We have previously characterized an infectious mouse mammary tumor virus [(MMTV(SW)] which induces a strong superantigen response in vivo. Here we describe the isolation and characterization of MMTV(C4) which was derived from milk of mice implanted with hyperplastic alveolar nodules. MMTV(C4) stimulates V beta 2 expressing T cells after local injection in vivo. Comparison with known open reading frame (orf) sequences revealed high homology to Mtv-6, an endogenous virus interacting with V beta 3-expressing T cells. The carboxyl-terminal amino acids were, however, altered. High homology including the carboxyl-terminal orf amino acids were found with MMTV(C3H-K). We show here that MMTV(C3H-K) has lost its superantigen function. Sequence comparisons permitted the characterization of few key amino acids which could be important for T cell receptor interaction and superantigen processing.

Sementes de feijoeiro (Phaseolus vulgaris L.) com alto teor de P, obtidas de plantas que receberam adubação foliar, foram utilizadas em um experimento em casa de vegetação para avaliar os efeitos do teor de P na semente, no crescimento e na nodulação do feijoeiro no estádio vegetativo. O experimento tinha arranjo fatorial 2x2x3x3 em blocos ao acaso com quatro repetições: dois cultivares (Carioca e Rio Tibagi), dois teores de P na semente (baixo e alto, correspondentes a 4,3 e 5,6mgg-1, em Carioca, e 4,8 e 6,3mgg-1, em Rio Tibagi), três doses de P aplicado ao solo (0, 30 e 60mgkg-1, respectivamente, P0, P30 e P60) e três épocas de coleta (20, 30 e 40dias após emergência - DAE). O alto teor de P na semente aumentou a matéria seca das plantas de feijoeiro em P0 nas três coletas, e em P30, aos 40DAE. Plantas originadas de sementes com alto teor de P mostraram menor dependência do suprimento de P no solo, quanto à produção de matéria seca, do que aquelas oriundas de sementes com baixo P. O alto teor de P na semente aumentou o número de nódulos em Carioca em P0, aos 20 e 30DAE, e aumentou a massa de nódulos em P30, aos 20DAE nos dois cultivares e, aos 40DAE, em P30 em Carioca e em P60 em Rio Tibagi. A acumulação de N na parte aérea aos 20DAE foi superior nas plantas originadas de sementes com alto teor de P. Segundo os resultados, sementes com alto teor de P obtido via adubação foliar podem aumentar o crescimento, a nodulação e a acumulação de N do feijoeiro em estádios iniciais de crescimento, particularmente sob baixas doses de P aplicado ao solo.

Contractile tension of alveolar epithelial cells plays a major role in the force balance that regulates the structural integrity of the alveolar barrier. The aim of this work was to study thrombin-induced contractile forces of alveolar epithelial cells. A549 alveolar epithelial cells were challenged with thrombin, and time course of contractile forces was measured by traction microscopy. The cells exhibited basal contraction with total force magnitude 55.0 ± 12.0 nN (mean ± SE, n = 12). Traction forces were exerted predominantly at the cell periphery and pointed to the cell center. Thrombin (1 U/ml) induced a fast and sustained 2.5-fold increase in traction forces, which maintained peripheral and centripetal distribution. Actin fluorescent staining revealed F-actin polymerization and enhancement of peripheral actin rim. Disruption of actin cytoskeleton with cytochalasin D (5 µM, 30 min) and inhibition of myosin light chain kinase with ML-7 (10 µM, 30 min) and Rho kinase with Y-27632 (10 µM, 30 min) markedly depressed basal contractile tone and abolished thrombin-induced cell contraction. Therefore, the contractile response of alveolar epithelial cells to the inflammatory agonist thrombin was mediated by actin cytoskeleton remodeling and actomyosin activation through myosin light chain kinase and Rho kinase signaling pathways. Thrombin-induced contractile tension might further impair alveolar epithelial barrier integrity in the injured lung.

