The objective of the current study was to analyze the effects of rhinoseptoplasty on internal nasal dimensions and speech resonance of individuals with unilateral cleft lip and palate, estimated by acoustic rhinometry and nasometry, respectively. Twenty-one individuals (aged 15-46 years) with previously repaired unilateral cleft lip and palate were analyzed before (PRE), and 6 to 9 (POST1) and 12 to 18 months (POST2) after surgery. Acoustic rhinometry was used to measure the cross-sectional areas (CSAs) of segments corresponding to the nasal valve (CSA1), anterior portion (CSA2), and posterior portion (CSA3) of the lower turbinate, and the volumes at the nasal valve (V1) and turbinate (V2) regions at cleft and noncleft sides, before and after nasal decongestion with a topical vasoconstrictor. Nasometry was used to evaluate speech nasalance during the reading of a set of sentences containing nasal sounds and other devoid of nasal sounds. At the cleft side, before nasal decongestion, there was a significant increase (P < 0.05) in mean CSA1 and V1 values at POST1 and POST2 compared with PRE. After decongestion, increased values were also observed for CSA2 and V2 at POST2. No significant changes were observed at the noncleft side. Mean nasalance values at PRE, POST1, an POST2 were not different from each other in both oral and nasal sentences. The measurement of CSAs and volumes by acoustic rhinometry revealed that rhinoseptoplasty provided, in most cases analyzed, a significant increase in nasal patency, without concomitant changes in speech resonance, as estimated by nasalance assessment.

Aim. The aim of this study was to describe, explore and explain the concept of sustainability in nursing. Background. Although researchers in nursing and medicine have emphasised the issue of sustainability and health, the concept of sustainability in nursing is undefined and poorly researched. A need exists for theoretical and empirical studies of sustainability in nursing. Design. Concept analysis as developed by Walker and Avant. Method. Data were derived from dictionaries, international healthcare organisations and literature searches in the CINAHL and MEDLINE databases. Inclusive years for the search ranged from 1990 to 2012. A total of fourteen articles were found that referred to sustainability in nursing. Results. Sustainability in nursing involves six defining attributes: ecology, environment, future, globalism, holism and maintenance. Antecedents of sustainability require climate change, environmental impact and awareness, confidence in the future, responsibility and a willingness to change. Consequences of sustainability in nursing include education in the areas of ecology, environment and sustainable development as well as sustainability as a part of nursing academic programs and in the description of the academic subject of nursing. Sustainability should also be part of national and international healthcare organisations. The concept was clarified herein by giving it a definition. Conclusion. Sustainability in nursing was explored and found to contribute to sustainable development, with the ultimate goal of maintaining an environment that does not harm current and future generations' opportunities for good health. This concept analysis provides recommendations for the healthcare sector to incorporate sustainability and provides recommendations for future research.

BACKGROUND: People living at home who lack ability to manage their medicine are entitled to assistance to improve adherence provided by a home care assistant employed by social care. AIM: The aim was to describe how older people with chronic diseases, living at home, experience the use and assistance of administration of medicines in the context of social care. DESIGN: A qualitative descriptive study. METHODS: Ten participants (age 65+) living at home were interviewed in the participants' own homes. Latent content analysis was used. FINDINGS: The assistance eases daily life with regard to practical matters and increases adherence to a medicine regimen. There were mixed feelings about being dependent on assistance; it interferes with self-sufficiency at a time of health transition. Participants were balancing empowerment and a dubious perception of the home care assistants' knowledge of medicine and safety. Physicians' and district nurses' professional knowledge was a safety guarantee for the medicine process. CONCLUSIONS: Assistance eases daily life and medicine regimen adherence. Dependence on assistance may affect self-sufficiency. Perceived safety varied relating to home care assistants' knowledge of medicine. RELEVANCE TO CLINICAL PRACTICE: A well-functioning medicine assistance is crucial to enable older people to remain at home. A person-centred approach to health- and social care delivery is efficient and improve outcome for the recipient of care.

