Os conídios de Mycosphaerella fijiensis agente causal da Sigatoka-negra da bananeira (Musa spp.) podem ser disseminados a longas distâncias, aderidos em diversos materiais, como tecidos e caixas. Neste contexto, o presente trabalho teve como objetivo selecionar desinfestantes eficientes para inibir a germinação de conídios do patógeno. Foram testados benomil, amônia quaternária, digluconato de chlorhexidina, formaldeído, óleo essencial de pimenta longa (OEPL), Ecolife-40, thiabendazole e hipoclorito de sódio, nas concentrações de 1, 5, 10, 25, 50 e 100 mg/l. Os conídios do isolado LPM 472 foram produzidos em BDA. Para os testes transferiu-se 1 ml de uma suspensão de 10(5) conídios/ml de M. fijiensis para cada tubo de ensaio com 1ml de desinfestante na sua respectiva concentração. Após 30 h de incubação à temperatura ambiente, com o auxílio de um microscópio óptico, quantificou-se a viabilidade de 100 conídios, computando-se apenas os germinados. A amônia quaternária, o benomil, o Ecolife-40 e o thiabendazole a 100 mg/l inibiram totalmente a germinação. Esses mesmos produtos, aplicados em frutos colhidos em área com a doença, apresentaram a mesma eficiência, via imersão ou pulverização, nas concentrações de 100 e 200 mg/l.

Subcellular changes are relevant to understand plant organogenesis and embryogenesis in the early stages of cell development. The cytology during cell development in tissue culture is however still poorly characterized. This study aimed to characterize the ultrastructural differences related to callogenesis of anthers, ovaries, leaf and nodal segments of Inga vera Willd. subsp. Affinis (DC.) T.D. Penn. Flower buds, nodal segments and leaves were disinfected and inoculated in test tubes containing MS medium with 3% sucrose and 4.5µM 2.4-D, except for leaf callogenesis, where 9µM of this auxin was used, and for the callogenesis of anthers and ovaries, where the culture medium was enriched with 0.25% activated charcoal and 90µM PVP. After 45 days in culture medium, the anther, ovary, leaf and nodal segment calli were fixed in Karnovisky and prepared for visualization by scanning and transmission electron microscopy. Ultrastructural differences were observed among the callus cells of anthers, ovaries, segments and leaves. There was no evidence of somatic embryo formation in the anther, leaf and nodal segment calli, in spite of some embryogenic characteristics in the cells. The ovary calli, with indications of embryo formation, seem to be the most responsive explant source for embryogenesis.

In this article, a methodology is used for the simultaneous determination of the effective diffusivity and the convective mass transfer coefficient in porous solids, which can be considered as an infinite cylinder during drying. Two models are used for optimization and drying simulation: model 1 (constant volume and diffusivity, with equilibrium boundary condition), and model 2 (constant volume and diffusivity with convective boundary condition). Optimization algorithms based on the inverse method were coupled to the analytical solutions, and these solutions can be adjusted to experimental data of the drying kinetics. An application of optimization methodology was made to describe the drying kinetics of whole bananas, using experimental data available in the literature. The statistical indicators enable to affirm that the solution of diffusion equation with convective boundary condition generates results superior than those with the equilibrium boundary condition.

Changes in the polyamine content were analyzed in different embryo developmental stages and tissues during seed development in the conifer Araucaria angustifolia (Bert.) O. Ktze. Free polyamine contents varied according to the tissue and stage of embryo development, the highest levels occurring in the embryonic axis at the early stages, when putrescine and spermidine were most abundant. The levels of spermidine were higher from the stage where cotyledons arise, whereas putrescine decreases. The putrescine/spermine+spermidine ratio was higher during the initial phases of seed development, corresponding to cell multiplication and elongation, with a decrease in the final stages, corresponding to stabilization of the dry matter content.

Gene therapy for hypertension is needed for the next generation of antihypertensive drugs. Current drugs, although effective, have poor compliance, are expensive and short-lasting (hours or one day). Gene therapy offers a way to produce long-lasting antihypertensive effects (weeks, months or years). We are currently using two strategies: a) antisense oligodeoxynucleotides (AS-ODN) and b) antisense DNA delivered in viral vectors to inhibit genes associated with vasoconstrictive properties. It is not necessary to know all the genes involved in hypertension, since many years of experience with drugs show which genes need to be controlled. AS-ODN are short, single-stranded DNA that can be injected in naked form or in liposomes. AS-ODN, targeted to angiotensin type 1 receptors (AT1-R), angiotensinogen (AGT), angiotensin converting enzyme, and ß1-adrenergic receptors effectively reduce hypertension in rat models (SHR, 2K-1C) and cold-induced hypertension. A single dose is effective up to one month when delivered with liposomes. No side effects or toxic effects have been detected, and repeated injections can be given. For the vector, adeno-associated virus (AAV) is used with a construct to include a CMV promoter, antisense DNA to AGT or AT1-R and a reporter gene. Results in SHR demonstrate reduction and slowing of development of hypertension, with a single dose administration. Left ventricular hypertrophy is also reduced by AAV-AGT-AS treatment. Double transgenic mice (human renin plus human AGT) with high angiotensin II causing high blood pressure, treated with AAV-AT1-R-AS, show a normalization of blood pressure for over six months with a single injection of vector. We conclude that ODNs will probably be developed first because they can be treated like drugs for the treatment of hypertension with long-term effects. Viral vector delivery needs more engineering to be certain of its safety, but one day may be used for a very prolonged control of blood pressure.