Bactérias endofíticas promotoras de crescimento podem aumentar a eficiência nutricional das plantas, favorecendo sua produção. O objetivo deste trabalho foi avaliar a influência de 10 isolados de bactérias endofíticas, previamente selecionados como agentes promotores do crescimento de plantas, sobre a eficiência de absorção, utilização e translocação de nutrientes em plantas de tomateiros em casa de vegetação. Para a introdução das bactérias endofíticas em plântulas de tomateiro cv. Santa Clara, utilizou-se o corte do hipocótilo. Cinqüenta e cinco dias após o transplantio das seções de parte área, as plantas foram coletadas para a determinação da matéria seca da parte aérea e dos teores de macro e micronutrientes. Os teores de N, P, K, Ca, Mg, Cu e Zn na parte aérea e os de N, P, Mg e Mn nas raízes das plantas inoculadas diferiram da testemunha sem inoculação. As bactérias endofíticas Micrococcus sp. (UFLA 11-LS) e Brevundimonas sp. (UFV-E49), identificadas por meio do seqüenciamento do gene 16S do DNA ribossômico, propiciaram a maior eficiência de absorção de P em relação à testemunha. A bactéria endofítica Micrococcus sp. apresentou maior eficiência na utilização de N, P, K, Ca, Mg, S, Cu, Fe e Zn. Os maiores teores de N, P, K, Mg e Zn foram encontrados na parte aérea das plantas inoculadas com Brevundimonas sp. Os resultados deste trabalho indicam que estes isolados de bactérias endofíticas podem aumentar a eficiência nutricional de plantas de tomate.

Recent publications have demonstrated that the protease caspase-1 is responsible for the processing of pro-interleukin 18 (IL-18) into the active form. Studies on cell lines and murine macrophages have shown that the bacterial invasion factor SipB activates caspase-1, triggering cell death. Thus, we investigated the role of SipB in the activation and release of IL-18 in human alveolar macrophages (AM), which are the first line of defense against inhaled pathogens. Under steady-state conditions, AM are a more important source of IL-18 than are dendritic cells (DC) and monocytes. Cytokine production by AM and DC was compared after both types of cells had been infected with a virulent strain of Salmonella enterica serovar Typhimurium and an isogenic sipB mutant, which were used as an infection model. Infection with virulent Salmonella led to marked cell death with features of apoptosis while both intracellular activation and release of IL-18 were demonstrated. In contrast, the sipB mutant did not induce such cell death or the release of active IL-18. The specific caspase-1 inhibitor Ac-YVAD-CMK blocked the early IL-18 release in AM infected with the virulent strain. However, the type of Salmonella infection did not differentially regulate IL-18 gene expression. We concluded that the bacterial virulence factor SipB plays an essential posttranslational role in the intracellular activation of IL-18 and the release of the cytokine in human AM.

O uso agrícola e o manejo de solos de Cerrado frequentemente causam sua compactação, reduzindo sua porosidade e disponibilidade de água e nutrientes, com efeito negativo sobre o crescimento e o desenvolvimento das culturas. O presente trabalho teve por objetivo avaliar os efeitos da densidade do solo e de doses de P no crescimento da soja e do eucalipto em solos com diferentes texturas: um Latossolo Vermelho (LV) muito argiloso e outro Latossolo Vermelho-Amarelo (LVA) textura média. Em vasos plásticos de 1,8 L, com 1,6 dm³ de solo, foram testadas, para o LV, as densidades de 0,90, 1,10 e 1,30 kg dm-3 e as doses de 0, 100, 200, 400 e 800 mg dm-3 de P, e para o LVA, as densidades de 1,30, 1,50 e 1,70 kg dm-3 e as doses de 0, 75, 150, 300 e 600 mg dm-3 de P. As plantas de soja e de eucalipto foram colhidas 40 e 80 dias após a semeadura, respectivamente. Foram avaliadas: matéria seca da parte aérea (MSPA) e de raízes (MSR), acúmulo de P na MSPA (P-PA) e MSR (P-R), nas duas espécies, e altura do eucalipto. Os resultados mostraram que o aumento da densidade do solo influenciou negativamente no crescimento da soja e do eucalipto, especialmente no solo LV e nas maiores doses de P; a resposta das plantas às doses de P aplicadas foi menor com o aumento da densidade do solo; e o aumento da densidade foi mais prejudicial ao crescimento do eucalipto - espécie mais eficiente na recuperação do P aplicado aos solos. De modo geral, o efeito negativo do aumento da densidade dos solos sobre a produção de MSPA das espécies, nas doses menores de P, é compensado quando as maiores doses de P são aplicadas.

