An implementable nonlinear control design approach is presented for a supersonic air-breathing ramjet engine. The primary objective is to ensure that the thrust generated by the engine tracks the commanded thrust without violating the operational constraints. An important constraint is to manage the shock wave location in the intake so that it neither gets detached nor gets too much inside the intake. Both the objectives are achieved by regulating the fuel flow to the combustion chamber and by varying the throat area of the nozzle simultaneously. The design approach accounts for the nonlinear cross-coupling effects and nullifies those. Also, an extended Kalman filter has been used to filter out the sensor and process noises as well as to make the states available for feedback. Furthermore, independent control design has been carried out for the actuators. To test the performance of the engine for a realistic flight trajectory, a representative trajectory is generated through a trajectory optimization process, which is augmented with a newly-developed finite-time state dependent Riccati equation technique for nullifying the perturbations online. Numerical simulation of separated flows in rocket nozzles is challenging because existing turbulence models are unable to predict it correctly. This paper addresses this issue with the Spalart-Allmaras and Shear Stress Transport (SST) eddy-viscosity models, which predict flow separation with moderate success. Their performances have been compared against experimental data for a conical and two contoured subscale nozzles. It is found that they fail to predict the separation location correctly, exhibiting sensitivity to the nozzle pressure ratio (NPR) and nozzle type. A careful assessment indicated how the model had to be tuned for better, consistent prediction. It is learnt that SST model's failure is caused by limiting of the shear stress inside boundary layer according to Bradshaw's assumption, and by over prediction of jet spreading rate. Accordingly, SST's coefficients were empirically modified to match the experimental wall pressure data. Results confirm that accurate RANS prediction of separation depends on the correct capture of the jet spreading rate, and that it is feasible over a wide range of NPRs by modified values of the diffusion coefficients in the turbulence model. Camundongos C57BL/6 machos com oito semanas de idade alimentados com diferentes dietas durante 16 semanas: de alta densidade energética (ADE, 26% das calorias de carboidrato, 60% de gordura e 14% de proteína) ou dieta padrão (CO, 76% das calorias de carboidrato, 10% de gordura e 14% de proteína). Comparado ao grupo CO, o grupo ADE apresentou maior ganho de massa e maior depósito de tecido adiposo, bem como maiores níveis plasmáticos de triglicerídeos, LDL-c, ALT, AST e fosfatase alcalina e com maiores níveis de corticosterona plasmática, glicose de jejum e insulina com uma consequente resistência à insulina (avaliado pelo HOMA-IR). No TOTG, a glicose plasmática aumentou ao máximo após 15 min. da administração de glicose oral em ambos os grupos. Entretanto os níveis de glicose foram maiores no grupo ADE que no grupo CO (P<0.0001). O clearance de glicose no grupo ADE foi reduzido, permanecendo aumentado após 120 min. (P<0.001), caracterizando intolerância a glicose no grupo ADE. O teste intraperitoneal de tolerância à insulina mostrou uma rápida redução na glicose plasmática após 15 minutos da administração de insulina em ambos os grupos, mas significativamente aumentada no grupo ADE (P<0.0001), permanecendo desta forma até os 120 min. após a administração. Concluindo, camundongos C57BL/6 respondem a dieta ADE desenvolvimento os sinais e sintomas associados à síndrome metabólica observada em humanos. Por conseguinte, este modelo animal poderá ajudar-nos a compreender melhor as alterações em órgãos alvos associadas com a síndrome metabólica, assim como a possibilidade de tratamentos diferentes.

