The comet assay is a method of DNA damage analysis widely used to quantify oxidative damage, crosslinks of DNA, apoptosis and genotoxicity of chemicals substances as chemical, pharmaceuticals, agrochemicals products, among others. This technique is suitable to detect DNA strand breaks, alkali-labile sites and incomplete excision repair sites and is based on the migration of DNA fragments by microeletroforesis, DNA migrates for the anode forming a “tail”, and the formed image has the appearance of a comet. The slides can be stained with fluorescence or silver, having differences in the microscopy type used for the analysis and the possibility of storage of the slides, moreover, the first one is a stained-method with more difficulties of accomplishment. The image analysis can be performed by a visual way, however, there is a disadvantage as the subjectivity on the results, that can be minimized by an automated method of digital analysis. This process was studied in this report with the aim to perceive the validation of the digital analysis turning it a quantitative method with larger reproductibility, minimizing the variability and imprecision due to the subjective analysis. For this validation we selected 50 comets photographed in a standardized way and printed, afterwards, pictures were submitted to three experienced appraisers, who quantified them manually. Later, the images were processed by free software ImageJ 1.38x, printed and quantified manually by the same appraisers. The intraclass correlation was higher to comet measures after image processing. Following, an algorithm of automated digital analysis from the measures of the comet was developed; the values obtained were compared with those 12 estimated manually after the processing resulting high correlation among the measures. Chrysomya megacephala (Fabricius), has considerable medical, veterinary and forensic importance. In insects in general, the ability to track the food substrate is due to highly sensitive organs adapted for detecting odors, particularly in the case of blowflies, which is one of carcass main consumers. Eggs of blowflies are usually aggregated in function of the behavior of other pheromone mediated by females laying eggs in place on the same substrate. One of the main reasons for studying oviposition behavior is because it may indirectly affect individual qualification, population dynamics and community structure. It has been suggested that female blowflies can evaluate the number of eggs on a substrate, decreasing the size of oviposition in very saturated substrates with larvae or eggs leading to a lower intraspecific competition. The present study investigated the oviposition behavior of females of C. megacephala, and wished to consider whether they are able to distinguish quantitatively (size of the mass) eggs previously put into a place of posture and hence indicate whether there is a limit beneficial (to avoid competition among larvae) for its oviposition on the same substrate or if that place would already be saturated, requiring find another substrate without eggs or less for futures ovipositions. With 20 females and five males, five treatments were applied, repeated ten times and analyzed using the Mann-Whitney test. Each treatment consists in placing, inside the cage, a flask with 50g of ground beef and another one in the other side, with the same amount of substrate with a certain amount of prior oviposition for a period of 5 hours. The masses prior eggs were used 0.03 g, 0.05 g, 0.1 g, 0.2 g and 0.3 g (respectively, T1, T2, T3, T4 and T5). This study investigated the effect of slope and antecedent soil moisture on the water depth stored and percolated on extensive green roofs built in pilot scale. For this purpose, slopes of 10, 20 and 30% were investigated. Moisture was measured before and after each test to determine the differential moisture (ΔU). A rainfall simulator was designed and tested; the best arrangement presents a Christiansen Uniformity Coefficient of 56%. The results yielded that the slopes of 10% presented the highest values of water depth stored (59,88%), water depth percolated (77,15%), the slope of 30% presented the highest water depth drained (48,22%.) and there was no correlation between slope, water depth stored, water depth percolated and ΔU. It was verified the effect of dilution on the modules i10% between water depth percolated and apparent color (0.71%) in i20% between water depth drained and apparent color (-0.748%) and in the module i30% between water depth drained and turbidity (-0.76%) and water depth drained and apparent color (-0.93%). In i10% there was high correlation between pH and water depth percolated (0.94%) and between water depth drained and pH (0.71%). The present study evaluated the use of semicircular bending test (SCB) as an alternative to conventional bending test to examine the effect of thermal shock. Still, studies the behavior of fracture surfaces generated by monofractal behavior analysis, which allows us to evaluate the contributions of the microstructure and the mechanical context in forming reliefs during the crack propagation. The fractal analysis is made from elevation maps obtained by reconstruction method by extension of the focus stacks of digital images acquired in microscope. The specimens used were samples semicircular pressed TiO2 (rutile) with and without heat shock, prepared for testing mode I loading. Were also produced, specimens in the form of bars for Weibull statistical analysis. From the three-point bending test, we found the variation of fracture toughness between the samples after the heat shock and natural condition. The SCB test was feasible for the analysis of thermal shock resistance. The results showed that the value of the fracture toughness decreases as the heat shock treatment

