During the process of the salt production, the first the salt crystals formed are disposed of as industrial waste. This waste is formed basically by gypsum, composed of calcium sulfate dihydrate (CaSO4.2H2O), known as carago cru or malacacheta . After be submitted the process of calcination to produce gypsum (CaSO4.0,5H2O), can be made possible its application in cement industry. This work aims to optimize the time and temperature for the process of calcination of the gypsum (carago) for get beta plaster according to the specifications of the norms of civil construction. The experiments involved the chemical and mineralogical characterization of the gypsum (carago) from the crystallizers, and of the plaster that is produced in the salt industry located in Mossoró, through the following techniques: x-ray diffraction (XRD), x-ray fluorescence (FRX), thermogravimetric analysis (TG/DTG) and scanning electron microscopy (SEM) with EDS. For optimization of time and temperature of the process of calcination was used the planning three factorial with levels with response surfaces of compressive mechanical tests and setting time, according norms NBR-13207: Plasters for civil construction and x-ray diffraction of plasters (carago) beta obtained in calcination. The STATISTICA software 7.0 was used for the calculations to relate the experimental data for a statistical model. The process for optimization of calcination of gypsum (carago) occurred in the temperature range from 120° C to 160° C and the time in the range of 90 to 210 minutes in the oven at atmospheric pressure, it was found that with the increase of values of temperature of 160° C and time calcination of 210 minutes to get the results of tests of resistance to compression with values above 10 MPa which conform to the standard required (> 8.40) and that the X-ray diffractograms the predominance of the phase of hemidrato beta, getting a beta plaster of good quality and which is in accordance with the norms in force, giving a by-product of the salt industry employability in civil construction

The gonads and the germinative cells of 3 male hinnies were studied with light and transmission electron microscopy with the aim to observe the development of germ cells and verify the morphological modifications due to the hybridization. The hinny seminiferous epithelium presented Sertoli cells and spermatogonia with normal features and anomalous spermatocytes I. The other cells from the spermatogenic sequence were not seen. Most of the alterations began to occur in the cytes I, which presented nuclear vacuolization and deposits of amorphous material between the carioteca and the nuclear lamina, forming vesicles, or exaggerated chromatin condensation, resulting in pyknosis. In the cytoplasm vacuolization was also observed, besides organelle destruction.The arrest of meiosis due to lock of chromosome homologies leads to germinative cell degeneration and, therefore, the spermatogenesis arrest. This fact causes a profound alteration in the seminiferous epithelium morphology in comparison with the parental species.

Esta dissertação baseia-se na análise da estratégia competitiva adotada no mercado de vergalhões de aço para construção civil em Belém, capital do Estado do Pará, pela Copala Indústrias Reunidas S/A. Existe então o interesse de avaliar as possibilidades da empresa no sentido de permanecer atuando com êxito no setor, em que pese a concorrência exercida por siderúrgicas de grande porte, cujos interesses são da abrangência nacional e internacional. Foram realizadas pesquisas considerando-se o mercado de aço para construção civil de um modo amplo, sendo consultados construtores, comerciantes e consumidores desse material. Outrossim, a própria Copala foi objeto de avaliação no que se refere a sua atuação tanto como adquirente de insumos para fabricação de aço, quanto como produtora e vendedora do produto.

A grande quantidade de resíduos sólidos, oriundos da indústria de mineração e metalurgia, constitui um sério problema sócio-ambiental. As características físicoquímicas destes resíduos despertam grande interesse para outras indústrias. A indústria cerâmica pode ser uma interessante consumidora da maioria deste material, para suprir a grande escassez das reservas de matérias-primas atual. Neste contexto, este trabalho mostra os estudos realizados para a reciclagem da lama vermelha, como matéria-prima na produção de agregado sintético, visando à construção civil. A lama vermelha, principal rejeito industrial da fabricação de alumina, mostrou-se um insumo de grande interesse na fabricação de diferentes tipos de agregado para ser utilizado na produção de concreto, para a construção civil. Pelas suas características físico-químicas e a grande quantidade que é produzida anualmente (cerca de 10.000.000 t em duas fábricas, só no Norte do Brasil). Estudos realizados neste trabalho mostram a possibilidade de fabricação de agregados, com diferentes propriedades e possibilidades de aplicação, na indústria da construção civil. Estas propriedades dependem do controle de parâmetros, como o teor de sílica livre e argila, a granulometria e a temperatura de sinterização. Tais variáveis permitem controlar perfeitamente a formação de fase vítrea que é a responsável pelas propriedades dos agregados como: porosidade, resistência mecânica e densidade. Este material pode ser utilizado em concreto convencional ou em concretos especiais, para atender a demanda da construção civil.