Durante a temporada de nidação, fêmeas de tartarugas marinhas costumam reduzir ou cessar por completo a ingestão de alimentos. Este fato sugere que o armazenamento de energia e nutrientes para a reprodução ocorra durante o período que antecede a migração para os sítios reprodutivos, enquanto estes animais ainda se encontram nas áreas de alimentação. Do ponto de vista fisiológico, tartarugas em atividade reprodutiva são capazes de permanecer longos períodos em jejum. Fatores neuroendócrinos vêm sendo recentemente apontados como os mais relevantes para a manutenção da homeostase energética de todos os vertebrados; entre eles, a leptina (hormônio anorexígeno) e a grelina (peptídeo orexígeno). Com o objetivo de compreender o mecanismo de fome e saciedade nas tartarugas marinhas, investigamos os níveis séricos destes hormônios e de outros indicadores nutricionais em fêmeas de Eretmochelys imbricata desovando no litoral do Rio Grande do Norte, Brasil. Foram coletadas amostras de sangue de 41 tartarugas durante as temporadas reprodutivas de 2010/2011 e 2011/2012. Os níveis séricos de leptina diminuíram significativamente ao longo do período de nidação, de modo a explicar a busca por alimentos ao término da temporada. Ao mesmo tempo, registramos uma tendência crescente nos níveis séricos de grelina, fator este que também justifica a remigração para as áreas de alimentação no fim do período. Não foram observadas tendências lineares para alguns dos parâmetros avaliados, entre eles: hematócrito, alanina aminotransferase (ALT), aspartato aminotransferase (AST), fosfatase alcalina (FA), gama glutamil transferase (GGT), lipoproteínas de baixa densidade (LDL) e lipoproteínas de alta densidade (HDL). É possível que a maior parte dos indicadores nutricionais tenha apresentado redução gradativa devido ao estresse fisiológico decorrente da vitelogênese e de repetidas oviposições. No entanto, é valido ressaltar que o quadro de restrição calórica por tempo prolongado é o principal responsável pelas alterações em índice de massa corpórea e padrões bioquímicos nestes animais.

A Floresta Tropical Atlântica apresenta uma enorme biodiversidade, e está atualmente sujeita a inúmeras pressões como a perda de área pela intensa ocupação humana, agricultura, pecuária, urbanização e industrialização. Esses impactos têm provocado desmatamento e fragmentação florestal, processos que interferem na manutenção das populações animais, inclusive afetando os ciclos silvestres de parasitas e microorganismos. Didelphis aurita é um marsupial da Mata Atlântica com alta capacidade adaptativa a ambientes perturbados. Esta espécie onívora é tolerante à fragmentação florestal, podendo sobreviver em ambientes silvestres, rurais, suburbanos e urbanos, tendo importância na conexão dos ciclos silvestres e urbanos de diversos agentes. Este trabalho teve por objetivo descrever aspectos hematológicos, bioquímicos e de hemoparasitas em Didelphis aurita de duas áreas da Serra dos Órgãos/ RJ, uma área fragmentada e outra de mata contínua. Entre julho de 2011 e fevereiro de 2012 foram capturados 61 animais que tiveram amostras de sangue avaliadas. Os resultados expressos como média desvio padrão foram: Volume Globular 38,66 % ( 4,97); Hemácias 5,40 ( 0,75) x106/mm3; Hemoglobina 12,78 ( 1,68) g/dL; VGM 71,69 ( 3,56) fl; CHGM 33,01 ( 0,63) %; Plaquetas 514,70 ( 323,10) x 103/mm3; Leucócitos 19.678,52 ( 10.152,26)/mm3; Basófilos 0,59 ( 0,72) %; Eosinófilos 13,79 ( 6,94)%; Bastonetes 0,77 ( 2,04) %; Segmentados 41,12 ( 13,95) %; Linfócitos 41,97 ( 12,97) %; Monócitos 1,75 ( 1,51)%. Para parâmetros bioquímicos encontramos os seguintes resultados: Proteínas totais 8,50 ( 1,68); albumina 3,03 ( 0,69); globulina 5,44 ( 1,66); uréia 83,57 ( 20,11); creatinina 0,44 ( 0,13); ALT 85,01 ( 65,65); AST 314,55 ( 130,58); FA 420,38 ( 371,89); GGT 19,40 ( 8,51). Os parâmetros hematócrito, hemoglobina, hematimetria, ALT, AST e FA foram maiores nos machos do que nas fêmeas. Adultos apresentaram valores de proteína plasmática total, leucócitos, hematócrito, hemoglobina, hematimetria, albumina, proteínas totais, creatinina e GGT maiores do que jovens, e o inverso ocorreu para plaquetas, globulina e FA. Animais do Fragmento apresentaram valores de massa corporal e albumina menores do que os do Garrafão, e o inverso ocorreu para GGT e globulina. Babesiasp. ocorreu em 26,6% da população, sendo mais freqüente em adultos. Estes resultados são os primeiros parâmetros de referência para Didelphis aurita na Serra dos Órgãos, contribuindo para o estudo desta espécie.

