Con el objetivo de evaluar la influencia de las bandejas de celdas sobre la producción de tomate tipo italiano en el campo, se realizó este trabajo en Piracicaba, SP, Brasil, de mayo a agosto del 2005. Los tratamientos consistieron en cuatro volúmenes de recipiente, tres bandejas de poliestireno expandido de 121,2; 34,6 y 12,0 cm³ y de una bandeja de plástico rígido de 14,0 cm³ (72, 128, 288 y 450 celdas, respectivamente) combinadas con cuatro edades para el trasplante (19, 24, 29 y 34 días después de la siembra). El delineamiento para la producción de mudas fue completamente al azar, con cinco plantas por parcela y tres repeticiones. Se analizaron área foliar, altura, masa verde y seca de la parte aérea y raíz y la calidad de las mudas. En la producción a campo, el delineamiento fue en bloques al azar con diez plantas por parcela y tres repeticiones. Fueron evaluadas la precocidad para inicio de cosecha, producción comercial y total por planta. Volúmenes mayores de recipiente (121,2 y 34,6 cm³) presentaron mejor calidad de mudas. En la producción de frutos, el número comercial y total de frutos por planta fue superior en la muda de 24 días de edad, sin embargo, en la producción total de frutos, no hubo diferencia entre los tratamientos. Por otro lado, también se obtuvo precocidad para la cosecha en los tratamientos realizados en los volúmenes de 121,2 y 34,6 cm³. El volumen de recipiente de 14,0 cm³ (450 celdas) resultó en mudas de calidad muy inferior, alongadas y raquíticas.

In the recent years, consumers became more aware and sensible in respect to environment and food safety matters. They are more and more interested in organic agriculture and markets and tend to prefer ‘organic’ products more than their traditional counterparts. To increase the quality and reduce the cost of production in organic and low-input agriculture, the 6FP-European “QLIF” project investigated the use of natural products such as bio-inoculants. They are mostly composed by arbuscular mycorrhizal fungi and other microorganisms, so-called “plant probiotic” microorganisms (PPM), because they help keeping an high yield, even under abiotic and biotic stressful conditions. Italian laws (DLgs 217, 2006) have recently included them as “special fertilizers”. This thesis focuses on the use of special fertilizers when growing tomatoes with organic methods in open field conditions, and the effects they induce on yield, quality and microbial rhizospheric communities. The primary objective was to achieve a better understanding of how plant-probiotic micro-flora management could buffer future reduction of external inputs, while keeping tomato fruit yield, quality and system sustainability. We studied microbial rhizospheric communities with statistical, molecular and histological methods. This work have demonstrated that long-lasting introduction of inoculum positively affected micorrhizal colonization and resistance against pathogens. Instead repeated introduction of compost negatively affected tomato quality, likely because it destabilized the ripening process, leading to over-ripening and increasing the amount of not-marketable product. Instead. After two years without any significant difference, the third year extreme combinations of inoculum and compost inputs (low inoculum with high amounts of compost, or vice versa) increased mycorrhizal colonization. As a result, in order to reduce production costs, we recommend using only inoculum rather than compost. Secondly, this thesis analyses how mycorrhizal colonization varies in respect to different tomato cultivars and experimental field locations. We found statistically significant differences between locations and between arbuscular colonization patterns per variety. To confirm these histological findings, we started a set of molecular experiments. The thesis discusses preliminary results and recommends their continuation and refinement to gather the complete results.

