HIGHLIGHTS FOR FY 2004 1. Completed the second of a 3-year Gulf sturgeon population estimate on the Escambia River, Florida. 2. Completed the first of a 2-year Gulf sturgeon population estimate on the Apalachicola River, Florida. 3. Conducted Gulf sturgeon presence-absence surveys in three other Florida river systems. 4. Documented Gulf sturgeon marine habitat use in the near shore waters of the Gulf of Mexico. 5. Identified environmental threats to Gulf sturgeon spawning habitat in the Choctawhatchee River, Florida. 6. Initiated a study to document Gulf sturgeon spawning with the collection of fertilized eggs in the Yellow River, Florida. 7. Implemented Gulf Striped Bass Restoration Plan by coordinating the 21st Annual Morone Workshop, leading the technical committee, transporting broodfish, and coordinating the stocking on the Apalachicola-Chattahoochee-Flint (ACF) river system. 8. Over 86,000 Phase II Gulf striped bass were marked with sequential coded wire tags and stocked in Lake Seminole and the Apalachicola River. Post-stocking evaluations were conducted at 31 sites. 9. Drafted updates to Apalachicola-Chattahoochee-Flint Striped Bass Restoration and Evaluation Five-Year Plan with partners. 10. Fishery surveys were conducted on Tyndall Air Force Base and St. Marks and St. Vincent National Wildlife Refuges. 11. Habitat evaluations and population surveys were completed at 153 Okaloosa darter stream sites. 12. Aquatic insect biomonitoring and identification of over 39,000 individual aquatic macroinvertebrates was completed and provided to Eglin Air Force Base. 13. Ten years of fishery data from Okefenokee and Banks Lake National Wildlife Refuges was analyzed with recommendations incorporated into the refuge Comprehensive Conservation Plan. 14. A draft mussel sampling protocol was tested in wadeable streams in northwest Florida and southwest Georgia. 15. Implemented recovery plan and candidate conservation actions for 14 listed and candidate freshwater mussels in the Northeast Gulf Watersheds. 16. Worked with partners in developing the Spring Creek Watershed Partnership in the Flint River basin, Georgia. 17. Multiple stream restoration and watershed management projects were initiated or completed. A total of 6.8 stream miles were restored for stream fishes, along with 56.4 miles of coastline were enhanced for sea turtle lighting. A total of 135 acres of wetlands and 58 acres of understory habitat were restored. 18. Multiple outreach projects were completed to detail aquatic resources conservation needs and opportunities. Participated in National Fishing Week event, BASS ProShops event, several festivals, and school outreach.

A new transition prediction model is introduced, which couples the intermittency effect into the turbulence transport equations and takes the characteristics of fluid transition into consideration to mimic the exact process of transition. Test cases include a two-dimensional incompressible plate and a two-dimensional NACA0012 airfoil. Performance of this transition model for incompressible flows is studied, with numerical results consistent to experimental data. The requirement of grid resolution for this transition model is also studied.

Using conventional methods, a laser pulse can be focused down to around 6-8 mu m, but further reduction of the spot size has proven to be difficult. Here it is shown by particle-in-cell simulation that with a hollow cone an intense laser pulse can be reduced to a tiny, highly localized, spot of around 1 mu m radius, accompanied by much enhanced light intensity. The pulse shaping and focusing effect is due to a nonlinear laser-plasma interaction on the inner surface of the cone. When a thin foil is attached to the tip of the cone, the cone-focused light pulse compresses and accelerates the ions in its path and can punch through the thin target, creating highly localized energetic ion bunches of high density.

A novel technique of controlling the evolution of the filamentation was experimentally demonstrated in an argon gas-filled tube. The entrance of the filament was heated by a furnace and the other end was cooled with air, which resulted in the temperature gradient distribution along the tube. The experimental results show that multiple filaments are merged into a single filament and then no filament by only increasing the temperature at the entrance of the filament. Also, the filament can appear and disappear after increasing the local temperature and input pulse energy in turn. This technique offers another degree of freedom to control the filamentation and opens a new way for multi-mJ level monocycle pulse generation through filamentation in the noble gas.

We demonstrate passive Q-switching of short-length double-clad Tm3+-doped silica fiber lasers near 2 mu m pumped by a laser diode array (LDA) at 790 nm. Polycrystalline Cr2+:ZnSe microchips with thickness from 0.3 to 1 mm are adopted as the Q-switching elements. Pulse duration of 120 ns, pulse energy over 14 mu] and repetition rate of 53 kHz are obtained from a 5-cm long fiber laser. As high as 530 kHz repetition rate is achieved from a 50-cm long fiber laser at similar to 10-W pump power. The performance of the Q-switched fiber lasers as a function of fiber length is also analyzed. (c) 2008 Elsevier B.V. All rights reserved.

