Conventional threading operations involve two distinct machining processes: drilling and threading. Therefore, it is time consuming for the tools must be changed and the workpiece has to be moved to another machine. This paper presents an analysis of the combined process (drilling followed by threading) using a single tool for both operations: the tap-milling tool. Before presenting the methodology used to evaluate this hybrid tool, the ODS (operating deflection shapes) basics is shortly described. ODS and finite element modeling (FEM) were used during this research to optimize the process aiming to achieve higher stable machining conditions and increasing the tool life. Both methods allowed the determination of the natural frequencies and displacements of the machining center and optimize the workpiece fixture system. The results showed that there is an excellent correlation between the dynamic stability of the machining center-tool holder and the tool life, avoiding a tool premature catastrophic failure. Nevertheless, evidence showed that the tool is very sensitive to work conditions. Undoubtedly, the use of ODS and FEM eliminate empiric decisions concerning the optimization of machining conditions and increase drastically the tool life. After the ODS and FEM studies, it was possible to optimize the process and work material fixture system and machine more than 30,000 threaded holes without reaching the tool life limit and catastrophic fail.

Unmanned air vehicles (UAVs) and micro air vehicles (MAVs) constitute unique application platforms for vibration-based energy harvesting. Generating usable electrical energy during their mission has the important practical value of providing an additional energy source to run small electronic components. Electrical energy can be harvested from aeroelastic vibrations of lifting surfaces of UAVs and MAVs as they tend to have relatively flexible wings compared to their larger counterparts. In this work, an electromechanically coupled finite element model is combined with an unsteady aerodynamic model to develop a piezoaeroelastic model for airflow excitation of cantilevered plates representing wing-like structures. The electrical power output and the displacement of the wing tip are investigated for several airflow speeds and two different electrode configurations (continuous and segmented). Cancelation of electrical output occurs for typical coupled bending-torsion aeroelastic modes of a cantilevered generator wing when continuous electrodes are used. Torsional motions of the coupled modes become relatively significant when segmented electrodes are used, improving the broadband performance and altering the flutter speed. Although the focus is placed on the electrical power that can be harvested for a given airflow speed, shunt damping effect of piezoelectric power generation is also investigated for both electrode configurations.

Vibration-based energy harvesting has been investigated by several researchers over the last decade. The goal in this research field is to power small electronic components by converting the waste vibration energy available in their environment into electrical energy. Recent literature shows that piezoelectric transduction has received the most attention for vibration-to-electricity conversion. In practice, cantilevered beams and plates with piezoceramic layers are employed as piezoelectric energy harvesters. The existing piezoelectric energy harvester models are beam-type lumped parameter, approximate distributed parameter and analytical distributed parameter solutions. However, aspect ratios of piezoelectric energy harvesters in several cases are plate-like and predicting the power output to general (symmetric and asymmetric) excitations requires a plate-type formulation which has not been covered in the energy harvesting literature. In this paper. an electromechanically coupled finite element (FE) plate model is presented for predicting the electrical power output of piezoelectric energy harvester plates. Generalized Hamilton`s principle for electroelastic bodies is reviewed and the FE model is derived based on the Kirchhoff plate assumptions as typical piezoelectric energy harvesters are thin structures. Presence of conductive electrodes is taken into account in the FE model. The predictions of the FE model are verified against the analytical solution for a unimorph cantilever and then against the experimental and analytical results of a bimorph cantilever with a tip mass reported in the literature. Finally, an optimization problem is solved where the aluminum wing spar of an unmanned air vehicle (UAV) is modified to obtain a generator spar by embedding piezoceramics for the maximum electrical power without exceeding a prescribed mass addition limit. Ao longo dos últimos anos, acompanhada da evolução tecnológica, da dificuldade da inspeção visual e da consciencialização dos efeitos de uma má inspeção, verificou-se uma maior sensibilidade para a importância da monitorização estrutural, principalmente nas grandes infra-estruturas de engenharia civil. Os sistemas de monitorização estrutural permitem o acompanhamento contínuo do comportamento de uma determinada estrutura de tal forma que com os dados obtidos, é possível avaliar alterações no comportamento da mesma. Com isso, tem-se desenvolvido e implementado estratégias de identificação de danos estruturais com o intuito de aumentar a fiabilidade estrutural e evitar precocemente que alterações na condição da estrutura possam evoluir para situações mais severas. Neste contexto, a primeira parte desta dissertação consiste numa introdução à monitorização estrutural e à deteção de dano estrutural. Relativamente à monitorização, são expostos os seus objetivos e os princípios da sua aplicação. Conjuntamente são apresentados e descritos os principais sensores e são explicadas as funcionalidades de um sistema de aquisição de dados. O segundo tema aborda a importância da deteção de dano introduzindo os métodos estudados neste trabalho. Destaca-se o método das linhas de influência, o método da curvatura dos modos de vibração e o método da transformada de wavelet. Na segunda parte desta dissertação são apresentados dois casos de estudo. O primeiro estudo apresenta uma componente numérica e uma componente experimental. Estuda-se um modelo de viga que se encontra submetida a vários cenários de dano e valida-se a capacidade do método das linhas de influência em detetar e localizar essas anomalias. O segundo estudo consiste na modelação numérica de uma ponte real, na posterior simulação de cenários de dano e na análise comparativa da eficácia de cada um dos três métodos de deteção de dano na identificação e localização dos danos simulados. Por último, são apresentadas as principais conclusões deste trabalho e são sugeridos alguns tópicos a explorar na elaboração de trabalhos futuros.