Dada a escassez de informação (principalmente de vazão) nas sub-bacias urbanas, os parâmetros das metodologias usadas no dimensionamento de uma rede de macrodrenagem são estimados a partir de características físicas, introduzindo incertezas sobre os valores corretos do parâmetro. Neste trabalho são apresentados resultados que indicam o impacto que as incertezas nos parâmetros têm sobre os valores da vazão e custo da rede, e custo estimado para redução das mesmas. Foram analisados os parâmetros das metodologias dos blocos alternados utilizada na determinação da chuva de projeto; da “Curva Número” do SCS utilizada para determinação da precipitação efetiva; e de Clark e Hidrograma Unitário Triangular (SCS) empregadas para a propagação de vazão. Também foi analisado o efeito da variabilidade espacial da chuva. Da análise de sensibilidade dos parâmetros determinou-se que para simulação distribuída da bacia, o CN resultou o parâmetro mais sensível. Por exemplo, para um incremento de uma unidade no CN foram encontradas variações de entre 8 e 10% na vazão, o que representa um incremento de 5% no custo. Também foram significativas as diferenças na chuva de projeto obtidas a partir de diferentes IDFs de postos pluviométricos próximos (diferenças de até 80% na vazão ou seja 40% no custo. Mudanças no valor do tempo de concentração somente tiveram efeito significativo na simulação concentrada da bacia; já variações do parâmetro de forma no HTA do método de Clark não tiveram efeitos sobre a vazão ou custo. Por outro lado, os custos de medição de chuva e vazão, necessários para geração da informação que permitiria reduzir a incerteza resultam menores ao impacto produzido por um erro 1% no CN, ressaltando a importância de efetuar coleta contínua de dados de vazão.

Although numerous studies have reported the production of skeletal muscle alpha -tropomyosin in E. coli, the protein needs to be modified at the amino terminus in order to be active. Without these modifications the protein does not bind to actin, does not exhibit head-to-tail polymerization, and does not inhibit the actomyosin Mg2+-ATPase in the absence of troponin. on the other hand, the protein produced in insect cells using baculovirus as an expression vector (Urbancikova, M., and Hitchcock-DeGregori, S. E., J. Biol. Chem., 269, 24310-24315, 1994) is only partially acetylated at its amino terminal and therefore is not totally functional. In an attempt to produce an unmodified functional recombinant muscle alpha -tropomyosin for structure-function correlation studies we have expressed the chicken skeletal alpha -tropomyosin cDNA in the yeast Pichia pastoris. Recombinant protein was produced at a high level (20 mg/L) and was similar to the wild type muscle protein in its ability to polymerize, to bind to actin and to regulate the actomyosin S1 Mg2+-ATPase. Yeasts are attractive hosts for heterologous protein production as they follow the general eukaryotic post-translational modification pattern. The well-known Saccharomyces cerevisiae has been used to produce a large variety of foreign proteins. The proper function of muscle tropomyosin depends on a specific modification at its N-terminus. Although tropomyosin has been produced in different expression systems, only the recombinant protein produced in the yeast Pichia pastoris has native-like functional properties. In this paper we describe the production of functional skeletal muscle tropomyosin in the yeast S. cerevisiae. The recombinant protein was produced in high amounts and production was strongly affected by genetic and environmental factors, including plasmid copy number, promoter strength, and growth media composition. O presente trabalho foi conduzido com o objetivo de avaliar a degradabilidade ruminal da matéria seca (MS), fibra em detergente neutro (FDN) e amido, além de pH, amônia e ácidos graxos voláteis ruminais, em bovinos alimentados com silagens de milho (SMi), de raspa de mandioca com polpa cítrica (SRp), de casca de mandioca com polpa cítrica (SCc) e de cana-de-açúcar com polpa cítrica (SCn). Foram utilizados quatro novilhos, mestiços, castrados, canulados no rúmen e duodeno, em quatro períodos experimentais, com 11 dias de adaptação à dieta e oito dias de coleta. O delineamento experimental foi o quadrado latino 4x4. Foram adotados oito horários para a incubação das silagens: 3, 6, 12, 24, 48, 72, 96 e 120 horas. A SRp apresentou maior degradação efetiva (Kp 5%) da MS e da FDN (48,44 e 45,78%, respectivamente), quando comparada com a SMi (45,50 e 23,75%), a SCc (43,87 e 24,20%) e a SCn (40,76 e 25,78%). Para todos os tratamentos, o pH e a concentração de N-NH3 ruminal foram adequados para o crescimento dos microrganismos ruminais. Os valores de AGV para os tratamentos de SMi, SRp e SCc foram semelhantes entre si e superiores aos do tratamento com SCn.

