OBJETIVO: avaliar a prevalência dos HPV 16, 18, 31 e 45 em amostras de raspado cervical de mulheres com alterações celulares e/ou colposcopia sugestiva de lesão de alto grau ou lesão de baixo grau persistente submetidas à conização. MÉTODOS: Foram incluídas 120 mulheres. A análise histológica dos cones cervicais revelou 7 casos de cervicite, 22 de NIC1, 31 de NIC2, 54 de NIC3 e 6 carcinomas invasores. Foram analisadas as amostras de raspado cervical coletadas antes da conização para a presença do DNA-HPV por PCR com os primers de consenso, PGMY09/11. As amostras positivas para DNA de HPV foram testadas para presença do HPV16, 18, 31 e 45 utilizando-se primers tipo específico para esses HPV. RESULTADOS: O DNA-HPV foi detectado em 67,5% das mulheres. O HPV 16 (40%) foi o tipo mais prevalente na maioria das lesões, seguido dos HPV 31 (13,3%), 45 (13,3%) e 18 (4,1%). Infecções múltiplas ocorreram em 15% dos casos e as infecções por outros tipos de HPV foram detectadas em 14% da amostra. CONCLUSÕES: as infecções pelos HPV 16 e 18 nem sempre ocorrem de maneira solitária (infecção única), estando associadas a outros tipos de HPV em diversas ocasiões.

We performed a systematic review and meta-analysis of randomized controlled trials that studied the conservative management of stress urinary incontinence (SUI). There were 1058 results after the initial searches, from which 37 studies were eligible according to previously determined inclusion criteria. For the primary outcomes, pelvic floor muscle training (PFMT) was more efficacious than no treatment in improving incontinence-specific quality of life (QoL) scales (SMD = [1]1.24SDs; CI 95% = [1]1.77 to [1]0.71SDs). However, its effect on pad tests was imprecise. Combining biofeedback with PFMT had an uncertain effect on QoL (MD = [1]4.4 points; CI 95% = [1]16.69 to 7.89 points), but better results on the pad test, although with elevated heterogeneity (MD = 0.9g; 95%CI = 0.71 to 1,10g); group PFMT was not less efficacious than individual treatment, and home PFMT was not consistently worse than supervised PFMT. Both intravaginal and superficial electrical stimulation (IES and SES) were better than no treatment for QoL and pad test. Vaginal cones had mixed results. The association of IES with PFMT may improve the efficacy of the latter for QoL and pad test, but the results of individual studies were not consistent. Thus, there is evidence of the use of PFMT on the treatment of SUI, with and without biofeedback.

During endodontic therapy (pulpectomy, root canal debridement and root canal filling) microbiological management is a major concern. Bacteria present in dentine tubules, apical foramina and apical delta are causally related to failure of the procedure. Studies have shown that during single session endodontic treatment bacteria remain within dental structures. The aim of the present study was to evaluate endodontic treatment performed as two sessions, using temporary endodontic dressing materials for different periods in four groups of experimental dogs. A total of 80 roots of second and third upper premolar teeth and second, third and fourth lower premolar teeth were divided into four groups. The pulp chamber was opened with burrs and the pulp exposed for 60 days to induce pulpal inflammation and necrosis. Groups II, III and IV were treated with calcium hydroxide plus camphorated paramono-chlorophenol (PMCC) for 7, 15 and 30 days, respectively. In all groups, the root canals were filled with zinc oxide-eugenol and gutta-percha cones. Clinical and radiographical measurements were performed every 2 weeks. After 60 days a small block section containing the teeth, surrounding periapical tissues and the periodontium was removed for histological and microbiological study. Histological analysis revealed intense inflammatory response in all groups. Microbiological analysis showed microbial reduction inversely proportional to the period of time that the intracanal temporary medicament was left in place.