In this study the authors used the Elisa-based antigen detection tests that distinguish E. histolytica from E. dispar to examine the prevalence of E. histolytica infection in individuals from an urban slum in Fortaleza, Northeastern, Brazil. This test has a sensitivity and specificity that is comparable to PCR and isoenzyme analysis, which is the gold standard. Single stools samples were obtained from 735 individuals. The prevalence of E. histolytica infection was 14.9% (110/735) and 25.4%(187/735) for E. dispar-E. histolytica complex. The most affected age group for E. histolytica /E. histolytica-E. dispar infection was the 1-5 year olds but there was no remarkable decrease with age. There was no significant difference in colonization rates between males and females. The results from this survey demonstrate that E. histolytica is highly prevalent in the Community studied. Furthermore, it offers promise for the antigen detection test as a sensitive and technically simple tool for detecting E. histolytica infection in the field.

National malaria control programmes have the responsibility to develop a policy for malaria disease management based on a set of defined criteria as efficacy, side effects, costs and compliance. These will fluctuate over time and national guidelines will require periodic re-assessment and revision. Changing a drug policy is a major undertaking that can take several years before being fully operational. The standard methods on which a decision can be taken are the in vivo and the in vitro tests. The latter allow a quantitative measurement of the drug response and the assessment of several drugs at once. However, in terms of drug policy change its results might be difficult to interpret although they may be used as an early warning system for 2nd or 3rd line drugs. The new WHO 14-days in vivo test addresses mainly the problem of treatment failure and of haematological parameters changes in sick children. It gives valuable information on whether a drug still `works'. None of these methods are well suited for large-scale studies. Molecular methods based on detection of mutations in parasite molecules targeted by antimalarial drugs could be attractive tools for surveillance. However, their relationship with in vivo test results needs to be established

Time-resolved measurements of tissue autofluorescence (AF) excited at 405 nm were carried out with an optical-fiber-based spectrometer in the bronchi of 11 patients. The objectives consisted of assessing the lifetime as a new tumor/normal (T/N) tissue contrast parameter and trying to explain the origin of the contrasts observed when using AF-based cancer detection imaging systems. No significant change in the AF lifetimes was found. AF bronchoscopy performed in parallel with an imaging device revealed both intensity and spectral contrasts. Our results suggest that the spectral contrast might be due to an enhanced blood concentration just below the epithelial layers of the lesion. The intensity contrast probably results from the thickening of the epithelium in the lesions. The absence of T/N lifetime contrast indicates that the quenching is not at the origin of the fluorescence intensity and spectral contrasts. These lifetimes (6.9 ns, 2.0 ns, and 0.2 ns) were consistent for all the examined sites. The fact that these lifetimes are the same for different emission domains ranging between 430 and 680 nm indicates that there is probably only one dominant fluorophore involved. The measured lifetimes suggest that this fluorophore is elastin.

Kunnossapidon kuluista suurimman osan aiheuttavat koneiden ennakoimattomat rikkoutumiset ja niiden tuomat tuotannonpysäytykset. Kunnossapidon suunnitelmallisuutta ja vikojen ennustettavuutta pyritään parantamaan erilaisin kunnonvalvonnallisin keinoin. Havaitsemalla alkava vikaantuminen ajoissa ja seuraamalla sen kehittymistä luodaan mahdollisuus korjaustoimenpiteiden ja -ajankohdan hallittuun suunnitteluun ja tätä kautta kustannussäästöihin. Tässä työssä on tutkittu kompressoriyksiköiden (kaasuturbiini ja keskipakokompressori) ja niiden apulaitteiden kunnossapidon menetelmiä, niiden hyödynnettävyyttä sekä mahdollisuuksia kehittää mekaanista kunnonhallintaa. Työssä päädyttiin johtopäätökseen, että käytettävien menetelmien hyödynnettävyydessä on kehitettävää. Kunnonvalvonnan ohjeistuksia tarkennettiin vastaamaan tämän päivän tavoitteita. Todettiin myös, että värähtelynvalvontaan perustuva kunnonvalvontajärjestelmä soveltuu hyvin kompressoriyksiköiden kunnonvalvonnan järjestelmäksi. Värähtelynvalvontaan perustuva kunnonvalvontajärjestelmä luo mahdollisuuden turvallisen käyttöajan ennustamiseen. Kunnonvalvonnan kehittämisen ja käyttövarmuuden parantamisen kannalta jatkotutkimuksen tekeminen kompressoriyksiköiden mekaanistenlaitteiden turvallisen käyttöajan ennustamiseksi on perusteltua.