To provide data for conservation, selection, and expansion programs of buffalo herds, this study evaluated the history of a population of Murrah buffaloes based on population structure and the effect of inbreeding on accumulated 305-d milk yield (MY), fat yield (FY), protein yield (PY), mozzarella production (MProd), and somatic cell score (SCS). The usefulness of including the individual inbreeding coefficient (F) or individual increase in inbreeding coefficient (Delta F) in the model to describe inbreeding depression was evaluated. Pedigree information from 8,054 animals born between 1976 and 2008 and 4,497 lactation records obtained from 12 herds were used. The realized effective population size was 40.10 +/- 1.27, and the mean F of the entire population was 2.14%. The ratio between the number of founders and ancestors demonstrated the existence of a bottleneck in the pedigree of this population, which may contribute to a reduction of genetic diversity. The effect of F on MY, FY, PY, MProd, and SCS was -1.005 kg, -0.299 kg, -0.246 kg, -1.201 kg, and -0.002 units, and the effect of Delta F transformed to equivalent F (%) for a mean of 2.57 equivalent generations was -4.287 kg, -0.581 kg, -0.383 kg, -2.001 kg, and -0.007 units, respectively. The inbreeding depression observed may have important economic repercussions for production systems. The Delta F can be considered the better of the two indicators of inbreeding depression due to its properties that prevent underestimation of this effect. A designed mating system to avoid inbreeding may be applied to this population to maintain genetic diversity.

Visando estudar as associações genéticas e fenotípicas entre a contagem de células somáticas (CCS) e a produção de leite até 305 dias (P305) e avaliar as perspectivas de utilização da CCS como critério de seleção em bovinos leiteiros, utilizaram-se dados obtidos pelo Programa de Análise da Eficiência de Rebanhos Leiteiros e da Qualidade do Leite, desenvolvido pelo Laboratório de Qualidade do Leite da ESALQ - USP. Por não apresentar distribuição normal, a contagem de células somáticas (CCS) foi transformada para uma escala logarítmica (ECS). Foram estimados os parâmetros genéticos para a P305 dias e a média dos escores de células somáticas (MECS). A média da produção de leite até 305 dias foi de 7.519,51 kg e a da MECS, de 4,04. As herdabilidades estimadas para P305 e MECS foram de 0,22 e 0,13, respectivamente, nas análises univariadas. Valores semelhantes foram estimados pela análise bivariada. A correlação genética entre a P305 e MECS foi de -0,16. Por apresentar correlação genética negativa, mas de baixa magnitude, a seleção para produção de leite não ocasionaria, a princípio, aumento indesejável da MECS.

Avaliou-se a ocorrência da murcha-de-curtobacterium em lavouras de feijoeiro comum em algumas localidades do Estado de Santa Catarina, nas safras 2002/03 e 2003/04, e o comportamento dos genótipos BRS Valente, Carioca, CHC 97-29, CHP 97-26, CNPF 8104, Diamante Negro, Empasc 201 - Chapecó, IAPAR 44, IPR Graúna, IPR Juriti, IPR Uirapuru, LP 9728, Pérola, SCS 202-Guará, Sel. CP 9310635, TPS Bionobre, TPS Bonito, TPS Magnífico, TPS Nobre, TPS Soberano e Xamego perante Curtobacterium flaccumfaciens pv. flaccumfaciens (Cff), em condições de casa-de-vegetação. As cultivares IAC Carioca Akytã, IAC Carioca Aruã e IAC Carioca Pyatã foram empregadas como padrões de resistência a Cff. As avaliações dos sintomas ocorreram aos 5, 10, 15, 20 e 25 dias após a inoculação (DAI) e, posteriormente, foi estimada a área abaixo da curva de progresso da murcha-de-curtobacterium (AACPMC), em cada genótipo. A doença esteve presente nos municípios de Campos Novos, Faxinal dos Guedes, Guatambu, Ipuaçu, Ponte Serrada e Tigrinhos e que, aos 10 DAI, as cultivares SCS 202 - Guará e IPR Juriti mostraram baixa severidade. Porém, aos 25 DAI, somente as cultivares padrões foram resistentes e apresentaram menor AACPMC.