Com o aumento do número de intervenções cirúrgicas para a remoção da catarata em cães, observa-se a necessidade de exames específicos que devem ser realizados antes da indicação cirúrgica. A catarata é uma das principais causas de cegueira em cães. Nos estágios mais avançados, impossibilita o exame fundoscópico e inviabiliza a observação de alterações retinianas como a atrofia progressiva da retina (APR), degeneração retiniana hereditária de acometimento bilateral que, quando diagnosticada, contra-indica a cirurgia. Com o intuito de reestabelecer a visão, o eletrorretinograma de campo total (Full field ERG) torna-se indispensável na avaliação pré-cirúrgica da remoção da catarata. Como os cães da raça Cocker Spaniel Inglês são predispostos à catarata e a degenerações retinianas, objetivamos neste estudo avaliar as respostas dos ERGs realizados nestes animais. Foram avaliados 136 eletrorretinogramas de cães da raça Cocker Spaniel Inglês (62 machos e 74 fêmeas, com idades entre 3 e 15 anos) no período de Setembro de 2004 a Maio de 2009. Todos os animais apresentavam baixa de visão e catarata durante o exame. O diagnóstico de degeneração retiniana foi baseado nos valores de amplitude pico a pico e tempo de culminação da onda-b nas 3 respostas (resposta escotópica de bastonetes, máxima resposta e resposta fotópica de cones), idade do paciente e estágio de maturação da catarata. Nos cães sem degeneração retiniana, a média da amplitude e do tempo de culminação nas três respostas obtidas foram, respectivamente: 71,55mV/65,15ms; 149,17mV/33,03ms; 31,06mV/27,90ms. Nos cães com degeneração retiniana, 38 animais apresentaram ERG extinto. Dentre os restantes que apresentavam baixas respostas, a média da amplitude e do tempo de culminação nas três respostas obtidas foram, respectivamente: 12,88mV/65,04ms; 24,16mV/36,25ms; 8,36mV/31,38ms. Foi observado que em 122 animais (89,7%) os exames eram compatíveis com diagnóstico de atrofia progressiva da retina. Frente aos resultados obtidos, conclui-se que os cães da raça Cocker Spaniel Inglês portadores de catarata devem ser submetidos ao eletrorretinograma de campo total antes da remoção cirúrgica devido à alta incidência de degeneração retiniana nesta raça.

Periodontitis in cattle is an infectious purulent progressive disease associated with strict anaerobic subgingival biofilm and is epidemiologically related to soil management at several locations of Brazil. This study aimed to detect Treponema species in periodontal pockets of cattle with lesions deeper than 5mm in the gingival sulcus of 6 to 24-month-old animals considered periodontally healthy. We used paper cones to collect the materials, after removal of supragingival plaques, and kept frozen (at -80°C) up to DNA extraction and polymerase chain reaction (PCR) using T. amylovorum, T. denticola, T. maltophilum, T. medium and T. vincentii primers. In periodontal pocket, it was possible to identify by PCR directly, the presence of Treponema amylovorum in 73% of animals (19/26), T. denticola in 42.3% (11/26) and T. maltophilum in 54% (14/26). Among the 25 healthy sites, it was possible to identify T. amylovorum in 18 (72%), T. denticola in two (8%) and T. maltophilum in eight (32%). Treponema medium and T. vincentii were not detected over all 51 evaluated samples. The presence of Treponema amylovorum, T. maltophilum and, in particular, the widely recognized T. denticola in subgingival microflora brings an original and potencially important contribution in studies of the bovine periodontitis.

Abstract: The paired oviducal glands of immature and mature females of Mustelus schmitti were examined macro and microscopically. Findings indicate that these glands possessed the same zonation as in most chondrichthyans from anterior to posterior: club, papillary, baffle and terminal zones. The whole gland is composed by simple tubular glands that connect with transverse grooves all along the organ. The club zone presents a typical indian club shape with a simple columnar and ciliated epithelium including secretory cells PAS (+) and AB (+). The papillary zone is characterized by lamella forming small and long cones in numbers of three. The epithelium of this zone contains ciliated cells with apical nuclei and secretory cells with basal nuclei that stain AB (+)The baffle zone consists of apically flattened lamellae alternating with spinnerets which are small projections disposed by both sides of the plateau. This whole structure is present in number of 8 or 9 units. A simple columnar ciliated epithelium covers the plateau and spinnerets and no AB or PAS staining is observed. The epithelium of the terminal zone is PAS (-) and AB (+), and elongated tubules, that run adjacent to the baffle zone are the site where groups of spermatozoa are clearly observed in the lumen. The epithelium of the sperm storage tubules do not stain with any of the dyes tested. Sperm was also observed in the baffle zone, presumably in its way to the fecundation in the oviduct because it displays no aggregation pattern and was between the folds of the epithelium. By scanning electron microscopy sperm was observed in the club and baffle zones in a gland which belonged to a pregnant female.

