Hygrophila ( Hygrophila polysperma (Roxb.) T. Anderson) is a plants which forms serious aquatic weed problems. Both submerged and emergent growth forms occur. Nutritional studies with a controlled release fertilizer and sediments collected from hygrophila-infested areas were conducted with the emergent growth habit to provide insights into growth of this introduced plant. Plant dry weights for experimental 16- week culture periods with low average temperatures were associated with low amounts of hygrophila biomass as compared to culture periods with high average temperatures. Hygrophila cultured in sand rooting media with the controlled release fertilizer produced as much as 20 times more dry weight than plants cultured in sediments only. First-degree linear regression statistics showed hygrophila dry weights were highly related to ammonia nitrogen, magnesium, sodium, and pH values in the sediments. These findings show the close relationship of the emergent growth habit of hygrophila to sediment nutrients. Analyses for certain sediment characteristics may provide an indication of the potential growth that may be expected for weed infestations of this plant. Hygrophila grows year round in south Florida; however, visual observations of canals and other bodies of water indicate that lower amounts of hygrophila plants occur during the cooler months of year than during the summer season. These findings show the seasonal growth of emergent hygrophila occurs with biomass dependent on both sediment nutrients and temperature.

Thirty sites were sampled in southern Biscayne Bay and Manatee Bay in December 1999 to determine the extent of toxicity in sediments. Analyses and assays included: pesticides and phenols in seawater; chemical contaminants in sediment; amphipod mortality, HRGS P450, sea urchin sperm fertilization and embryology, MicrotoxTM, MutatoxTM, grass shrimp AChE and juvenile clam mortality assays; sea urchin sperm, amphipod and oyster DNA damage; and benthic community assessment. Sediment sites near the mouth of canals showed evidence of contamination. Contaminant plumes and associated toxicity do not appear to extend seaward of the mouth of the canals in an appreciable manner. Concentrations of contaminants in the sediments in open areas of Biscayne and Manatee Bays are generally low. (PDF contains 140 pages)

