The study is focused on structural aspects of interaction between silencing suppressor p19 and CUG-repeating small RNAs. The work involves crystal structure determination of a protein-unbound RNA form and RNA fragments of various lengths (19, 20, 21 nucleotides) complexed with p19-suppressor. Results prove the ability of silencing suppressor p19 to bind CUG-repeating small RNAs, as well as reveal features of U•U mismatches flanked by Watson-Crick C•G base pairs in p19-bound and p19-unbound states. In addition, structural data reveal a p19 specific site for anchoring extra nucleotides in small RNAs. In general, the study extends our knowledge about the mechanism of small RNA recognition by silencing suppressor p19.

ENGLISH: The average linear growth rate of skipjack in the eastern Pacific is less than 1 mm per day except for fish 375 to 424 mm in length at release. The growth rate shows a decrease with increasing length and increasing time at liberty. The growth rate of fish in the length range of about 43 to 57 cm is apparently more rapid in the eastern Pacific than in the western Pacific. Dsing data for the northeastern and southeastern Pacific combined, K and ~ were estimated to be 0.658 (on an annual basis) and 885 mm, respectively, by the ungrouped method and 0.829 and 846 mm, respectively, by the grouped method. Sensitivity analyses have shown however, that the estimates of these parameters are poorly determined by the sum of squares method used to derive them. Estimates of K and ~ for the eastern Pacific tend to be lower and higher, respectively, than those for the western Pacific. The average linear growth rate of yellowfin in the eastern Pacific is a little less than 1 mm per day for fish between about 25 and 100 cm in length at release. The growth appears to be most rapid in Area 2 (Revillagigedo Islands) and slowest in Areas 1 (Baja California), 5 (Central America- Colombia), and 6 (Ecuador-Peru). There is considerable variation in the growth rates of individual fish. The growth does not show a decrease with increasing length or increasing time at liberty so realistic estimates of the parameters of the von Bertalanffy or other similar equations cannot be calculated from these data. If realistic estimates of these parameters are to be secured larger fish must be tagged and released or many more long-term returns from fish to about 100 cm in length at release must be obtained. The growth patterns for the eastern Pacific, central Pacific and eastern Atlantic found by most other investigators differ from one another and from those found in the present study. Some of these differences may be real and others may be due to deficiencies in the data or the methods of analysis. Estimates obtained from tagging data are believed to be realistic provided the tags do not inhibit the growth of the fish. It appears that the growth rates of single- and double-tagged fish are the same; this indicates, though not unequivocally, that the tags do not inhibit the growth. La tasa de crecimiento disminuye a medida que aumenta la longitud y el tiempo en libertad. La tasa de crecimiento de peces de entre unos 43 y 57 cm de longitud parece ser mayor en el Pacífico oriental que en el occidental. A partir de datos del Pacífico nororiental y suroriental combinados, se estimaron K y loo en 0.658 (anual) y 885mm, respectivamente, usando el método no agrupado, y 0.829 y 846mm, respectivamente, usando el método agrupado. Para obtener estimaciones realistas de estos parámetros sería necesario marcar peces mayores u obtener muchas más devoluciones a largo plazo de marcas de peces de unos 100cm de longitud de liberación. Los patrones de crecimiento correspondientes al Pacífico oriental, Pacífico central, y Atlántico oriental descubiertos por la mayoría de los investigadores son diferentes entre síy también de los del presente estudio. Es posibleque algunas de estas diferencias sean verdaderas, mientras que otras se deban a faltas en los datos on en los métodos analíticos utilizados. Se considera que las estimaciones obtenidas a partir de los datos de marcado son realistas, suponiendo siempre que las marcas no impidan el crecimiento de los peces. Parece ser que las tasas de crecimiento de peces con una marca y con dos son idénticas, lo cual indica, aunque sin certeza total, que las marcas no ejercen tal efecto. (PDF contains 76 pages.)