O presente trabalho refere-se ao estudo taxonômico dos gêneros Chaetomorpha e Rhizoclonium ocorrentes no litoral da Bahia. As espécies foram delimitadas com base no hábito, presença ou ausência de rizóides laterais, tipo de célula basal, forma e dimensões celulares. A presença de inclusões cristalinas nas células também foi verificada. Foram identificadas nove espécies, sete para o gênero Chaetomorpha (C. aerea (Dillwyn) Kütz., C. antennina (Bory) Kütz., C. brachygona Harv., C. clavata Kütz., C. crassa (C. Agardh) Kütz., C. minima Collins & Herv. e C. nodosa Kütz.) e duas para o gênero Rhizoclonium (R. africanum Kütz. and R. riparium (Roth) Kütz. ex Harv.). Chaetomorpha crassa, C. minima e C. nodosa são novas adições para o litoral nordeste do Brasil e C. aerea, C. clavata e R. africanum são adições para o litoral da Bahia. Três tipos de cristais foram encontrados: 1. cristais de sílica, em forma de finas agulhas agrupadas, presentes em C. aerea, C. antennina, C. brachygona, C. clavata e C. crassa; 2. cristais de oxalato de cálcio, em forma de octaedros, encontrados em C. antennina e C. clavata e, 3. cristais de carbonato de cálcio, em forma de cones formando agregados globosos, observados em C. clavata. Talos férteis de C. antennina, C. clavata, C. nodosa, R. africanum e R. riparium são ilustrados pela primeira vez para o Brasil. Descrições detalhadas das estruturas vegetativas e reprodutivas das espécies e comparações com táxons afins são apresentadas.

As a result of recent investigations, the cytoskeleton can be viewed as a cytoplasmic system of interconnected filaments with three major integrative levels: self-assembling macromolecules, filamentous polymers, e.g., microtubules, intermediate filaments and actin filaments, and supramolecular structures formed by bundles of these filaments or networks resulting from cross-bridges between these major cytoskeletal polymers. The organization of this biological structure appears to be sensitive to fine spatially and temporally dependent regulatory signals. In differentiating neurons, regulation of cytoskeleton organization is particularly relevant, and the microtubule-associated protein (MAP) tau appears to play roles in the extension of large neuritic processes and axons as well as in the stabilization of microtubular polymers along these processes. Within this context, tau is directly involved in defining neuronal polarity as well as in the generation of neuronal growth cones. There is increasing evidence that elements of the extracellular matrix contribute to the control of cytoskeleton organization in differentiating neurons, and that these regulations could be mediated by changes in MAP activity. In this brief review, we discuss the possible roles of tau in mediating the effects of extracellular matrix components on the internal cytoskeletal arrays and its organization in growing neurons.

A consistência é um aspecto funcional importante das gorduras plásticas, que são misturas de cristais de gordura sólida e óleo líquido. A relação entre as duas fases e o caracter cristalino da fase sólida determinam a consistência e a firmeza das amostras. O objetivo do trabalho foi determinar a consistência de duas gorduras vegetais hidrogenadas e suas misturas binárias e ternárias, com a presença ou não de óleo de soja, a partir dos resultados do analisador de textura (TA XT2), variando as profundidades de penetração e o ângulo dos cones. Aplicou-se um modelo de regressão múltipla, do tipo cúbico especial para as misturas de óleo e gorduras, utilizando o aplicativo MIXPLOT para a construção de diagrama triangular. A força de compressão variou de 0,0080 a 5,459 kg, dependendo da amostra e da profundidade de penetração do cone. Para os cones de ângulos 30, 40 e 45º, as consistências foram todas estatisticamente semelhantes, a p< 0,05. Para algumas amostras, os resultados empregando o cone de 20º foram estatisticamente diferentes. Tais diferenças de resultados podem ser justificadas pelo fato da geometria do cone de 20º ser diferente, em função do truncamento de sua ponta. A análise do diagrama triangular mostra que o componente óleo de soja não contribuiu estatisticamente para a consistência. Os coeficientes relativos às interações foram sempre negativos, demonstrando efeito antagônico para a consistência, característico das interações eutéticas entre gorduras. Este efeito eutético ocorre devido à incompatibilidade no estado sólido entre os componentes da mistura.