Neben Tomatensaft wurde eine Vielzahl von Säften und Blattextrakten als Medienzusätze auf Wachstumsförderung bei 30 verschiedenen Oenococcus oeni-Stämmen getestet. Es zeigte sich eine breite Wachstumsförderung bei allen Zusätzen mit Ausnahme von Zitronensaft, sodass die Wachstumsfaktoren keine tomatenspezifischen Inhaltsstoffe sein können und eher ubiquitär in der Pflanzenwelt vorkommen. Das Ausmaß der Wachstumsförderung war stammabhängig sehr unterschiedlich und Tomatensaft stellte keineswegs für alle Stämme den optimalen Medienzusatz dar. Durch Schälen der Früchte war eine für die Analytik hilfreiche Abtrennung schalenspezifischer Inhaltsstoffe möglich, wobei auch die Schalenextrakte großes Potential für die Suche nach Wachstumsfaktoren offenbarten und die Wichtigkeit einer Auftrennung der Frucht in die verschiedenen Fruchtbereiche betonte. Aus Tomatensaft konnte analytisch der anorganische Wachstumsfaktor Mangan identifiziert werden. Die größten Zelldichten der Oenokokken-Stämme wurden hierbei bei 67 µM und 34 mM Manganzusatz erreicht. Bei 13 von 20 getesteten Oenokokkenstämmen konnte bei Zusatz von 34 mM Mangan der Tomatensaft ersetzt werden, bei 4 Stämmen (z. B. Stamm B2) fehlten jedoch noch weitere Wachstumsfaktoren und bei 3 Stämmen (z. B. Stamm B120) kam es zu einem verfrühten Absterben. Da weitere Mineralstoffe sowie veraschte Säfte und Blattextrakte keinen positiven Einfluß auf die Oenokokken-Zelldichte hatten, wurde mittels semipräparativer HPLC nach zusätzlichen organischen Wachstumsfaktoren für den Stamm B2 gesucht. Hierzu wurde der nachfolgende Wachstums-Assay miniaturisiert und erfolgreich auf Microtiterplatten etabliert. Es gelang die Isolierung und Identifizierung eines wachstumsfördernden Trisaccharides aus Mangoschalen-Extrakt, das aus den Zuckern Glucose, Rhamnose und Arabinose bestand. Von den monomeren Zuckern erhöhte lediglich die Arabinose die Zelldichte, das Optimum lag bei 1,5 g/l. Auch aus Zitronenmesokarp-Extrakt war die Isolierung eines wachstumsfördernden arabinosehaltigen Disaccharides möglich, die Menge reichte jedoch noch nicht für eine genaue Identifizierung aus. Desweiteren erwies sich 1,5 g/l Cystein als wachstumsstimulierend. Ein Zusatz aller gefundenen Wachstumsfaktoren (34 mM Mangan, 1,5 g/l Arabinose und 1,5 g/l Cystein) ersetzte den Tomatensaft bei weiteren Oenokokken-Stämmen (z.B. Stamm B120) komplett, wobei bei allen Stämmen sogar eine schnellere Anzucht erfolgte. Neben dem Tomatensaft war auch der Zusatz von Hefeextrakt zum Grundmedium nicht mehr nötig, sodass ein neues vereinfachtes Medium für die Anzucht von Oenokokken mit komplexen Nährstoffansprüchen vorgeschlagen werden konnte. Lediglich beim Stamm B2 zeigte sich noch ein OD-Unterschied von 0,2 in der stationären Phase, der nach Adsorptionsversuchen an Polyvinylpolypyrrolidon auf noch unidentifizierte Polyphenole im Tomatensaft zurückzuführen ist. Aus grünem Tee erwies sich das Polyphenol Epigallocatechingallat (EGCG) konzentrationsabhängig sowohl als Hemmstoff (>550 mg/l EGCG) als auch Wachstumsfaktor (400-500 mg/l EGCG) für den Oenokokken-Stamm B2. Der hemmende als auch der fördernde Einfluss auf das Wachstum wurde mittels Sytox/DAPI-Färbung bestätigt. Der sogenannte „Tomatensaft-Faktor“ ist also nicht eine spezielle Substanz, sondern das synergistische Zusammenwirken mehrerer einfacher Substanzen wie Mineralstoffe, Aminosäuren, Kohlenhydrate und Polyphenole. Auch sind die Oenokokken-Stämme bezüglich ihres Nährstoffbedarfes sehr unterschiedlich, sodass für jeden Stamm einzeln das optimale Substratspektrum ermittelt werden muss.

