Current cattlemen must seek methods to maximum reproductive efficiency of their beef herds, making the business competitive and enhancing greater profitability. For the cow-calf producers, efficiency translates into more cows producing one calf every year. Fixed-time artificial insemination (TAI) is one of several technologies that producers utilize to reach this goal. Postpartum anestrus is one of the biggest obstacles to overcome in order to obtain suitable results at the end of the breeding season. The TAI protocols are efficient in re-establishing cyclicity in noncycling cows during the postpartum period, streamlining the use of labor on farms, allowing the use of artificial insemination (AI) on a large scale, and introducing superior genetics to the herd. The protocols that are most commonly used in Brazil are based on progesterone (P4) releasing devices that prevent premature estrus and ovulation; and estradiol (E2) to synchronize the initiation of a new follicular wave. In such protocols, administration of equine chorionic gonadotropin (eCG) helps the development of the follicle, increases ovulation rate, improves the endocrine and uterine environment during proestrus and diestrus, and improves fertility. The use of eCG in TAI protocols for suckled cows, non-lactating cows and heifers increases the pregnancy rate allowing more calves at the end of the breeding season and higher profitability to the cattlemen

In the last years Brazil has achieved a leading position in worldwide cattle breeding. This country is consolidating the practice of an intensive and technical level cattle breeding, due to greater emphasis on reproductive efficiency and genetic improvement. The attention given to reproductive efficiency became to enhance the genetic improvement process, by exploiting the full reproductive potential of the animals. The biotechniques applied to reproduction arised to expand the reproductive potential for greater economic return. Are the most important reproductive biotechniques the FTAI and in vivo and in vitro embryo production for embryo transfer. Different hormonal treatments are consolidating to interfere with normal physiology in order to achieve the desired goals. The FTAI works to increase the reproductive rates of cattle through pre-booked artificial insemination in all the females in reproduction, without the need for estrous detection. The in vivo and in vitro embryo production aims to distribute the genetics of males and females genetically improved by transferring their embryos to genetically inferior females. Superovulation treatments designed to prevent the mechanism of dominance, which results in many follicles ovulating simultaneously for in vivo embryo production. To the in vitro embryo production, the technique of ovarian puncture guided by transvaginal ultrasound allows to aspirate a number of quality oocytes. The application of biotech is based on knowledge of the female and ovarian reproductive physiology and knowledge of their acting in different breeds and environments.

To assess the possibility of shifting the sex ratio at birth, since this can contribute to enhancing the genetic gain and producity in cattle, this work is aimed at raising the factors that may influence sex determination of creates. This study is based on research conducted in the area of reproduction and production of beef and dairy cattle, some quotes in humans, mouse, dogs and a study in pigs. This was due to back of data and studies in the bovine species. It was noted that stress during pregnancy, intake of vitamin C or others substances and the time of artificial insemination are the factors that may influence the determination of the type of product. Human studies, concluded that woman more stressed are more likely to produce female children of the quietest. Furthermore, sows that received ascorbic a ad orally for seven days and were inseminated during this period, produce more female then those who did not receive the vitamin. These studies may suggest that cows can also suffer influence of stress and food for the determination of sex of calf. There are also studies suggesting that cows inseminated at the earliest time of ovulation ted to produce more male calves than those inseminated a few hours before that time. Currently, Brazil has one of the largest cattle herds worldwide. In order to keep that milk and meat were introduced reproductive biotechnologies such as artificial insemination, embryo transfer and in vitro fertilization. In certain situations the technique may have undesired effect, for example, the production of calve calves due to the very large increase in the gestation period when performed in vitro fertilization. To avoid this problem we perform the induction of labor in order to prevent the product is longer the womb. This induction can also be made in case of diseases that compromise the life of the mother, twin pregnancy an abnormal size calf. The administration of short acting steroids, prostaglandins, association of short acting steroids and prostaglandins and association of short acting steroids, prostaglandins and long-acting corticosteroids are some of the possibilities of induction

In the last decades several hormonal treatments to induce multiple ovulation and embryo transfer (MOET) have been developed. Tight control of the time of ovulation allowed the use of fixed-time artificial insemination (FTAI) in bovine embryos donors, facilitating animal management. Although, protocols that allow FTAI have evolved and yield as much embryo as conventional protocols that requires estrus detection, substantial increase in viable embryo production has not been observed in superestimulated bovine cattle. The present review put emphasis on superestimulatory protocols in wich the last two doses of pFSH are replaced by eCG or LH. Recent results indicate that an extra LH stimulus (using eCG or LH), on the last day of P-36 superestimulatory treatment, seems to improve transferable embryo yield in both Bos taurus and Bos indicus cattle.

