The paper contains a brief review of the studies on the life histories of Indian species of prawns chiefly belonging to the family Penaeidae. References to similar work carried out outside India are furnished where significant variations have been observed. The three main larval stages viz., Nauplius, Protozoea and Zoea (Mysis) and their important characteristics, including modes of locomotion, are described. The post-larval development of one species that has been studied in detail (Metapenaeus dobsoni) is indicated in outline. Some aspects of the bionomics of these prawns, especially breeding and migration, are also briefly dealt with in view of their relevance in their life cycle. An outline of the life histories of some Palaemonid prawns of both fresh water and marine habitats is added at the end and the need for well- planned investigations in regard to species of such economic value as Palaemon carcinus (Macrobrachium rosenbergii) is indicated.

Performances of fingerling size and water area was studied. Seven species of fish included 25% of Hypophthalmichthys molitrix, 15% of Gada catla, 20% of Labeo rohita, 10% of Cirrhinus mrigala, 10% of Ctenophyringodon idella and 20% of Macrobrachium rosenbergii were stocked at the rate of 8,000 individual/ha along Barbodes gonionoms at 6.0g weight size with a stocking density of 2,000 indl./ ha. Survival of all species was in the range of 39.9 to 96.3% except M. rosenbergii The highest survival was obtained from L. rohita (96.3%) followed by C mrigala (96.0 %)., B. gonionotus (92.3%), C idella (90.6%), H. molitrix (84.0%), C cada (81.7%) respectively. While the survival of M. rosenbergiiwas 13.1%. The highest production of 4912 kg/ha/yr was obtained from the bigger ponds (0.4 ha) stocked with 20-32g size of fingerlings, while lowest production (3123 kg/ha/yr) from small pond (0.1 ha) stocked with 3-10 g fingerlings. Economic analysis shows the highest net profit in bigger ponds stocked with bigger fish seed.

Disease is the main restraining factor for the development of shrimp sector in Bangladesh. Both brackish water shrimp bagda (Penaeus monodon) and freshwater prawn golda, (Macrobrachium rosenbergii) farming in Bangladesh have been facing disease problem. A study on disease outbreak in both brackish water and freshwater small-scale shrimp farms (gher) in Khulna district was carried out through interviewing randomly selected 3-5% of shrimp farmers with a structured questionnaire during March to December, 2002. The study showed that 97% bagda farming ghers and 80% golda farming ghers were affected by disease. White spot disease was the severe disease for bagda, whereas, antenna rot was the main disease for freshwater prawn. Change of water and liming were carried out as control measures of disease for bagda shrimp farming, while it was only liming for golda farming. A small number of shrimp farmers (15%) used chemicals for treatment of shrimp diseases. Development of suitable farming technology to prevent disease contamination and innovation of proper treatments for diseases are required to overcome the disease problems for sustainable shrimp farming in Bangladesh.

A study was conducted to examine the culture practices of freshwater giant prawn, Macrobrachium rosenbergii in some selected areas of Mymensingh. Based on a sample of 100 farmers from three different upazila, namely Phulpur, Gouripur and Ishwargonj in Mymensingh district, 94% of farmers cultured prawn with fish in their pond. Only 6% of farmers cultured prawn, fish and dike crops for higher economic return. The culture period is typically nine months; hatchery produced post-larvae were stocked from May to June and harvested from November to January. Per hectare production of prawn, 375 kg/yr was very low because the farmers followed simple culture method. Most of the farmers made a profit of Tk. 68,403/ha/yr and the major costs incurred were for purchasing prawn seed and feed. The culture of prawn in pond system is technically possible under different conditions though expansion of small-scale prawn farming mainly depends on reduction of production costs. Future targets could be integration of pond prawn culture with other agricultural activities especially dike cropping and rice production in the monsoon.

An investigation was carried out in Phulpur upazila, Mymensingh to examine the current production practices of freshwater giant prawn, Macrobrachium rosenbergii and its marketing systems with sustainable livelihood approach. The livelihoods of a considerable number of rural poor are associated with prawn production in Phulpur upazila. Based on a sample of 50 farmers, about 94% farmers were found to culture prawn with fish in their ponds. Only 4% and 2% farmers were found to culture prawn-fish-dike crops and only prawn respectively. Prawn marketing is almost exclusively a preserve of the private sector where the livelihoods of a large number of people are associated with its distribution and marketing systems. The market chain from producers to consumers passes through a number of intermediaries. About 40% of the produced prawns are exported and the rest 60% are sold to local markets. The price of prawn depends on quality, size and weight. The average farm-gate price of prawn varied from Tk. 110 to 160/kg, whereas it's [sic] market price varied from Tk. 150 to 350/kg. Most of the farmers and traders have improved their socio-economic conditions through prawn farming and marketing activities. However, concerns arise about the long-term sustainability of prawn farming and marketing systems due to lack of technical knowledge of prawn farming, poor road and transport facilities, higher transport cost, poor supply of ice, lack of cash and credit facilities. It is therefore essential to provide institutional and organizational support and credit facilities for sustainable prawn production and marketing systems.