89 ripe female brooders of the catfish, Clarias anguillaris (Body wt. Range 150g-1, 200g) were induced to spawn by hormone (Ovaprim) induced natural spawning technique over a period of 10 weeks. Matching ripe males were used for pairing the females at the ratio of two males to a female. Six ranges of brood stock body weights were considered as follows; <200g; 200g-399g; 400g-599g; 600-799g; 800g-999g; > 1000g and the number of fry produced by each female brooder was scored/recorded against the corresponding body weight range. The number of fry per unit quantity of hormone and the cost of production a fry based on the current price of Ovaprim (hormon) were determined so as to ascertain most economic size range. The best and most economic size range was between 400g-599g body weight with about 20,000 fry per ml of hormone and N0.028 per fry, while the females above 1000g gave the poorest results of 9,519 fry per ml of hormone and N0.059 per fry. For optimum production of Clarias anguillaris fry and maximum return on investment female brooders of body weights ranging between 400g-599g are recommended for hormone induced natural breeding exercises

Borno State possesses great potentials for fish production both from inland fisheries and aquaculture. The socio-economic and environmental production factors are suitable for fish production. If the potential of the State were well harnessed, it would be playing significant roles in achieving self-sufficiency in fish production in Nigeria. But the situation at the moment is that its fisheries potentials are not being optimally utilized. While the inland waters of Lake Chad are currently being recklessly exploited, aquaculture development is given little or no attention. It is evident that there is a missing link between research results and the potential end users. Because information in fish production variables is a pre-requisite for fisheries development, the gap that exists between two poles must be bridged, fisheries Extension provides this important link between research result and the end users of research findings. The paper examines the importance of extension services as the key to unlock fish production information that are usually consigned to the pages of academic journals and research publications

A Hohlraum-like configuration is proposed for realizing a simple compact source for neutrons. A laser pulse enters a tiny thin-shelled hollow-sphere target through a small opening and is self-consistently trapped in the cavity. The electrons in the inner shell-wall region are expelled by the light pressure. The resulting space-charge field compresses the local ions into a thin layer that becomes strongly heated. An inward expansion of ions into the shell cavity then occurs, resulting in the formation at the cavity center of a hot spot of ions at high density and temperature, similar to that in inertial electrostatic confinement.

The epidemic of HIV/AIDS in the United States is constantly changing and evolving, starting from patient zero to now an estimated 650,000 to 900,000 Americans infected. The nature and course of HIV changed dramatically with the introduction of antiretrovirals. This discourse examines many different facets of HIV from the beginning where there wasn't any treatment for HIV until the present era of highly active antiretroviral therapy (HAART). By utilizing statistical analysis of clinical data, this paper examines where we were, where we are and projections as to where treatment of HIV/AIDS is headed.

Chapter Two describes the datasets that were used for the analyses. The primary database utilized was collected by myself from an outpatient HIV clinic. The data included dates from 1984 until the present. The second database was from the Multicenter AIDS Cohort Study (MACS) public dataset. The data from the MACS cover the time between 1984 and October 1992. Comparisons are made between both datasets.

Chapter Three discusses where we were. Before the first anti-HIV drugs (called antiretrovirals) were approved, there was no treatment to slow the progression of HIV. The first generation of antiretrovirals, reverse transcriptase inhibitors such as AZT (zidovudine), DDI (didanosine), DDC (zalcitabine), and D4T (stavudine) provided the first treatment for HIV. The first clinical trials showed that these antiretrovirals had a significant impact on increasing patient survival. The trials also showed that patients on these drugs had increased CD4+ T cell counts. Chapter Three examines the distributions of CD4 T cell counts. The results show that the estimated distributions of CD4 T cell counts are distinctly non-Gaussian. Thus distributional assumptions regarding CD4 T cell counts must be taken, into account when performing analyses with this marker. The results also show the estimated CD4 T cell distributions for each disease stage: asymptomatic, symptomatic and AIDS are non-Gaussian. Interestingly, the distribution of CD4 T cell counts for the asymptomatic period is significantly below that of the CD4 T cell distribution for the uninfected population suggesting that even in patients with no outward symptoms of HIV infection, there exists high levels of immunosuppression.