The transgenic application of green fluorescent protein (GFP) as fetal cell marker on cattle cloned placenta could provide an exclusive model for studying the morphologic and immunologic maternal-fetal interactions, providing information about its mapping, distinguishing the fetal from maternal cells. This model will have direct application, mainly because these animals present problems during its development. With this model's support, we intend to verify the substances transport between mother and fetus during endocytosis, through the immunolocalization of protein named caveolae. For these, we used 06 cloned bovine and 30 cattle samples of artificial insemination (AI) with 90 days of pregnancy, which had been their development interrupted by humanitarian slaughter of the recipient and recovery of the pregnant uterus. We collected the placentome and the chorion. A part of the samples was cut and fixed, by immersion, on a solution containing 4% of parafomaldehyde or 10% of formaldehyde on a sodium phosphate buffer (PBS), at 0,1 M pH 7.4, Zamboni solution (4% of paraformaldehyde, 15% of picric acid, on sodium phosphate buffer 0,1 M pH 7.4), metacarn (60% of metanol, 30% of chloroform, and 10% glacial acetic acid), for morphologic and immunohistochemistry verification for caveolinas proteins -1 and -2 (CAV -1 and CAV-2). The caveolins -1 were found in fetal and maternal villi, but its strongest staining was observed in the endometrial stroma. The caveolins -2 had positive staining in trophoblast and chorioallantoic membrane, and specifically in giant trophoblastic binucleated cell. Therefore the results were compared between cloned cattle and from AI or natural mating, for assisting on detection of the reason of many placental alterations, embryonic losses, spontaneous abortion, post-natal mortality and large offspring syndrome on laboratory-manipulated animals. The result suggests that the proteins caveolins -1 and -2 (CAV-1 and CAV-2) are part of the caveolae composition and important structures related to the molecule transfer to the fetus, nourish it through endocytosis and pinocytosis.

The present study evaluated the use of PCR for Histophilus somni detection in bovine semen. Semen samples were experimentally infected with H. somni at dilutions ranging from 107 to 101 bacteria/mL and subjected to DNA extraction by the phenol/chloroform method, followed by PCR amplification. The amplification products were analyzed by electrophoresis in 8% acrylamide gel. The oligonucleotide primers used yielded an amplification fragment of 400 base pairs from the bacterial DNA. Positive amplification was obtained even for the 101 bacteria/mL dilution. PCR proved to be an efficient method for the detection of H. somni. The results obtained in this study have brought relevant information for the diagnosis of H. somni, justifying the need for the diagnosis of this bacterium in bulls, especially in semen samples that should be free of contamination. The PCR method has shown to be a useful tool for the quality control of semen produced in artificial insemination centers.

The aim of this work was to examine both the influence of anatomical and technical aspects on fertility rate of sheep based on the performance of transcervical artificial insemination (TCAI). Transcervical artificial insemination was performed with traction of the cervix in 122 ewes using frozen semen from 11 rams, both Santa Ines breed. The data collected were: type of external cervical opening (CO) (P - papilla; FL - flap; DB - duckbill, S - spiral; RO - rosette), duration of cervical manipulation (2-3, 4-5 and 6-7 minutes), degree of difficulty in cervical transposition (low, moderate, high) and presumed semen deposition site (SC - superficial cervical; DC - deep cervical; IU - intrauterine). The influence of these variables on pregnancy rate was evaluated. Cervical opening type and duration of cervical manipulation had no influence (p>0.05) on fertility. The degree of difficulty in cervical manipulation influenced (p<0.05) pregnancy rate, since insemination classified as low grade had 52% of pregnancy, while those classified as high recorded only 20%. The presumed site of semen deposition influenced significantly (p<0.05) fertility. Pregnancy rates of deposition at each site were: UI – 45.8%, DC – 25.7%; SC – 15.4%. As expected, deeper depositions resulted in higher fertility. In conclusion, the performance of TCAI did not depend on the anatomical classification of external cervical opening of ewe and the duration of cervical manipulation within the range tested (2-7 minutes). The TCAI may have higher fertility rates if difficulties in the application were reduced and the semen deposition was deeper.