Study the semi-quantitative and quantitative technique in the diagnosis of catheter-related infections in newborns and to determine oxacillin resistance in Staphylococcus isolated. It was analyzed 353 catheter tips from 273 newborns in the Neonatal Unit of Hospital FMB. To confirm the diagnosis of infection, were analyzed the clinical data of newborns, the presence of at least one positive blood culture and growth of ≥ 1000 CFU/mL on quantitative culture and/or ³15 UFC on semiquantitative culture, with the same microorganism isolation (species and drug sensitivity) in blood culture and no other focus of infection except the catheter. The disk diffusion technique was used to check similarity of strains and resistance to oxacillin. Of the 353 tips analyzed, 39 were included in this study as the inclusion criteria. The semiquantitative culture was positive in 26 (66.7%) catheters and quantitative culture was positive in 24 (61.5%). Of 273 patients, 19 (6.9%) had a diagnosis of catheter-related bloodstream Infection (CR-BSI). Of the 19 episodes of CR-BSI, S. epidermidis was the predominant etiological agent (84.2%). The resistance to the antibiotic methicillin was found in 14 (73.7%) strains of Staphylococcus. The semiquantitative method was more sensitive (79%) compared with the quantitative method (63%). The use of antibiotics may have influenced the sensitivity of the quantitative method as the microorganisms present in the lumen are exposed to higher concentrations of antibiotics administered via the catheter

As abelhas da espécie Apis mellifera são conhecidas por sua organização social, comportamento devotado e aparente complexidade comportamental, sendo também consideradas excelentes sistemas-modelo para estudos de comunicação, aprendizado e formação de memória. Uma vez que o comportamento social dos insetos é resultante de interações complexas em diferentes níveis de organização biológica, torna-se importante estudar os peptídeos envolvidos nesse processo, já que estes são indispensáveis em muitos processos fisiológicos, funcionando como neurotransmissores, hormônios, toxinas, antibióticos, defensinas, neuromoduladores e neurohormônios. Desta forma, o objetivo do presente trabalho foi padronizar uma metodologia analítica visando à determinação do perfil peptídico do cérebro de abelhas da espécie Apis mellifera utilizando como ferramentas a espectrometria de massas e a cromatografia líquida. Foram coletadas operárias de Apis mellifera com 20 dias de idade, cujos cérebros foram dissecados e devidamente processados visando a posterior separação e identificação dos peptídeos. Foram testadas duas estratégias distintas para a separação e identificação dos peptídeos, denominadas on-line e off-line. Ambas baseavam-se na utilização de um sistema de cromatografia líquida sob fase reversa e um espectrômetro de massas do tipo electrospray/Ion Trap-Time of Flight, mas na primeira essas ferramentas estavam acopladas, enquanto na segunda os dois processos foram realizados separadamente. Tais procedimentos permitiram identificar 19 peptídeos pertencentes às principais famílias de neuropeptídeos de cérebros de abelhas já descritos na literatura, entre elas as Alatostatina, Apidaecina, Corazonina e Taquicininas. The main purpose of a business is to serve customers, but to be successful must take into account other factors such as survival, profitability, growth and operational standpoint to use their resources effectively. The use of tools to support the process of analysis of management decision-making is gaining importance in the context of competitiveness in the world market. The mathematical models are critical to determine theoretical prices of options and analyze whether they are overrated or underrated. This information strongly influence in operations carried out by the investor. Therefore, it is necessary that the employee model present high degree of reliability and be consistent with the reality of investment to which it is intended. In this sense, this dissertation aims to apply the steps of mathematical modeling in the Pricing of options for decision making in the investment of a hydroelectric power plant. Was used a Monte Carlo simulation, with the Latin Hypercube Method, to determine the volatility of returns of the project. In order to validate the proposed model, compared to the results found by the Binomial Model, which is one of the models most used in this type of investment. The results reinforce the hypothesis that the mathematical modeling with the Binomial Model is critical to investment decision-making in hydroelectric power

