Antiophidic activity from decoct of Jatropha gossypiifolia L. leaves against Bothrops jararaca venom. Snakebites are a serious worldwide public health problem. In Latin America, about 90 % of accidents are attributed to snakes from Bothrops genus. Currently, the main available treatment is the antivenom serum therapy, which has some disadvantages such as inability to neutralize local effects, risk of immunological reactions, high cost and difficult access in some regions. In this context, the search for alternative therapies to treat snakebites is relevant. Jatropha gossypiifolia L., a medicinal plant popularly known in Brazil as “pinhão-roxo”, is very used in folk medicine as antiophidic. So, the aim of this study is to evaluate the antiophidic properties of this species against enzymatic and biological activities from Bothrops jararaca snake venom. The aqueous leaf extract of J. gossypiifolia was prepared by decoction. The inhibition studies were performed in vitro, by pre-incubation of a fixed amount of venom with different amounts of extract from J. gossypiifolia for 60 min at 37 °C, and in vivo, through oral or intraperitoneal treatment of animals, in different doses, 60 min before venom injection. The proteolytic activity upon azocasein was efficiently inhibited, indicating inhibitory action upon metalloproteinases (SVMPs) and/or serine proteases (SVSPs). The extract inhibited the fibrinogenolytic activity, which was also confirmed by zymography, where it was possible to observe that the extract preferentially inhibits fibrinogenolytic enzymes of 26 and 28 kDa. The coagulant activity upon fibrinogen and plasma were significantly inhibited, suggesting an inhibitory action upon thrombin-like enzymes (SVTLEs), as well as upon clotting factor activators toxins. The extract prolonged the activated partial thromboplastin time (aPTT), suggesting an inhibitory action toward not only to SVTLEs, but also against endogenous thrombin. The defibrinogenating activity in vivo was efficiently inhibited by the extract on oral route, confirming the previous results. The local hemorrhagic activity was also significantly inhibited by oral route, indicating an inhibitory action upon SVMPs. The phospholipase activity in vitro was not inhibited. Nevertheless, the edematogenic and myotoxic activities were efficiently inhibited, by oral and intraperitoneal route, which may indicate an inhibitory effect of the extract upon Lys49 phospholipase (PLA2) and/ or SVMPs, or also an anti-inflammatory action against endogenous chemical mediators. Regarding the possible action mechanism, was observed that the extract did not presented proteolytic activity, however, presented protein precipitating action. In addition, the extract showed significant antioxidant activity in different models, which could justify, at least partially, the antiophidic activity presented. The metal chelating action presented by extract could be correlated with SVMPs inhibition, once these enzymes are metal-dependent. The phytochemical analysis revealed the presence of sugars, alkaloids, flavonoids, tannins, terpenes and/or steroids and proteins, from which the flavonoids could be pointed as major compounds, based on chromatographic profile obtained by thin layer chromatography (TLC). In conclusion, the results demonstrate that the J. gossypiifolia leaves decoct present potential antiophidic activity, including action upon snakebite local effects, suggesting that this species may be used as a new source of bioactive molecules against bothropic venom.