Paroxysmal nocturnal hemoglobinuria (PNH) is an acquired clonal syndrome characterized by intravascular hemolysis mediated by complement, thrombotic events and alterations in hematopoiesis. Basically, the molecular events which underlie the complexity of the syndrome consist of the absence of the glycosylphosphatidylinositol (GPI) anchor as a consequence of somatic mutations in the PIG-A gene, located on the X chromosome. The GPI group is responsible for the attachment of many proteins to the cytoplasmic membrane. Two of them, CD55 and CD59, have a major role in the inhibition of the action of complement on the cellular membrane of blood cells. The absence of GPI biosynthesis can lead to PNH. Since mutations in the PIG-A gene are always present in patients with PNH, the aim of this study was to characterize the mutations in the PIG-A gene in Brazilian patients. The analysis of the PIG-A gene was performed using DNA samples derived from bone marrow and peripheral blood. Conformation-sensitive gel electrophoresis was used for screening the mutation and sequencing methods were used to identify the mutations. Molecular analysis permitted the identification of three point mutations in three patients: one G->A transition in the 5' portion of the second intron, one T->A substitution in the second base of codon 430 (Leu430->stop), and one deletion deltaA in the third base of codon 63. This study represents the first description of mutations in the PIG-A gene in a Brazilian population.

We investigated the somatic maturation of neonate rats treated during the suckling period with citalopram, a selective serotonin reuptake inhibitor. Groups with 6 male neonates were randomly assigned to different treatments 24 h after birth. Each litter was suckled by one of the dams until the 21st postnatal day. Body weight, head axis and tail length were measured daily from the 1st to the 21st postnatal day. Time of ear unfolding, auditory conduit opening, incisor eruption, and eye opening was determined. Pups received 5 mg (Cit5), 10 mg (Cit10) or 20 mg/kg (Cit20) citalopram sc, or saline (0.9% NaCl, w/v, sc). Compared to saline, body weight was lower (24.04%, P < 0.01) for Cit10 from the 10th to the 21st day and for Cit20 from the 6th to the 21st day (38.19%, P < 0.01). Tail length was reduced in the Cit20 group (15.48%, P < 0.001) from the 8th to the 21st day. A reduction in mediolateral head axis (10.53%, P < 0.05) was observed from the 11th to the 21st day in Cit10 and from the 6th to the 21st day in Cit20 (13.16%, P < 0.001). A reduction in anteroposterior head axis was also observed in the Cit20 group (5.28%, P < 0.05) from the 13th to the 21stday. Conversely, this axis showed accelerated growth from the 12th to the 21stday in the Cit5 group (13.05%, P < 0.05). Auditory conduit opening was delayed in the Cit5 and Cit20 groups and incisor eruption was delayed in all citalopram groups. These findings show that citalopram injected during suckling to rats induces body alterations and suggest that the activity of the serotoninergic system participates in growth mechanisms.

The objective of the present study was to determine the frequency of somatic chromosomal anomalies and Y chromosomal microdeletions (azoospermia factor genes, AZF) in infertile males who seek assisted reproduction. These studies are very important because the assisted reproduction techniques (mainly intracytoplasmic sperm injection) bypass the natural selection process and some classical chromosomal abnormalities, microdeletions of AZF genes or some deleterious genic mutations could pass through generations. These genetic abnormalities can cause in the offspring of these patients male infertility, ambiguous external genitalia, mental retardation, and other birth defects. We studied 165 infertile men whose infertility was attributable to testicular problems (60 were azoospermic, 100 were oligospermic and 5 were asthenospermic). We studied 100 metaphases per patient with GTG banding obtained from temporary lymphocyte culture for chromosomal abnormality detection and performed a genomic DNA analysis using 28 Y chromosome-specific sequence-tagged sites for Y AZF microdeletion detection. Karyotyping revealed somatic anomalies in 16 subjects (16/165 = 9.6%). Of these 16, 12 were in the azoospermic group (12/60 = 20%) and 4 were in the oligospermic group (4/100 = 4%). The most common chromosomal anomaly was Klinefelter syndrome (10/165 = 6%). Microdeletions of AZF genes were detected in 12 subjects (12/160 = 7.5%). The frequencies detected are similar to those described previously. These results show the importance of genetic evaluation of infertile males prior to assisted reproduction. Such evaluation can lead to genetic counseling and, consequently, to primary and secondary prevention of mental retardation and birth defects.

