O uso de plantas medicinais é uma tendência generalizada na medicina popular. O género Plectranthus pertence à família das Lamiaceae tal como a salva (Salvia), o manjericão (Ocimum) e a hortelã (Mentha), que apresentam uma grande diversidade de usos etnobotânicos. No presente trabalho, foram avaliadas as atividades antimicrobiana e antioxidante de extractos de cinco espécies do género Plectranthus: P. hadiensis (vd), P. madagascarensis, P. neochilus, P. verticillatus e P. barbatus. Os extractos foram obtidos por diversos métodos: infusão, decocção, ultra-sons, micro-ondas e maceração (10% m/v de planta seca) com dois solventes diferentes (água e acetona). O método de infusão permitiu obter a maior quantidade de extrato seco a partir da planta Plectranthus barbatus (22 ± 1,0 – 33 ± 3,0 mg/mL). A actividade antimicrobiana dos extratos obtidos foi determinada usando o método da difusão em poço e foi testada contra três bactérias Gram-positivas e duas bactérias Gram-negativas. Só os extratos acetónicos de P. madagascarensis, P. verticillatus e P. hadiensis (vd) demonstraram ser ativos e exclusivamente em bactérias Gram-positivas. O extrato de P. madagascarensis demonstrou a maior actividade antibacteriana com halos de inibição entre 23 e 36 mm. A atividade antioxidante dos extratos foi determinada qualitativamente usando o método do radical 2,2-difenil-1-picril-hidrazina (DPPH) em ccd obtiveram-se resultados positivos na maioria dos extractos estudados.

Gliomas são os mais comuns e devastadores tumores primários do sistema nervoso central. Os nucleotídeos da adenina são moléculas sinalizadoras no meio extracelular, envolvidas em importantes condições fisiológicas e patológicas. O ATP, neurotransmissor excitatório, e a adenosina, neuromoduladora, entre outros efeitos, podem induzir proliferação celular em linhagens de gliomas. Os eventos induzidos pelos nucleotídeos extracelulares são controlados pela ação das E-NTPDases, que hidrolisam o ATP até adenosina extracelularmente. Recentes estudos epidemiológicos têm sugerido que os flavonóides derivados da dieta, em particular a quercetina, apresentam um papel benéfico em prevenir ou inibir a tumorigênese. Assim, primeiramente nós avaliamos o efeito antiproliferativo da quercetina em linhagem de glioma humano U138MG. O estudo demonstrou que este flavonóide induziu em cultura de gliomas: (1) diminuição da proliferação e da viabilidade celular; (2) morte celular via necrose e apoptose; (3) parada no ciclo celular na fase G2 e (4) diminuição do índice mitótico. Além disso, nós demonstramos que a quercetina, enquanto promoveu regressão tumoral, protegeu culturas organotípicas hipocampais do dano isquêmico. Em conjunto, esses dados sugerem que a quercetina exibe efeitos antiproliferativos direcionados para as células tumorais e reduzida citotoxicidade para células normais, características altamente desejáveis na quimioterapia. Dados do nosso laboratório demonstram que o metabolismo extracelular das purinas encontra-se alterado em linhagens de gliomas com relação a culturas de astrócitos, sugerindo que mudanças no sistema purinérgico podem ser uma característica dos gliomas que potencialmente podem contribuir para o seu fenótipo de malignidade. Assim, o passo seguinte desse trabalho foi investigar o perfil de secreção dos derivados da adenina, o metabolismo extracelular do AMP e a ação da quercetina sobre o sistema purinérgico. As culturas de glioma apresentaram secreção de ATP, o qual foi detectado em maiores níveis com relação as outras moléculas avaliadas, ADP, AMP, adenosina e inosina. O AMP extracelular foi eficientemente metabolizado pelos gliomas, demonstrando uma ecto-5’-NT/CD73 muito ativa. Adicionalmente, quercetina interagiu com o sistema purinérgico, inibindo não-competitivamente a atividade da ecto-5’-NT/CD73 e modulando negativamente a sua expressão. Nós sugerimos que a inibição da atividade da ecto-5’-NT/CD73 pode resultar em um decréscimo na disponibilidade de adenosina extracelular, uma promotora tumoral. Tal efeito pode estar correlacionado com a inibição da proliferação promovida pela quercetina nessa linhagem de glioma. Nossos dados sugerem que a quercetina pode ter uma função importante na inibição da proliferação dos gliomas, atuando em diferentes vias de sinalização, incluindo o sistema purinérgico. Assim, esse estudo abre novas perspectivas para as potenciais aplicações dos flavonóides na prevenção e tratamento de tumores cerebrais.