Retinoic acid, a derivative of vitamin A, is known to play diverse roles in development and regeneration. Previous research in the mollusc Lymnaea stagnalis has shown that a gradient of all-trans retinoic acid attracts the growth cones of cultured neurons. The present study investigates the sub-cellular mechanisms within the growth cones of Lymnaea pedal A neurons which mediate the attractive response to a gradient of alltrans retinoic acid. In this study, the mechanism of growth cone turning is shown to be local, as neurites mechanically isolated from their cell body retain the capacity to turn towards an exogenous gradient of all-trans retinoic acid. The turning response is dependent on the initiation of protein synthesis and calcium influx, but does not appear to involve signaling through protein kinase C (PKC). The retinoid X receptor (RXR), which classically functions as a transcription factor, was also shown to be involved in the turning response, functioning locally through a non-genomic pathway. These data show, for the first time in any species, that all-trans retinoic acid's chemotropic action involves a local mechanism involving non-genomic signaling through the RXR. As retinoic acid is known to playa role in regeneration, understanding the mechanisms underlying retinoic acid signaling may lead to further advances in regenerative neuroscience.

In vertebrates, signaling by retinoic acid (RA) is known to play an important role in embryonic development, as well as organ homeostasis in the adult. In organisms such as adult axolotls and newts, RA is also important for regeneration of the CNS, limb, tail, and many other organ systems. RA mediates many of its effects in development and regeneration through nuclear receptors, known as retinoic acid receptors (RARs) and retinoid X receptors (RXRs). This study provides evidence for an important role of the RA receptor, RAR~2, in ,( '. regeneration ofthe spinal cord and tail of the adult newt. It has previously been proposed that the ability of the nervous system to regenerate might depend on the presence or absence of this RAR~2 isoform. Here, I show for the very first time, that the regenerating spinal cord of the adult newt expresses this ~2 receptor isoform, and inhibition of retinoid signaling through this specific receptor with a selective antagonist inhibits tail and spinal cord regeneration. This provides the first evidence for a role of this receptor in this process. Another species capable of CNS ~~generation in the adult is the invertebrate, " Lymnaea stagnalis. Although RA has been detected in a small number of invertebrates (including Lymnaea), the existence and functional roles of the retinoid receptors in most invertebrate non-chordates, have not been previously studied. It has been widely believed, however, that invertebrate non-chordates only possess the RXR class of retinoid receptors, but not the RARs. In this study, a full-length RXR cDNA has been cloned, which was the first retinoid receptor to be discovered in Lymnaea. I then went on to clone the very first full-length RAR eDNA from any non-chordate, invertebrate species. The functional role of these receptors was examined, and it was shown that normal molluscan development was altered, to varying degrees, by the presence of various RXR and RAR agonists or antagonists. The resulting disruptions in embryogenesis ranged from eye and shell defects, to complete lysis of the early embryo. These studies strongly suggest an important role for both the RXR and RAR in non-chordate development. The molluscan RXR and RAR were also shown to be expressed in the adult, nonregenerating eNS, as well as in individual motor neurons regenerating in culture. More specifically, their expression displayed a non-nuclear distfibution, suggesting a possible non-genomic role for these 'nuclear' receptors. It was shown that immunoreactivity for the RXR was present in almost all regenerating growth cones, and (together with N. Farrar) it was shown that this RXR played a novel, non-genomic role in mediating growth cone turning toward retinoic acid. Immunoreactivity for the novel invertebrate RAR was also found in the regenerating growth cones, but future work will be required to determine its functional role in nerve cell regeneration. Taken together, these data provide evidence for the importance of these novel '. retinoid receptors in development and regeneration, particularly in the adult nervous system, and the conservation of their effects in mediating RA signaling from invertebrates to vertebrates.

