The present work was developed in order to obtain data on bromatologic composition of caged hen manure (CHM) without and with hydrolised sugarcane bagasse (HSB) stocked during 10, 20 and 30 days on the ground under the cage. CHM as well as HSB remained under the cage for 10, 20 and 30 days. CHM Stocking for 30 days on the ground under the cage resulted in lower values of nutrients. The presence of HSB avoided loss of nutrients during the stocking period of CHM and preserved CHM accumulated under the cage.

An experiment was conducted to study the in vitro digestibility of dry matter (IVDMD) and gross energy (IVGED) of hydrolyzed sugarcane bagasse with different storage times and two periods of adaptation of the bovine to the diet. A completely randomized design with four treatments and five replications was used. Treatments A,B,C, and D which corresponded to the times of 0, 15, 30, and 45 days of storage time period of the bagasse, respectively. Each treatment was studied at 7 and 45 after adaptation of the bovine ruminal donor to the diet. After 30 days storage, the bagasse IVDMD was inpaired for both 7 and 45 days of adaptation. Averages values of 25.8; 25.9; 23.1 and 23.3% were obtained for IVDMD with seven days of adaptation, and 25.04; 21.6; 12.5 and 11.4% with 45 days of adaptation for storage periods of 0; 15; 30 and 45 days, respectively. For IVGED, the averages obtained were 33.4; 27.8; 30.9; and 27.2% with seven days, and 30; 26.6; 20.6 and 14.5% with 45 days of adaptation for storage periods of 0, 15, 30 and 45 days, respectively.

Aiming to study the biogas produced by 5 substrates from both quality and quantity point of view, this research was conducted at the Rural Engineering Department of FCAV/UNESP - Brazil, State of São Paulo. The substractes that were used in the anaerobic digestion were characterized as: 1 - Slaughter fowls' manure with napier grass bed (MFNG); 2 - Slaughter fowls' manure with triturate napier grass bed (MFNGT); 3 - Suine manure (SM); 4 - Bovine manure (BM) and 5 - Bovine manure mixed with 50% of sugarcane bagasse (BM50S). From the data collected it was concluded that: the substract (MFNG) and the substract containing SM produced higher and lower volumes of biogas respectively, when compared to the others; the mixture of sugarcane bagasse in the substract containing Bovine manure damages the accumulated biogas production and its quality; 57 days after filling the biodigestors, all substrates produced biogas with a methane level higher than 48% except for the substrates containing Suine manure; the triturate substrates (MFNGT) did not present such distinct characteristics from the non-triturate substrates (MFNG); the quality of biogas at the maximum production stage was similar, for all the studied substrates; the biogas which presented higher quantity of methane in its composition was the one produced with BM, overcoming the value up to 17.7% over the MFNGT production.

The objective of this work was to evaluate the mycelial growth of the Coprinus comatus strain CCO 01/01 in culture based on organic residues of Saccharum officinarum (sugarcane bagasse), Citrus sinensis (orange bagasse), Ananas comosus (pineapple residues) and Musa sp. (banana leaf), supplemented with wheat bran in the proportions of 0, 10 and 20%, kept at 27 degrees C. The mycelial growth of C. comatus was evaluated daily by measurement of the diameter of the colony during seven days of incubation. The banana leaf was considered the best residue for the cultivation of the C. comatus even without supplementation, meaning lower production costs. The supplementation of pineapple residues with 10% of wheat bran favored fungi growth. Sugar-cane bagasse was suitable for the growth of the C. comatus provided it is enriched with wheat bran. The orange bagasse, without pH correction, was not appropriate for the mycelial growth of C. comatus.