The toxicity of sediments in Biscayne Bay and many adjoining tributaries was determined as part of a bioeffects assessments program managed by NOAA’s National Status and Trends Program. The objectives of the survey were to determine: (1) the incidence and degree of toxicity of sediments throughout the study area; (2) the spatial patterns (or gradients) in chemical contamination and toxicity, if any, throughout the study area; (3) the spatial extent of chemical contamination and toxicity; and (4) the statistical relationships between measures of toxicity and concentrations of chemicals in the sediments. The survey was designed to characterize sediment quality throughout the greater Biscayne Bay area. Surficial sediment samples were collected during 1995 and 1996 from 226 randomly-chosen locations throughout nine major regions. Laboratory toxicity tests were performed as indicators of potential ecotoxicological effects in sediments. A battery of tests was performed to generate information from different phases (components) of the sediments. Tests were selected to represent a range in toxicological endpoints from acute to chronic sublethal responses. Toxicological tests were conducted to measure: reduced survival of adult amphipods exposed to solid-phase sediments; impaired fertilization success and abnormal morphological development in gametes and embryos, respectively, of sea urchins exposed to pore waters; reduced metabolic activity of a marine bioluminescent bacteria exposed to organic solvent extracts; induction of a cytochrome P-450 reporter gene system in exposures to solvent extracts; and reduced reproductive success in marine copepods exposed to solid-phase sediments. Contamination and toxicity were most severe in several peripheral canals and tributaries, including the lower Miami River, adjoining the main axis of the bay. In the open basins of the bay, chemical concentrations and toxicity generally were higher in areas north of the Rickenbacker Causeway than south of it. Sediments from the main basins of the bay generally were less toxic than those from the adjoining tributaries and canals. The different toxicity tests, however, indicated differences in severity, incidence, spatial patterns, and spatial extent in toxicity. The most sensitive test among those performed on all samples, a bioassay of normal morphological development of sea urchin embryos, indicated toxicity was pervasive throughout the entire study area. The least sensitive test, an acute bioassay performed with a benthic amphipod, indicated toxicity was restricted to a very small percentage of the area. Both the degree and spatial extent of chemical contamination and toxicity in this study area were similar to or less severe than those observed in many other areas in the U.S. The spatial extent of toxicity in all four tests performed throughout the bay were comparable to the “national averages” calculated by NOAA from previous surveys conducted in a similar manner. Several trace metals occurred in concentrations in excess of those expected in reference sediments. Mixtures of substances, including pesticides, petroleum constituents, trace metals, and ammonia, were associated statistically with the measures of toxicity. Substances most elevated in concentration relative to numerical guidelines and associated with toxicity included polychlorinated biphenyls, DDT pesticides, polynuclear aromatic hydrocarbons, hexachloro cyclohexanes, lead, and mercury. The toxicity of sediments in Sabine Lake, Texas, and adjoining Intracoastal Waterway canals was determined as part of bioeffects assessment studies managed by NOAA’s National Status and Trends Program. Tests were selected to represent a range in toxicological endpoints from acute to chronic sublethal responses. Toxicological tests were conducted to measure: reduced survival of adult amphipods exposed to solid-phase sediments; impaired fertilization success and abnormal morphological development in gametes and embryos, respectively, of sea urchins exposed to pore waters; reduced metabolic activity of a marine bioluminescent bacteria exposed to organic solvent extracts; and induction of a cytochrome P-450 reporter gene system in exposures to solvent extracts of the sediments. Chemical analyses were performed on portions of each sample to quantify the concentrations of trace metals, polynuclear aromatic hydrocarbons, and chlorinated organic compounds. Correlation analyses were conducted to determine the relationships between measures of toxicity and concentrations of potentially toxic substances in the samples. Based upon the compilation of results from chemical analyses and toxicity tests, the quality of sediments in Sabine Lake and vicinity did not appear to be severely degraded. Chemical concentrations rarely exceeded effects-based numerical guidelines, suggesting that toxicant-induced effects would not be expected in most areas. None of the samples was highly toxic in acute amphipod survival tests and a minority (23%) of samples were highly toxic in sublethal urchin fertilization tests. Although toxic responses occurred frequently (94% of samples) in urchin embryo development tests performed with 100% pore waters, toxicity diminished markedly in tests done with diluted pore waters. Microbial bioluminescent activity was not reduced to a great degree (no EC50 <0.06 mg/ml) and cytochrome P-450 activity was not highly induced (6 samples exceeded 37.1 ug/g benzo[a]pyrene equivalents) in tests done with organic solvent extracts. Urchin embryological development was highly correlated with concentrations of ammonia and many trace metals. Cytochrome P450 induction was highly correlated with concentrations of a number of classes of organic compounds (including the polynuclear aromatic hydrocarbons and chlorinated compounds). [EN] Panic disorder is a highly prevalent neuropsychiatric disorder that shows co-occurrence with substance abuse. Here, we demonstrate that TrkC, the high-affinity receptor for neurotrophin-3, is a key molecule involved in panic disorder and opiate dependence, using a transgenic mouse model (TgNTRK3). Constitutive TrkC overexpression in TgNTRK3 mice dramatically alters spontaneous firing rates of locus coeruleus (LC) neurons and the response of the noradrenergic system to chronic opiate exposure, possibly related to the altered regulation of neurotrophic peptides observed. Notably, TgNTRK3 LC neurons showed an increased firing rate in saline-treated conditions and profound abnormalities in their response to met5-enkephalin. Behaviorally, chronic morphine administration induced a significantly increased withdrawal syndrome in TgNTRK3 mice. In conclusion, we show here that the NT-3/TrkC system is an important regulator of neuronal firing in LC and could contribute to the adaptations of the noradrenergic system in response to chronic opiate exposure. Moreover, our results indicate that TrkC is involved in the molecular and cellular changes in noradrenergic neurons underlying both panic attacks and opiate dependence and support a functional endogenous opioid deficit in panic disorder patients.

Irrigation in the eleven River Basin Authorities and in particular in the South Chad Irrigation Project and the Baga Polder Project is increasing and demands high yearly investments for aquatic weed control in canals and drains. If the weeds are biologically controlled by the grass carp (Ctenopharyngodon idella Val.), the costs will be turned into profit, particularly when the fish production (resident fish plus grass carp) is harvested and sold for food. The use of irrigation canals and drains for aquaculture in the form of fish polyculture may be a wise step towards increased fish production. This paper highlights the concept of fish production from aquatic weed control and concludes that it is a proven profitable venture in several countries

