The availability and uptake of Cd by lettuce (Lactuca sativa L.) in two common tropical soils (before and after liming) were studied in order to derive human health-based risk soil concentration. Cadmium concentrations ranging from 1 to 12 mg kg(-1) were added to samples from a clayey Oxisol and a sandy-loam Ultisol under glasshouse conditions. After incubation, a soil sample was taken from each pot, the concentration of Cd in the soil was determined, lettuce was grown during 36 d, and the edible parts were harvested and analyzed for Cd. A positive linear correlation was observed between total soil Cd and the Cd concentration in lettuce. The amount of Cd absorbed by lettuce grown in the Ultisol was about twice the amount absorbed in the Oxisol. Liming increased the soil pH and slightly reduced Cd availability and uptake. CaCl2 extraction was better than DTPA to reflect differences in binding strength of Cd between limed and unlimed soils. Risk Cd concentrations in the Ultisol were lower than in the Oxisol, reflecting the greater degree of uptake from the Ultisol. The derived risk Cd values were dependent on soil type and the exposure scenario.

Se evaluó la eficacia de Tagetes spp. como cultivo intercalar y extracto natural sobre la fluctuación poblacional de áfidos en el cultivo de Lactuca sativa (L.). Se realizó un diseño en bloques completamente aleatorizado, siendo la planta la unidad de muestreo. Se consideraron tres tratamientos: testigo (T), cultivo intercalado con Tagetes sp. (CI) y cultivo pulverizado con extracto natural de Tagetes sp. (CP) (n=6). Los datos obtenidos se sometieron a un ANOVA y test de Tukey, según los lineamientos de ensayos repetidos en el tiempo. La densidad poblacional de áfidos fue mayor en el testigo y menor en el cultivo pulverizado, comprobándose una densidad media en el cultivo intercalar.

En el norte de la provincia de Entre Ríos se ha desarrollado un modelo de producción de arroz basado en el uso de agua superficial mediante la construcción de presas de tierra: existen más de 60 con superficies variables. El uso de agroquímicos en esa zona podría introducir un factor de contaminación en el suelo y en el agua de las represas. El objetivo de este trabajo fue determinar la calidad del agua de 19 represas para riego del centro norte de Entre Ríos, mediante bioensayos de germinación de Lactuca sativa var. mantecosa. Los bioensayos se realizaron en cajas de Petri, con papel de filtro en la base humedecido con 3 ml de agua de la muestra correspondiente. Se sembraron 20 semillas por caja, distribuyéndose los tratamientos en bloques al azar con 4 repeticiones, en cámara de germinación con alternancia de luz y oscuridad. Se registró el porcentaje de germinación y la longitud promedio de la raíz; se calcul un índice de germinación. El porcentaje de germinación promedio de los tratamientos fue de 96,07% y de 97,9% en el testigo. El índice de germinación en todos los casos fue superior al 60% y no se detectó toxicidad en el agua proveniente de las distintas represas.