O gênero Baccharis pertence a subtribo Baccharidinae, tribo Astereae, família Asteraceae. Foram investigadas oito espécies de Baccharis pertencentes a seção Caulopterae, do ponto de vista químico e biológico. As espécies B. articulata, B. crispa, B. cf. milleflora, B. microcephala, B. myriocephala, B. stenocephala, B. trimera e B. usterii foram analisadas quanto a composição química de seu óleo volátil por CG e CG/EM. A maioria das espécies apresentou predominância de sesquiterpenos, com exceção das duas espécies bialadas estudadas, B. articulata e B. stenocephala, as quais apresentaram quantidades apreciáveis do monoterpeno -pineno, e da espécie farmacopeica, B. trimera, com a predominância de acetato de carquejila. Devido a dificuldade na diferenciação das espécies B. trimera e B. crispa por meio de caracteres morfo-anatômicos, estas tiveram a composição química de seu volátil acompanhada ao longo de diferente épocas e locais de coleta. Os extratos hexânicos destas espécies foram também analisados por CG e CG/EM a fim de detectar a presença de acetato de carquejila em amostras coletadas no Brasil e Argentina. Os resultados sugerem a presença de acetato de carquejila como um marcador da espécie B. trimera. Os óleos voláteis de B. articulata, B. crispa, B. microcephala, B. myriocephala, B. trimera e B. usterii foram investigados quanto a atividade antimicrobiana pelo método da bioautografia e todas elas apresentaram alguma atividade. Os óleos voláteis de B. articulata, B. crispa e B. trimera apresentaram fraca atividade antinflamatória in vitro pelo método da câmara de Boyden modificada. A DL50 do óleo de B. trimera foi investigada via intraperitoneal em camundongos e calculada em 0,915 g/kg A partir do extrato aquoso de B. usterii, foram isolados seis ácidos cafeoilquínicos. Quando comparados por CLAE acoplada a técnicas de identificação "on-line", os extratos aquosos de B. trimera, B. crispa e B. usterii apresentaram os seis ácidos em comum. O extrato aquoso de B. trimera diferenciou-se dos demais pela presença de flavonóides, em especial, nepetina. Os três extratos aquosos apresentaram atividade antioxidante frente a DPPH, sendo esta atividade também detectada nos seis ácidos cafeoilquínicos isoladamente.

In this study, a BCR-ABL expressing human chronic myelogenous leukaemia cell line (K562) was used to investigate the antitumoral potential of a novel lectin (CvL) purified from the marine sponge Cliona varians. CvL inhibited the growth of K562 cells with an IC50 value of 70 g/ml, but was ineffective to normal human peripheral blood lymphocytes in the same range of concentrations tested (180 g/ml). Cell death occurred after 72 h of exposure to the lectin and with sign of apoptosis as analysed by DAPI staining. Investigation of the possible effectors of this process showed that cell death occurred in the presence of Bcl-2 and Bax expression, and involved a caspase-independent pathway. Confocal fluorescence microscopy indicated a major role for the lysosomal protease cathepsin B in mediating cell death. Accordingly, pre-incubation of K562 cells with the cathepsin inhibitor L-trans-epoxysuccinyl-L-leucylamido-(4-guanidino)butane (E-64) abolished the cytotoxic effect of CvL. Furthermore, we found upregulation of tumor necrosis factor receptor 1 (TNFR1) and down-modulation of p65 subunit of nuclear factor kappa B (NFB) expression in CvL-treated cells. These effects were accompanied by increased levels of p21 and downmodulation of pRb, suggesting that CvL is capable of cell cycle arrest. Collectively, these findings suggest that cathepsin B acts as death mediator in CvL-induced cytotoxicity possibly in a still uncharacterized connection with the membrane death receptor pathway