The reproductive performance of cattle may be influenced by several factors, but mineral imbalances are crucial in terms of direct effects on reproduction. Several studies have shown that elements such as calcium, copper, iron, magnesium, selenium, and zinc are essential for reproduction and can prevent oxidative stress. However, toxic elements such as lead, nickel, and arsenic can have adverse effects on reproduction. In this paper, we applied a simple and fast method of multi-element analysis to bovine semen samples from Zebu and European classes used in reproduction programs and artificial insemination. Samples were analyzed by inductively coupled plasma spectrometry (ICP-MS) using aqueous medium calibration and the samples were diluted in a proportion of 1:50 in a solution containing 0.01% (vol/vol) Triton X-100 and 0.5% (vol/vol) nitric acid. Rhodium, iridium, and yttrium were used as the internal standards for ICP-MS analysis. To develop a reliable method of tracing the class of bovine semen, we used data mining techniques that make it possible to classify unknown samples after checking the differentiation of known-class samples. Based on the determination of 15 elements in 41 samples of bovine semen, 3 machine-learning tools for classification were applied to determine cattle class. Our results demonstrate the potential of support vector machine (SVM), multilayer perceptron (MLP), and random forest (RF) chemometric tools to identify cattle class. Moreover, the selection tools made it possible to reduce the number of chemical elements needed from 15 to just 8.

In order to provide information that may help researchers to understand the main cause(s) of differences in bull fertility frequently observed in field trials, this study aimed to investigate conception rates as well as several in vitro sperm characteristics of different sires of unknown fertility utilized in a Timed-AI (TAI) program. Suckled Nelore cows submitted to the same TAI protocol were allocated into eight breeding groups of approximately 120 animals each. Frozen semen doses from three Angus bulls and three different batches from each bull were utilized. Approximately 100 doses from each batch were used in TAI. Sires, batches and AI technicians were equally distributed across breeding groups. Cows were examined for pregnancy diagnosis 40 d after TAI. For in vitro sperm analyses, the same thawing procedure was repeated in the laboratory to mimic field conditions. The following in vitro sperm characteristics were assessed: computerized motility, thermal resistance, plasma and acrosomal membrane integrity, lipid peroxidation, morphology, morphometry and chromatin structure. No effect of breeding group, body condition score, AI technician and sire was observed. However, some significant differences among bulls were detected in laboratory analyses. Semen from sire presenting numerically lower (P > 0.05) pregnancy/AI also presented lower (P < 0.05) values in all sperm characteristics analyzed in thermal resistance test at 4 h (Total Motility, Progressive Motility, Average Path Velocity, Straight-Line Velocity, Curvilinear Velocity, Amplitude of Lateral Head Displacement, Beat Cross Frequency, Straightness, Linearity, and Percentage of Rapidly Moving Cells), higher (P < 0.05) Major and Total Defects in sperm morphological test, lower (P < 0.05) Length, Ellipticity and Fourier parameter (Fourier 0) in sperm morphometric analysis as well as higher (P < 0.05) chromatin heterogeneity. It was concluded that, although no bull effect was observed in the field experiment, the sire that presented numerically lower pregnancy/AI also presented lower semen quality according to the laboratory analyses performed. One hundred eighty-nine mixed breed beef heifers from 13 consignors enrolled in the MACEP heifer development project were utilized in this study. Heifers were synchronized by feeding 0.5 mg melengestrol acetate (MGA) per head per day for 14 days followed by an injection of prostaglandin F2a (PGF2a; 25 mg Lutalyse®) 17 days after the last MGA feeding. Each heifer was fitted with a Heatwatch® transmitter on the morning of PGF2a administration to facilitate detection of estrus. Vaginal conductivity measurements were taken using an Ovatec® probe every 12 hours for 96 hours beginning at the time of PGF2a injection. Heifers randomly assigned to produce a female calf were inseminated near the onset of estrus (as indicated by probe values of £ 55 on the decline). Heifers randomly assigned to produce a male calf were inseminated approximately 24 hours after the onset of estrus (as indicated by probe values of ³ 60 on the incline). All heifers not inseminated by 96 hours after PGF2a were mass inseminated in an attempt to impregnate as many heifers as possible. Heifers that were diagnosed as pregnant as a result of the artificial insemination were subjected to ultrasonography for fetal sex determination. Only 70 of the 189 heifers (37.0%) exhibited estrus according to Heatwatch® and incidence of estrus was influenced by heifer average daily gain, reproductive tract score, and disposition score. Heifers receiving a disposition score of 3 (78.7) had a higher (P<.05) probe reading at AI than those receiving a disposition score of 1 or 2 (70.8 and 72.5, respectively). Heifers with probe readings at insemination of 80 - 84 and > 84 had lower (P<.05) pregnancy rates to AI (13.6 and 0.0%, respectively) than heifers with probe readings in the ranges of < 60, 60 - 64, 65 - 69, 70 - 74, and 75 - 79 (35.7, 40.9, 31.4, 35.3, and 26.9% respectively). Heifers that were bred when probe values were increasing had a lower (P<.05) percentage of male fetuses (34.4%) than those bred during a period of decreasing probe values (69.2% male fetuses). These results demonstrate that a vaginal conductivity probe may be a useful tool to determine an insemination time that could potentially alter calf sex ratio.