The number of molecular diagnostic assays has increased tremendously in recent years.Nucleic acid diagnostic assays have been developed, especially for the detection of human pathogenic microbes and genetic markers predisposing to certain diseases. Closed-tube methods are preferred because they are usually faster and easier to perform than heterogenous methods and in addition, target nucleic acids are commonly amplified leading to risk of contamination of the following reactions by the amplification product if the reactions are opened. The present study introduces a new closed-tube switchable complementation probes based PCR assay concept where two non-fluorescent probes form a fluorescent lanthanide chelate complex in the presence of the target DNA. In this dual-probe PCR assay method one oligonucleotide probe carries a non-fluorescent lanthanide chelate and another probe a light absorbing antenna ligand. The fluorescent lanthanide chelate complex is formed only when the non-fluorescent probes are hybridized to adjacent positions into the target DNA bringing the reporter moieties in close proximity. The complex is formed by self-assembled lanthanide chelate complementation where the antenna ligand is coordinated to the lanthanide ion captured in the chelate. The complementation probes based assays with time-resolved fluorescence measurement showed low background signal level and hence, relatively high nucleic acid detection sensitivity (low picomolar target concentration). Different lanthanide chelate structures were explored and a new cyclic seven dentate lanthanide chelate was found suitable for complementation probe method. It was also found to resist relatively high PCR reaction temperatures, which was essential for the PCR assay applications. A seven-dentate chelate with two unoccupied coordination sites must be used instead of a more stable eight- or nine-dentate chelate because the antenna ligand needs to be coordinated to the free coordination sites of the lanthanide ion. The previously used linear seven-dentate lanthanide chelate was found to be unstable in PCR conditions and hence, the new cyclic chelate was needed. The complementation probe PCR assay method showed high signal-to-background ratio up to 300 due to a low background fluorescence level and the results (threshold cycles) in real-time PCR were reached approximately 6 amplification cycles earlier compared to the commonly used FRET-based closed-tube PCR method. The suitability of the complementation probe method for different nucleic acid assay applications was studied. 1) A duplex complementation probe C. trachomatis PCR assay with a simple 10-minute urine sample preparation was developed to study suitability of the method for clinical diagnostics. The performance of the C. trachomatis assay was equal to the commercial C. trachomatis nucleic acid amplification assay containing more complex sample preparation based on DNA extraction. 2) A PCR assay for the detection of HLA-DQA1*05 allele, that is used to predict the risk of type 1 diabetes, was developed to study the performance of the method in genotyping. A simple blood sample preparation was used where the nucleic acids were released from dried blood sample punches using high temperature and alkaline reaction conditions. The complementation probe HLA-DQA1*05 PCR assay showed good genotyping performance correlating 100% with the routinely used heterogenous reference assay. 3) To study the suitability of the complementation probe method for direct measurement of the target organism, e.g., in the culture media, the complementation probes were applied to amplificationfree closed-tube bacteriophage quantification by measuring M13 bacteriophage ssDNA. A low picomolar bacteriophage concentration was detected in a rapid 20- minute assay. The assay provides a quick and reliable alternative to the commonly used and relatively unreliable UV-photometry and time-consuming culture based bacteriophage detection methods and indicates that the method could also be used for direct measurement of other micro-organisms. The complementation probe PCR method has a low background signal level leading to a high signal-to-background ratio and relatively sensitive nucleic acid detection. The method is compatible with simple sample preparation and it was shown to tolerate residues of urine, blood, bacteria and bacterial culture media. The common trend in nucleic acid diagnostics is to create easy-to-use assays suitable for rapid near patient analysis. The complementation probe PCR assays with a brief sample preparation should be relatively easy to automate and hence, would allow the development of highperformance nucleic acid amplification assays with a short overall assay time.