Pectate lyase (PL) was produced by the filamentous fungus Penicillium viridicatum RFC3 in solid-state cultures of a mixture of orange bagasse and wheat bran (1 : 1 w/w), or orange bagasse, wheat bran and sugarcane bagasse (1 : 1 : 0.5 w/w), and in a submerged liquid culture with orange bagasse and wheat bran (3%) as the carbon source. PL production was highest (1,500U mL -1 or 300Ug -1 of substrate) in solid-state fermentation (SSF) on wheat bran and orange bagasse at 96 hours. PL production in submerged fermentation (SmF) was influenced by the initial pH of the medium. With the initial pH adjusted to 4.5, 5.0, and 5.5, the peak activity was observed after 72, 48, and 24 hours of fermentation, respectively, when the pH of the medium reached the value 5.0. PL from SSF and SmF were loaded on Sephadex-G75 columns and six activity peaks were obtained from crude enzyme fromSSF and designated PL I, II, III, IV, V, andVI, while five peaks were obtained fromcrude enzyme fromSmF and labeled PL I', II', III', IV', and VII'. Crude enzyme and fraction III from each fermentative process were tested further. The optimum pH for crude PL from either process was 5.5, while that for PL III was 8.0. The maximum activity of enzymes from SSF was observed at 35°C, but crude enzyme was more thermotolerant than PL III, maintaining its maximum activity up to 45°C. Crude enzyme from SmF and PL III' showed thermophilic profiles of activity, with maximum activity at 60 and 55°C, respectively. In the absence of substrate, the crude enzyme from SSF was stable over the pH range 3.0-10.0 and PL III was most stable in the pH range 4.0-7.0. Crude enzyme from SmF retained 70%-80% of its maximum activity in the acid-neutral pH range (4.0-7.0), but PIII showed high stability at alkaline pH (7.5-9.5). PL from SSF was more thermolabile than that from SmF. The latter maintained 60% of its initial activity after 1 h at 55°C. Lipase production by Trichoderma harzianum was evaluated in submerged fermentation (SF) and solid-state fermentation (SSF) using a variety of agro-industrial residues. Cultures in SF showed the highest activity (1.4 U/mL) in medium containing 0.5 % (w/v) yeast extract, 1 % (v/v) olive oil and 2.5 C:N ratio. This paper is the first to report lipase production by T. harzianum in SSF. A 1:2 mixture of castor oil cake and sugarcane bagasse supplemented with 1 % (v/w) olive oil showed the best results among the cultures in SSF (4 U/g ds). Lipolytic activity was stable in a slightly acidic to neutral pH, maintaining 50 % activity after 30 min at 50 C. Eighty percent of the activity remained after 1 h in 25 % (v/v) methanol, ethanol, isopropanol or acetone. Activity was observed with vegetable oils (olive, soybean, corn and sunflower) and long-chain triacylglycerols (triolein), confirming the presence of a true lipase. The results of this study are promising because they demonstrate an enzyme with interesting properties for application in catalysis produced by fermentation at low cost. © 2012 Springer-Verlag and the University of Milan.

The structural polysaccharides contained in plant cell walls have been pointed to as a promising renewable alternative to petroleum and natural gas. Ferulic acid is a ubiquitous component of plant polysaccharides, which is found in either monomeric or dimeric forms and is covalently linked to arabinosyl residues. Ferulic acid has several commercial applications in food and pharmaceutical industries. The study herein introduces a novel feruloyl esterase from Aspergillus clavatus (AcFAE). Along with a comprehensive functional and biophysical characterization, the low-resolution structure of this enzyme was also determined by small-angle X-ray scattering. In addition, we described the production of phenolic compounds with antioxidant capacity from wheat arabinoxylan and sugarcane bagasse using AcFAE. The ability to specifically cleave ester linkages in hemicellulose is useful in several biotechnological applications, including improved accessibility to lignocellulosic enzymes for biofuel production. © 2012 Springer-Verlag Berlin Heidelberg.

The stagnant effective thermal conductivities (K0) of sugar cane bagasse (SCB), wheat bran (WB), orange pulp and peel (OPP) and their combination (weight proportion 1:2:2 SCB/OPP/WB) were obtained using the line heat source method. These solid materials were applied to pectinase production via solid-state fermentation. The moisture content ranged from 4 to 80% (w.b.). A conduction mechanism through the porous media was observed, along with conduction through a liquid film and contact thermal resistance between the samples and the probe. K0 was low for intermediate moisture contents and approached the molecular conductivity of water for high moisture contents. © 2013 Copyright Taylor and Francis Group, LLC.
