This article describes a new test method for the assessment of the severity of environmental stress cracking of biomedical polyurethanes in a manner that minimizes the degree of subjectivity involved. The effect of applied strain and acetone pre-treatment on degradation of Pellethane 2363 80A and Pellethane 2363 55D polyurethanes under in vitro and in vivo conditions is studied. The results are presented using a magnification-weighted image rating system that allows the semi-quantitative rating of degradation based on distribution and severity of surface damage. Devices for applying controlled strain to both flat sheet and tubing samples are described. The new rating system consistently discriminated between. the effects of acetone pre-treatments, strain and exposure times in both in vitro and in vivo experiments. As expected, P80A underwent considerable stress cracking compared with P55D. P80A produced similar stress crack ratings in both in vivo and in vitro experiments, however P55D performed worse under in vitro conditions compared with in vivo. This result indicated that care must be taken when interpreting in vitro results in the absence of in vivo data. (C) 2001 Elsevier Science Ltd. All rights reserved.

The prevalence of type 2 diabetes among Australian residents is 7.5%; however, prevalence rates up to six times higher have been reported for indigenous Australian communities. Epidemiological evidence implicates genetic factors in the susceptibility of indigenous Australians to type 2 diabetes and supports the hypothesis of the thrifty genotype, but, to date, the nature of the genetic predisposition is unknown. We have ascertained clinical details from a community of indigenous Australian descent in North Stradbroke Island, Queensland. In this population, the phenotype is characterized by severe insulin resistance. We have conducted a genomewide scan, at an average resolution of 10 cM, for type 2 diabetes-susceptibility genes in a large multigeneration pedigree from this community. Parametric linkage analysis undertaken using FASTLINK version 4.1p yielded a maximum two-point LOD score of +2.97 at marker D2S2345. Multipoint analysis yielded a peak LOD score of +3.9

Many of the asexual stage Plasmodium falciparum proteins that are the targets of host protective responses are markedly polymorphic. The full repertoire of diversity is not defined for any antigen. Most studies have focused on the genes encoding merozoite surface proteins 1 and 2 (MSP1, MSP2). We explored the extent of diversity of some of the less studied merozoite surface antigens and analyzed the degree of complexity of malaria field isolates by deriving nucleotide sequences of several antigens. We have determined the genotype of apical membrane antigen 1 (AMA1) in a group of 30 field samples, collected over 29 months, from individuals living in an area of intense malaria transmission in Irian Jaya, identifying 14 different alleles. AMA1 genotyping was combined with previously determined MSP2 typing. AMA1 had the greatest power in distinguishing between isolates but methodological problems, especially when mixed infections are present, suggest it is not an ideal typing target. MSP1, MSP3, and glutamate-rich protein genotypes were also determined from a smaller group of samples, and all results were combined to derive an extended antigenic haplotype. Within this subset of 10 patients, nine different genotypes could be discerned; however, five patients were all infected with the same strain. This strain was present in individuals from two separate villages and was still present 12 months later. This strain was predominant at the first time point but had disappeared at the fourth time point. This significant change in malaria genotypes could be due to strain-specific immunity developing in this population.

Quantifying the analgesic effect of placebo electrotherapy is an important part of understanding the placebo response in physiotherapy. This repeated measures study of induced ischaemic pain compared reports of pain threshold, pain tolerance, and pain endurance under three conditions: control, placebo interferential, and placebo TENS. Both of the placebo conditions significantly delayed the report of pain threshold. Placebo interferential also delayed pain tolerance. Each placebo condition reduced pain intensity in the 6th minute. Only placebo TENS reduced pain at the 9th minute of ischaemic pain. The nature of pain reduction in the placebo conditions suggests that analgesia was due to learned expectancies and endogenous opioid release. Further research into the impact of positive expectancies of pain relief in our patients could clarify the efficacy of physiotherapy outcomes for pain.