Various modern aquaculture practices applied in fish production especially in Asia are reviewed. The vast Nigerian aquatic medium of numerous water bodies like rivers, streams, lakes reservoirs, flood plains, irrigation canals, coastal swamps offer great potentials for aquaculture production, if optimally utilized. Constraints to modernization of aquaculture in Nigeria among other factors are: 1) a serious shortage of trained manpower; 2) lack of knowledge on profitability of aquaculture as an industry; 3) limited availability of fund (or capital); 4) non-recognition of indigenous trained aquaculture personnel; 5) inadequate data base on the biology and ecological requirements of endemic fish species with aquaculture potentials; 6) insufficient data on production and management techniques; and 7) lack of rational aquaculture development planning. Recommendations are made towards combating these constraints

Details are given of a study carried out in Nigeria, to introduce the practice of fish-cum-rice culture, using Sarotherodon galilaeus. Two plots each measuring 360m super(2) were used for this study and were compared with the farmer's two plots measuring 300m super(2) and 350m super(2). The plots were modified and had two central canals. Rice seedlings were transplanted into the plots after 19 days using a planting distance of 20 x 20cm. Three rice seedlings were planted per hole, using rice variety FARO 40, and grown for 90 days. About 240 and 180 S. galilaeus fingerlings of mean weight of 30g and 26g were stocked in the two experimental plots, respectively. They were fed with pelleted feed of 25% C.P. and monitored for 100 days. A yield of 22.8kg was obtained in plot A while 15.66kg was obtained in plot B. A rice yield of 250kg (i.e 5 bags) was obtained in each of the plots. The results obtained were compared with plots with no fish

As background to a study of the application of astracods in environmental archaeology, a number of sites in South Wales were visited and sampled. Sites included seven broad environmental categories consisting of lakes, permanent ponds, non-permanent ponds, semi-static canals and reens (drainage ditches), non-permanent small lotic water-bodies, permanent fast-flowing waters and wells. In all, twenty-three species were recorded, and with one exception all belonged to the predominately freshwater Cypridoidea. Overall the most commonly encountered species in South Wales was Cypria ophthalmica. Comparing finds with earlier records, it would appear that Ilyocypris bradyi, Candona pratensis, Eucypris lilljeborgi, Herpetocypris chevreuxi, Potamocypris variegata, P. similis and P. pallida are new additions to the Welsh fauna.

O presente estudo objetivou testar experimentalmente a existência da possível correlação entre a penetração de cimento nos túbulos dentinários e a qualidade do selamento. Foram utilizados 60 incisivos centrais superiores humanos que formaram um único grupo experimental. Após a eliminação das porções coronárias, as raízes foram padronizadas em 13 mm de comprimento. A instrumentação dos canais foi realizada no sentido coroa-ápice, e o comprimento de trabalho estabelecido a 1 mm aquém do forame apical. Como solução irrigadora foi empregado o NaOCl a 5,25% e ao final, EDTA a 17%. Em seguida, todos os canais foram obturados com guta-percha e cimento AH Plus marcado com um corante fluorescente. Para determinar a qualidade do selamento das obturações endodônticas, as amostras foram submetidas ao modelo de infiltração de glicose sob pressão. As raízes foram montadas em um dispositivo de dupla-câmara selada para permitir a infiltração da glicose. Como controle negativo foram utilizados 4 dentes hígidos, e como controle positivo, 2 dentes instrumentados porém, não obturados. Foram utilizados 0,75 mL de solução de glicose a 1 mol/L na câmara superior e 0,75 mL de água deionizada na câmara inferior. Os dispositivos foram conectados a um sistema de distribuição de pressão desenvolvido com o objetivo de permitir a infiltração de 32 amostras em uma mesma etapa. A solução de glicose foi forçada apicalmente sob uma pressão de 15 psi durante 1 hora. Uma alíquota de 50 L foi coletada da câmara inferior para quantificar a glicose infiltrada. A concentração de glicose foi determinada através de um método enzimático com o auxílio do Kit Glucose HK e de um espectrofotômetro em um comprimento de onda de 340 nm. Na sequência, as amostras foram desacopladas dos corpos de prova, embutidas em resina epóxi e cortadas em 3 secções transversais. Uma sequência de preparação metalográfica padrão foi realizada para permitir a observação da penetração de cimento nos túbulos dentinários por meio de microscopia confocal e óptica. Os dados obtidos nos 2 experimentos foram cruzados pelo teste de correlação de Spearman, o qual revelou a inexistência de qualquer possibilidade de correlação (r = 0,12). Com base nesses resultados, o presente trabalho concluiu que, dentro das condições experimentais usadas, a quantidade de cimento presente dentro dos túbulos dentinários não teve relação com a qualidade do selamento produzido.