Chapter Four discusses where we are at present. HIV quickly grew resistant to reverse transcriptase inhibitors which were given sequentially as mono or dual therapy. As resistance grew, the positive effects of the reverse transcriptase inhibitors on CD4 T cell counts and survival dissipated. As the old era faded a new era characterized by a new class of drugs and new technology changed the way that we treat HIV-infected patients. Viral load assays were able to quantify the levels of HIV RNA in the blood. By quantifying the viral load, one now had a faster, more direct way to test antiretroviral regimen efficacy. Protease inhibitors, which attacked a different region of HIV than reverse transcriptase inhibitors, when used in combination with other antiretroviral agents were found to dramatically and significantly reduce the HIV RNA levels in the blood. Patients also experienced significant increases in CD4 T cell counts. For the first time in the epidemic, there was hope. It was hypothesized that with HAART, viral levels could be kept so low that the immune system as measured by CD4 T cell counts would be able to recover. If these viral levels could be kept low enough, it would be possible for the immune system to eradicate the virus. The hypothesis of immune reconstitution, that is bringing CD4 T cell counts up to levels seen in uninfected patients, is tested in Chapter Four. It was found that for these patients, there was not enough of a CD4 T cell increase to be consistent with the hypothesis of immune reconstitution.

In Chapter Five, the effectiveness of long-term HAART is analyzed. Survival analysis was conducted on 213 patients on long-term HAART. The primary endpoint was presence of an AIDS defining illness. A high level of clinical failure, or progression to an endpoint, was found.

Chapter Six yields insights into where we are going. New technology such as viral genotypic testing, that looks at the genetic structure of HIV and determines where mutations have occurred, has shown that HIV is capable of producing resistance mutations that confer multiple drug resistance. This section looks at resistance issues and speculates, ceterus parabis, where the state of HIV is going. This section first addresses viral genotype and the correlates of viral load and disease progression. A second analysis looks at patients who have failed their primary attempts at HAART and subsequent salvage therapy. It was found that salvage regimens, efforts to control viral replication through the administration of different combinations of antiretrovirals, were not effective in 90 percent of the population in controlling viral replication. Thus, primary attempts at therapy offer the best change of viral suppression and delay of disease progression. Documentation of transmission of drug-resistant virus suggests that the public health crisis of HIV is far from over. Drug resistant HIV can sustain the epidemic and hamper our efforts to treat HIV infection. The data presented suggest that the decrease in the morbidity and mortality due to HIV/AIDS is transient. Deaths due to HIV will increase and public health officials must prepare for this eventuality unless new treatments become available. These results also underscore the importance of the vaccine effort.

The final chapter looks at the economic issues related to HIV. The direct and indirect costs of treating HIV/AIDS are very high. For the first time in the epidemic, there exists treatment that can actually slow disease progression. The direct costs for HAART are estimated. It is estimated that the direct lifetime costs for treating each HIV infected patient with HAART is between $353,000 to $598,000 depending on how long HAART prolongs life. If one looks at the incremental cost per year of life saved it is only $101,000. This is comparable with the incremental costs per year of life saved from coronary artery bypass surgery.

Policy makers need to be aware that although HAART can delay disease progression, it is not a cure and HIV is not over. The results presented here suggest that the decreases in the morbidity and mortality due to HIV are transient. Policymakers need to be prepared for the eventual increase in AIDS incidence and mortality. Costs associated with HIV/AIDS are also projected to increase. The cost savings seen recently have been from the dramatic decreases in the incidence of AIDS defining opportunistic infections. As patients who have been on HAART the longest start to progress to AIDS, policymakers and insurance companies will find that the cost of treating HIV/AIDS will increase.