Freshwater giant prawn, Macrobrachium rosenbergii fry produced during late season can not withstand low temperature thus the prawn culture programme during winter is hampered. To overcome this problem, late season (August-September) prawn juveniles (0.9-6.8 g) were stocked at a density of 1.43 to 3.57/square meter in 350-476 square-meter ponds in Pabna and Mymensingh districts during October 2000 and cultured till May 2001. Monthly average water temperature during the winter months (December-February) varied from 16 to 22 °C and gradually increased to 32 °C in May. The prawn fry showed fast growth rate and attained an average weight of 60-70 g within eight months including three winter months. Growth compensation was observed during summer months. Survival rate was 60-79%. After extrapolation of the present growth rate more than 1,600 kg/ha production can be achieved in better-managed ponds. Extrapolated cost of production was Tk. 268,000 and 200,000 Tk./ha in two best ponds, sale value was Tk. 644,9146 [sic] and 528,466 and gross profit was Tk. 376,000-410,000, suggesting a higher economic feasibility of farming freshwater prawn with over-wintered juveniles.

The effect of AC and DC electric stimulations on the heart-rate and the entire body of Heteropneustis fossillis, Tilapia mossambica and Macrobrachium rosenbergii were studied and presented in kymograph tracings. The reaction of spinal cord in Puntius ticto, Heteropneustis fossilis and Tilapia mossambica to D. C. field was observed to find out its role in electric shocks. A test-check of the electrical resistance of a few species was also conducted. The effect of D. C. and A.C. on the body muscle was found to be the same as that in the case of frog. Different degrees of cardiac slowing were observed in AC and DC. Unbalanced galvanotropic movements were also noticed in spinal fishes.

Decapsulated cysts of Artemia salina were experimentally fed to the larvae of Penaeus monodon, Metapenaeus ensis, M. endeavouri and Macrobrachium rosenbergii and were found to be comparable to freshly hatched brine shrimp nauplii.