The lung possesses specific transport systems that intra- and extracellularly maintain salt and fluid balance necessary for its function. At birth, the lungs rapidly transform into a fluid (Na(+))-absorbing organ to enable efficient gas exchange. Alveolar fluid clearance, which mainly depends on sodium transport in alveolar epithelial cells, is an important mechanism by which excess water in the alveoli is reabsorbed during the resolution of pulmonary edema. In this review, we will focus and summarize on the role of ENaC in alveolar lung liquid clearance and discuss recent data from mouse models with altered activity of epithelial sodium channel function in the lung, and more specifically in alveolar fluid clearance. Recent data studying mice with hyperactivity of ENaC or mice with reduced ENaC activity clearly illustrate the impaired lung fluid clearance in these adult mice. Further understanding of the physiological role of ENaC and its regulatory proteins implicated in salt and water balance in the alveolar cells may therefore help to develop new therapeutic strategies to improve gas exchange in pulmonary edema.

Sodium transport via epithelial sodium channels (ENaC) expressed in alveolar epithelial cells (AEC) provides the driving force for removal of fluid from the alveolar space. The membrane-bound channel-activating protease 1 (CAP1/Prss8) activates ENaC in vitro in various expression systems. To study the role of CAP1/Prss8 in alveolar sodium transport and lung fluid balance in vivo, we generated mice lacking CAP1/Prss8 in the alveolar epithelium using conditional Cre-loxP-mediated recombination. Deficiency of CAP1/Prss8 in AEC induced in vitro a 40% decrease in ENaC-mediated sodium currents. Sodium-driven alveolar fluid clearance (AFC) was reduced in CAP1/Prss8-deficient mice, due to a 48% decrease in amiloride-sensitive clearance, and was less sensitive to beta(2)-agonist treatment. Intra-alveolar treatment with neutrophil elastase, a soluble serine protease activating ENaC at the cell surface, fully restored basal AFC and the stimulation by beta(2)-agonists. Finally, acute volume-overload increased alveolar lining fluid volume in CAP1/Prss8-deficient mice. This study reveals that CAP1 plays a crucial role in the regulation of ENaC-mediated alveolar sodium and water transport and in mouse lung fluid balance.

Diffuse alveolar hemorrhage (DAH) is defined by the presence of red blood cells originating from the lung capillaries or venules within the alveoli. The diagnosis is established on clinical features, radiological pattern, and especially bronchoalveolar lavage. Diffuse alveolar hemorrhage may have many immune or non-immune causes. Immune causes of DAH include vasculitides, connective tissue diseases, especially systemic lupus erythematosus, and antiglomerular basement membrane antibody disease (Goodpasture's syndrome). Treatment is both supportive and causal, often based on high dose corticosteroids and immunosuppressive therapy (especially intravenous cyclophosphamide). Plasma exchanges are performed in antiglomerular basement membrane antibody disease and systemic lupus erythematosus, and are considered in systemic vasculitis. Non-immune causes of DAH mainly include heart diseases, coagulation disorders, infections, drug toxicities and idiopathic DAH. Treatment of non-immune DAH is that of its cause. Whatever the cause, DAH is an emergency requiring prompt assessment and early treatment.

O objetivo deste trabalho foi avaliar a tolerância de juvenis de pirarucu, Arapaima gigas, a concentrações crescentes de amônia na água. Oito peixes (2,6±0,4 kg e 70,5±4,1 cm de comprimento) foram distribuídos aleatoriamente em quatro tanques (0,8x0,9x2,6 m) com 410 L de água tamponada com Na2PO4 e HCl, ambos a 0,1 M, onde ficaram por 33 dias. As concentrações de amônia no plasma sanguíneo dos peixes e na água foram determinadas pelo método do salicilato. A glicose plasmática e os níveis de amônia aumentaram proporcionalmente à concentração de amônia na água. Juvenis de pirarucu toleram altas concentrações de amônia na água.