A ETA Guandu é de vital importância para o estado do Rio de Janeiro por ser responsável pelo abastecimento de 8,5 milhões de pessoas na Região Metropolitana. Este trabalho tem como objetivo efetuar uma análise qualitativa de sua vulnerabilidade, em relação à qualidade e quantidade de água captada, perante diversos fatores de estresse, visando dimensionar a importância da variabilidade climática na composição de sua vulnerabilidade. Adotou-se o conceito de vulnerabilidade como função da exposição (grau em que um sistema experimenta estresses), da sensibilidade (grau em que um sistema é afetado pelo estresse) e capacidade adaptativa (capacidade de um sistema de se ajustar, moderar ou lidar com as conseqüências de um estresse). Dessa forma, foi possível desenvolver uma metodologia de análise qualitativa de vulnerabilidade, específica ao caso da ETA Guandu, perante quatro fatores de estresse considerados como os mais determinantes para a vulnerabilidade atual: i) variabilidade climática; ii) transposição; iii) condições ambientais e qualidade da água e iv) acidentes ambientais. Os resultados evidenciaram que o maior grau de vulnerabilidade da ETA Guandu se relaciona à transposição, já que diversas partes de sua infraestrutura não dispõem de ações de manutenção preventiva. A vulnerabilidade devido às condições ambientais é também intensa, principalmente a turbidez no período chuvoso; entre 2000 e 2010, a ETA Guandu foi parcialmente paralisada 22 vezes em função do grande aporte de sedimentos. Não identificamos nenhum registro de paralisação total da ETA Guandu em função de acidentes ambientais, embora algumas vezes estes tenham imposto a paralisação da transposição. O componente variabilidade climática (intensificação de eventos hidrológicos extremos) revelou-se como o estressor menos determinante da vulnerabilidade atual.

We compare the effectiveness of six exchange/correlation functional combinations (Becke/Lee, Yang and Parr; Becke-3/Lee, Yang and Parr; Becke/Perdew-Wang 91; Becke-3/Perdew-Wang 91; Becke/Perdew 86; Becke-3/Perdew 86) for computing C-N, O-O and N-NO2 dissociation energies and dipole moments of five compounds. The upconversion properties of Er3+/Yb3+ codoped tellurite glasses and glass fibers with D-shape cladding under 980 mu excitation were investigated. Intense emission bands centered at 531, 546 and 658 nm corresponding to the transitions Er3+: H-2(11/2) -> I-4(15/2) , S-4(3/2) -> I-4(15/2) and F-4(9/2) -> I-4(15/2), respectively, were observed at room temperature. Compared with that in Er3+/Yb3+ codoped tellurite bulk glass, the upconversion luminescence becomes more efficient in the fiber geometry. The dependence of upconversion intensities on fiber geometry and possible upconversion mechanism are discussed and evaluated. The presented Er3+/Yb3+ codoped tellurite fibers with intense upconversion luminescence can be used as potential host materials for upconversion fiber lasers. (c) 2005 Elsevier B.V. Data from ichthyoplankton surveys conducted in 1972 and from 1977 to 1999 (no data were collected in 1980) by the Alaska Fisheries Science Center (NOAA, NMFS) in the western Gulf of Alaska were used to examine the timing of spawning, geographic distribution and abundance, and the vertical distribution of eggs and larvae of flathead sole (Hippoglossoides elassodon). In the western Gulf of Alaska, flathead sole spawning began in early April and peaked from early to mid-May on the continental shelf. It progressed in a southwesterly direction along the Alaska Peninsula where three main areas of flathead sole spawning were indentified: near the Kenai Peninsula, in Shelikof Strait, and between the Shumagin Islands and Unimak Island. Flathead sole eggs are pelagic, and their depth distribution may be a function of their developmental stage. Data from MOCNESS tows indicated that eggs sink near time of hatching and the larvae rise to the surface to feed. The geographic distribution of larvae followed a pattern similar to the distribution of eggs, only it shifted about one month later. Larval abundance peaked from early to mid-June in the southern portion of Shelikof Strait. Biological and environmental factors may help to retain flathead sole larvae on the continental shelf near their juvenile nursery areas.
