfusion en 2012, décrit l'étude des antigènes de groupe sanguins à la surface des microparticules générées à partir de concentrés érythrocytaires après ajout de calcium ionophore. Les résultats de cette étude indiquent que les antigènes de groupes sanguins appartenant aux systèmes RH, KEL, JK, FY, MNS, LE et LU sont présents à la surface des microparticules. Ces antigènes pourraient potentiellement être source d'allo-immunisation après transfusion.

Many currently used and candidate vaccine adjuvants are particulate in nature, but their mechanism of action is not well understood. Here, we show that particulate adjuvants, including biodegradable poly(lactide-co-glycolide) (PLG) and polystyrene microparticles, dramatically enhance secretion of interleukin-1beta (IL-1beta) by dendritic cells (DCs). The ability of particulates to promote IL-1beta secretion and caspase 1 activation required particle uptake by DCs and NALP3. Uptake of microparticles induced lysosomal damage, whereas particle-mediated enhancement of IL-1beta secretion required phagosomal acidification and the lysosomal cysteine protease cathepsin B, suggesting a role for lysosomal damage in inflammasome activation. Although the presence of a Toll-like receptor (TLR) agonist was required to induce IL-1beta production in vitro, injection of the adjuvants in the absence of TLR agonists induced IL-1beta production at the injection site, indicating that endogenous factors can synergize with particulates to promote inflammasome activation. The enhancement of antigen-specific antibody production by PLG microparticles was independent of NALP3. However, the ability of PLG microparticles to promote antigen-specific IL-6 production by T cells and the recruitment and activation of a population of CD11b(+)Gr1(-) cells required NALP3. Our data demonstrate that uptake of microparticulate adjuvants by DCs activates the NALP3 inflammasome, and this contributes to their enhancing effects on innate and antigen-specific cellular immunity.

Microparticles are phospholipid vesicles shed mostly in biological fluids, such as blood or urine, by various types of cells, such as red blood cells (RBCs), platelets, lymphocytes, endothelial cells. These microparticles contain a subset of the proteome of their parent cell, and their ready availability in biological fluid has raised strong interest in their study, as they might be markers of cell damage. However, their small size as well as their particular physico-chemical properties makes them hard to detect, size, count and study by proteome analysis. In this review, we report the pre-analytical and methodological caveats that we have faced in our own research about red blood cell microparticles in the context of transfusion science, as well as examples from the literature on the proteomics of various kinds of microparticles.

Sulfur in the soil occurs in two basic forms, organic and inorganic S. The organic form accounts for 95 % of S in most soils. The effectiveness of organic S to oxidate to sulfate was evaluated for total S determination in soil samples by wet (acid) and dry-ash (alkaline) oxidation methods. To evaluate the wet method and the possible use as a reference when evaluating the dry method proposed here, a reference standard from the US National Institute of Standards and Technology (NIST) was used (Montana Soil - NIST 2710). The dry-ash oxidation process with alkaline oxidizing agents is one of the simplest oxidation methods of organic S to the sulfate form and was compared with the wet process. The objective of the study was to develop a dry method that would be easy to apply and allow the complete conversion of organic S to sulfate in soil samples and later detection by turbidimetry. The effectiveness of organic S oxidation to sulfate was evaluated by means of three alkaline oxidation mixtures: NaHCO3 + Ag2O, Eschka mixture (17 % Na2CO3, 66 % MgO, and 17 % K2CO3), and NaHCO3 + CuO. The procedure to quantify the sulfate concentration was based on the reaction with barium chloride and turbidimetric detection. Sulfur quantification in the standard sample by the wet method proved adequate, precise and accurate. It should also be pointed out that no significant differences were found (95 % reliability) between the wet and dry processes (NaHCO3 and Ag2O oxidation mixture) in six different Brazilian soils. The proposed dry method can therefore be used in the preparation of soil samples for total S determination.