A segurança pública, de acordo com a Constituição Federal Brasileira de 1988, é dever do Estado, direito e responsabilidade de todos. É exercida nas diversas esferas de atribuições pelas instituições policiais bem como por outras organizações com o objetivo de preservar a ordem pública e a incolumidade das pessoas e do patrimônio, assegurando a plenitude do Estado Democrático de Direito. Na esfera estadual, a Polícia Civil é um dos órgãos responsáveis pela garantia da proteção da sociedade, possuindo uma estrutura organizacional e uma gerência voltada para o cumprimento de sua missão institucional. Diante de uma carência de pesquisas em relação às instituições do estado do Pará, principalmente aquelas voltadas para a segurança pública, este trabalho se propõe a investigar o modelo policial adotado na Polícia Civil do Pará no que tange a estrutura organizacional e a gestão do órgão bem como as políticas institucionais realizadas com o propósito de controlar a violência e a criminalidade no estado. Como resultado observa-se que Polícia Civil do Pará se organiza estruturalmente em diretorias, assessorias e divisões, alicerçando o exercício da atividade policial, tendo como representação máxima do órgão a figura do Delegado Geral de Polícia Civil. Em relação ao gerenciamento da Instituição, sugestiona-se uma centralização bem como uma burocratização na execução de suas atividades. Por outro lado, políticas institucionais, como o Fundo Rotativo Policial e a Política Integracionista, contribuem para uma evolução gerencial em relação à descentralização das atividades. Demonstra-se que a estrutura organizacional e a gerência como também as políticas institucionais influenciam no desenvolvimento do trabalho policial, já que essas variáveis são as principais bases para a execução da atividade fim da Instituição: a proteção da dignidade da pessoa humana. Este trabalho caracteriza-se como inédito e pioneiro, podendo servir de base para implantação de políticas públicas mais assertivas e adaptadas às reais necessidades da sociedade.

As fibras ópticas são utilizadas em diferentes áreas e nas mais variadas aplicações. Na Engenharia Civil começaram a ser utilizadas no monitoramento de estruturas como alternativa de substituição dos tradicionais sensores elétricos. Este trabalho tem como objetivo estudar a aplicação e viabilidade dos sensores a base de fibra óptica no monitoramento de grandes estruturas da engenharia civil. Para avaliação dos resultados, foram realizados três diferentes tipos de testes experimentais onde corpos-de-prova foram instrumentados com extensômetros elétricos e ópticos. O primeiro tipo de teste correspondeu a ensaios de tração simples em barras de aço com carregamento monotônico até o rompimento de barras de aço. No segundo tipo de teste foi estudada a automação de um medidor triortogonal de junta que é um instrumento tipicamente utilizado para a instrumentação de barragens de concreto. Finalmente o último tipo de testes foi constituído por ensaios de cilindros de concreto à compressão para a determinação do módulo de elasticidade. Com relação à utilização dos sensores ópticos, durante a realização dos testes surgiram muitos problemas relacionados com a unidade óptica de aquisição de dados sendo necessária a utilização de três diferentes modelos de unidade de aquisição para a conclusão dos testes. No que se refere à exatidão dos resultados, observou-se que os resultados apresentados pelos sensores ópticos foram compatíveis com os resultados dos sensores elétricos. Entretanto, quando os sensores ópticos foram solicitados por grandes deformações implicaram em perda de sinal devido à interrupção do fluxo de luz, inabilitando o sensor para leituras.

This research deals with the behaviour of grouted dowels used in beam-to-column connections in precast concrete structures. The research focuses primarily on the theoretical and experimental analysis of the resistance mechanism of the dowels. The experimental programme included 15 models for analysing the following variations in dowel parameters: a) dowel diameters of 16, 20 and 25 mm, b) dowel inclinations of 0 degrees (i.e. perpendicular to the interface), 45 degrees and 60 degrees, c) compressive strength of classes C35 and C50 for the concrete adjacent to the dowels, and d) the absence or presence of compressive loads normal to the interface. The experimental results indicate that the ultimate capacity and shear stiffness of the inclined dowels are significantly higher than those of the perpendicular dowels. Based on these results, an analytical model is proposed that considers the influence of the parameters studied regarding the capacity of the dowel.