Two common lung-related complications in the neonate are respiratory distress syndrome, which is associated with a failure to generate low surface tension at the air-liquid interface because of pulmonary surfactant insufficiency, and bronchopulmonary dysplasia (BPD), a chronic lung injury with reduced alveolarization. Surfactant phosphatidylcholine (PC) molecular species composition during alveolarization has not been examined. Mass spectrometry analysis of bronchoalveolar lavage fluid of rodents and humans revealed significant changes in surfactant PC during alveolar development and BPD. In rats, total PC content rose during alveolarization, which was caused by an increase in palmitoylmyristoyl-PC (16:0/14:0PC) concentration. Furthermore, two animal models of BPD exhibited a specific reduction in 16:0/14:0PC content. In humans, 16:0/14:0PC content was specifically decreased in patients with BPD and emphysema compared with patients without alveolar pathology. Palmitoylmyristoyl-PC content increased with increasing intrinsic surfactant curvature, suggesting that it affects surfactant function in the septating lung. The changes in acyl composition of PC were attributed to type II cells producing an altered surfactant during alveolar development. These data are compatible with extracellular surfactant 16:0/14:0PC content being an indicator of alveolar architecture of the lung.

Impairment of lung liquid absorption can lead to severe respiratory symptoms, such as those observed in pulmonary oedema. In the adult lung, liquid absorption is driven by cation transport through two pathways: a well-established amiloride-sensitive Na(+) channel (ENaC) and, more controversially, an amiloride-insensitive channel that may belong to the cyclic nucleotide-gated (CNG) channel family. Here, we show robust CNGA1 (but not CNGA2 or CNGA3) channel expression principally in rat alveolar type I cells; CNGA3 was expressed in ciliated airway epithelial cells. Using a rat in situ lung liquid clearance assay, CNG channel activation with 1 mM 8Br-cGMP resulted in an approximate 1.8-fold stimulation of lung liquid absorption. There was no stimulation by 8Br-cGMP when applied in the presence of either 100 μM L: -cis-diltiazem or 100 nM pseudechetoxin (PsTx), a specific inhibitor of CNGA1 channels. Channel specificity of PsTx and amiloride was confirmed by patch clamp experiments showing that CNGA1 channels in HEK 293 cells were not inhibited by 100 μM amiloride and that recombinant αβγ-ENaC were not inhibited by 100 nM PsTx. Importantly, 8Br-cGMP stimulated lung liquid absorption in situ, even in the presence of 50 μM amiloride. Furthermore, neither L: -cis-diltiazem nor PsTx affected the β(2)-adrenoceptor agonist-stimulated lung liquid absorption, but, as expected, amiloride completely ablated it. Thus, transport through alveolar CNGA1 channels, located in type I cells, underlies the amiloride-insensitive component of lung liquid reabsorption. Furthermore, our in situ data highlight the potential of CNGA1 as a novel therapeutic target for the treatment of diseases characterised by lung liquid overload.

O objetivo deste trabalho foi avaliar a eficiência de tratamentos térmicos no aumento do potencial de frigoconservação de nêsperas 'Fukuhara' e os efeitos desses tratamentos na qualidade e nas propriedades físico-químicas e bioquímicas dos frutos. Os tratamentos de condicionamento térmico foram: armazenamento a 1ºC durante 60 dias (controle); armazenamento a 5ºC durante 60 dias (padrão); armazenamento a 5ºC, durante 7 dias, e 1ºC durante 53 dias; armazenamento a 10ºC, durante 7 dias, e 1ºC durante 53 dias; aquecimento intermitente, em ciclos de 6 dias a 1ºC + 1 dia a 15ºC, durante 60 dias; condicionamento dos frutos a 37ºC, durante 3 horas, e a 1ºC durante 60 dias; e condicionamento dos frutos a 37ºC, durante 6 horas, e a 1ºC durante 60 dias. Os frutos foram mantidos a 85-90% de umidade relativa, durante todo o armazenamento. Foram determinados: firmeza da polpa, índice de escurecimento, acidez titulável, pH, teor de sólidos solúveis, teor de ácido ascórbico, teor de compostos fenólicos, atividade das enzimas fenilalanina amônia-liase (PAL), polifenoloxidase (PPO) e peroxidase (POD) após 15, 30, 45 e 60 dias de armazenamento. Houve correlação entre firmeza de polpa e atividade de POD, após 60 dias de armazenamento refrigerado. O aquecimento intermitente e o aquecimento a 37ºC, durante 3 horas, foram eficientes no controle do escurecimento interno. Os tratamentos térmicos não evitaram o enrijecimento de polpa. Tratamentos térmicos aumentam o potencial de armazenamento de nêsperas sem alterar as características físico-químicas dos frutos.
