Background: In higher primates, during non-pregnant cycles, it is indisputable that circulating LH is essential for maintenance of corpus luteum (CL) function. On the other hand, during pregnancy, CL function gets rescued by the LH analogue, chorionic gonadotropin (CG). The molecular mechanisms involved in the control of luteal function during spontaneous luteolysis and rescue processes are not completely understood. Emerging evidence suggests that LH/CGR activation triggers proliferation and transformation of target cells by various signaling molecules as evident from studies demonstrating participation of Src family of tyrosine kinases (SFKs) and MAP kinases in hCG-mediated actions in Leydig cells. Since circulating LH concentration does not vary during luteal regression, it was hypothesized that decreased responsiveness of luteal cells to LH might occur due to changes in LH/CGR expression dynamics, modulation of SFKs or interference with steroid biosynthesis. Methods: Since, maintenance of structure and function of CL is dependent on the presence of functional LH/CGR its expression dynamics as well as mRNA and protein expressions of SFKs were determined throughout the luteal phase. Employing well characterized luteolysis and CL rescue animal models, activities of SFKs, cAMP phosphodiesterase (cAMP-PDE) and expression of SR-B1 (a membrane receptor associated with trafficking of cholesterol ester) were examined. Also, studies were carried out to investigate the mechanisms responsible for decline in progesterone biosynthesis in CL during the latter part of the non-pregnant cycle. Results and discussion: The decreased responsiveness of CL to LH during late luteal phase could not be accounted for by changes in LH/CGR mRNA levels, its transcript variants or protein. Results obtained employing model systems depicting different functional states of CL revealed increased activity of SFKs pSrc (Y-416)] and PDE as well as decreased expression of SR-B1correlating with initiation of spontaneous luteolysis. However, CG, by virtue of its heroic efforts, perhaps by inhibition of SFKs and PDE activation, prevents CL from undergoing regression during pregnancy. Conclusions: The results indicated participation of activated Src and increased activity of cAMP-PDE in the control of luteal function in vivo. That the exogenous hCG treatment caused decreased activation of Src and cAMP-PDE activity with increased circulating progesterone might explain the transient CL rescue that occurs during early pregnancy.

Long-term batch cultures of Escherichia coli grown in nutrient-rich medium accumulate mutations that provide a growth advantage in the stationary phase (GASP). We have examined the survivors of prolonged stationary phase to identify loci involved in conferring a growth advantage and show that a mutation in the hns gene causing reduced activity of the global regulator H-NS confers a GASP phenotype under specific conditions. The hns-66 allele bears a point mutation within the termination codon of the H-NS open reading frame, resulting in a longer protein that is partially functional. Although isolated from a long-term stationary-phase culture of the parent carrying the rpoS819 allele that results in reduced RpoS activity, the hns-66 survivor showed a growth disadvantage in the early stationary phase (24 to 48 h) when competed against the parent. The hns-66 mutant is also unstable and reverts at a high frequency in the early stationary phase by accumulating second-site suppressor mutations within the ssrA gene involved in targeting aberrant proteins for proteolysis. The mutant was more stable and showed a moderate growth advantage in combination with the rpoS819 allele when competed against a 21-day-old parent. These studies show that H-NS is a target for mutations conferring fitness gain that depends on the genetic background as well as on the stage of the stationary phase.