Sorbitol é um poliol encontrado em produtos para fins especiais, como diet e light, Dentro deste contexto, incluem-se as lactantes como grandes consumidoras, almejando o retorno mais rápido ao peso pré-gestacional. Devido à grande carência em dados referentes às consequências metabólicas do consumo excessivo destes tipos de produtos, o objetivo deste trabalho foi avaliar os possíveis efeitos da ingestão materna de sorbitol na lactação sobre os perfis nutricional, bioquímico e toxicológico nas proles amamentadas. O teste da Salmonella/microssoma foi utilizado, inicialmente, na avaliação mutagênica e citotóxica com linhagens de S. enterica sorovar Typhimurium (TA97, TA98, TA100, TA102, TA104 e TA1535). Verificamos a capacidade de reversão da mutação (I.M.) e sobrevivência (%), em diferentes concentrações (0,4; 4; 40; 400; 4000 e 5000 μg/placa). Os resultados foram considerados positivos para valores de I.M. ≥ 2,0. Ratas wistar lactantes (6 por grupo experimental), cada uma com 6 filhotes, receberam sorbitol (0,00015 mg/g/dia; 0,0015 mg/g/dia e 0,15 mg/g/dia), nos primeiros 14 dias da lactação. Neste período, avaliamos a biometria das mães e proles, consumo de ração e ingestão hídrica das mães. Após a lactação, as ratas mães foram ordenhadas, e, junto com as proles, sacrificadas por punção cardíaca, para coleta de sangue total. Os fígados das proles foram submetidos ao método de perfusão, para obtenção de hepatócitos em cultura primária, ao final dos 14 dias de lactação. Os fêmures das proles foram retirados, para obtenção da medula óssea. A bioquímica de sangue das proles (glicose, triglicerídeo, colesterol total, LDL, proteínas totais, albumina, ALT, AST, cálcio total e ionizado) foi analisada, assim como a bioquímica do leite ordenhado (triglicerídeos). Os testes do micronúcleo em medual óssea e hepatócitos, assim como o teste Cometa em sangue total, foram utilizados para avaliação genotóxica e citotóxica, de acordo com as diretrizes da OECD. Os resultados mostraram que, em concentrações mais baixas (0,00015 e 0,0015 mg/g), o sorbitol induziu o ganho de peso, principalmente na menor concentração (0,00015 mg/g), e alterações no perfil lipídico do sangue em todas as concentrações. As quantidades de triglicerídeo no leite variaram em função da dose ingerida, reduzida na maior concentração (0,15 mg/g) e aumentada na menor (0,00015 mg/g). A maior concentração (0,15 mg/g) resultou em perda de peso das mães e proles, diminuição de proteínas viscerais totais, albumina e aumento de enzimas hepáticas (ALT e AST) nas proles. Os resultados do teste da Salmonella/microssoma não indicaram mutagenicidade, entretanto, uma relação de dependência entre dose e Índice de Mutagenicidade (I.M.). Ambos os testes, micronúcleos de medula óssea e hepatócitos, apresentaram uma citotoxicidade dose dependente, estatisticamente significativa em relação ao grupo controle, corroborando com a genotoxicidade do teste Cometa e dependência de dose encontrada no teste da Salmonella/microssoma. Em concentrações mais baixas, parece existir modulação das vias lipogênicas, em contrapartida, nas mais altas a toxicidade parece dificultar tais alterações, induzindo o efeito contrário. Concluimos que o consumo excessivo de sorbitol resulta em alterações metabólicas e toxicológicas em lactentes, mesmo sendo considerado seguro pelo FDA e ANVISA.

The sublethal exposure (0.24 ppm) of Nuvan on some biochemical compositions such as serum protein, blood glucose, glutamate oxaloacetate transaminase (AST) and glutamate pyruvate transaminase (ALT) and on some hematological parameters such as red blood corpuscles (RBC), white blood corpuscles (WBC), hemoglobin content (Hb), mean corpuscular concentration (MCV), mean corpuscular hemoglobin concentration (MCHC) of Catla catla fingerlings were studied. The hematological and biochemical parameters evoked a significant reduction (excepting MCV, ALT and AST which is significantly increased) with increasing days of Nuvan exposure.

Traditionally tubers of cyperus (Cyperus rotundus) and its extracts have been used for alluring fish during harvesting in India. An experiment was conducted to evaluate its feeding stimulatory activity and effect on the growth of a commercially important freshwater fish, Cirrhinus mrigala. Three isonitrogenous and isocaloric formulated diets viz. plant ingredient based control and control supplemented with cyperus tuber (CS) at 1% and 5% levels were fed to the fingerlings of mrigal, C. mrigala (2.68+0.20 g) for a period of 45 days. The growth performance and the activity of metabolic enzymes, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), in liver, gill and muscle tissues of mrigal were studied during every 15 days interval. Highest relative growth (72.28%) was obtained in the mrigal fed with the diet containing 5% cyperus (5% CS), while the relative growths were 66.18% and 43.40% for the fish fed with the 1% CS diet and control respectively. The activities of AST and ALT were significantly higher (p<0.01) in both 1% and 5% CS diets as compared to the control in all the tissues studied. Higher aminotransferase activities were observed in the tissues of 5% CS group than in those of 1% CS group throughout the experimental period. The observed higher enzymatic activity was concomitant with the higher growth rate in fish. The results suggested that cyperus tuber supplementation increased feed palatability and growth.