Bovine congenital pseudomyotonia (PMT) is a genetic disease in Chianina and other breeds of cattle that induces muscular stiffness. PMT in the Chianina breed is caused by a missense mutation in exon 6 of the ATP2A1 gene, which encodes the SERCA1 pump. In this study, the prevalence of PMT carriers and the frequency of the deleterious PMT allele in selected subpopulations of the Chianina breed were estimated. The prevalence of PMT carriers among ranked Chianina sires used for artificial insemination in the years 2007-2011 was 13.6%. The frequency of PMT carriers in young bull calves born in the period January 2007 to June 2011 selected for a performance testing programme was 13.4%. Selective breeding against this genetic defect is restricted to males only and therefore is predicted to require at least seven generations to eradicate PMT.

Switzerland is officially free from bovine Tritrichomonas foetus. While bulls used for artificial insemination (AI) are routinely examined for this pathogen, bulls engaged in natural mating, as well as aborted fetuses, are only very sporadically investigated, indicating that the disease awareness for bovine tritrichomoniasis is low. Natural mating in cattle is becoming increasingly popular in Switzerland. Accordingly, a re-introduction/re-occurrence of T. foetus in cattle seems possible either via resurgence from a yet unknown bovine reservoir, or via importation of infected cattle. The low disease awareness for bovine tritrichomoniasis might favor an unnoticed re-establishment of T. foetus in the Swiss cattle population. The aim of our study was thus to search for the parasite, and if found, to assess the prevalence of bovine T. foetus in Switzerland. We included (1) bulls over two years of age used in natural mating and sent to slaughter, (2) bulls used for natural service in herds with or without fertility problems and (3) aborted fetuses. Furthermore, the routinely examined bulls used for AI (4) were included in this study. In total, 1362 preputial samples from bulls and 60 abomasal fluid samples of aborted fetuses were analyzed for the presence of T. foetus by both in vitro cultivation and molecular analyses. The parasite could not be detected in any of the samples, indicating that the maximal prevalence possibly missed was about 0.3% (95% confidence). Interestingly, in preputial samples of three bulls of category 1, apathogenic Tetratrichomonas sp. was identified, documenting a proof-of-principle for the methodology used in this study.

Este trabajo analiza las creencias mitológicas, agüeros o supersticiones asociadas con las actividades agropecuarias y de conservación en nueve unidades productivas rurales de la cuenca media del río La Vieja, Colombia. Plantea que las gentes de antaño reconocen la influencia de múltiples relatos sobre condiciones productivas y ambientales que redundan en el éxito o el fracaso de las actividades rurales. Las creencias se clasificaron según su funcionalidad y las actividades que regulan. Se describieron así 14 mitos que influyen en las actividades de cultivo, cría, conservación y regulación social. El principal elemento de discusión fue la funcionalidad, considerando la distribución y aplicación en los subsistemas de producción en la finca, la relación con la edad y el género y con la posición jerárquica en la estructura familiar. El flujo de información entre sistemas expertos y sistemas de creencias tradicionales ayuda a construir sistemas adaptados. A manera de ejemplo, se puede mencionar un mayordomo de un hato ganadero que combina la creencia en las fases de la luna con el empleo de la técnica genética de la inseminación artificial para asegurar no sólo la preñez de la madre sino también el género femenino de la cría.