Yeast Rpb4, a subunit of RNA pol II is not essential for viability but is involved in multiple cellular phenotypes such as temperature sensitivity, enhanced pseudohyphal morphology, and decreased sporulation. Both in vivo and in vitro studies strongly support involvement of Rpb4 in transcription initiation, while its role in transcription elongation is not entirely consistent. Here we show that Rpb4 is not required for recruitment of RNA pol II on the coding region of YLR454w, a representative long gene. Yet we find strong genetic interaction of rpb4 Delta with mutants in many transcription elongation factors such as Paf1, Spt4, Dst1, Elp3 and Rpb9. We demonstrate that, Rpb4 interacts functionally with Paf1 to affect the transcription elongation of the FKS1 gene. Our results suggest that while Rpb4 is not required for general transcription elongation, it could support transcription elongation for specific of class of genes by interaction with other elongation factors. Among the human factors that influence safe driving, visual skills of the driver can be considered fundamental. This study mainly focuses on investigating the effect of visual functions of drivers in India on their road crash involvement. Experiments were conducted to assess vision functions of Indian licensed drivers belonging to various organizations, age groups and driving experience. The test results were further related to the crash involvement histories of drivers through statistical tools. A generalized linear model was developed to ascertain the influence of these traits on propensity of crash involvement. Among the sampled drivers, colour vision, vertical field of vision, depth perception, contrast sensitivity, acuity and phoria were found to influence their crash involvement rates. In India, there are no efficient standards and testing methods to assess the visual capabilities of drivers during their licensing process and this study highlights the need for the same.

During systemic disease in mice, Salmonella enterica grows intracellularly within discrete foci of infection in the spleen and liver. In concomitant infections, foci containing different S. enterica strains are spatially separated. We have investigated whether functional interactions between bacterial populations within the same host can occur despite the known spatial separation of the foci and independence of growth of salmonellae residing in different foci. In this study we have demonstrated that bacterial numbers of virulent S. enterica serovar Typhimurium C5 strain in mouse tissues can be increased by the presence of the attenuated aroA S. Typhimurium SL3261 vaccine strain in the same tissue. Disease exacerbation does not require simultaneous coinjection of the attenuated bacteria. SL3261 can be administered up to 48 hr after or 24 hr before the administration of C5 and still determine higher tissue numbers of the virulent bacteria. This indicates that intravenous administration of a S. enterica vaccine strain could potentially exacerbate an established infection with wild-type bacteria. These data also suggest that the severity of an infection with a virulent S. enterica strain can be increased by the prior administration of a live attenuated vaccine strain if infection occurs within 48 hr of vaccination. Exacerbation of the growth of C5 requires Toll-like receptor 4-dependent interleukin-10 production with the involvement of both Toll/interleukin-1 receptor-domain-containing adaptor inducing interferon-beta and myeloid differentiation factor 88.

Five fishing villages in Lake Chad Basin region of Borno State (Nigeria) were assessed for the roles of children in fishing activities in the area. The villages surveyed include: Bundaram, Yobe, Daba masara, Dumba and Doro. The results show that the children were largely between 12-18 years of age in the villages. Generally, the younger children (less than 12 years) participate in activities that require no technicality and little physical strength, while the older children (12 years and above) engage in skillful fabrication of gear and fishing activities. Some activities in the surveyed villages were gender specific. Such activities include fish processing (smoking) which is exclusive for female and few male children, who carry out preliminary cleaning of fish before any processing method is applied. 80% of the children in the five fishing villages claimed proper understanding of the techniques and procedure involved in most fishing activities. About 65% of the children sampled showed willingness to become full time fishermen while 22% were uncertain and claimed that they do not know what the future holds for them. 15% of them resolved to migrate to town so that they could live a city life

The extent of rural women fisherfolks contribution to fisheries was grossly under estimated and certainly under valued. The basis of their involvement in fisheries activities is to make themselves equal partners to men productive and self reliant participants in the process of improving their own and their family living standard and to enable them realize their full potentials. This survey is informed a system of data collection with the aid of questionnaires and analysis. Ninety-six of the questionnaires were administered to women fisher folks in eight fishing communities selected at random. Thirty seven (97) percent of the women fisher folks are between the ages of 25-35 years, 59.4% of them can only read and write in Arabic language and 21.9% only are literature in western education, which is a general characteristic of the rural population in Nigeria. 24.0% of the respondents are in full time fishing activities while 65-5% are fishmongers. They belong to cooperative societies but only 26% indicated to have benefited from loan and credit facilities. 84.4% of the respondents are sustained by the business. The major problems facing women fisherfolks includes poor market price, fish spoilage, high transport cost and lack of access to loan and credit facilities and extension assistance. Solution to these problems will increase their status benefit and development