Envenomation caused by venomous animals, mainly scorpions and snakes, are a serious matter of public health. Tityus serrulatus is considered the most venomous scorpion in South America because of the high level of toxicity of its venom. It is responsible for causing serious accidents, mainly with kids. The species Bothrops jararaca is a serpent that has in its venom a complex mixture of enzyme, peptides and other molecules. The toxins of the venom of B. jararaca induce local and systemic inflammatory responses. The treatment chosen to serious cases of envenomation is the intravenous administration of the specific antivenom. However, the treatment is not always accessible to those residents in rural areas, so that they use medicinal plant extracts as the treatment. In this context, aqueous extracts, fractions and isolated compounds of Aspidosperma pyrifolium (pereiro) and Ipomoea asarifolia (salsa, salsa-brava), used in popular medicine, were studied in this research to evaluate the anti-inflammatory activity in the peritonitis models induced by carrageenan and peritonitis induced by the venom of the T. serrulatus (VTs), and in the local oedema model and inflammatory infiltrate induced by the venom of the B. jararaca, administrated intravenously. The results of the assays of cytotoxicity, using the MTT, showed that the aqueous extracts from the plant species presented low toxicity to the cells that came from the fibroblast of the mouse embryo (3T3).The chemical analysis of the extracts by High Performance Liquid Chromatography revealed the presence of the rutin flavonoid, in A. pyrifoliu, and rutin, clorogenic acid and caffeic acid, in I. asarifolia. Concerning the pharmacological evaluation, the results showed that the pre-treatment using aqueous extracts and fractions reduced the total leukocyte migration to the abdominal cavity in the peritonitis model caused by the carrageenan and in the peritonitis model induced by the T. serulatus venom. Yet, these groups presented anti-oedematous activity, in the local oedema model caused by the venom of the B. jararaca, and reduced the inflammatory infiltrate to the muscle. The serum (anti-arachnid and anti-bothropic) specific to each venom acted inhibiting the inflammatory action of the venoms and were used as control. The compounds identified in the extracts were also tested and, similar to the plant extracts, showed meaningful anti-inflammatory effects, in the tested doses. Thus, these results are indicating the potential anti-inflammatory activity of the plants studied. This is the first research that evaluated the possible biological effects of the A. pyrifolium and I. asarifolia, showing the biological potential that these species have.

The local effects (pain, edema, hemorrhage and myonecrosis) and systemic (cardiovascular alterations, shock and blood clotting disorders) caused by the venom of Bothrops are due to the numerous protein and non-protein components, which are part of the constitution of the poison. The only form of therapy is scientifically validated antivenom serum therapy which, however, is not effective with respect to local effects produced, risk of immunological reactions, high cost and difficult access in some regions. Thus, the search for new alternatives to serum therapy becomes important, and in this context, many medicinal plants have been highlighted by the popular use as antiophidic. Among these plants, we can mention the species Jatropha mollissima (Euphorbiaceae) which has popular use in traditional medicine as antiophidic, anti-inflammatory, antimicrobial and antipyretic. Therefore, this study aims to evaluate the neutralizing potential of local effects induced by the venom of Bothrops erythromelas and Bothrops jararaca with the aqueous extract of the leaves of J. mollissima. The leaf extracts were prepared by decoction, fractionated (by liquid-liquid partition) and characterized by thin layer chromatography (TLC) and High Performance Liquid Chromatography (HPLC). Antiophidic activity of the extract was evaluated in model of paw edema, peritonitis, bleeding and myotoxicity induced by venoms of B. jararaca and B. erythromelas. In all models, the extract was evaluated by intraperitoneal route at the doses of 50, 100 and 200 mg/kg, administered 30 minutes prior to injection of the venom (pretreatment protocol). Stains suggestive of the presence of flavonoids: apigenin, luteolin, orientin, isoorientin, vitexin and vitexin-2-O-rhamnoside were detected in the extract by co-CCD. By means of HPLC were identified isoorientin, orientin, vitexin and isovitexin. All tested doses of J. mollissima extract reduced the paw edema induced by the venom with intensity similar to dexamethasone. The aqueous extract of J. mollissima leaves on all evaluated doses, inhibited cell migration induced by B. jararaca and B. erythromelas promoting inhibition of recruitment of mononuclear cells and the polymorphonuclear cells. Local bleeding induced by B. jararaca venom was significantly inhibited by the extract. Both venoms were inhibited by the extract in myotoxic activity. These results indicate that the aqueous extract of J. mollissima leaves have snakebite potential, particularly with respect to local effects, which may justify the use of this plant in traditional medicine and complementary therapy as anti-venom serum.

