The aim of this study was to investigate levels of expression of two major genes, the odorant binding protein Gp-9 (general protein-9) and foraging, that have been shown to be associated with behavioural polymorphisms in ants. We analysed workers and young nonreproductive queens collected from nests of the monogyne (single reproductive queen per nest) and polygyne (multiple reproductive queens) social forms of Solenopsis invicta. In workers but not young queens, the level of foraging expression was significantly associated with social form and the task performed (ie localization in the nest or foraging area). The level of expression of Gp-9 was also associated with social form and worker localization. In addition there was a higher level of expression of the Gp-9(b) allele compared with the Gp-9(B) allele in the heterozygote workers and the young nonreproductive queens. Finally, in the polygyne colonies the level of expression of foraging was not significantly associated with the Gp-9 genotype for either workers or young nonreproductive queens, suggesting that both genes have independent non-epistatic effects on behaviour in S. invicta.

Narcolepsy is a rare sleep disorder characterized by excessive daytime sleepiness and cataplexy. Familial narcolepsy accounts for less than 10% of all narcolepsy cases. However, documented multiplex families are very rare and causative mutations have not been identified to date. To identify a causative mutation in familial narcolepsy, we performed linkage analysis in the largest ever reported family, which has 12 affected members, and sequenced coding regions of the genome (exome sequencing) of three affected members with narcolepsy and cataplexy. We successfully mapped a candidate locus on chromosomal region 6p22.1 (LOD score ¼ 3.85) by linkage analysis. Exome sequencing identified a missense mutation in the second exon of MOG within the linkage region. A c.398C>G mutation was present in all affected family members but absent in unaffected members and 775 unrelated control subjects. Transient expression of mutant myelin oligodendrocyte glycoprotein (MOG) in mouse oligodendrocytes showed abnormal subcellular localization, suggesting an altered function of the mutant MOG. MOG has recently been linked to various neuropsychiatric disorders and is considered as a key autoantigen in multiple sclerosis and in its animal model, experimental autoimmune encephalitis. Our finding of a pathogenic MOG mutation highlights a major role for myelin and oligodendrocytes in narcolepsy and further emphasizes glial involvement in neurodegeneration and neurobehavioral disorders. Los resultados del comportamiento de las redes de arrastre de fondo y pelágica utilizadas en el crucero de evaluación del stock de Merluza en otoño de 1997, BIC Humboldt 9705-06, muestran, según el análisis de los modelos de regresión lineal, una buena correlación entre la abertuda vertical y la abertura horizontal entre alas de la boca de la red, en todos los estratos de profundidad; y una ligera variación en el estrato II, de la relación entre la profundidad y la longitud de cable de arrastre principal; existiendo una buena correlación en los estratos I y III debido a la configuración del fondo, velocidad de arrastre, condiciones de las corrientes submarinas, medio ambiente, etc. Así tenemos que la abertura vertical y la abertura horizontal en función a la velocidad de arrastre, fue inversa y directamente proporcional con un buen grado de correlación. La selectividad de la red de arrastre de fondo Granton 400/130 con copo de 80 mm y sobre copo de 13 mm de tamaño de malla, utilizada en la evaluación del recurso merluza se realizó mediante el método de copo cubierto. Se obtuvieron ojivas naturales de retención y escape, curvas de selección por subáreas. El factor de selección para la merluza en forma general fue de 4,0 y los rangos de selección se determinaron entre 31,1 cm a 36 cm y la ojiva natural al 50% fue de 33,20 cm.

Foi realizado um experimento em casa de vegetação do Departamento de Ciência do Solo da Universidade Federal de Lavras, com o objetivo de verificar a influência dos atributos de quatro solos de várzea sobre a distribuição de algumas frações de fósforo nas folhas de feijoeiro, determinar suas respectivas concentrações críticas e a relação dessas frações com a atividade da fosfatase ácida foliar e a relação entre a atividade da fosfatase ácida determinada "In vitro" e "in vivo". O delineamento experimental foi o inteiramente casualizado, em esquema fatorial 4 x 5 x 2, com quatro repetições, sendo quatro solos [Glei Húmico (GH), Orgânico (O), Aluvial (A) e Glei Pouco Húmico (GP)], cinco doses de P (75, 150, 300, 500 e 800 mg dm-3 de P) e dois níveis de calagem (sem e com). Cada parcela foi constituída por um vaso com três dm³, onde foram cultivadas três plantas de feijoeiro. Uma das plantas foi colhida no florescimento, determinando-se, nas folhas, as concentrações de P total (Pt), P total solúvel em ácido (Pts), P orgânico (Po) e P inorgânico solúvel em ácido (Pi). Nas demais plantas, colhidas no final do ciclo, foi avaliada a produção de grãos. Os níveis críticos das frações de P no feijoeiro cultivado nos solos GH, O, A e GP, sem calagem, foram, respectivamente: Pt = 2,3, 2,2, 2,0, e 1,3 mg g-1; Pts = 110, 67, 78 e 63; Po = 32, 19, 20 e 21 e Pi = 77, 47, 58 e 43 µg g-1. Com calagem, os níveis críticos respectivos foram: Pt = 2,8, 3,0, 3,5 e 4,2 mg g-1; Pts = 105, 96, 130 e 154; Po = 47, 38, 42 e 52 e Pi = 59, 58, 88 e 103 µg g-1. A atividade da fosfatase ácida não foi uma boa característica de diagnose nutricional de fósforo em condições de severa deficiência desse elemento.