Foi avaliada a influência de doses de nitrogênio (N) na severidade da murcha-de curtobacterium do feijoeiro, causada por Curtobacterium flaccumfaciens pv. flaccumfaciens, nas cultivares IAC Pyatã, IPR 88 Uirapuru e SCS 202 Guará, em condições de casa-de-vegetação. Os tratamentos foram a dose recomendada de N e outras 25 e 50 % abaixo e acima desta, empregando-se uréia. O aumento de doses de N influenciou positivamente o progresso da murcha-de-curtobacterium somente na cultivar IAC Carioca Pyatã, incrementou a quantidade de N na parte aérea e não interferiu no peso da matéria seca das plantas de todas as cultivares avaliadas.

O objetivo deste trabalho foi avaliar o efeito de doses de potássio (K) na severidade da murcha-de-curtobacterium em três cultivares de feijoeiro (IAC Carioca Pyatã, IPR 88 - Uirapuru e SCS 202 - Guará), em condições de casa-de-vegetação. Os tratamentos foram 135,0; 112,5; 90,0; 67,5 e 45,0 kg.ha-1 de K2O, na forma de cloreto de potássio. As avaliações ocorreram aos 5, 10, 15, 20 e 25 dias após a inoculação e foi estimada a área abaixo da curva de progresso da murcha-de-curtobacterium (AACPMC). Não foi verificada influência das doses de K2O na AACPMC e na quantidade de K na parte aérea de plantas das cultivares IAC Carioca Pyatã e IPR 88 - Uirapuru. Conforme o aumento das doses de K2O, somente houve incremento na massa da matéria seca das plantas não inoculadas da cultivar SCS 202-Guará.

Astyanax scabripinnis possesses a widespread polymorphism for metacentric B chromosomes as large as the largest chromosome pair in the A complement. on the basis of C-banding pattern, it was hypothesized that these B chromosomes are isochromosomes that have arisen by means of centromere misdivision and chromatid nondisjunction. In the present paper we test this hypothesis by analysing (i) the localization of a repetitive DNA sequence on both B chromosome arms, and (ii) synaptonemal complex formation, in order to test the functional homology of both arms. Genomic DNA digested with KpnI and analysed by gel electrophoresis showed fragments in a ladder-like pattern typical of tandemly repetitive DNA. These fragments were cloned and their tandem organization in the genome was confirmed. A 51-bp long consensus sequence, which was AT-rich (59%) and contained a variable region and two imperfect reverse sequences, was obtained. Fluorescence in situ hybridization (FISH) localized this repetitive DNA into noncentromeric constitutive heterochromatin which encompasses the terminal region of some acrocentric chromosomes, the NOR region, and interstitial polymorphic heterochromatin in chromosome 24. Most remarkably, tandem repeats were almost symmetrically placed in the two arms of the B chromosome, with the exception of two additional small clusters proximally located on the slightly longer arm. Synaptonemal complex (SC) analysis showed 26 completely paired SCs in males with 1B. The ring configuration of the B univalent persisting until metaphase I suggests that the two arms formed chiasmata. All these data provided strong support for the hypothesis that the B chromosome is an isochromosome.

We analyzed the quality of raw milk from eight dairy farms in Rio Grande do Norte stored in a cooling tank , in order to evaluate methods for determining somatic cell counts (SCC). The Somaticell® kit and a portable Direct Cell Counter (DCC) were compared with each other and with the MilkoScanTM FT+ (FOSS Denmark), which uses Fourier Transform Infrared (FTIR) spectroscopy). Direct cell counter data were processed for somatic cell scores (log-transformed somatic cell count) and analyzed with the SAS®, statistical package , Statistical Analysis System, (SAS, INSTITUTE, 1998). Comparison of means and correlation of somatic cell scores were conducted using Pearson s correlation coefficient and the Tukey Test at 1 %. No significant difference was observed for comparison of means. The correlation between somatic cell scores was significant, that is, 0.907 and 0.876 between the MilkoScanTM FT+ and the Somaticell® kit and Direct Cell Count (DCC) respectively, and 0.943 between the Somaticell® kit and Direct Cell Count (DCC). The methods can be recommended for monitoring the quality of raw milk kept in a cooling tank in the production unit