O estudo teve como objetivo descrever a biologia reprodutiva e a dieta de populações de Thamnodynastes hypoconia em ambientes úmidos subtemperados do extremo sul brasileiro. Para avaliação da reprodução foram feitas análises macroscópicas das gônadas. Para avaliação da ecologia alimentar foram empregadas análises de conteúdo estomacal e de isótopos estáveis (δ13C e δ15N). Os machos apresentaram maior comprimento rostro-cloacal que as fêmeas e atingiram a maturidade sexual com menor tamanho corporal. O ciclo reprodutivo das fêmeas foi sazonal, com vitelogênese secundária ocorrendo entre o inverno e a primavera. Os machos apresentaram ciclo contínuo. Aparentemente a cópula ocorre entre o final da primavera e o início do verão, quando foram detectados embriões nas fêmeas e a parturição ocorreu no verão. Não houve relação entre o tamanho da ninhada e o tamanho da fêmea, e a frequência reprodutiva de 40% foi considerada relativamente baixa. Todavia, por ser uma espécie dominante em banhados subtemperados do extremo sul do Brasil sua estratégia reprodutiva parece ser eficiente. Dessa forma, o ciclo contínuo dos machos, bem como a viviparidade possam potencializar o deslocamento do pico reprodutivo da população de modo a ajustar-se de acordo com os picos de temperatura do ambiente. A análise de conteúdo estomacal mostrou uma dieta predominantemente anurófaga, exceto por um único lagarto. Hylidae foi a família mais representativa, com predomínio de Hypsiboas pulchellus, seguida de Leptodactylidae, onde Leptodactylus latrans teve maior importância. As análises de isótopos estáveis corroboraram a importância dessas espécies na alimentação da serpente. Apesar de ser uma serpente terrestre, T. hypoconia apresentou assinatura isotópica mais próxima das fontes primárias oriundas do ambiente aquático, o que indica a importância das áreas alagáveis para a subsistência de organismos terrestres de habitats adjacentes.

La evolución ha permitido que varios animales produzcan secreciones para su supervivencia: los venenos, aquellos que son dañinos y letales cuando ejercen sus efectos en el organismo, modifican las respuestas normales como por ejemplo la coagulación y el proceso inflamatorio. Presentamos una revisión bibliográfica actualizada de los venenos que pueden ser empleados y así explicar cómo estas sustancias pueden representar una terapéutica en el futuro; de la misma manera describimos la situación de nuestro país que puede ser fuente de investigación en este tema

La presente recopilación bibliográfica especializada en Toxicología Veterinaria, es un esfuerzo académico que refleja los logros en esta área de la ciencia por diversos investigadores, el cual va encaminado a fortalecer los conocimientos de los estudiantes de Medicina Veterinaria y como fuente de consulta para profesionales de las ciencias veterinarias. El clínico es quien debe enfrentarse a diario con pacientes potencialmente envenenados, y decidir si el diagnóstico de “envenenamiento” es verdadero y, si es así, cual de entre los miles de sustancias químicas o naturales (plantas tóxicas y animales de ponzoña) existentes puede ser la responsable del caso que lo ocupa. Con este documento no se pretende sustituir las diversas publicaciones existentes sobre este tema, pero que sirva de un medio de orientación en el campo de la toxicología veterinaria, ya que resalta los problemas toxicológicos que se presentan comúnmente en la producción de alimentos de naturaleza animal y en mantenimiento de animales de compañía. Se incluye información básica sobre dosisrespuesta y tipos de comprobaciones toxicológicas a que se someten los nuevos compuestos para determinar su seguridad y toxicidad, de forma que el estudiante tenga una idea del tipo de información disponible y de cómo se obtiene. Al organizar este documento se puso énfasis en las intoxicaciones por especie que se observan comúnmente en la práctica diaria. Se espera que el estudiante, veterinario toxicólogo se beneficien al disponer de una información adecuada y útil para tratar las intoxicaciones clínicas y de campo que se observan diariamente. La toxicología, es la ciencia que estudia los venenos o agentes tóxicos, incluyendo sus propiedades químicas, identificación, efectos biológicos y los posibles tratamientos de los efectos que producen. El toxicólogo veterinario requiere de entrenamiento especializado, así como de experiencia en el manejo de varias sustancias venenosas sintéticas o naturales (producida por plantas o animales). Debe además diferenciar las enfermedades infecciosas de las condiciones metabólicas causadas por venenos, también debe conocer la gran variedad de productos químicos agrícolas, aditivos de alimentos, contaminantes ambientales, radiaciones diversas, gases venenosos y venenos de origen vegetal y animal que puedan afectar la salud de los animales. La farmacología y la toxicología comparten muchos intereses, incluyendo mecanismo de absorción y eliminación, mecanismo de acción, principios de tratamiento y relaciones dosis–respuesta. Algunos medicamentos pueden actuar como venenos en ciertas condiciones, por lo tanto el farmacólogo como el toxicólogo comparten un interés por las reacciones adversas de los fármacos. La toxicología se divide en dos aspectos: La toxicología general. La toxicología específica.