BACKGROUND AND OBJECTIVE: Protease inhibitors are highly bound to orosomucoid (ORM) (alpha1-acid glycoprotein), an acute-phase plasma protein encoded by 2 polymorphic genes, which may modulate their disposition. Our objective was to determine the influence of ORM concentration and phenotype on indinavir, lopinavir, and nelfinavir apparent clearance (CL(app)) and cellular accumulation. Efavirenz, mainly bound to albumin, was included as a control drug. METHODS: Plasma and cells samples were collected from 434 human immunodeficiency virus-infected patients. Total plasma and cellular drug concentrations and ORM concentrations and phenotypes were determined. RESULTS: Indinavir CL(app) was strongly influenced by ORM concentration (n = 36) (r2 = 0.47 [P = .00004]), particularly in the presence of ritonavir (r2 = 0.54 [P = .004]). Lopinavir CL(app) was weakly influenced by ORM concentration (n = 81) (r2 = 0.18 [P = .0001]). For both drugs, the ORM1 S variant concentration mainly explained this influence (r2 = 0.55 [P = .00004] and r2 = 0.23 [P = .0002], respectively). Indinavir CL(app) was significantly higher in F1F1 individuals than in F1S and SS patients (41.3, 23.4, and 10.3 L/h [P = .0004] without ritonavir and 21.1, 13.2, and 10.1 L/h [P = .05] with ritonavir, respectively). Lopinavir cellular exposure was not influenced by ORM abundance and phenotype. Finally, ORM concentration or phenotype did not influence nelfinavir (n = 153) or efavirenz (n = 198) pharmacokinetics. CONCLUSION: ORM concentration and phenotype modulate indinavir pharmacokinetics and, to a lesser extent, lopinavir pharmacokinetics but without influencing their cellular exposure. This confounding influence of ORM should be taken into account for appropriate interpretation of therapeutic drug monitoring results. Further studies are needed to investigate whether the measure of unbound drug plasma concentration gives more meaningful information than total drug concentration for indinavir and lopinavir.

A monoclonal antibody CC92 (IgM), raised against a fraction of rat liver enriched in Golgi membranes, recognizes a novel Endo H-resistant 74-kD membrane glycoprotein (gp74). The bulk of gp74 is confined to the cis-Golgi network (CGN). Outside the Golgi gp74 is found in tubulovesicular structures and ER foci. In cells incubated at 37 degrees C the majority of gp74 is segregated from the intermediate compartment (IC) marker p58. However, in cells treated with organelle perturbants such as low temperature, BFA, and [AIF4]- the patterns of the two proteins become indistinguishable. Both proteins are retained in the Golgi complex at 20 degrees C and in the IC at 15 degrees C. Incubation of cells with BFA results in relocation of gp74 to p58 positive IC elements. [AIF4]- induces the redistribution of gp74 from the Golgi to p58-positive vesicles and does not retard the translocation of gp74 to IC elements in cells treated with BFA. Disruption of microtubules by nocodazol results in the rapid disappearance of the Golgi elements stained by gp74 and redistribution of the protein into vesicle-like structures. The responses of gp74 to cell perturbants are in sharp contrast with those of cis/middle and trans-Golgi resident proteins whose location is not affected by low temperatures or [AIF4]-, are translocated to the ER upon addition of BFA, and stay in slow disintegrating Golgi elements in cells treated with nocodazol. The results suggest that gp74 is an itinerant protein that resides most of the time in the CGN and cycles through the ER/IC following the pathway used by p58.

The Gp-9 gene in fire ants represents an important model system for studying the evolution of social organization in insects as well as a rich source of information relevant to other major evolutionary topics. An important feature of this system is that polymorphism in social organization is completely associated with allelic variation at Gp-9, such that single-queen colonies (monogyne form) include only inhabitants bearing B-like alleles while multiple-queen colonies (polygyne form) additionally include inhabitants bearing b-like alleles. A recent study of this system by Leal and Ishida (2008) made two major claims, the validity and significance of which we examine here. After reviewing existing literature, analyzing the methods and results of Leal and Ishida (2008), and generating new data from one of their study sites, we conclude that their claim that polygyny can occur in Solenopsis invicta in the U.S.A. in the absence of expression of the b-like allele Gp-9(b) is unfounded. Moreover, we argue that available information on insect OBPs (the family of proteins to which GP-9 belongs), on the evolutionary/population genetics of Gp-9, and on pheromonal/behavioral control of fire ant colony queen number fails to support their view that GP-9 plays no role in the chemosensory-mediated communication that underpins regulation of social organization. Our analyses lead us to conclude that there are no new reasons to question the existing consensus view of the Gp-9 system outlined in Gotzek and Ross (2007).

Sera from transgenic mice (TM) carrying human genes of alpha 1-acid glycoprotein (orosomucoid or ORM) have been analyzed by isoelectrofocusing and subsequent immunoblotting with antihuman ORM antibodies. With this technique it is possible to reveal selectively the human protein secreted in the TM sera. Orosomucoid bands present in TM sera have been compared with those of the most common human ORM phenotypes to correlate the products of specific genes to previously identified genetic variants. In this paper, we report the identification of the genes encoding for variants ORM1 F1 and ORM2 A, which are genes AGP-A and AGP-B/B' respectively. The nucleotide sequences of these genes are known; therefore a direct correlation between variants and specific amino acid sequences can be established.