The vitamin A metabolite, retinoic acid (RA), is known to play a crucial role in several developmental processes including axial patterning and differentiation. More recently, RA has been implicated in the regenerative process acting through its classical signaling pathway, the nuclear receptors, retinoic acid receptor (RAR) and retinoid X receptor (RXR), to mediate gene transcription. Moreover, RA has been shown to act as a guidance molecule for growth cones of regenerating motorneurons of the pond snail, Lymnaea stagnalis. Our lab has recently shown that RA can induce this morphological response independent of nuclear transcription, however, the role of the retinoid receptors in RA-induced chemoattraction is still unknown. Here, I show that the retinoid receptors, RXR and RAR, may mediate the growth cones response to the metabolically active retinoic acid isomers, all-trans and 9-cis RA, in Lymnaea stagnalis. Data presented here show that both an RXR and RAR antagonist can block growth cone turning in response to application of both isomers. Because no prior investigations have shown growth cone turning of individual vertebrate neurons, I aimed to show that both retinoic acid isomers were capable of inducing growth cone turning of embryonic spinal cord neurons in the frog, Xenopus laevis. For the first time in Xenopus, I showed that both all-trans and 9-cis RA were able to induce significantly more neurite outgrowth from cultured embryonic spinal cord neurons and induce positive growth cone turning of individual growth cones. In addition, I showed that the presence of the RXR antagonist, HX531, blocked 9-cis RA-induced growth cone turning and the RARβ antagonist, LE135, blocked all-trans RA-induced growth cone turning in this species. Evidence provided here shows for the first time, conservation of retinoic acid-induced growth cone turning in a vertebrate model system. In addition, these data show that the receptors involved in this morphological response may be the same in vertebrates and invertebrates.

Les effets de la marijuana, un médicament utilisé par l’homme depuis des millénaires, sur le système visuel sont peu connus. Une meilleure connaissance de la distribution du système endocannabinoïde (eCB) de la rétine pourrait expliquer comment cette drogue affecte la vision. Cette étude vise à caractériser la distribution du récepteur cannabinoïde CB1 (CB1R) et de l’enzyme de dégradation FAAH (“fatty acid amide hydrolase”) des ligands du CB1R dans la rétine du singe Vert (Chlorocebus sabaeus). De plus, elle vise à déterminer quelles sous-populations cellulaires de la rétine expriment ces composantes. La plupart des études à ce jour ont été conduites surtout sur les rongeurs et peu de travaux ont été réalisés chez le singe. Notre étude vient donc combler cette carence. Par le biais de méthodes immunohistochimiques, nous avons investigué la localisation du CB1R et de l’enzyme FAAH à différentes excentricités rétiniennes, de la fovéa centralis vers la périphérie. Nos résultats, en accord avec notre hypothèse de travail, démontrent que CB1R et FAAH sont exprimés à travers toute la rétine mais avec, cependant, des différences notoires. Au niveau de la couche des photorécepteurs, CB1R est exprimé préférentiellement dans les cônes et ce patron d’expression suit la distribution des photorécepteurs centre-périphérie. De plus, CB1R se retrouve surtout dans les pédicules des cônes de la couche plexiforme externe. CB1R et FAAH sont abondants dans les cellules bipolaires tant au centre qu’en périphérie. Le soma et l’axone des cellules ganglionnaires expriment aussi CB1R et FAAH. Ces données suggèrent que le système eCB est présent à travers toute la rétine du primate et pourrait expliquer les perturbations visuelles entrainées par la marijuana, telles la photosensibilité et la vision des couleurs.