Building facilities have become important infrastructures for modern productive plants dedicated to services. In this context, the control systems of intelligent buildings have evolved while their reliability has evidently improved. However, the occurrence of faults is inevitable in systems conceived, constructed and operated by humans. Thus, a practical alternative approach is found to be very useful to reduce the consequences of faults. Yet, only few publications address intelligent building modeling processes that take into consideration the occurrence of faults and how to manage their consequences. In the light of the foregoing, a procedure is proposed for the modeling of intelligent building control systems, considersing their functional specifications in normal operation and in the of the event of faults. The proposed procedure adopts the concepts of discrete event systems and holons, and explores Petri nets and their extensions so as to represent the structure and operation of control systems for intelligent buildings under normal and abnormal situations. This paper describes the applicability of a Hg-electroplated-Pt ultramicroelectrode in the quantification of elemental sulphur in naphtha samples by square-wave voltammetry. A reproducible deposition methodology was studied and is reported in this paper. This methodology is innovative and relies on the quality of the mercury stock solution to obtain reproducible surfaces required for the analytical methodology. All analyses were performed using a Hg-electroplated-Pt ultramicroelectrode (Hg-Pt UME) due to the low sensibility of such devices to ohmic drops in resistive solutions. The responses of the peak areas in voltammetric experiments were linear in all of the range studied. The method developed here is accurate and reproducible, with a detection limit of 0.010 mg L-1 and a good recovery range for both standard solutions of elemental sulphur (85 to 99%) and real naphtha sample (79%). These results attest to the potential for the application of this electroanalytical methodology in determining elemental sulphur in naphtha samples containing mercaptans and disulphides.

This paper presents preliminary results to determine small displacements of a global positioning system (GPS) antenna fastened to a structure using only one L1 GPS receiver. Vibrations, periodic or not, are common in large structures, such as bridges, footbridges, tall buildings, and towers under dynamic loads. The behavior in time and frequency leads to structural analysis studies. The hypothesis of this article is that any large structure that presents vibrations in the centimeter-to-millimeter range can be monitored by phase measurements of a single L1 receiver with a high data rate, as long as the direction of the displacement is pointing to a particular satellite. Within this scenario, the carrier phase will be modulated by antenna displacement. During a period of a few dozen seconds, the relative displacement to the satellite, the satellite clock, and the atmospheric phase delays can be assumed as a polynomial time function. The residuals from a polynomial adjustment contain the phase modulation owing to small displacements, random noise, receiver clock short time instabilities, and multipath. The results showed that it is possible to detect displacements of centimeters in the phase data of a single satellite and millimeters in the difference between the phases of two satellites. After applying a periodic nonsinusoidal displacement of 10 m to the antenna, it is clearly recovered in the difference of the residuals. The time domain spectrum obtained by the fast Fourier transform (FFT) exhibited a defined peak of the third harmonic much more than the random noise using the proposed third-degree polynomial model. Embryonic carcinoma cells are widely used models for studying the mechanisms of proliferation and differentiation occurring during early embryogenesis. We have now investigated how down-regulation of P2X2 and P2X7 receptor expression by RNA interference (RNAi) affects neural differentiation and phenotype specification of P19 embryonal carcinoma cells. Wild-type P19 embryonal carcinoma cells or cells stably expressing shRNAs targeting P2X2 or P2X7 receptor expression were induced to differentiate into neurons and glial cells in the presence of retinoic acid. Silencing of P2X2 receptor expression along differentiation promoted cell proliferation and an increase in the percentage of cells expressing glial-specific GFAP, while the presence of beta-3 tubulin-positive cells diminished at the same time. Proliferation induction in the presence of stable anti-P2X2 receptor RNAi points at a mechanism where glial proliferation is favored over growth arrest of progenitor cells which would allow neuronal maturation. Differently from the P2X2 receptor, inhibition of P2X7 receptor expression during neural differentiation of P19 cells resulted in a decrease in cell proliferation and GFAP expression, suggesting the need of functional P2X7 receptors for the progress of gliogenesis. The results obtained in this study indicate the importance of purinergic signaling for cell fate determination during neural differentiation, with P2X2 and P2X7 receptors promoting neurogenesis and gliogenesis, respectively. The shRNAs down-regulating P2X2 or P2X7 receptor gene expression, developed during this work, present useful tools for studying mechanisms of neural differentiation in other stem cell models. Ao disciplinar a reparação de dano do qual resulta incapacidade laboral, o Código Civil Brasileiro de 2002 trouxe, como alternativa à pensão vitalícia, a possibilidade de indenização paga de uma só vez. Diante disso, o primeiro impulso hermenêutico foi apurar tal montante multiplicando o valor da renda mensal do ofendido pela sua expectativa de sobrevida. o presente artigo busca, fundamentalmente, questionar a adequação epistemológica desta fórmula, bem como esquadrinhar as diretrizes processuais aplicáveis ao dispositivo. O fim colimado exigirá, inter alia, perscrutar o significado da expressão “arbitramento”, a extensão do conceito de “reparação integral” e as consequências jurídicas e econômicas do recebimento antecipado. Posteriormente, pretende-se minudenciar a titularidade, a efetiva existência e o momento da opção pelo prejudicado, à luz da cláusula do “devido processo”.