En la actualidad la horticultura ocupa un espacio predominante en la producción agropecuaria, proveyendo productos indispensables para la alimentación humana, ya sea para ser consumidos en fresco o bien procesados con distintas tecnologías. La lechuga (Lactuca sativa L.) es una hortaliza de consumo mundial y su producción ha ido en aumento. En Mendoza, la comercialización de lechuga como ensalada, tiene gran importancia, a lo largo de todo el año. El contenido de nitratos en hoja es un aspecto importante de su calidad. Según especie, cultivar, parte de la planta, época y ambiente de cultivo, fertilización nitrogenada, esta hortaliza puede presentar contenidos de nitratos elevados, ocasionando indirectamente daños a la salud del consumidor. El objetivo de este trabajo es evaluar los contenidos de nitratos en lechuga cultivada con agua de riego superficial, durante cuatro ciclos agrícolas, en la 3ra Zona de Riego del Río Mendoza y verificar si existe correlación entre tales contenidos y los tenores de nitratos del agua de riego utilizada. La zona de estudio comprendió las localidades de Los Corralitos, La Primavera, Kilmetro 8, Mundo Nuevo y Las Violetas, correspondiente al área administrada por la Asociación de Inspecciones de Cauce de la 3ª Zona de Riego del Río Mendoza. Durante cuatro ciclos agrícolas, se realizó en forma periódica, la obtención de plantas de lechuga y de agua de riego, en cada uno de los puntos de muestreo establecidos previamente. En el laboratorio, las plantas de lechuga fueron secadas y molidas; posteriormente se determinó el contenido de nitratos mediante método colorimétrico. En el caso de las muestras de agua la determinación de nitratos se realizó en espectrofotómetro Hach. La distribución de los contenidos de nitratos en el total de las muestras de lechuga analizadas mostró que el 90% de los datos se encuentran entre 280 y 1744 g NO3 g-1 p.f. Los tipos comerciales mantecosa y morada presentaron tenores mayores que el tipo arrepollada, sin embargo no se encontraron diferencias significativas entre ellos. Con respecto a la evolución estacional, los contenidos de nitratos en otoño e invierno fueron significativamente superiores a los determinados en primavera. Los tenores encontrados, no superaron los lmites máximos establecidos por la Unión Europea, en hortalizas de hoja para consumo en fresco (Reglamento CE N°1258/2011) Los valores de nitratos hallados en la totalidad de las muestras de agua de riego fueron siempre inferiores al lmite fijado por la legislación vigente, observándose una alta variabilidad entre ellos. En los puntos de muestreo correspondientes al tramo final del recorrido del agua, se encontraron tenores mayores, resultando tales variaciones estadísticamente significativas. En el período invernal se detectaron concentraciones de nitratos más elevadas con respecto a los meses estivales, si bien las diferencias observadas entre las estaciones no resultaron significativas. No se halló correlación significativa entre los contenidos de nitratos en lechuga y los tenores de nitratos en el agua de riego. De los resultados obtenidos, se puede concluir que las condiciones edafoclimáticas y culturales de los agroecosistemas del Cinturón Verde de Mendoza, no favorecen la acumulación de nitratos en lechuga, si bien esta especie presenta una marcada tendencia a alcanzar altos contenidos de este compuesto. En nuestra región, es de suma importancia realizar acciones de información y divulgación del problema de la contaminación por nitratos de origen agrario, debido no sólo a los aspectos de calidad sino también a temas vinculados al impacto ambiental y al cuidado de los recursos hídricos.

Los objetivos principales del presente trabajo fueron: a) Estudiar cuál es el uso adecuado de las distintas herramientas para mejorar la mineralización del rastrojo de la cosecha de arroz (Oryza sativa L.) bajo siembra directa y b) Determinar, para períodos cortos, la cantidad de rastrojo depositado superficialmente y su tiempo de mineralización al utilizar las distintas técnicas de manejo del mismo. Los tratamientos fueron: cosechadora con desparramador deflector (T1DD), cosechadora sin desparramador triturador (T2SDT), cosechadora con desparramador centrífugo de caucho (T3DC) y cosechadora con desparramador metálico (T4DM). Alternativas implementadas: a) desmalezado b) rolo cuchilla y c) quemado de rastrojo. En las primeras dos alternativas, se aplicaron tres dosis de urea. Los resultados principales fueron: a) La T2SDT produjo, una mayor deposición de material en el centro de la máquina (21680 kg ha-1), b) la distribución de rastrojo de la (T3DC) fue más uniforme que para los otros tratamientos. Las principales conclusiones fueron: 1) Los desparramadores centrífugos de caucho y metálico produjeron mejor distribución del material, 2) El uso de desmalezadora o rolo cuchilla sobre rastrojo de los tratamientos T3DC y T4DM acelela mineralización de los mismos. 3) Las dosis de urea causó poco incremento en la mineralización del rastrojo.

The evapotranspiration (ETc) of sprinkler-irrigated rice was determined for the semiarid conditions of NE Spain during 2001, 2002 and 2003. The surface renewal method, after calibration against the eddy covariance method, was used to obtain values of sensible heat flux (H) from high-frequency temperature readings. Latent heat flux values were obtained by solving the energy balance equation. Finally, lysimeter measurements were used to validate the evapotranspiration values obtained with the surface renewal method. Seasonal rice evapotranspiration was about 750–800 mm. Average daily ETc for mid-season (from 90 to 130 days after sowing) was 5.1, 4.5 and 6.1 mm day−1 for 2001, 2002 and 2003, respectively. The experimental weekly crop coefficients fluctuated in the range of 0.83–1.20 for 2001, 0.81–1.03 for 2002 and 0.84–1.15 for 2003. The total growing season was about 150–160 days. In average, the crop coefficients for the initial (Kcini), mid-season (Kcmid) and late-season stages (Kcend) were 0.92, 1.06 and 1.03, respectively, the length of these stages being about 55, 45 and 25 days, respectively.