Il pomodoro è una delle colture principali del panorama agro-alimentare italiano e rappresenta un ingrediente base della tradizione culinaria nazionale. Il pomodoro lavorato dall’industria conserviera può essere trasformato in diverse tipologie merceologiche, che si differenziano in base alla tecniche di lavorazione impiegate ed alle caratteristiche del prodotto finito. la percentuale di spesa totale destinata all’acquisto di cibo fuori casa è in aumento a livello globale e l’interesse dell’industria alimentare nei confronti di questo canale di vendita è quindi crescente. Mentre sono numerose le indagine in letteratura che studiano i processi di acquisto dei consumatori finali, non ci sono evidenze di studi simili condotti sugli operatori del Food Service. Obiettivo principale della ricerca è quello di valutare le preferenze dei responsabili acquisti del settore Food Service per diverse tipologie di pomodoro trasformato, in relazione ad una gamma di attributi rilevanti del prodotto e di caratteristiche del cliente. La raccolta dei dati è avvenuta attraverso un esperimento di scelta ipotetico realizzato in Italia e alcuni mercati esteri. Dai risultati ottenuti dall’indagine emerge che i Pelati sono la categoria di pomodoro trasformato preferita dai responsabili degli acquisti del settore Food Service intervistati, con il 35% delle preferenze dichiarate nell'insieme dei contesti di scelta proposti, seguita dalla Polpa (25%), dalla Passata (20%) e dal Concentrato (15%). Dai risultati ottenuti dalla stima del modello econometrico Logit a parametri randomizzati è emerso che alcuni attributi qualitativi di fiducia (credence), spesso impiegati nelle strategie di differenziazione e posizionamento da parte dell’industria alimentare nel mercato Retail, possono rivestire un ruolo importante anche nell’influenzare le preferenze degli operatori del Food Service. Questo potrebbe quindi essere un interessante filone di ricerca da sviluppare nel futuro, possibilmente con l'impiego congiunto di metodologie di analisi basate su esperimenti di scelta ipotetici e non ipotetici.

Tomatoes are the most common crop in Italy. The production cycle requires operations in the field and factory that can cause musculoskeletal disorders due to the repetitive movements of the upper limbs of the workers employed in the sorting phase. This research aims to evaluate these risks using the OCRA (occupational repetitive actions) index method This method is based firstly on the calculation of a maximum number of recommended actions, related to the way the operation is performed, and secondly on a comparison of the number of actions effectively carried out by the upper limb with the recommended calculated value. The results of the risk evaluation for workers who manually sort tomatoes during harvest showed a risk for the workers, with an exposure index greater than 20; the OCRA index defines an index higher than 3.5 as unacceptable. The present trend of replacing manual sorting onboard a vehicle with optical sorters seems to be appropriate to reduce the risk of work-related musculoskeletal disorders (WMSDs) and is supported from both a financial point of view and as a quality control measure.

Expansins are members of a multigene family of extracellular proteins, which increase cell wall extensibility in vitro and thus are thought to be involved in cell expansion. The major significance of the presence of this large gene family may be that distinctly expressed genes can independently regulate cell expansion in place and time. Here we report on LeExp9, a new expansin gene from tomato, and compare its expression in the shoot tip with that of LeExp2 and LeExp18. LeExp18 gene is expressed in very young tissues of the tomato shoot apex and the transcript levels are upregulated in the incipient primordium. LeExp2 mRNA accumulated in more mature tissues and transcript levels correlated with cell elongation in the elongation zone. In situ hybridization experiments showed a uniform distribution of LeExp9 mRNA in submeristematic tissues. When gibberellin-deficient mutant tomatoes that lacked elongation of the internodes were treated with gibberellin, the phenotypic rescue was correlated with an increase in LeExp9 and LeExp2, but not LeExp18 levels. We propose that the three expansins define three distinct growing zones in the shoot tip. In the meristem proper, gibberellin-independent LeExp18 mediates the cell expansion that accompanies cell division. In the submeristematic zone, LeExp9 mediates cell expansion at a time that cell division comes to a halt. LeExp9 expression requires gibberellin but the hormone is not normally limiting. Finally, LeExp2 mediates cell elongation in young stem tissue. LeExp2 expression is limited by the available gibberellin. These data suggest that regulation of cell wall extensibility is controlled, at least in part, by differential regulation of expansin genes.