In species of Cladocera not forming ephippia, the latent eggs have a sheath formed by glands of the reproductive canals. Representatives of the families Daphniidae and Moinidae Goulden, 1968, in connection with the formation of their complex-structured ephippia, lost these glands. It was investigated whether there are such glands in species the latent eggs of which are enclosed in primitive ephippia. For this, with the help of histological methods, 55 females of Acroperus elongatus (Sars) (Chydoridea) and 88 females of Lathonura rectirostris (O. F. Muller), (Macrothricidae) were collected near Leningrad, were studied.

O propósito desse trabalho foi verificar a retenção de pinos fibro resinosos cimentados em canais radiculares alargados simulando raízes extensamente comprometidas, fabricados por duas técnicas usadas para diminuir sua desadaptação; comparando-as ao pino fixado somente com cimento. Foram utilizados vinte e quatro raízes de dentes humanos unirradiculares, padronizadas com 15.0 mm de comprimento e 5.0 0.3 mm de diâmetro. As raízes foram incluídas em resina acrílica e divididas em grupos de acordo com a técnica usada: grupo I - pino DC White Post no2 cimentado com sistema adesivo quimicamente ativado e com cimento resinoso dual; grupo II mesmo pino reanatomizado com resina composta para copiar a anatomia do canal radicular, cimentado da mesma forma; e grupo III mesmo pino associado a três pinos acessórios, cimentados do mesmo modo. O canal radicular teve seu diâmetro padronizado pela broca no2 para o pino DC em uma profundidade de 12.0 mm e alargado com uma broca tronco cônica em uma profundidade de 10.0 mm. As oito raízes de cada grupo foram seccionadas transversalmente em três discos de 3.0 mm, a partir da cervical para a execução de um ensaio de extrusão, descartando-se os últimos 2.0 mm, que serviram somente para centralizar o pino. Os valores de retenção foram registrados e tratados estatisticamente por ANOVA e pelo teste SNK (p<0.05). Diferenças significativas foram observadas entre três porções radiculares investigadas em todos os grupos, com os valores de retenção diminuindo da cervical para apical. A retenção na porção apical do grupo com pinos customizado com resina foi estatisticamente maior que na mesma região dos demais grupos. Nenhuma diferença foi encontrada entre o grupo com pinos acessórios e o grupo somente com pino e cimento nessa parte da raiz. Nenhuma diferença foi observada comparando as porções cervical e média dos diferentes grupos. Os tipos de falha após o teste de extrusão foram observados em microscópio eletrônico de varredura com aumento de 200, 600 e 1000 vezes. Elas ocorreram exclusivamente entre o pino e o cimento ou a resina composta. A camada de adesão (camada híbrida) foi mais facilmente observada nas porções cervical e média de todos os grupos. Isso sugere que a retenção nas porções apicais da raiz é predominantemente friccional. Uma vez que pinos acessórios somente alcançam até a porção média do canal radicular, a retenção do pino não é aumentada com essa técnica.