The paper presents the radiometric parameters determined by the medical physicist during routine radiotherapy planning service in cases of breast cancer . The contours of the breast volume in patients undergoing radiation breast tumors at the Hospital das Clinicas, Faculty of Medicine , UNESP, Botucatu ( HCFMB ) during the year 2012 were analyzed . In order to analyze the influence of physical and radiometric parameters for the determination of the dose distribution of irradiated breast volume , four measurements of isodose curves were prepared in four different heights breast , and compared with the isodose curves plotted computationally . In the routine of planning , the medical physicist must determine the isodose curve that gives the best dose distribution homogeneity in the irradiated volume . The choice of the treatment plan can be done by dedicated computer systems , which require significantly costly investments available services having better financial support . In the Service of Medical Physics , Department of Radiotherapy , HC FMB , we use a two-dimensional software for determination of isodose curves , however , this software is out of date and frequently becomes inoperable due to the lack of maintenance and it is a closed system without feasibility of interference from computer professionals . This fact requires manual preparation of isodose curves , which are subject to uncertainties due to the subjectivity in the clinical interpretation of medical radiation oncologist and medical physicist responsible for planning , plus dispendiar significant calculation time . The choice of the optimal isodose curve depends on the energy of the radiation beam , the geometry and dimensions of the irradiated area . Given the competitive reality of the globalized market, companies are compelled to develop strategic aspects of the management process, in order to ensure their survival. Among the strategic aspects is the price formation process, whose reasoning occurs mainly of adoption through cost system. Knowledge of costs and expenses provides conditions for the company to position itself strategically in the market, being able to follow the price of competition and sometimes even surpass them, called, more and more attention with regard to price formation. This study aims to identify the best price to be transferred to the market. To this end, were approached concept of pricing and its components, their strategies and pricing objectives, as well as the influence of demand on price formation, as well as the added value the brand and quality; and even a careful analysis of the price established by the said company in the service offered. We used the method of literature review and case study in the company ESA international aeronautical industry. O tratamento clareador se destaca por ser uma técnica conservadora, que pode proporcionar elevação da autoestima e melhorar o convívio social das pessoas. No entanto, pacientes expostos a essa terapia experimentam diferentes níveis de ocorrência e de intensidade de sensibilidade dentária. Essa sintomatologia é preocupante, pois tem sido apontada como o reflexo de uma eventual resposta pulpar frente ao tratamento realizado. Por outro lado, a presença de dor é desestimulante aos pacientes, prejudicando a evolução do tratamento e reduzindo o sentimento de satisfação. Apesar dessa importância, a maior parte dos estudos de sensibilidade dentária ainda emprega métodos subjetivos, dificultando a reprodutibilidade e interpretação dos resultados. Objetivo: nesse sentido, o presente artigo objetiva analisar a resposta neurossensorial proporcionada pela aplicação de estímulos térmicos gerados pelo equipamento TSA II (Medoc TSA II Neurosensory Analyzer, Israel), em um paciente submetido ao tratamento clareador, seguido pelo uso ou não uso de um dessensibilizante. Métodos: as análises foram realizadas antes do clareamento (AC), depois do clareamento (DC) e depois da aplicação ou não do dessensibilizante (DD) em cada hemiarcada estudada. Os dados foram submetidos ao teste t de Student, com nível de significância de 5%. Resultados: observou-se que o clareamento dentário alterou o limiar de sensação ao frio, provocando aumento da sensibilidade dentária, e que o uso do dessensibilizante foi efetivo. Conclusão: o uso de testes sensoriais quantitativos mostrou-se promissor para o estudo da sensibilidade dentária, podendo colaborar no estabelecimento de terapias seguras e confortáveis.

A cinomose é uma doença de desafio diagnóstico, especialmente quando não há histórico de vacinação. O objetivo deste estudo foi detectar e quantificar partículas virais de cinomose em diferentes fluidos e tecidos biológicos de um cão, determinando o melhor tecido para diagnóstico viral ante mortem na fase de viremia. Atendeu-se um cão adulto com manifestações clínicas inespecíficas e corpúsculos de Sinegaglia Lentz em linfócitos. Amostras post mortem foram submetidas a PCR em tempo real (qPCR), que demonstrou RNA viral em concentrações de (x105) em líquor (1.216), bexiga (1.009), cérebro (605), sangue (572), cerebelo (523), rins (373), fígado (257), pulmões (191), estômago (154), terceira pálpebra (70) e urina (2,1). A técnica de qPCR permitiu confirmar a infecção pelo vírus, descartando vacinação recente. A amostra de líquor mostrou-se representativa para diagnóstico molecular de fase aguda de cinomose no animal estudado.