Empirical research on discriminatory attitudes and behaviour grapples with the social undesirability of its object. In many studies using regular survey methods, estimates are biased, and the social context of discrimination is not taken into account. Several methods have been developed, especially to deal with the first problem. In this regard, the estimation of the ‘true value’ of discriminatory attitudes is at the centre of interest. However, methodological contributions focusing on the social context of attitude communication and discriminatory behaviour, as well as the correlation between both, are rare. We present two experimental methods which address those issues: factorial surveys and stated choice experiments. In a first study, the usefulness of factorial surveys is demonstrated with data on German anti-Semitism (N=279). We show that the rate of approval with anti-Semitic statements increases if (a) respondents are told that the majority of fellows agree with such statements, (b) the term “Jews” is replaced by the term “Israelis”, and (c) reference to the Holocaust is made. Apart from the main effects of these experimental factors, significant interaction effects regarding the political attitudes and social status of respondents are observed. In a second study, a stated choice experiment on the purchase of olive oil and tomatoes was conducted in Germany (N=440). We find that respondents prefer Italian and Dutch products (control treatment) compared to Israeli and Palestinian ones (discrimination treatments). There are no significant differences between preferences for a so called ‘Peace product’ (which is produced jointly by Israelis and Palestinians) and products from Italy as well as the Netherlands. Yet, taking discriminatory attitudes (anti-Semitic and anti-Arabic attitudes) into account, a strong correlation between those attitudes and stated behaviour (purchase of Israeli, Palestinian and jointly produced products) can be found. This adds support to the hypothesis that discriminatory attitudes hold behavioural consequences.

Background. The prevalence of obesity and overweight children has been an ongoing health epidemic in the US for the last several decades. The problem has consistently worsened and has disproportionately been the most prevalent among low socioeconomic status (SES) populations. Food availability in the home has been suggested to be a potential factor related to overweight and obesity, as availability is likely associated with intake. Food availability of low SES preschool aged children has not been well examined. The purpose of this study was to explore the food environment of the Harris County Department of Education (HCDE) Head Start population, and describe reported frequency of intake of particular food groups. The effect of food availability on reported intake was also examined.^ Methods. This was a cross-sectional study of secondary data analysis. Data obtained from 17 HCDE Head Start Centers was analyzed using PASW 18 Statistical Software. Demographic analyses included population, age, gender, race, parent occupation, type of home, and language spoken in the home. Descriptive statistics included reported availability of foods in the home as well as frequency of intake.^ Regression analysis examined the relationship of availability of foods on intake. The food categories included were: dark leafy green and orange vegetables, other vegetables, fruits, soda, salty snacks, and sweet snacks. For both vegetable categories reported intake of fresh, frozen, and canned vegetables were included. For the fruit category, intake of fresh, frozen, canned, and dried fruits were reported.^ Results. Results showed that 90-95% of parents reported having vegetables and fruits available in the home. However, the only significant relationship between availability and intake was for fresh fruit and dried fruit. No associations were seen among the vegetable groups. Other vegetables (bell peppers, eggplant, tomatoes, onions, iceberg lettuce, asparagus) that were frozen, approached significance for availability on intake, however once adjusted for confounders the relationship was no longer present. Among soda, salty snacks, and sweet snacks the only significant relationship was seen for soda availability and intake. Salty snacks and sweet snacks presence in the home was not a predictor of increased frequency of intake.^ Conclusions. This research supported the hypothesis that availability of foods has an impact on intake for fresh fruits, dried fruits and soda. No associations were seen for vegetables, salty snacks and sweet snacks. Additionally, most of the parents reported having fruits and vegetables in the home, but reported intakes were not meeting the Dietary Guidelines for Americans recommendations. Strengths of the study included the large sample size taken from numerous HCDE Head Start Centers. Limitations included questionable reliability of participant’s responses, ability to generalize to other populations, and the use of secondary data rather than prospectively collected data.^