Over the past 20 years, the incidence of cutaneous malignant melanoma (CMM) has increased dramatically worldwide. A positive family history of the disease is among the most established risk factors for CMM; it is estimated that 10% of CMM cases result from an inherited predisposition. Although mutations in two genes, CDKN2A and CDK4, have been shown to confer an increased risk of CMM, they account for only 20%-25% of families with multiple cases of CMM. Therefore, to localize additional loci involved in melanoma susceptibility, we have performed a genomewide scan for linkage in 49 Australian pedigrees containing at least three CMM cases, in which CDKN2A and CDK4 involvement has been excluded. The highest two-point parametric LOD score (1.82; recombination fraction [theta] 0.2) was obtained at D1S2726, which maps to the short arm of chromosome 1 (1p22). A parametric LOD score of 4.65 (theta = 0) and a nonparametric LOD score of 4.19 were found at D1S2779 in nine families selected for early age at onset. Additional typing yielded seven adjacent markers with LOD scores 13 in this subset, with the highest parametric LOD score, 4.95 (theta = 0) ( nonparametric LOD score 5.37), at D1S2776. Analysis of 33 additional multiplex families with CMM from several continents provided further evidence for linkage to the 1p22 region, again strongest in families with the earliest mean age at diagnosis. A nonparametric ordered sequential analysis was used, based on the average age at diagnosis in each family. The highest LOD score, 6.43, was obtained at D1S2779 and occurred when the 15 families with the earliest ages at onset were included. These data provide significant evidence of a novel susceptibility gene for CMM located within chromosome band 1p22.

Fatty acids inhibit insulin-mediated glucose metabolism in skeletal muscle, an effect largely attributed to defects in insulin-mediated glucose transport. Insulin-resistant mice transgenic for the overexpression of lipoprotein lipase (LPL) in skeletal muscle were used to examine the molecular mechanism(s) in more detail. Using DNA gene chip array technology, and confirmation by RT-PCR and Western analysis, increases in the yeast Sec1p homolog Munc18c mRNA and protein were found in the gastrocnemius muscle of transgenic mice, but not other tissues. Munc18c has been previously demonstrated to impair insulin-mediated glucose transport in mammalian cells in vitro. Of interest, stably transfected C2C12 cells overexpressing LPL not only demonstrated increases in Munc18c mRNA and protein but also in transcription rates of the Munc18c gene. jlr To confirm the relevance of fatty acid metabolism and insulin resistance to the expression of Munc18c in vivo, a 2-fold increase in Munc18c protein was demonstrated in mice fed a high-fat diet for 4 weeks. Together, these data are the first to implicate in vivo increases in Munc18c as a potential contributing mechanism to fatty acid-induced insulin resistance.

Pili of pathogenic Neisseria are major virulence factors associated with adhesion, cytotoxicity, twitching motility, autoaggregation, and DNA transformation. Pili are modified posttranslationally by the addition of phosphorylcholine. However, no genes involved in either the biosynthesis or the transfer of phosphorylcholine in Neisseria meningitidis have been identified. In this study, we identified five candidate open reading frames (ORFs) potentially involved in the biosynthesis or transfer of phosphorylcholine to pilin in N. meningitidis. Insertional mutants were constructed for each ORF in N. meningitidis strain C311#3 to determine their effect on phosphorylcholine expression. The effect of the mutant ORFs on the modification by phosphorylcholine was analyzed by Western analysis with phosphorylcholine-specific monoclonal antibody TEPC-15. Analysis of the mutants showed that ORF NMB0415, now defined as pptA (pilin phosphorylcholine transferase A), is involved in the addition of phosphorylcholine to pilin in N. meningitidis. Additionally, the phase variation (high frequency on-off switching of expression) of phosphorylcholine on pilin is due to changes in a homopolymeric guanosine tract in pptA.

Camundongos normais, utilizados como filtros biológicos, foram inoculados endovenosamente, com tripomastigotas sangüícolas da derivada RCL da cepa RC do T. cruzi. Decorridas 48 horas, os animais foram submetidos ao xenodiagnóstico e à punção cardíaca para semeadura do sangue em meio de Warren. No estômago dos triatomíneos e em cultura a 28°C, os tripomastigotas diferenciaram-se em formas arredondadas (esferomastigotas e/ou amastigotas). Esse comportamento das formas sangüícolas largas tem sido observado com freqüência e nos leva a inferir que a biologia do T. cruzi não estaria apenas relacionada com a cepa, mas, eventualmente, com populações do parasita.