A series of binary borosilicate glasses prepared by the sol-gel method are shown to be bioactive. Tetraethyl orthosilicate (TEOS) and trimethylborate (TMB) in acidic medium are used to prepare xB(2)O(3)center dot(1-x)SiO2 glass systems for x = 0.045-0.167. The formation of a layer of apatite-like mineral on the glass surface becomes apparent after soaking in simulated body fluid for 48 h. We have measured the B-11-B-11 homonuclear second moments of the borosilicate glasses and inferred that no macroscopic phase separation occurred in our glasses. The B-11 chemical shift data also show that the formation of clustered boroxol rings is negligible in our glass system. Although the bioactivity of our borosilicate glasses is less than that of CaO-SiO2 sol-gel glasses, these simple binary systems could be taken as reference glass systems for the search of new bioactive borosilicate glasses. Southeast Bering Sea Carrying Capacity (SEBSCC, 1996–2002) was a NOAA Coastal Ocean Program project that investigated the marine ecosystem of the southeastern Bering Sea. SEBSCC was co-managed by the University of Alaska Fairbanks, NOAA Alaska Fisheries Science Center, and NOAA Pacific Marine Environmental Laboratory. Project goals were to understand the changing physical environment and its relationship to the biota of the region, to relate that understanding to natural variations in year-class strength of walleye pollock (Theragra chalcogramma), and to improve the flow of ecosystem information to fishery managers. In addition to SEBSCC, the Inner Front study (1997–2000), supported by the National Science Foundation (Prolonged Production and Trophic Transfer to Predators: Processes at the Inner Front of the S.E. Bering Sea), was active in the southeastern Bering Sea from 1997 to 1999. The SEBSCC and Inner Front studies were complementary. SEBSCC focused on the middle and outer shelf. Inner Front worked the middle and inner shelf. Collaboration between investigators in the two programs was strong, and the joint results yielded a substantially increased understanding of the regional ecosystem. SEBSCC focused on four central scientific issues: (1) How does climate variability influence the marine ecosystem of the Bering Sea? (2) What determines the timing, amount, and fate of primary and secondary production? (3) How do oceanographic conditions on the shelf influence distributions of fish and other species? (4) What limits the growth of fish populations on the eastern Bering Sea shelf? Underlying these broad questions was a narrower focus on walleye pollock, particularly a desire to understand ecological factors that affect year-class strength and the ability to predict the potential of a year class at the earliest possible time. The Inner Front program focused on the role of the structural front between the well-mixed waters of the coastal domain and the two-layer system of the middle domain. Of special interest was the potential for prolonged post-spring-bloom production at the front and its role in supporting upper trophic level organisms such as juvenile pollock and seabirds. Of concern to both programs was the role of interannual and longer-term variability in marine climates and their effects on the function of sub-arctic marine ecosystems and their ability to support upper trophic level organisms.

This report, "Harmful Algal Bloom Management and Response: Assessment and Plan" reviews and evaluates Harmful Algal Bloom (HAB) management and response efforts, identifies current prevention, control, and mitigation programs for HABs, and presents an innovative research, event response, and infrastructure development plan for advancing the response to HABs. In December 2004, Congress enacted and the President signed into law the Harmful Algal Bloom and Hypoxia Amendments Act of 2004, (HABHRCA 2004). The reauthorization of HABHRCA acknowledged that HABs are one of the most scientifically complex and economically damaging coastal issues challenging our ability to safeguard the health of our Nation’s coastal ecosystems. The Administration further recognized the importance of HABs as a high priority national issue by specifically calling for the implementation of HABHRCA in the President’s U.S. Ocean Action Plan. HABHRCA 2004 requires four reports to assess and recommend research programs on HABs in U.S. waters. This document comprises two linked reports specifically aimed at improving HAB management and response: the Prediction and Response Report and the follow-up plan, the National Scientific Research, Development, Demonstration, and Technology Transfer (RDDTT) Plan on Reducing Impacts from Harmful Algal Blooms. This document was prepared by the Interagency Working Group on Harmful Algal Blooms, Hypoxia, and Human Health, which was chartered through the Joint Subcommittee on Ocean Science and Technology of the National Science and Technology Council and the Interagency Committee on Ocean Science and Resource Management Integration. This report complements and expands upon HAB-related priorities identified in Charting the Course for Ocean Science in the United States for the Next Decade: An Ocean Research Priorities Plan and Implementation Strategy, recently released by the Joint Subcommittee on Ocean Science and Technology. It draws from the contributions of numerous experts and stakeholders from federal, state, and local governments, academia, industry, and non-governmental organizations through direct contributions, previous reports and planning efforts, a public comment period, and a workshop convened to develop strategies for a HAB management and response plan. Given the importance of the Nation’s coastal ocean, estuaries, and inland waters to our quality of life, our culture, and the economy, it is imperative that we move forward to better understand and mitigate the impacts of HABs which threaten all of our coasts and inland waters. This report is an effort to assess the extent of federal, state and local efforts to predict and respond to HAB events and to identify opportunities for charting a way forward.