La utilización de ADR en el ámbito de las reclamaciones de consumo tiene un marco legal específico en España -y también en la UE- desde 1993, que deriva de sus características especiales. Algunas son de cuño exclusivamente jurídico: se trata de reclamaciones en las que el marco legal aplicable es el denominado derecho de consumo; otras poseen un soporte básicamente fáctico: suelen tener poca entidad económica, es decir, entran en la categoría de lo que denominamos small claims, y el hecho de que las reclamaciones -en algunos supuestos que van en aumento- sean transfronterizas condiciona la opción entre jurisdicción tradicional y ADR, como también tendremos ocasión de analizar. Hay que añadir a estos elementos jurídicos y fácticos un elemento importante de política legislativa en un ámbito de la UE; los ADR se entienden como un instrumento básico para garantizar el acceso de los consumidores a la justicia, pero al mismo tiempo, en el ámbito del comercio electrónico, son un ele- mento de gran trascendencia en la creación de la denominada e-confidence. Por este motivo, se explo- ran continuamente formas de ODR (On-line Dispute Resolution). Los ODR pretenden la mayor eficacia ofreciendo un soporte técnico capaz de solucionar una controversia con o sin la intervención de un tercero, y dentro o fuera de la organización del empresario. De este modo, se usa un mismo expediente técnico para poner en marcha sucesivamente más de un ADR, o se potencian los mecanismos automáticos que prescinden de los conceptos jurídicos y, en medio de la exploración constante, España apuesta por el arbitraje electrónico de consumo en el RD 236/2008. A estos temas vamos a referirnos a continuación.

Paroxysmal nocturnal hemoglobinuria (PNH) is a rare, acquired stem cell disorder, with its primary clinical manifestations being hemolytic anemia, marrow failure and thrombophilia. Chronic hemolysis, failures of the fibrinolytic system, increased leukocyte-derived tissue factor levels in plasma, procoagulant microparticles generated through complement-mediated damage of platelets and venous endothelium are related to the acquired hypercoagulable state. Visceral thrombosis (including hepatic veins and mesenteric veins), cerebrovascular events and pulmonary embolism predict a poor outcome. Thrombosis is also associated with significant morbidity during pregnancy. Depending on the sites of thrombosis, a score-based probability to predict outcome can be assigned. Abdominal vein thromboses account for the majority of morbidity and mortality related to thrombosis, and time-dependent trends suggest that mortality rates tend to decline, with the advent of evolution of therapeutic and diagnostic strategies. In contrast, mortality rates from cerebrovascular events display no significant decline. Prompt diagnosis requires both clinical suspicion and sophisticated imaging techniques, along with multidisciplinary therapeutic intervention. In the eculizumab era, a significant reduction of thrombotic events was observed during therapy, and long-term follow up is needed to establish any benefit in rates and pattern of this complication. However, up to now, only bone marrow transplantation permanently abolishes the coagulation defect.

Las circunstancias del hallazgo de esta moneda son completamente fortuitas. Se encontró junto al embalse de La Torrassa (La Guingueta), cerca de la localidad de Esterri d'Aneu, Pallars Sobira, provincia de LIeida, siendo sus descubridores los hijos de don Pascual Massanet. Las características de la moneda son las siguientes: es de oro y muy poco desgastada, prácticamente «flor de cuño», conservando casi su peso originario que es de 4,110 gr., siendo su diámetro de 21 mm. Las leyendas del anverso y del reverso se leen perfectamente.

Se han caracterizado químicamente diferentes tipos de vidrios arquitectónicos originales de la iglesia del Monestir de Pedralbes, Barcelona (realizados hacia 1330). Esta nota expone los resultados obtenidos mediante el empleo de microsonda electrónica en vidrios de varios colores (blanco, verde, rosado, azul, amarillo, rojo) de la vidriera de Sant Pere y Sant Jaume. Corresponden esencialmente a vidrios sódicos (dentro del rango 13-19 % de Na2O) de coloración y composición homogénea, con elementos que actúan como colorante (p.e., CuO en rango 1.6-6 % en los vidrios verdes). El vidrio rojo, potásico, es una excepción notable (16.5 % K2O, 14 % CaO), tecnológicamente diferente (vidrio"plaqué" blanco con una intercalación roja de composición análoga pero sensiblemente más rica en CuO). Los vidrios sódicos no aparecen substancialmente dañados, los vidrios potásicos presentan corrosiones (moderadas) y pérdida parcial de transparencia. El predominio en esta vidriera de vidrios sódicos (inusuales para la época por lo que conocemos) ha permitido una excelente conservación. La composición química es una característica que permite diagnosticar precozmente el estado de deterioro y evaluar los costes de restauración en una vidriera, independientemente de los recubrimientos, pátinas y morteros que ésta presente.

L’objecte d’aquest projecte és determinar la temperatura de descomposició de l’YBCO perun rang de diferents pressions parcials d’oxigen en l’atmosfera que es troba. Per obteniraquests resultats, en primer lloc, hem d’aconseguir generar de forma sistemàtica diversespressions parcials dins del forn i, en segon lloc, buscar quin és el mètode experimental mésadequat que ens doni els resultats més fiables utilitzant com a referència la descomposiciódel CuO. A més de realitzar els calibratges necessàries dels aparells utilitzats per poderobtenir uns bons resultats