The acute toxicity and effects of diazinon on some haematological parameters of kutum (Rutilus frisii kutum, Kamensky, 1901) weighing 613.33 g±157.06 g were studied under static water quality conditions at 15°C ± 2ºC in winter and spring 2009. The effective physical and chemical parameters of water were pH= 7-8.2, dh= 300mg/L (caco3), DO= 7 ppm and T= 15°C±2ºC. The first test was primarily to determine the effects of acute toxicity (LC5096 h) of the agricultural toxicant diazinon (emulsion 60%) on kutum male brood stocks. For this purpose, 4 treatments were used to test toxicity; each treatment was repeated in 3 tanks with 9 fish per treatment and with 180 litres water capacity. After obtaining the final results, the information was analysed statistically with Probit version 1.5 (USEPA, 1985), and we determined the LC10, LC50 and LC90 values at 24 hours, 48 hours, 72 hours and 96 hours; the maximum allowable concentration value (LC5096 h divided by 10) (TRC, 1984); and the degree of toxicity. The second stage of testing consists of four treatments: LC0= 0 as experimental treatment, treatment A with a concentration of LC1= 0.107 mg/L, treatment B with concentration of LC5= 0.157 mg/L, treatment C with concentration of MAC value= 0.04 mg/L. Male brood stocks of kutum were treated with these concentrations for 45 days. Experiments were carried out under static conditions based on the standard TRC, 1984 method over 45 days. Our results show that long-term exposure to diazinon causes a decrease in the erythrocyte count (RBC), haemoglobin (Hb), haematocrit (PCV), mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), leucocyte count (WBC), lymphocyte, testosterone, iron (Fe), sodium (Na), lactate dehydrogenase (LDH), and cholinesterase (CHeS). In addition, diazinon also causes an increase in prolymphocyte, aspartate aminotransferase (AST), cholesterol, alkaline phosphatase (ALP) and adrenaline (P<0.05). There are no significant effects on monocyte, eosinophil, magnesium (Mg), chloride (Cl), glucose (BS), urea (BUN), uric acid (U.A), triglyceride (TG), calcium (Ca), albumin (Alb), total protein (TP), cortisol, noradrenaline and high density lipoprotein (HDL) levels in kutum male brood stocks (P>0.05). Pathology results showed toxin diazinon no effect on average weight and fish body length, the average weight of heart, brain, spleen, liver, kidney and liver index but caueses decrease of gonad weigth and gonad index and also, cause complications of tissue necrosis, vascular congestion, inflammation in the liver, a sharp reduction in the number of glomeruli, necrosis, vascular congestion and haemorage in the kidney, capsule thickening and fibrosis, atrophy, vascular congestion, macrophages release increased, increasing sediment Hemosiderine and thickening of artery walls in the spleen, atrophy, fibrosis and necrosis in testis , vascular congestion, increased distance between the myocardium and fibrous string in heart and neuronal loss, vascular congestion and edema in the brain of kutum male brood stocks.

为了揭示含有微囊藻毒素水产品的食用安全性,以雄性BALB/C小鼠为模式生物,通过为期13周的灌胃染毒实验,研究了鱼肉结合态MCLR对小鼠的亚慢性毒性作用,并与水溶态MCLR的毒性进行了对比.结果表明,68.75μg/kg(以单位体重计)剂量水溶态MCLR可引起小鼠肝脏显著系数增加(p<0.05),ALT、AST活性增强(p<0.01),引起肝细胞出现有空泡状病变;而肾脏酶学指标BUN、Cr及肾脏组织病理学观察均未发现明显异常;与之相比,同剂量鱼肉结合态MCLR对小鼠各项指标的影响并不明显,仅在实验第1周出