Em Julho de 2005, o Prémio Nobel da Medicina ou Fisiologia foi atribuído a dois australianos, o patologista Robin Warren e o médico Barry Marshall, pelo seu trabalho sobre a bactéria Helicobacter pylori e a sua relação com patologias comuns da região gástrica. Para alguns, a distinção não era senão o reconhecimento, há muito merecido, de uma contribuição de grande relevância para a medicina. Para outros, ela significou o triunfo de um estilo de investigação em medicina com raízes nos trabalhos pioneiros da microbiologia do século XIX. Para outros, ainda, o significado da distinção residia no facto de, dessa vez, o Prémio Nobel consagrar um trabalho com um impacto visível e significativo nas vidas e no bem-estar de milhões de doentes pelo mundo fora. Não deixou de ser notado que, na ocasião, pouco se falou de genes, de genomas ou de temas ou tecnologias “de ponta”. Warren e Marshall haviam procedido à identificação, isolamento e cultura de um agente infeccioso e demonstrado as relações causais entre este e algumas patologias comuns do estômago, como a gastrite crónica ou a úlcera péptica, abrindo assim caminho ao diagnóstico e a terapias eficazes dessas patologias. Os dois australianos mostraram que havia caminhos modestos que também levavam ao Nobel, sem a necessidade de ceder às modas científicas ou de procurar aparecer nas manchetes. Em todo o caso, tratava-se do coroar de um longo processo, iniciado em finais da década de 1970, que levou a bactéria Helicobacter pylori a tornar-se num ponto de passagem obrigatório para os gastroenterologistas e para os que eram afectados por doenças gástricas. Warren e Marshall tornavam-se, assim, os principais porta-vozes da bactéria e das associações que permitiam que o que outrora fora considerado como uma entidade inexistente se tornasse uma entidade biomédica real.

The higher education system in Europe is currently under stress and the debates over its reform and future are gaining momentum. Now that, for most countries, we are in a time for change, in the overall society and the whole education system, the legal and political dimensions have gained prominence, which has not been followed by a more integrative approach of the problem of order, its reform and the issue of regulation, beyond the typical static and classical cost-benefit analyses. The two classical approaches for studying (and for designing the policy measures of) the problem of the reform of the higher education system - the cost-benefit analysis and the legal scholarship description - have to be integrated. This is the argument of our paper that the very integration of economic and legal approaches, what Warren Samuels called the legal-economic nexus, is meaningful and necessary, especially if we want to address the problem of order (as formulated by Joseph Spengler) and the overall regulation of the system. On the one hand, and without neglecting the interest and insights gained from the cost-benefit analysis, or other approaches of value for money assessment, we will focus our study on the legal, social and political aspects of the regulation of the higher education system and its reform in Portugal. On the other hand, the economic and financial problems have to be taken into account, but in a more inclusive way with regard to the indirect and other socio-economic costs not contemplated in traditional or standard assessments of policies for the tertiary education sector. In the first section of the paper, we will discuss the theoretical and conceptual underpinning of our analysis, focusing on the evolutionary approach, the role of critical institutions, the legal-economic nexus and the problem of order. All these elements are related to the institutional tradition, from Veblen and Commons to Spengler and Samuels. The second section states the problem of regulation in the higher education system and the issue of policy formulation for tackling the problem. The current situation is clearly one of crisis with the expansion of the cohorts of young students coming to an end and the recurrent scandals in private institutions. In the last decade, after a protracted period of extension or expansion of the system, i. e., the continuous growth of students, universities and other institutions are competing harder to gain students and have seen their financial situation at risk. It seems that we are entering a period of radical uncertainty, higher competition and a new configuration that is slowly building up is the growth in intensity, which means upgrading the quality of the higher learning and getting more involvement in vocational training and life-long learning. With this change, and along with other deep ones in the Portuguese society and economy, the current regulation has shown signs of maladjustment. The third section consists of our conclusions on the current issue of regulation and policy challenge. First, we underline the importance of an evolutionary approach to a process of change that is essentially dynamic. A special attention will be given to the issues related to an evolutionary construe of policy analysis and formulation. Second, the integration of law and economics, through the notion of legal economic nexus, allows us to better define the issues of regulation and the concrete problems that the universities are facing. One aspect is the instability of the political measures regarding the public administration and on which the higher education system depends financially, legally and institutionally, to say the least. A corollary is the lack of clear strategy in the policy reforms. Third, our research criticizes several studies, such as the one made by the OECD in late 2006 for the Ministry of Science, Technology and Higher Education, for being too static and neglecting fundamental aspects of regulation such as the logic of actors, groups and organizations who are major players in the system. Finally, simply changing the legal rules will not necessary per se change the behaviors that the authorities want to change. By this, we mean that it is not only remiss of the policy maker to ignore some of the critical issues of regulation, namely the continuous non-respect by academic management and administrative bodies of universities of the legal rules that were once promulgated. Changing the rules does not change the problem, especially without the necessary debates form the different relevant quarters that make up the higher education system. The issues of social interaction remain as intact. Our treatment of the matter will be organized in the following way. In the first section, the theoretical principles are developed in order to be able to study more adequately the higher education transformation with a modest evolutionary theory and a legal and economic nexus of the interactions of the system and the policy challenges. After describing, in the second section, the recent evolution and current working of the higher education in Portugal, we will analyze the legal framework and the current regulatory practices and problems in light of the theoretical framework adopted. We will end with some conclusions on the current problems of regulation and the policy measures that are discusses in recent years.