The repressor element 1-silencing transcription factor (REST) was first identified as a protein that binds to a 21-bp DNA sequence element (known as repressor element 1 (RE1)) resulting in transcriptional repression of the neural-specific genes [Chong et al., 1995; Schoenherr and Anderson, 1995]. The original proposed role for REST was that of a factor responsible for restricting neuronal gene expression to the nervous system by silencing expression of these genes in non-neuronal cells. Although it was initially thought to repress neuronal genes in non-neuronal cells, the role of REST is complex and tissue dependent. In this study I investigated any role played by REST in the induction and patterning of differentiation of SH-SY5Y human neuroblastoma cells exposed to IGF-I. and phorbol 12- myristate 13-acetate (PMA) To down-regulate REST expression we developed an antisense (AS) strategy based on the use of phosphorothioate oligonucleotides (ODNs). In order to evaluate REST mRNA levels, we developed a real-time PCR technique and REST protein levels were evaluated by western blotting. Results showed that nuclear REST is increased in SH-SY5Y neuroblastoma cells cultured in SFM and exposed to IGF-I for 2-days and it then declines in 5-day-treated cells concomitant with a progressive neurite extension. Also the phorbol ester PMA was able to increase nuclear REST levels after 3-days treatment concomitant to neuronal differentiation of neuroblastoma cells, whereas, at later stages, it is down-regulated. Supporting these data, the exposure to PKC inhibitors (GF10923X and Gö6976) and PMA (16nM) reverted the effects observed with PMA alone. REST levels were related to morphological differentiation, expression of growth coneassociated protein 43 (GAP-43; a gene not regulated by REST) and of synapsin I and βIII tubulin (genes regulated by REST), proteins involved in the early stage of neuronal development. We observed that differentiation of SH-SY5Y cells by IGF-I and PMA was accompanied by a significant increase of these neuronal markers, an effect that was concomitant with REST decrease. In order to relate the decreased REST expression with a progressive neurite extension, I investigated any possible involvement of the ubiquitin–proteasome system (UPS), a multienzymatic pathway which degrades polyubiquinated soluble cytoplasmic proteins [Pickart and Cohen, 2004]. For this purpose, SH-SY5Y cells are concomitantly exposed to PMA and the proteasome inhibitor MG132. In SH-SY5Y exposed to PMA and MG 132, we observed an inverse pattern of expression of synapsin I and β- tubulin III, two neuronal differentiation markers regulated by REST. Their cytoplasmic levels are reduced when compared to cells exposed to PMA alone, as a consequence of the increase of REST expression by proteasome inhibitor. The majority of proteasome substrates identified to date are marked for degradation by polyubiquitinylation; however, exceptions to this principle, are well documented [Hoyt and Coffino, 2004]. Interestingly, REST degradation seems to be completely ubiquitin-independent. The expression pattern of REST could be consistent with the theory that, during early neuronal differentiation induced by IGF-I and PKC, it may help to repress the expression of several genes not yet required by the differentiation program and then it declines later. Interestingly, the observation that REST expression is progressively reduced in parallel with cell proliferation seems to indicate that the role of this transcription factor could also be related to cell survival or to counteract apotosis events [Lawinger et al., 2000] although, as shown by AS-ODN experiments, it does not seem to be directly involved in cell proliferation. Therefore, the decline of REST expression is a comparatively later event during maturation of neuroroblasts in vitro. Thus, we propose that REST is regulated by growth factors, like IGF-I, and PKC activators in a time-dependent manner: it is elevated during early steps of neural induction and could contribute to down-regulate genes not yet required by the differentiation program while it declines later for the acquisition of neural phenotypes, concomitantly with a progressive neurite extension. This later decline is regulated by the proteasome system activation in an ubiquitin-indipendent way and adds more evidences to the hypothesis that REST down-regulation contributes to differentiation and arrest of proliferation of neuroblastoma cells. Finally, the glycosylation pattern of the REST protein was analysed, moving from the observation that the molecular weight calculated on REST sequence is about 116 kDa but using western blotting this transcription factor appears to have distinct apparent molecular weight (see Table 1.1): this difference could be explained by post-translational modifications of the proteins, like glycosylation. In fact recently, several studies underlined the importance of O-glycosylation in modulating transcriptional silencing, protein phosphorylation, protein degradation by proteasome and protein–protein interactions [Julenius et al., 2005; Zachara and Hart, 2006]. Deglycosilating analysis showed that REST protein in SH-SY5Y and HEK293 cells is Oglycosylated and not N-glycosylated. Moreover, using several combination of deglycosilating enzymes it is possible to hypothesize the presence of Gal-β(1-3)-GalNAc residues on the endogenous REST, while β(1-4)-linked galactose residues may be present on recombinant REST protein expressed in HEK293 cells. However, the O-glycosylation process produces an immense multiplicity of chemical structures and monosaccharides must be sequentially hydrolyzed by a series of exoglycosidase. Further experiments are needed to characterize all the post-translational modification of the transcription factor REST.