The planktivorous filter-feeding silver carp (Hypophthalmichthys molitrix) and bighead carp (Aristichthys nobilis) are the attractive candidates for bio-control of plankton communities to eliminate odorous populations of cyanobacteria. However, few studies focused on the health of such fishes in natural water body with vigorous toxic blooms. Blood parameters are useful and sensitive for diagnosis of diseases and monitoring of the physiological status of fish exposed to toxicants. To evaluate the impact of toxic cyanobacterial blooms on the planktivorous fish, 12 serum chemistry variables were investigated in silver carp and bighead carp for 9 months, in a large net cage in Meiliang Bay, a hypereutrophic region of Lake Taihu. The results confirmed adverse effects of cyanobacterial blooms on two phytoplanktivorous fish, which mainly characterized with potential toxicogenomic effects and metabolism disorders in liver, and kidney dysfunction. In addition, cholestasis was intensively implied by distinct elevation of all four related biomarkers (ALP, GGT, DBIL, TBIL) in bighead carp. The combination of LDH, AST activities and DBIL, URIC contents for silver carp, and the combination of ALT. ALP activities and TBIL, DBIL. URIC concentrations for bighead carps were found to most strongly indicate toxic effects from cyanobacterial blooms in such fishes by a multivariate discriminant analysis. (C) 2009 Elsevier B.V. Some species of the genera Anabaena can produce various kinds of cyanotoxins, which may pose risks to environment and human health. Anabaena has frequently been observed in eutrophic freshwater of China in recent years, but its toxicity has been reported only in a few studies. In the present study, the toxicity of an Anabaena flos-aquae strain isolated from Lake Dianchi was investigated. Acute toxicity testing was performed by mouse bioassay using crude extracts from the lyophilized cultures. The mice exposed to crude extracts showed visible symptoms of toxicity and died within 10-24 h of the injection. Serum biochemical parameters were evaluated by the use of commercial diagnostic kits. Significant alterations were found in the serum biochemical parameters: alkaline phosphatase (AKP), gamma-glutamyl transpepticlase (gamma-GT), aspartate amino transferase (AST), alanine amino transferase (ALT), AST/ALT ratio, total protein content, albumin content, albumin/globulin (A/G) ratio, blood urea nitrogen (BUN), serum creatinine (Ssr), and total antioxidative capacity (T-AOC). Histopathological observations were carried out with hematoxylin and eosin (HE) stain under light microscope. Severe lesions were seen in the livers, kidneys, and lungs of the mice injected with crude extracts. The alterations of biochemical parameters were in a dose-dependent manner, and the severities of histological lesions were in the same manner. Based on biochemical and histological studies, this research firstly shows the presence of toxin-producing Anabaena species in Lake Dianchi and the toxic effects of its crude extracts on mammals. Healthy crucian carp (Carassius auratus) were treated by intraperitoneal (i.p.) injection of crude cyanobacterial extracts at two doses, 50 and 200 mu g MC-LR equiv kg(-1) BW. High mortality (100%) was observed within 60 h post injection in the high-dose group. In the treated fish, activities of four plasma enzymes, alanine aminotransferase (ALT), alkaline phosphatase (ALP), aspartate aminotransferase (AST), and lactate dehydrogenase (LDH), all showed substantial increases, with both dose and time-dependent effects. These increases of enzyme activity indicate severe impairment occurred in the liver of crucian carp over time. Plasma concentrations of energy-related biomolecules including glucose (GLU), cholesterol (CHO), triglyceride (TG), and total protein (TP) showed marked changes in the high-dose group, possibly a nutritional imbalance correlated with the liver injury caused by intraperitoneal exposure to crude cyanobacterial extracts.

Up to now, in vivo studies on the toxic effects of microcystins (MCs) on the ultrastructures of fish liver have been very limited. The phytoplanktivorous silver carp was injected i.p. with extracted hepatotoxic microcystins (mainly MC-RR and -LR) at a dose of 1000 mu g MC-LReq. kg(-1) body weight, showing a time-dependent ultrastructural change in liver as well as significant increases in enzyme activity of plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH). We observed for the first time the occurrence of a large amount of activated secondary lysosomes, which might be an adaptive mechanism to eliminate or lessen cell damage caused by MCs through lysosome activation. Quantitative and qualitative determinations of MCs in the liver were conducted by HPLC and LC-MS2, respectively. MCs concentration in the liver reached the maximum (114.20 mu g g(-1) dry weight) after 3 h post-injection, and then rapidly dropped to 7.57 mu g g(-1) dry weight at 48 h, indicating a deputation of 99% accumulated MC-LReq. On the other hand, a decrease trend in glutathione (GSH) concentration was observed in the liver of silver carp while the activity of glutathione S-transferase (GST) increased significantly after injection. The high tolerance of silver carp to MCs might be due to the high basic GSH level in their liver, and/or an increased GSH synthesis. (C) 2007 Elsevier Inc. All rights reserved.