O presente trabalho teve por objetivo quantificar, comparativamente, a área de preenchimento de dois materiais obturadores sólidos, cones de guta-percha (GP) e cones de Resilon (R), no terço apical de incisivos inferiores humanos, ex vivo, obturados pela técnica da onda contínua de condensação. Os espécimes foram submetidos a um protocolo, desde a cirurgia de acesso até o final do preparo químico-mecânico e divididos aleatoriamente em dois grupos, de 21 dentes cada, de acordo com o material utilizado. Não foi utilizado cimento endodôntico em nenhuma das amostras. Após a obturação, as amostras foram seccionadas transversalmente em dois níveis, a 3 e a 5mm do ápice, e subdivididas em grupos de acordo com a altura de corte e do material obturador, sendo estabelecido: GP3 (guta-percha com corte a 3mm), GP5 (guta-percha com corte a 5mm), R3 (Resilon com corte a 3mm) e R5 (Resilon com corte a 5mm). Posteriormente, as amostras foram submetidas a um processo de lixamento e polimento e examinadas em microscópio óptico por reflexão com aumento de 50x a 100x. Para a análise e processamento digital das imagens, foi utilizado o sistema de imagens Axio Vision 4.6 para Windows, sendo obtidas as medidas para cada área observada em micrômetros (μm); uma da área da cavidade, e outra da área de material obturador. Foi aferido o grau de circularidade de cada amostra, por uma fórmula matemática utilizada automaticamente pelo programa, onde 1 (um) é considerado o círculo perfeito e, quanto mais achatado o canal, mais tendente a 0 (zero) nesta escala. Obteve-se a área do canal, a circularidade de 0 a 1, a área preenchida pelo material obturador e, a porcentagem da área de preenchimento do material obturador em relação à área do canal. Foi realizado o cruzamento dos grupos dois a dois pelo teste t de Student, sendo verificada diferença estatisticamente significante entre os grupos GP3 e R3, tendo o grupo R3 apresentado maior porcentagem de área do canal radicular preenchida pelo material obturador em suas amostras (p<0,05). Na relação da circularidade com a quantidade de preenchimento, com o teste de Correlação de Pearson, não foi observada forte correlação entre a forma final do canal (relação de circularidade) e a quantidade de preenchimento do canal radicular pelos materiais obturadores testados. Conclui-se que houve grande variação de preenchimento mínimo e máximo em todos os grupos testados e o Resilon apresentou maior porcentagem de preenchimento de área do canal radicular em suas amostras.

O objetivo deste estudo foi avaliar a espessura mínima radicular remanescente e o desgaste porcentual do terço cervical em raízes mesiais de molares inferiores, após a instrumentação com as técnicas ProTaper Universal e Lima única F2. Foram obtidos 100 primeiros molares inferiores com raízes completamente separadas. Desse total, foram selecionados e incluídos no estudo somente 22 que possuíam dois canais distintos na raiz mesial, comprimento entre 20 e 22 mm e grau de curvatura da raiz mesial com angulação variando entre 10 e 20. Destes, 8 foram eliminados por possuírem uma anatomia muito discrepante, o que limitava o processamento e análise digital das imagens (PADI). Os dentes foram acessados e a patência apical foi realizada em todos os canais determinando o comprimento de trabalho. Em cada dente, cada canal mesial foi instrumentado por uma técnica diferente. As amostras foram posicionadas em um dispositivo de montagem e digitalizadas através de microtomografia computadorizada antes e depois de serem completamente instrumentadas. O volume de interesse, correspondente à região de zona de risco, com uma grande margem de tolerância, tanto em direção apical quanto em direção cervical, foi determinado por 234 fatias, totalizando um comprimento vertical de 3,5 mm, para avaliação quantitativa comparativa. Através de PADI mediu-se, de forma automática, a espessura mínima radicular nos dois canais mesiais, antes e após a instrumentação, para todas as fatias de todos os dentes. A partir destes dados foi calculado o desgaste porcentual. Após o tratamento estatístico das mais de 6500 medidas obtidas, pôde-se concluir que não existiu diferença no desgaste da zona de risco produzido pelas duas técnicas de instrumentação testadas. Em todos os casos a espessura radicular remanescente permaneceu dentro de uma margem de segurança, não havendo, portanto, nenhum caso de rasgo ou perfuração. Dessa forma, ambas as técnicas estudadas foram consideradas seguras quanto ao desgaste da zona de risco da raiz mesial dos molares inferiores.

Neurons obtained directly from human somatic cells hold great promise for disease modeling and drug screening. Available protocols rely on overexpression of transcription factors using integrative vectors and are often slow, complex, and inefficient. We report a fast and efficient approach for generating induced neural cells (iNCs) directly from human hematopoietic cells using Sendai virus. Upon SOX2 and c-MYC expression, CD133-positive cord blood cells rapidly adopt a neuroepithelial morphology and exhibit high expansion capacity. Under defined neurogenic culture conditions, they express mature neuronal markers and fire spontaneous action potentials that can be modulated with neurotransmitters. SOX2 and c-MYC are also sufficient to convert peripheral blood mononuclear cells into iNCs. However, the conversion process is less efficient and resulting iNCs have limited expansion capacity and electrophysiological activity upon differentiation. Our study demonstrates rapid and efficient generation of iNCs from hematopoietic cells while underscoring the impact of target cells on conversion efficiency.