Polymers of mushroom cellular wall are recognized for presenting a lot of biological activities such as anti-inflammatory, antioxidant and anti-tumoral action. Polysaccharides from mushrooms of different molecular mass obtained mushrooms can activate leucocytes, stimulate fagocitic, citotoxic and antimicrobial activity including oxygen reactive species production. In this study were investigated chemical characteristics, in vitro antioxidant activity and anti-inflammatory action in an acute inflammation model of the polysaccharides extracted from Tylopilus ballouii. Results showed that were mainly extracted polysaccharides and that it primarily consisted of mannose and galactose with variable amounts of xylose and fucose. Infrared analysis showed a possible interation between this polysaccharides and proteins. In addition, molecular mass was about 140KDa. Antioxidant activity was tested by superoxide and hydroxyl radical scavenging assay, total antioxidant activity and lipid peroxidation assay. For superoxide and hydroxyl radical generation inhibition, polysaccharides have an IC50 of 2.36 and 0.36 mg/mL, respectively. Lipid peroxidation assay results showed that polysaccharides from Tylopilus ballouii present an IC50 of 3.42 mg/mL. Futhermore, anti-inflammatory assay showed that polysaccharides cause an paw edema decreasing in 32.8, 42 and 56% in 30, 50 and 70 mg/Kg dose, respectively. Thus, these results can indicate a possible use for these polysaccharides from Tylopilus ballouii as an anti-inflammatory and antioxidant.

A chymotrypsin inhibitor was purified from Erythrina velutina seeds by ammonium sulphate fractionation, affinities chromatographies on Trypsin-Sepharose, Quimotrypsin-Sepharose and reversed phase C-18 FPLC/AKTA system. The inhibitor, named EvCI, shown molecular mass of 17 kDa, as determined by SDSPAGE. 2D-PAGE showed four isoinhibitors with pI values of 4,42, 4,63, 4,83 and 5,06, with molecular mass of 17 kDa each. The aminoacid sequence of EvCI was determined by MALDI-TOF-MS and showed a high similarity with other Kunitz-type inhibitor of Erythrina variegata. EvCI competitively inhibited chymotrypsin, with Ki of 4 x10-8 M, but did not inhibited trypsin, pancreatic elastase, bromelain and papain. The inhibitory activity of EvCI was stable over wide pH and temperature ranges. In the presence of DTT 100 mM for 120 min, EvCI lost 50 % of activity. Cytotoxicity was studied in HeLa, MDA, HepG2, K562 and PC3 cells after 72-h incubation period. EvCl inhibited HeLa cells growth with an IC50 value of 50 μg/ml. Subsequent studies in HeLa cells analysis of cell death by annexin V/PI double-staining and cell cycle, using flow cytometry. The results provide evidence for a cytostatic activity of EvCl and support further studies on potential application of this inhibitors as an antiproliferative agent in combined therapy against cervical cancer

Galactans are polysaccharides sulfated present in the cell wall of red algae. Carrageenans are galactans well known in the food industry as gelling polysaccharides and for induce inflammatory process in rodents as animal model. The extraction of polysaccharides from A. multifida has been carried out by proteolysis and precipitation in different volumes of acetone, which produced three fractions (F1, F2, and FT). Chemical and physical analyses revealed that these fractions are sulfated galactan predominantly. Results of the antioxidant activity assays showed that all of these fractions have antioxidant activity and that was associated with sulfate content of the analysis of reducing power and total antioxidant capacity. However, these fractions were not effective against lipid peroxidation. The fraction FT presented higher activity on the APTT test at 200 μg (> 240 s). The assessment of the hemolytic activity showed that the FT fraction has the best activity, increasing lyses by the complement system to 42.3% (50 μg) (p< 0,001). The fraction FT showed the best yield, anticoagulant and hemolytic activity between the three fractions and therefore it was choose for the in vivo studies. The Inflammation assessment using the FT fraction (50 mg / kg MB) showed that the cellular migration and the IL-6 production increased 670.1% (p< 0,001) and 531.8% (p< 0,001), respectively. These results confirmed its use as an inflammation inducer in animal model. Cytotoxicity assay results showed that all fractions have toxic effects on 3T3 and HeLa cells after exposition of 48 hours, except when 100 μg for both F1 and FT were used. These results arise the discussion whether these polysaccharides it should be used as additive in foods, cosmetics and medicines.