The distribution of psychiatric disorders and of chronic medical illnesses was studied in a population-based sample to determine whether these conditions co-occur in the same individual. A representative sample (N = 1464) of adults living in households was assessed by the Composite International Diagnostic Interview, version 1.1, as part of the São Paulo Epidemiological Catchment Area Study. The association of sociodemographic variables and psychological symptoms regarding medical illness multimorbidity (8 lifetime somatic conditions) and psychiatric multimorbidity (15 lifetime psychiatric disorders) was determined by negative binomial regression. A total of 1785 chronic medical conditions and 1163 psychiatric conditions were detected in the population concentrated in 34.1 and 20% of respondents, respectively. Subjects reporting more psychiatric disorders had more medical illnesses. Characteristics such as age range (35-59 years, risk ratio (RR) = 1.3, and more than 60 years, RR = 1.7), being separated (RR = 1.2), being a student (protective effect, RR = 0.7), being of low educational level (RR = 1.2) and being psychologically distressed (RR = 1.1) were determinants of medical conditions. Age (35-59 years, RR = 1.2, and more than 60 years, RR = 0.5), being retired (RR = 2.5), and being psychologically distressed (females, RR = 1.5, and males, RR = 1.4) were determinants of psychiatric disorders. In conclusion, psychological distress and some sociodemographic features such as age, marital status, occupational status, educational level, and gender are associated with psychiatric and medical multimorbidity. The distribution of both types of morbidity suggests the need of integrating mental health into general clinical settings.

Recent advances have raised hope that transplantation of adherent somatic cells could provide dramatic new therapies for various diseases. However, current methods for transplanting adherent somatic cells are not efficient enough for therapeutic applications. Here, we report the development of a novel method to generate quasi-natural cell blocks for high-efficiency transplantation of adherent somatic cells. The blocks were created by providing a unique environment in which cultured cells generated their own extracellular matrix. Initially, stromal cells isolated from mice were expanded in vitro in liquid cell culture medium followed by transferring the cells into a hydrogel shell. After incubation for 1 day with mechanical agitation, the encapsulated cell mass was perforated with a thin needle and then incubated for an additional 6 days to form a quasi-natural cell block. Allograft transplantation of the cell block into C57BL/6 mice resulted in perfect adaptation of the allograft and complete integration into the tissue of the recipient. This method could be widely applied for repairing damaged cells or tissues, stem cell transplantation, ex vivo gene therapy, or plastic surgery.

A hidrólise do amido e a síntese de açúcares durante o amadurecimento da banana são transformações bioquímicas importantes, havendo evidências de que ocorrem de forma homogênea no fruto. Para confirmar este fato, amostras de banana nanicão (Musa spp.) colhidas aos 110 dias pós-antese, foram coletadas no decorrer do amadurecimento e foram determinados os teores de amido, hexoses e sacarose e a atividade das enzimas sacarose-fosfato sintase (SPS) e sacarose sintase (SS) em diferentes partes do fruto. Observou-se que na banana verde, existe mais amido na porção periférica (18%) do que na central (13%). Porém, a sua velocidade de degradação durante o amadurecimento é a mesma, o que resulta em teores diferenciados de amido residual na banana madura. Também o aparecimento e acúmulo de sacarose foi simultâneo nas duas regiões e coincidente com os valores máximos de atividade da SPS. Utilizando-se de técnica de identificação por anticorpos específicos para SS e SPS em tecidos verde e maduro, observou-se uma distribuição homogênea das enzimas e aparente correlação entre a cor desenvolvida e a variação de atividade.

Considerando-se as limitações tecnológicas dos processos tradicionais para produção de banana passa com uma qualidade padrão, a desidratação osmótica surge como uma opção à padronização deste processo. Neste trabalho visou-se determinar parâmetros do processo de desidratação osmótica da banana e avaliar a influência da concentração da solução osmótica nas características físico-químicas do fruto e construir as curvas de secagem da banana pré-tratada através de desidratação osmótica com e sem vácuo. Concluiu-se que é possível obter banana desidratada como produto de umidade intermediária, através de pré-tratamento osmótico seguido de secagem. As características do produto foram influenciadas pela concentração e proporção fruto:xarope do meio osmótico utilizado.