Single nucleotide-polymorphisms (SNPs) are a source of diversity among human population, which may be responsible for the different individual susceptibility to diseases and/or response to drugs, among other phenotypic traits. Several low penetrance susceptibility genes associated with malignant melanoma (MM) have been described, including genes related to pigmentation, DNA damage repair and oxidative stress pathways. In the present work, we conducted a candidate gene association study based on proteins and genes whose expression we had detected altered in melanoma cell lines as compared to normal melanocytes. The result was the selection of 88 loci and 384 SNPs, of which 314 fulfilled our quality criteria for a case-control association study. The SNP rs6854854 in ANXA5 was statistically significant after conservative Bonferroni correction when 464 melanoma patients and 400 controls were analyzed in a discovery Phase I. However, this finding could not be replicated in the validation phase, perhaps because the minor allele frequency of SNP rs6854854 varies depending on the geographical region considered. Additionally, a second SNP (rs6431588) located on ILKAP was found to be associated with melanoma after considering a combined set of 1,883 MM cases and 1,358 disease-free controls. The OR was 1.29 (95% CI 1.12-1.48; p-value= 4x10(-4)). Both SNPs, rs6854854 in ANXA5 and rs6431588 in ILKAP, show population structure, which, assuming that the Spanish population is not significantly structured, suggests a role of these loci on a specific genetic adaptation to different environmental conditions. Furthermore, the biological relevance of these genes in MM is supported by in vitro experiments, which show a decrease in the transcription levels of ANXA5 and ILKAP in melanoma cells compared to normal melanocytes.

Este estudo parte dos pressupostos de que o desenvolvimento humano em todos os seus aspectos depende de um outro social, de que a família é o primeiro ambiente socializador da criança, e suas práticas são essenciais para que a criança se desenvolva. Pressupõe-se também que: os bebês compartilham características universais, interações são constitutivas do desenvolvimento, o investimento parental é característica da espécie, e o formato que cada um desses aspectos assume está diretamente relacionado ao contexto sociocultural. A partir desses pressupostos pode-se pensar nas mudanças nos papéis sociais exigidas pelo sistema econômico da sociedade urbana ocidental, em que a mulher cada vez mais tem trabalhos remunerados, necessitando muitas vezes deixar os filhos em creches e levando o pai a ganhar espaço em relação aos cuidados com os filhos, inclusive tendo uma participação nestas instituições. Desta forma, o presente trabalho pretendeu investigar os diversos aspectos do envolvimento paterno com seus filhos, inclusive a sua participação na creche,, buscando compreendê-lo através da abordagem sociocultural. Fizeram parte da pesquisa empírica nesta tese três estudos distintos com pais e mães de crianças de 0 a 5 anos, incluindo entrevistas, aplicação do instrumento Estilo Paterno e análise da participação paterna em eventos e atividades escolares, com o intuito de investigar o nível de satisfação paterno e materno com o envolvimento do pai com os filhos e identificando em que aspectos essa participação é mais ou menos freqüente nos cuidados cotidianos, incluindo a escola. Os resultados encontrados indicam que, embora haja uma tendência dos pais em um maior engajamento nas atividades voltadas para o entretenimento com os filhos, estes se mostram cada vez mais disponíveis para o engajamento nas demais atividades diárias. Na creche há uma tendência de maior participação paterna quanto menor a idade do filho e enquanto não prevalecem os aspectos pedagógicos. Os pais participantes de uma forma geral estão satisfeitos com o seu exercício da paternidade, mas consideram que as mães, não só incentivam a sua participação, como desejam um maior engajamento dos pais. Ao término deste estudo podemos afirmar que, as famílias participantes desta amostra brasileira na cidade do Rio de Janeiro, tendem a não ter como prática o exercício da paternidade tradicional, porém ainda não vivem plenamente o exercício da paternidade não tradicional.