However, even though unrepaired UV-induced DNA damages are related to mutagenesis, cell death and tumorigenesis, they do not explain phenotypes such as neurodegeneration and internal tumors observed in patients with syndromes like Xeroderma Pigmentosum (XP) and Cockayne Syndrome (CS) that are associated with NER deficiency. Recent evidences point to a role of NER in the repair of 8-oxodG, a typical substrate of Base Excision Repair (BER). Since deficiencies in BER result in genomic instability, neurodegenerative diseases and cancer, it was investigated in this research the impact of XPC deficiency on BER functions in human cells. It was analyzed both the expression and the cellular localization of APE1, OGG1 e PARP-1, the mainly BER enzymes, in different NER-deficient human fibroblasts. The endogenous levels of these enzymes are reduced in XPC deficient cells. Surprisingly, XP-C fibroblasts were more resistant to oxidative agents than the other NER deficient fibroblasts, despite presenting the highest of 8-oxodG. Furthermore, subtle changes in the nuclear and mitochondrial localization of APE1 were detected in XP-C fibroblasts. To confirm the impact of XPC deficiency in the regulation of APE1 and OGG1 expression and activity, we constructed a XPC-complemented cell line. Although the XPC complementation was only partial, we found that XPC-complemented cells presented increased levels of OGG1 than XPC-deficient cells. The extracts from XPC-complemented cells also presented an elevated OGG1 enzimatic activity. However, it was not observed changes in APE1 expression and activity in the XPCcomplemented cells. In addition, we found that full-length APE1 (37 kDa) and OGG1- α are in the mitochondria of XPC-deficient fibroblasts and XPC-complemented fibroblasts before and after induction of oxidative stress. On the other hand, the expression of APE1 and PARP-1 are not altered in brain and liver of XPC knockout mice. However, XPC deficiency changed the APE1 localization in hypoccampus and hypothalamus. We also observed a physical interaction between XPC and APE1 proteins in human cells. In conclusion, the data suggest that XPC protein has a role in the regulation of OGG1 expression and activity in human cells and is involved mainly in the regulation of APE1 localization in mice. Aditionally, the response of NER deficient cells under oxidative stress may not be only associated to the NER deficiency per se, but it may include the new functions of NER enzymes in regulation of expression and cell localization of BER proteins

In the present study, six families of sulfated polysaccharides were obtained from seaweed Dictyopteris delicatula (Lamouroux, 1809) and their anticoagulant, antioxidant and antitumor activities were evaluated. All fractions showed anticoagulant activity on aPTT assay, but not on PT assay. Fractions also exhibited total antioxidant activity, superoxide radical scavenging capacity and ferric chelating property. Thus, six fractions (F0.5v, F0.7v, F1.0v, F1.3v, F1.5v e F2.0v) we obtained by proteolytic digestion, followed by acetone fractionation and molecular sieving on Sephadex G-100. Chemical analyses demonstrated that all polysaccharides contain heterofucans composed mainly of fucose, xylose, glucose, galactose, uronic acid, and sulfate. Any fractions changed the PT. However, all fractions were able on double the aPPT on a dose-dependent manner. The heterofucans F0.7v and F1.0v showed low anticoagulant activity while F1.5v presented the most prominent anticoagulant activity .When compared to Clexane®, a low molecular weight heparin, at same concentration F1.5v presented similar anticoagulant activity. The fucans F0.5v and F0.7v at 1.0 mg/mL showed high ferric chelating activity (~45%), whereas fucans F1.3v (0.5 mg/mL) showed considerable reducing power, about 53.2% of the activity of vitamin C. The fucan F1.5v presented the most prominent anticoagulant activity. The best antiproliferative activity was found with fucans F1.3v and F0.7v. However, F1.3v activity was much higher than F0.7v inhibiting almost 100% of HeLa cell proliferation. These fucans have been selected for further studies on structural characterization as well as in vivo experiments, which are already in progress