Numerous pulmonary schistosome egg granulomas were present in mice submitted to partial portal vein ligation (Warren's model). The granulomas were characterized by cellular aggregations formed within alveolar tissue. Main cellular types were macrophages (epithelioid cells), eosinophils, plasma cells and lymphocytes. These cells were supported by scanty fibrous stroma and exhibited close membrane contact points amongst themselves, but without forming specialized adhesion apparatus. When granulomas involved arterial structures, proliferation of cndothelial and smooth muscle cells occurred and fibrosis associated with angiogenesis became more evident. Granulomas formed around mature eggs in the pulmonary alveolar tissue presented approximately the same size and morphology regardless of the time of infection, the latter being 10, 18 and 25 weeks after cercarial exposure. This persistence of morphological appearance suggests that pulmonary granulomas do not undergo immunological modulation, as is the case with the granulomas in the liver and, to a lesser extent, in the intestines. Probably, besides general immunological factors, local (stromal) factors play an important role in schistosomal granuloma modulation.

Trabalho académico com o objetivo do autor desenvolver um estudo prévio e um projeto de uma travessia sobre o rio Lima, na cidade de Viana do Castelo constituída por uma ponte de tirantes rodoferroviária. O projeto académico visa, também, desenvolver e compreender: os conceitos básicos, as metodologias de conceção, e o funcionamento de estruturas desse género. O motivo principal da escolha do tema é a necessidade de uma alternativa à ponte Eiffel em Viana do Castelo, e juntando o facto de em Portugal não existir nenhuma obra de arte de tirantes rodoferroviária até ao presente, seria interessante estudar e projetar uma estrutura rodoferroviária de tirantes. Das diversas possibilidades de sistemas estruturais estudados, adotou-se uma ponte que acomodará 4 vias rodoviárias e 2 vias ferroviárias, com um desenvolvimento total de 660 metros, constituída por dois vãos laterais com 165 metros cada um, e com um vão central de 330 metros. A obra de arte será em semi-leque com dois planos de tirantes, ancorados a duas torres de betão em Y invertido de altura aproximadamente de 110 metros. O tabuleiro será duplo misto aço-betão, constituído por duas vigas trianguladas do tipo Warren, e por carlingas, afastadas entre si de 15 metros com secções tubulares metálicas de espessura variável. As carlingas ao nível superior suportam a laje de betão, que constitui a rodovia, e inferiormente, suportam outra laje de betão para a parte ferroviária. O trabalho inicia-se com o enquadramento conceptual geral da envolvente da obra de arte, seguidamente com apresentação da evolução histórica ao longo do tempo das pontes de tirantes, e à apresentação de algumas pontes rodoferroviárias de tirantes. É realizada uma análise preliminar, onde se estudam as restrições, as condicionantes, o local de implantação, e o sistema da configuração geométrica a adotar na conceção estrutural. São descritos todos os tipos de materiais, equipamentos a utilizar, bem como as suas características mecânicas necessárias para o cálculo estrutural. A quantificação das ações e das combinações de cálculo efetuaram-se de acordo com as normas em vigor nacionais e europeias, designadamente os Eurocódigos das várias especialidades e o Regulamento de Segurança e Ações para Estruturas de Edifícios e Pontes. Efetuou-se um pré-dimensionamento e uma otimização de vários sistemas estruturais possíveis de todos os elementos estruturais, tendo em conta variáveis de estudo como a economia e a resistência estrutural das secções, por forma a chegar à solução final. A estrutura foi discretizada e analisada num modelo estático tridimensional num programa de cálculo automático. A análise de resultados foi efetuada longitudinalmente para a verificação dos Estados Limites Últimos e Estados Limites de Utilização dos elementos estruturais que constituem a ponte. Foi ainda efetuada uma estimativa orçamental da ponte no rio Lima na cidade de Viana do Castelo.