A satellite DNA sequence, As120a, specific to the A-genome chromosomes in the hexaploid oat, Avena sativa L., was isolated by subcloning a fragment with internal tandem repeats from a plasmid, pAs120, that had been obtained from an Avena strigosa (As genome) genomic library. Southern and in situ hybridization showed that sequences with homology to sequences within pAs120 were dispersed throughout the genome of diploid (A and C genomes), tetraploid (AC genomes), and hexaploid (ACD genomes) Avena species. In contrast, sequences homologous to As120a were found in two A-genome species (A. strigosa and Avena longiglumis) and in the hexaploid A. sativa whereas this sequence was little amplified in the tetraploid Avena murphyi and was absent in the remaining A- and C-genome diploid species. In situ hybridization of pAs120a to hexaploid oat species revealed the distribution of elements of the As120a repeated family over both arms of 14 of 42 chromosomes of this species. By using double in situ hybridization with pAs120a and a C genome-specific probe, three sets of 14 chromosomes were revealed corresponding to the A, C, and D genomes of the hexaploid species. Simultaneous in situ hybridizations with pAs120a and ribosomal probes were used to assign the SAT chromosomes of hexaploid species to their correct genomes. This work reports a sequence able to distinguish between the closely related A and D genomes of hexaploid oats. This sequence offers new opportunities to analyze the relationships of Avena species and to explore the possible evolution of various polyploid oat species.

Rice has become a model cereal plant for molecular genetic research. Rice has the most comprehensive molecular linkage maps with more than 2000 DNA markers and shows synteny and colinearity with the maps of other cereal crops. Until now, however, no information was available about the positions of centromeres and arm locations of markers on the molecular linkage map. Secondary and telotrisomics were used to assign restriction fragment length polymorphism markers to specific chromosome arms and thereby to map the positions of centromeres. More than 170 restriction fragment length polymorphism markers were assigned to specific chromosome arms through gene dosage analysis using the secondary and telotrisomics and the centromere positions were mapped on all 12 linkage groups. The orientations of seven linkage groups were reversed to fit the "short arm on top" convention and the corrected map is presented.

Screening for drought resistance of rainfed lowland rice using drought score (leaf death) as a selection index has a long history of use in breeding programs. Genotypic variation for drought score during the vegetative stage in two dry season screens was examined among 128 recombinant inbred lines from four biparental crosses. The genotypic variation detected for drought score in the dry season was used to examine the reliability of the dry season screening method to estimate relative grain yield of genotypes under different types of drought stress in the wet season. Large genotypic variation for drought score existed in two experiments (A and B). However, there was no relationship between the drought scores of genotypes determined in these two experiments. Different patterns of development and severity of drought stress in these two experiments, i.e. slow development and mild plant water deficit in experiment A and fast development and severe plant water deficit in experiment B, were identified as the major factors contributing to the genotypes responding differently. Larger drought score in the dry season experiments was associated with lower grain yield under specific drought stress conditions in the wet season, but the association was weak to moderate and significant only in particular drought conditions. In most cases, a significant phenotypic and moderate genetic correlation between drought score in the dry season and grain yield in the wet season existed only when both drought score and grain yield of genotypes were affected by similar patterns and severity of drought stress in their respective experimental environments. The dry season environments used to measure genotypic variation for drought score should be managed to correspond to relevant types of drought environment that are frequent in the wet season. The efficiency of using the drought score as an indirect selection criterion for improving grain yield for drought conditions was lower than the direct selection for grain yield, and hence wet season screening with grain yield as a selection criterion would be more efficient. However, using drought score as a selection index, a larger number of genotypes can be evaluated than for wet season grain yield. Therefore, it is possible to apply higher selection intensities using the drought score system, and the selected lines can be further tested for grain yield in the wet season. (C) 2004 Elsevier B.V. Lucerne (Medicago sativa) has been suggested as an ideal refuge habitat as part of an integrated pest management (IPM) program because it harbours high numbers of beneficial arthropods. Whether or not cutting of lucerne encourages the movement of these beneficials into adjacent target crops is unknown. Vacuum samples were used to determine the effects of cutting lucerne on arthropod abundance (pests and predators) within lucerne and adjacent soybean (Glycine max) crops. Vacuum-sample collections of arthropods were conducted before and after lucerne cutting on seven occasions in four fields over two seasons. In the lucerne, 10 m by 1 m strips parallel to the crop interface were sampled at 5, 10, 15, 20 and 30 m from the interface. In the soybean, 10 m of row were sampled at the same distances from the crop interface. The abundance of predators in lucerne was reduced immediately after cutting at all distances from the interface. Predator abundance in soybean did not show any change. The cutting of lucerne significantly reduced pest numbers within the lucerne but had little effect on pest abundance in the adjacent soybean. The temporal pattern in pest and predator abundance was very different for each field sampled. Generally, arthropods decreased in abundance after cutting and gradually increased as the lucerne grew back. In soybeans, arthropod numbers fluctuated regardless of the cutting of the lucerne. Cutting of lucerne alone does not guarantee movement of predators into the adjacent target crop. The presence of lucerne fields within a cropping area may have some impact on regional predator populations, and so still be useful for IPM programs, but this has yet to be tested critically.