Dental pulp stem cells have been widely investigated because of their ability to differentiate into both dental and non-dental cells, with potential use in therapies involving tissue engineering. The technique of cell cryopreservation represents a viable alternative for the conservation of these cells, since it stops reversibly, in a controlled manner, all of cell biological functions in an ultra low temperature. The present study aimed to evaluate, using in vitro experiments, the influence of a cryopreservation protocol on the biologic acti vity of stem cells from human exfoliated deciduous teeth (SHED). Cells obtained from the pulp of three deciduous teeth on end-stage exfoliation or with indicated extraction were expanded in α-MEM culture medium supplemented with antibiotics and 15% fetal bovine serum. At second subculture (P2), a group of cells were submitted to cryopreservation for 30 days in 10% DMSO diluted in fetal bovine serum, at -80º C, while the remind cells continued under normal conditions of cell culture. Cell proliferation was evaluated in both groups (not cryopreserved or cryopreserved) by Trypan blue stain essay at intervals of 24, 48 and 72h after plating. Cell cycle analysis of SHEDs submitted or not to the cryopreservation protocol was performed in the same intervals. Events related to cell death were studied by Annexyn V and PI expression under flow cytometry at the intervals of 24 and 72h. The presence of nuclear morphological changes was evaluated by DAPI staining at 72h interval. It was observed that both groups exhibited an upward cell proliferation curve, without considerable changes in cell viability throughout the experiment. The distribution of cell in the cell cycle phasis was consistent with cell proliferation in both groups. There were no nuclear morphological damages in the end range of the experiment. therefore, it is concluded that the proposed cryopreservation protocol is efficient for storing the studied cell type, allowing its use in future experimental studies

Conduziu-se este trabalho, com o objetivo de avaliar a atividade antioxidante de diferentes extratos de cogumelo Agaricus blazei, bem como a estabilidade oxidativa do óleo de soja adicionado de extrato de cogumelo. O cogumelo seco em estufa a 55ºC e triturado (10 g) fui submetido à extração, à temperatura ambiente, com 100 mL de metanol e metanol:água (1:1) com duração de 6 e 12 horas para ambas as extrações. O extrato de maior atividade antioxidante, conforme o método DPPH, foi aplicado em óleo de soja na concentração de 0,1% de compostos fenólicos totais e, então, submetido ao método do Rancimat e ao teste acelerado em estufa a 60ºC por um período de 16 dias. Amostras de óleo foram retiradas da estufa cada 4 dias e analisadas quanto ao índice de peróxidos e dienos conjugados. Como parâmetros de comparação, foram utilizados os antioxidantes sintéticos BHT (100 mg/kg), TBHQ (50 mg/kg) e o óleo de soja isento de antioxidantes (controle). Os resultados demonstraram que o extrato metanólico:aquoso, com 6 horas de extração, apresentou maior atividade antioxidante. A aplicação desse extrato em óleo de soja proporcionou a seguinte ordem em relação à estabilidade oxidativa: TBHQ > extrato de cogumelo > BHT = óleo de soja (controle). O extrato de cogumelo também foi eficiente em relação à formação de peróxidos e dienos conjugados que, apesar de aumentarem ao longo do tempo, foi menor que o BHT, porém maior que o TBHQ. O extrato de cogumelo apresentou-se efetivo na proteção do óleo, podendo ser considerado um potencial antioxidante natural.

Bovine mastitis is considered important disease causing major economic losses in dairy herds. Antimicrobial drug can promote resistance, chemical residues in food and environmental contamination. The purpose of this study was to evaluate the antibacterial activity in vitro of pomegranate extract on Staphylococcus aureus isolated from bovine milk, evaluate its antioxidant activity, and quantify levels of total phenols and fravonoides of the different extracts used. Aqueous extracts were used in nature and dry, from the peel of the fruit (EAC) and leaves (EAF). Additionally, it was evaluated the antioxidant activity (AA%), total phenols and flavonoids. Milk samples were inoculated, incubated, and the colonies were characterized as Staphylococcus aureus were adjusted to 1.0x106 UFC/mL in the 6 standard of the MacFarland scale. The sensitivity of the microbial isolates were determined in quintuplicate, in disk diffusion test. The minimum inhibitory concentration was determined by visible inhibition zone greater than 15mm. The results were evaluated by ANOVA, Tukey 5%, using the program SISVAR 5.3 - DEX/UFLA. Results implied that the aqueous extract from the bark of dried fruit was capable of inhibiting bacterial growth at concentrations of 3%. The other treatments only showed this activity, from the concentrations of 15%, 20% and 30% for dry EAF, in nature EAC and in nature EAF, respectively. Regarding the action antioxidant of dry EAC, was not correlated with total phenols and flavonoids. Probably other alkaloids substances present in the extract studied, may have been responsible for this activity. It is concluded that extracts of Punica granatum L., especially those obtained by the shell of the fruit, showed inhibitory activity against S. aureus, indicating your use potential for the control of bovine mastitis.