Neste estudo, procuramos avaliar a composição e abundância de espécies de anfíbios e répteis em uma paisagem fragmentada de Mata Atlântica, no município de Cachoeiras de Macacu, Estado do Rio de Janeiro. Amostramos a herpetofauna da região na área contínua de floresta da Reserva Ecológica de Guapiaçu (REGUA), em 12 fragmentos do entorno com tamanhos e graus de isolamento diferentes, e áreas de pasto (matriz). Utilizamos para amostragem destes animais as metodologias de buscas ativas visuais e armadilhas de interceptação e queda, além de encontros ocasionais. No total foram registradas 55 espécies de anfíbios anuros pertencentes a 12 famílias e entre os répteis foram registradas 26 espécies, sendo uma espécie de anfisbena, uma de jacaré, nove de lagartos e 15 de serpentes. Para os anfíbios, houve uma dominância de espécies da família Hylidae, que representaram mais da metade do total de espécies encontradas no estudo. Já entre os répteis, houve uma predominância de espécies de serpentes da família Dipsadidae. Considerando apenas os registros feitos pelas metodologias empregadas, a área contínua de floresta da REGUA possui uma riqueza de espécies de anfíbios (N = 30) e de lagartos (N = 4) menor do que o conjunto de fragmentos (N = 36 e N = 8), mas superior ao que foi encontrado na matriz (N = 25 e N = 1). Entretanto, para os anfíbios, mais de um terço das espécies (N = 11) que ocorreu na mata contínua não ocorreu nos fragmentos ou na matriz, o que sugere que estas espécies podem ser mais sensíveis a alterações do hábitat. A maior riqueza de espécies encontrada no conjunto de fragmentos pode ser parcialmente explicada pelo fato de muitas espécies tanto de anfíbios quanto de lagartos que são típicas de áreas abertas, terem sua ocorrência favorecida neste tipo de condição de ambiente relativamente menos fechado dos fragmentos. Quando avaliamos o efeito de métricas da paisagem, observamos diferentes respostas entre os anfíbios e os lagartos. Enquanto para os anfíbios houve uma tendência de fragmentos mais distantes terem uma menor riqueza de espécies e de modos reprodutivos associados a estas espécies, para os lagartos a área dos fragmentos parece ser uma importante variável na estruturação das comunidades. Entretanto, por particularidades das características fisiológicas e ecológicas de anfíbios e lagartos, é possível que outros fatores expliquem a distribuição diferenciada das espécies. De forma geral, as áreas de matriz amostradas pareceram ser hostis a espécies florestais tanto de anfíbios quanto de lagartos. Além disso, para anfíbios, a presença de ambientes reprodutivos pode ser fator crucial para a ocorrência de algumas espécies. De forma similar ao encontrado em outros estudos, para a manutenção da diversidade de anfíbios e lagartos na paisagem fragmentada tratada neste estudo é necessário preservar o grande bloco florestal e aumentar a conectividade deste com os fragmentos, o que poderia permitir a área contínua servir de área-fonte de dispersores para os remanescentes florestais.