O amido de bananas tem sido pesquisado na área de nutrição a partir da introdução do conceito de Amido Resistente. O amido de Musa AAA-Nanicão e Musa AAB-Terra foram caracterizados quanto as suas respostas fisiológicas [12]. Em continuidade, o presente trabalho estudou características físicas e morfológicas dos grânulos de amido de ambas as espécies de banana comparando-as com amido nativo de milho comercial. O amido de bananas foi extraído segundo CHIANG, CHU & CHU [3]. A morfologia dos grânulos foi realizada após tratamento enzimático in vitro a 37°C/24h com alfa-amilase pancreática. Foram efetuados os respectivos amilogramas e difractogramas de raios-X. Os grânulos de amido da Musa Tipo AAA-Nanicão apresentaram comprimento entre 30-40µm. Em Musa AAB-Terra, os grânulos, também ovais e alongados, eram um pouco menores, 20-30µm. A corrosão enzimática in vitro iniciava-se sobre a superfície anteriormente lisa e formavam estrias superficiais e apicais. A Microscopia Eletrônica de Varredura (MEV) mostrou que a hidrólise in vitro por 24 horas foi pequena e ocorria sobretudo nas camadas amorfas dos grânulos de ambas as espécies. O padrão de corrosão demonstrou-se distinto daquele ocorrido no amido de milho. As suspensões de amido de bananas ao viscosímetro demonstraram forte capacidade de hidratação e menor capacidade de retrogradação em relação ao milho, sobretudo do amido de Musa Tipo AAA-Nanicão; o amido de Musa Tipo AAB-Terra apresentou maior estabilidade de pasta. Na análise de difração de raios-X, os grânulos de bananas apresentaram padrão tipo B e C para Musa Tipo AAA-Nanicão e Musa Tipo AAB-Terra, respectivamente. Conclui-se que os amidos de Musa AAA-Nanicão e Musa AAB-Terra são estruturalmente distintos, justificando as respostas fisiológicas distintas encontradas posteriormente pelos mesmos autores. As distinções das propriedades físicas e bioquímicas obtidas para os grânulos, embora moderadas entre si e bastante distintas em relação ao amido de milho, não explicaram a alta resistência dos amidos das bananas à digestão enzimática. Os parâmetros obtidos nas análises permitiram, pela primeira vez avaliar as características e diferenças entre grânulos de amido de espécies distintas de banana e suas relações ao amido de milho.

Bananas verdes [Nanicão (Musa sp., subgrupo Cavendish) e Prata (Musa sp., subgrupo Prata)] foram estudadas durante o seu amadurecimento. As propriedades físicas (firmeza), físico-químicas (pH, acidez total titulável e sólidos solúveis) e químicas (açúcares, compostos fenólicos e voláteis) foram analisadas e demonstraram diferenças significantes (p < 0,05) entre as bananas. A banana Prata apresentou valores mais altos de compostos fenólicos, sólidos solúveis, açúcares e firmeza do que a banana Nanicão. O método de coleta e análise dos compostos voláteis foram o headspace em dedo frio e a cromatografia gasosa. Os ésteres acetatos, butiratos, isobutiratos e isovaleratos foram predominantes. A banana Prata produziu maiores concentrações de voláteis do que a Nanicão, exceto para os acetatos. O comportamento das curvas de produção dos ésteres seguiu um aumento contínuo, até um pico, para em seguida apresentar uma diminuição no estágio de escurecimento das cascas dos frutos.

Objetivou-se avaliar o efeito do 1-metilciclopropeno (1-MCP) sobre a qualidade e extensão da vida pós-colheita de bananas-maçã. Frutas no grau 2 de coloração da casca foram tratadas com 50 nL.L-1 de 1-MCP durante 0 (controle), 6, 9, 12 e 24 horas. Posteriormente, foram armazenadas sob condição ambiente: 22± 2 ºC 85± 5% UR. Avaliaram-se as características sensoriais: aparência dos frutos no grau 4 (frutos mais verdes que amarelos) e grau 7 (frutos amarelos com pontuações marrons) de coloração da casca; descasque, aroma, sabor, firmeza da polpa e aspecto global nos frutos no grau 7, utilizando-se um teste de aceitabilidade através de escala hedônica de 9 pontos; e ainda a intenção de compra nos frutos nos graus 4 e 7, através de escala de 5 pontos. Avaliou-se também, visualmente, a extensão da vida pós-colheita, e, instrumentalmente, a coloração da casca e firmeza da polpa. Conclui-se que a aplicação de 50 nL.L-1 por um período de 12 horas é a mais adequada, pois promove extensão na vida pós-colheita de bananas-maçã, armazenadas em temperatura ambiente, em aproximadamente 11 dias, sem alterar a qualidade sensorial, a intenção de compra, coloração da casca e firmeza dos frutos quando maduros, comparados ao controle.