The effect of interspecific heterosis in crosses between Medicago sativa subsp. sativa and M. sativa subsp. falcata was assessed. Three sativa and 3 falcata plants were crossed in a diallel design. Progeny dry matter yield and natural plant height were assessed in a replicated field experiment at Gatton, Queensland. Yield data were analysed using the method of residual maximum likelihood (REML) and Griffing's model 1. There were significant differences between the reciprocal, general combining ability (GCA), and specific combining ability (SCA) effects. As expected, S-1 populations were lower yielding than their respective intraspecific cross and falcata x falcata crosses were significantly lower yielding than sativa x sativa crosses. Some of the interspecific crosses indicated substantial SCA effects, yielding at least as well as the best sativa x sativa crosses. We have demonstrated the potential usefulness of unselected M. sativa subsp. falcata as a heterotic group in the improvement of yield in northern Australian adapted lucerne material, and discuss how it could be incorporated into future breeding to overcome the yield stagnation currently being experienced in Australian programs.

Early work has shown variation in the grain yield of rice cultivars grown under water stress conditions to be associated with the plant water status, mainly with the maintenance of high leaf water potential (LWP) at flowering and grain filling stage. Considerable variation for LWP among rice varieties has been recorded. The present work was designed to investigate genotypic consistency in water potential within the plant and under canopy manipulation to vary plant water requirement. In a glasshouse experiment, with six rice genotypes, a consistent water potential gradient from stem base to leaf tip has been observed. Leaf tip water potential has been found as the minimum LWP that can be recorded at any time of stress. Genotypes with similar canopy size could maintain different levels of LWP under stress conditions. In a field experiment, with four selected lines, four canopy sizes and two canopy mixture treatments were introduced prior to the imposition of control, mild and severe water stress conditions. It was found that the line differences in LWP and relative water content (RWC) were expressed under both mild and severe stress conditions, regardless of canopy size, tiller number and whether they were mixed with another line with different capacity to maintain LWP. Although there were some differences among canopy size treatments for radiation interception in three water conditions, canopy manipulation (plant size) within a line did not affect the expression of LWP and hence genotypic variation in LWP was maintained. Under both glasshouse and field conditions, lines that maintained high LWP had larger xylem diameter and stem areas than those that had low LWP. Strategies to introduce genes into non-embryogenic plants for complementation of a mutation are described and tested on tetraploid alfalfa (Medicago sativa). Genes conditioning embryogenic potential, a mutant phenotype, and a gene to complement the mutation can be combined using several different crossing and selection steps. In the successful strategy used here, the M. sativa genotype MnNC-1008(NN) carrying the recessive non-nodulating mutant allele nn(1) was crossed with the highly embryogenic alfalfa line Regen S and embryogenic hybrid individuals were identified from the F1 progeny. After transformation of these hybrids with the wild-type gene (NORK), an F2 generation segregating for the mutation and transgene were produced. Plants homozygous for the mutant allele and carrying the wild-type NORK transgene could form root nodules after inoculation with Sinorhizobium meliloti demonstrating successful complementation of the nn(1) mutation.