The 1992 PACLIM meeting featured the Long-Term Ecological Research (LTER) program, sponsored by the National Science Foundation. Ranging from hot to cold and wet to dry climatic regimes, these 18 sites are attempting to understand the web of relationships in different locations as communities evolve over time scales of years to decades to centuries. During this time they are subject to external forcings, including those that vary smoothly and somewhat predictably, like the seasons, upon which are superimposed random "shocks" of various magnitudes.

H.J. Andrews Experimental Forest is a 6400 ha forest of Douglas fir, western hemlock, and Pacific silver fir located in, and typical of, the central portion of the western slope of the Cascade mountain range of Oregon. The forest is one of 19 sites in the Long-Term Ecological Research (LTER) program sponsored by the National Science Foundation. ... Because of the scientific significance of Andrews Forest, it is important to investigate the temporal variability of annual and seasonal temperature and precipitation values at the site and identify past times of anomalous climatic conditions. It is also important to establish quantitatively the relationships between the climate of Andrews Forest and that of its surrounding area and, hence, place the climate of Andrews Forest into its regional context.

Background: The emergence of agriculture about 10,000 years ago marks a dramatic change in human evolutionary history. The diet shift in agriculture societies might have a great impact on the genetic makeup of Neolithic human populations. The regionally restricted enrichment of the class I alcohol dehydrogenase sequence polymorphism (ADH1BArg47His) in southern China and the adjacent areas suggests Darwinian positive selection on this genetic locus during Neolithic time though the driving force is yet to be disclosed. Results: We studied a total of 38 populations (2,275 individuals) including Han Chinese, Tibetan and other ethnic populations across China. The geographic distribution of the ADH1B*47His allele in these populations indicates a clear east-to-west cline, and it is dominant in south-eastern populations but rare in Tibetan populations. The molecular dating suggests that the emergence of the ADH1B*47His allele occurred about 10,000 similar to 7,000 years ago. Conclusion: We present genetic evidence of selection on the ADH1BArg47His polymorphism caused by the emergence and expansion of rice domestication in East Asia. The geographic distribution of the ADH1B*47His allele in East Asia is consistent with the unearthed culture relic sites of rice domestication in China. The estimated origin time of ADH1B*47His allele in those populations coincides with the time of origin and expansion of Neolithic agriculture in southern China.

Much attention has been paid on amphibian peptides for their wide-ranging pharmacological properties, clinical potential, and gene-encoded origin. More than 300 antimicrobial peptides (AMPs) from amphibians have been studied. Peptidomics and genomics analysis combined with functional test including microorganism killing, histamine-releasing, and mast cell degranulation was used to investigate antimicrobial peptide diversity. Thirty-four novel AMPs from skin secretions of Rana nigrovittata were identified in current work, and they belong to 9 families, including 6 novel families. Other three families are classified into rugosin, gaegurin, and temporin family of amphibian AMP, respectively. These AMPs share highly conserved preproregions including signal peptides and spacer acidic peptides, while greatly diversified on mature peptides structures. In this work, peptidomics combined with genomics analysis was confirmed to be an effective way to identify amphibian AMPs, especially novel families. Some AMPs reported here will provide leading molecules for designing novel antimicrobial agents. (C) 2009 Elsevier Inc. All rights reserved

采用水平淀粉凝胶电泳技术对花鲈群体的遗传结构进行了研究,共检测了中国沿海花鲈146 尾和40 尾日本 东京湾花鲈。其群体的多态位点比例为012667 —015333 ,观测杂合度和预期杂合度分别为010211 —010515 和 010398 —010797。中日花鲈在LDH 3 , GPI21 3 , GPI22 3 基因位点上的等位基因接近完全置换。中国花鲈各群体之 间的根井遗传距离为010004 —010011 ,平均值约为010080 ;而中日花鲈间的根井遗传距离为011870 —011954 ,平均值 为011926。以上结果表明中国花鲈群体间遗传变异很小,中日花鲈间遗传变异远大于中国花鲈群体间的遗传距 离。