La rétine est constituée de plusieurs types de neurones incluant les cellules amacrines, ganglionnaires, bipolaires et les photorécepteurs. Les photorécepteurs, qui englobent les cônes et les bâtonnets, sont des neurones sensoriels hautement spécialisés qui permettent la conversion de la lumière en signaux électriques par le mécanisme de phototransduction. Les mécanismes moléculaires par lesquels les progéniteurs rétiniens (RPCs) se différencient en différents neurones spécialisés comme les photorécepteurs sont encore peu connus. Le gène Polycomb Bmi1 appartient à la famille des gènes Polycomb qui forment des complexes multimériques impliqués dans la répression de l’expression génique via le remodelage de la chromatine. Au niveau biologique, le gène Bmi1 régule, entre autre, le contrôle de la prolifération cellulaire, le métabolisme des radicaux libres, et la réparation de l’ADN. Récemment, il a été démontré que Bmi1 joue un rôle critique dans la prolifération et l’auto-renouvellement d’un groupe de RPCs immatures. De plus, Bmi1 est essentiel au développement post-natal de la rétine. L'objectif de cette étude est d'analyser le rôle de Bmi1 dans le développement et la survie des photorécepteurs chez la souris. Nos résultats révèlent un phénotype de dégénérescence des photorécepteurs de types cônes chez notre modèle de souris déficiente pour Bmi1. Les bâtonnets sont insensibles à la mutation. De plus, Bmi1 est exprimé de façon prédominante dans les cônes. Nos expériences de culture de cellules rétiniennes suggèrent que le phénotype est cellule-autonome. Par ailleurs, la co-délétion du gène Chk2, membre de la réponse aux dommages à l'ADN, permet de ralentir la progression du phénotype. Les rétines Bmi1-/- et Bmi1-/-Chk2-/- présentent une augmentation importante des dommages oxydatifs à l'ADN. Ces résultats suggèrent que le stress oxydatif pourrait jouer un rôle important dans la survie des cônes. L'étude du rôle du gène Polycomb Bmi1 dans les photorécepteurs est importante pour une meilleure compréhension des mécanismes contribuant à la survie des cônes et pourrait mener à la découverte de nouveaux traitements des maladies dégénératives des cônes.

Au cours des dernières années, un intérêt grandissant concernant les rôles physiologiques des endocannabinoïdes (eCBs) a été observé. Le système eCB est une cible attrayante pour la modulation du système immunitaire et de la douleur périphérique. Bien que le récepteur CB1 soit distribué dans le système nerveux, le récepteur CB2 est traditionnellement associé au système immunitaire. Ce dogme fait maintenant l’objet d’un débat depuis la découverte de l’expression du récepteur CB2 dans certains neurones. La rétine est un modèle important pour l’étude de processus neuronaux. La présence du récepteur CB1 y a été démontrée. Des études fonctionnelles rapportent que l’activation des récepteurs cannabinoïdes affecte le fonctionnement de plusieurs cellules rétiniennes. À ce jour, aucune étude ne s’est intéressée au rôle global des récepteurs CB1 et CB2 dans la rétine. Nous avons investigué les conséquences de l’élimination du récepteur CB1 (cnr1-/-) ou du récepteur CB2 (cnr2-/-) sur la fonction rétinienne mesurée par électrorétinographie. Nous avons également caractérisé la distribution du récepteur CB2 dans la rétine. Pour ce faire, nous avons comparé la spécificité de plusieurs anticorps dirigés contre le récepteur CB2. Seulement l’un des anticorps testés a montré une spécificité satisfaisante. Il a permis de détecter la présence du récepteur CB2 dans les cônes, les bâtonnets, les cellules horizontales, amacrines, bipolaires et ganglionnaires. Nos résultats d’électrorétinographie indiquent que seules les souris cnr2-/- présentent une amplitude accrue de l’onde a des ERG, en conditions scotopiques. En conditions photopiques, l’amplitude de l’onde b des souris cnr2-/- montre un schéma d’adaptation à la lumière différent des autres groupes. Aucun effet significatif n’a été observé chez les animaux cnr1-/-. Ces résultats permettent de conclure que les récepteurs CB1 et CB2 jouent des rôles différents dans le traitement visuel et que le récepteur CB2 semble être impliqué dans l’établissement des réponses rétiniennes.