This work deals with the nonlinear piezoelectric coupling in vibration-based energy harvesting, done by A. Triplett and D.D. Quinn in J. of Intelligent Material Syst. and Structures (2009). In that paper the first order nonlinear fundamental equation has a three dimensional state variable. Introducing both observable and control variables in such a way the controlled system became a SISO system, we can obtain as a corollary that for a particular choice of the observable variable it is possible to present an explicit functional relation between this variable one, and the variable representing the charge harvested. After-by observing that the structure in the Input-Output decomposition essentially changes depending on the relative degree changes, presenting bifurcation branches in its zero dynamics-we are able in to identify this type of bifurcation indicating its close relation with the Hartman - Grobman theorem telling about decomposition into stable and the unstable manifolds for hyperbolic points.

RACIONAL: A doença do refluxo gastroesofágico é a afecção digestiva de maior prevalência. Os portadores podem apresentar na evolução algumas complicações, sendo o esôfago de Barrett a de maior importância, tendo em vista seu potencial de malignidade. Todavia os processos inflamatórios do trato gastrointestinal podem apresentar degeneração maligna. OBJETIVOS: Avaliar os possíveis danos do DNA em portadores de esofagite de refluxo gastroesofágico de vários graus e verificar a aplicação do ensaio Cometa na detecção dos mesmos. MÉTODOS: Foram estudados 25 pacientes distribuídos em quatro grupos: controle (n=5), esofagite leve (n=8), esofagite severa (n=5) e câncer (n=7). O ensaio Cometa foi realizado no sangue periférico (linfócitos) e biópsia do terço distal do esôfago. RESULTADOS: O ensaio Cometa detectou danos no DNA nos pacientes com esofagite leve e severa (sangue periférico e biópsia), sendo que na esofagite severa a intensidade dos danos foi maior (p<0,05). Os danos do DNA dos pacientes com esofagite severa e câncer não mostraram diferença significativa e a intensidade dos mesmos corresponde ao ensaio Cometa classe 4 (maior que 95% de danos). CONCLUSÕES: 1) As frequências de quebras do DNA da mucosa esofágica e linfócitos estão diretamente relacionadas ao grau de inflamação; 2) a esofagite severa apresenta praticamente a mesma frequência de danos no DNA do câncer esofágico; 3) o ensaio Cometa mostrou-se muito sensível para a detecção dos danos do DNA.

This paperwork presents the complete circuitry used to build a microcontroller-based pH-meter. Key control software is also discussed. An industry-standard glass combination electrode has been employed for pH detection. Electrode parameter extraction procedure is presented. The comet assay has been conducted with numerous cell lines to assess in vitro genotoxicity. In order to use the comet assay as part of an in vitro test for evaluating genotoxicity, however, there are cell-specific factors that need to be better understood. In this present study we have evaluated some factors that may impact upon the DNA damage detected in whole blood (WB) cells and lymphocytes (ILs). Experiments were conducted comparing responses of both cells, and investigating the effects of the female hormonal cycle, and oral contraceptive (OC) use on DNA damage detection in the in vitro comet assay, at three sampling time. No significant differences were detected in the basal levels of DNA damage detected in ILs and WB cells from women OC users and non-users and from men. Basal DNA damage in ILs was unaffected by gender and stage of the menstrual cycle or the stage of the treatment schedule. Our results also indicated that the H2O2 induces DNA damage in human lymphocytes independently of gender, low-dose OC use and hormonal fluctuation. However, data showed that in 3rd sampling of menstrual cycle, lymphocytes were more resistant to H2O2-induced DNA damage than those from OC users and men. © 2007 Elsevier Ltd. All rights reserved.

The pathogens manifestation in plantations are the largest cause of damage in several cultivars, which may cause increase of prices and loss of crop quality. This paper presents a method for automatic classification of cotton diseases through feature extraction of leaf symptoms from digital images. Wavelet transform energy has been used for feature extraction while Support Vector Machine has been used for classification. Five situations have been diagnosed, namely: Healthy crop, Ramularia disease, Bacterial Blight, Ascochyta Blight, and unspecified disease. © 2012 Taylor & Francis Group.

The X-ray Fluorescence (XRF) analysis is a technique for the qualitative and quantitative determination of chemical constituents in a sample. This method is based on detection of the characteristic radiation intensities emitted by the elements of the sample, when properly excited. A variant of this technique is the Total Reflection X-ray Fluorescence (TXRF) that utilizes electromagnetic radiation as excitation source. In total reflection of X-ray, the angle of refraction of the incident beam tends to zero and the refracted beam is tangent to the sample support interface. Thus, there is a minimum angle of incidence at which no refracted beam exists and all incident radiation undergoes total reflection. In this study, we evaluated the influence of the energy variation of the beam of incident x-rays, using the MCNPX code (Monte Carlo NParticle) based on Monte Carlo method. © 2013 AIP Publishing LLC.