La antesis de una flor es un estadio crucial para el destino del ovario ya que estímulos endógenos -polinización o nivel hormonal elevado- o exógenos -aplicación hormonal- inducen el proceso de fructificación. En ausencia de estos estímulos el ovario deja de crecer e inicia un proceso de senescencia. Previamente se ha observado que, en ovarios no polinizados, la senescencia va acompañada por un incremento en la actividad/expresión de ciertas tiol-proteasas, parámetros que disminuyen durante la etapa temprana del desarrollo de frutos. El objetivo fue analizar la localización de proteasas y los cambios anatómicos asociados a los procesos de fructificación o senescencia que se producen durante la etapa temprana del desarrollo de los frutos de tomate obtenidos por polinización natural o por tratamiento con AG3. Durante el período en que los ovarios no polinizados mantienen su sensibilidad a la aplicación hormonal, se observó una acumulación de polipéptidos relacionados antigénicamente con la tiol-proteasa papaína, principalmente en la epidermis del pericarpio, sin evidenciarse alteraciones en la integridad de los tejidos. Frutos de 8 días de desarrollo obtenidos con AG3, en comparación con los polinizados, evidenciaron un incremento en el espesor del pericarpio relacionado principalmente con una mayor elongación de las células del mesocarpio.

Tuta absoluta (Meyrick) (Lepidoptera- Gelechiidae) es una plaga del tomate cultivado en Mendoza (Argentina) que afecta la calidad y la conservación de su producto. Su cría artificial permite conocer su bioetología para aplicarla a su control. Los objetivos de este trabajo fueron: • Adaptar la especie a la reproducción en condiciones artificiales. • Evaluar dietas para la cría de las larvas. La fundación de la cría partió de larvas salvajes extraídas de hojas y frutos dañados de un cultivo bajo cubierta, ubicado en Guaymallén (Mendoza). La cría desarrolló dentro de cámaras con condiciones controladas de temperatura, humedad y fotoperíodo. Para el apareamiento y oviposición de los adultos se diseñaron tres tipos de jaulas. Como sustratos de oviposición se utilizó papel apergaminado, entretela y algodón. Sobre papel no se obtuvieron posturas. La mayor cantidad de huevos se registró sobre algodón. Los huevos obtenidos se sembraron en dieta artificial y natural. Las larvas se lograron alimentar con dieta artificial pero los adultos -obtenidos con alto rendimiento: 67,58 % huevo a adulto- no dieron suficiente descendencia para mantener la cría.

El tomate de cáscara (Physalis ixocarpa Brot.) es un cultivo alimenticio de gran importancia económica en México. Sin embargo, es afectado por diversas plagas y enfermedades tales como los Thrips (Thysanoptera: Frankliniella occidentalis) y el virus de la marchitez manchada del tomate (TSWV) que llegan a causar hasta un 80% de pérdidas. El objetivo del presente trabajo fue modelizar la distribución espacial de huevos de Thrips mediante técnicas geoestadísticas y obtener, en consecuencia, mapas de incidencia por medio del Kriging. Se georreferenciaron 121 puntos de muestreo en cada una de las parcelas comerciales de los municipios de Luvianos, Jocotitlán e Ixtlahuaca, a través del método de transectos en tres etapas fenológicas del cultivo. Se contabilizó el número de huevos de Thrips en cada punto de muestreo. Los resultados mostraron que las poblaciones de huevos deThrips presentan una distribución agregada, identificándose varios centros de conglomeración a través de los mapas obtenidos. Los semivariogramas obtenidos de la distribución espacial se ajustaron principalmente a los modelos gaussianos y esféricos. La distribución de huevos de Thrips se presentó en centros de agregación dentro de las parcelas estudiadas, lo cual permitirá establecer estrategias y medidas de control o mitigación en términos de sitios específicos de infestación de huevos de Thrips.