Manganese superoxide dismutase (MnSOD) plays an important role in crustacean immune defense reaction by eliminating oxidative stress. Knowledge on MnSOD at molecular level allows us to understand its regulatory mechanism in crustacean immune system. A novel mitochondrial manganese superoxide dismutase (mMnSOD) was cloned from hepatopancreas of Chinese shrimp Fenneropenaeus chinensis by 3' and 5' rapid amplification of cDNA ends (RACE) PCR. The full-length cDNA consists of 1185 bp with a 660 bp open reading frame, encoding 220 amino acids. The deduced amino acid sequence contains a putative signal peptide of 20 amino acids. Sequence comparison showed that the mMnSOD of F. chinensis shares 88% and 82% identity with that of giant freshwater prawn Macrobrachium rosenbergii and blue crab Callinectes sapidus, respectively. mMnSOD transcripts were detected in hepatopancreas, hemocytes, lymphoid organ, intestine, ovary, muscle and gill by Northern blotting. RT-PCR analysis indicated that mMnSOD showed different expression profiles in shrimp hemocytes and hepatopancreas after artificial infection with while spot syndrome virus (WSSV). In addition, a fusion protein containing mMnSOD was produced in vitro. LC-ESI-MS analysis showed that two peptide fragments (-GDVNTVISLAPALK- and -NVRPDYVNAIWK-) of the recombinant protein were identical to the corresponding sequence of M. rosenbergii mMnSOD, and the enzyme activity of the refolded recombinant protein was also measured. 对虾病害在世界范围内的广泛传播,给水产养殖和沿海农村经济造成了重大损失。深入开展对虾免疫机制研究并在此基础上寻找对虾疾病防治的有效方法已成为当务之急。研究表明,当对虾等甲壳动物受到外界病原刺激时,其体内的吞噬细胞在吞噬活动中会激活磷酸己糖支路的代谢,引起呼吸爆发,产生多种活性氧分子。另外,受到病原侵染的对虾还会产生其他多种免疫反应,这些免疫反应将消耗大量的能量(ATP),产能的呼吸链会加速运转,由此也会引发大量活性氧的产生。这些活性氧分子可以杀灭入侵的病原微生物,但同时由于活性氧分子反应的非特异性,它们也会对宿主的细胞、组织和器官造成严重伤害,进而导致对虾生理机能的损伤和免疫系统的破坏。所以,消除对虾体内因过度免疫反应产生的过量氧自由基将能够增强其抵御病原侵染的能力,提高免疫力。本论文从中国明对虾体内克隆了线粒体型超氧化物歧化酶(mMnSOD)、胞质型超氧化物歧化酶(cMnSOD)、过氧化氢酶(Catalase)和过氧化物还原酶(Peroxiredoxin)等四种与免疫系统相关的抗氧化酶基因,分析了它们的分子结构特征,组织分布及应答不同病原刺激的表达变化模式,并对其中的mMnSOD基因和Peroxiredoxin基因进行了体外重组表达、分离纯化和酶活性分析。 采用RACE技术从中国明对虾血细胞中克隆了两个超氧化物歧化酶(SOD)基因,通过序列比对分析发现,其中一个为mMnSOD基因,另一个为cMnSOD基因。mMnSOD基因的cDNA全长为1185个碱基,其中开放阅读框为660个碱基,编码220个氨基酸,其中推测的信号肽为20个氨基酸。多序列比对结果显示中国明对虾mMnSOD基因的推导氨基酸序列与罗氏沼虾、蓝蟹的推导氨基酸序列同源性分别为88%和82%。Northern blot结果表明,该基因在对虾的肝胰脏、血细胞、淋巴器官、肠、卵巢、肌肉和鳃等组织中均有表达。半定量RT-PCR结果显示,对虾感染病毒3 h时,该基因在血细胞和肝胰脏中的转录水平显著升高。此外,通过构建原核表达载体,本研究对该基因进行了体外重组表达,并对纯化的重组蛋白进行了质谱鉴定和酶活分析。cMnSOD基因的cDNA全长为1284个碱基,其中开放阅读框为861个碱基,编码287个氨基酸。多序列比对结果显示中国明对虾cMnSOD基因的推导氨基酸序列与斑节对虾和凡纳滨对虾的同源性高达98%和94%。组织半定量结果显示,cMnSOD基因在对虾被检测的各个组织中均有表达。 另外,半定量RT-PCR结果表明,对虾感染病毒23h时,该基因在肝胰脏中的转录上升到正常水平的3.5倍;而感染后59 h时,该基因在血细胞中的转录上升到正常水平的2.5倍。 利用根据其他生物过氧化氢酶保守氨基酸序列设计的简并引物,结合RACE技术,从中国明对虾肝胰脏中克隆到了过氧化氢酶基因的部分片段,片段长1725个碱基。多序列比对结果发现目前所得中国明对虾Catalase基因部分片段的推导氨基酸序列与罗氏沼虾和皱纹盘鲍Catalase氨基酸序列的同源性分别达到95%和73%。通过实时荧光定量PCR技术对中国明对虾Catalase基因在各个组织中的分布情况及病毒感染后该基因在血细胞和肝胰脏中的转录变化进行了研究。结果发现,该基因在肝胰脏、鳃、肠和血细胞中表达水平较高,在卵巢、淋巴器官和肌肉中的表达水平相对较弱;感染病毒23 h和37 h时,对虾血细胞和肝胰脏中该基因mRNA的表达量分别出现显著性上升。 依据中国明对虾头胸部cDNA文库提供的部分片段信息,结合SMART-RACE技术,从中国明对虾肝胰脏中克隆到了过氧化物还原酶基因(Peroxiredoxin), 该基因的cDNA全长为942个碱基,其中开放阅读框为594个碱基,编码198个氨基酸。中国明对虾Peroxiredoxin基因的推断氨基酸序列与伊蚊、文昌鱼和果蝇等Peroxiredoxin基因的推断氨基酸序列同源性分别为77%、76%和73%。其蛋白理论分子量为22041.17 Da,pI为5.17。Northern blot结果表明,Peroxiredoxin基因在对虾的肝胰脏、血细胞、淋巴器官、肠、卵巢、肌肉和鳃等组织中均有表达。实时荧光定量PCR结果显示,弧菌感染后,该基因在对虾血细胞和肝胰脏中的转录水平都有明显变化并且表达模式不同。另外,对该基因进行了体外重组表达,并对纯化的重组蛋白进行了质谱鉴定和酶活性分析。酶活性分析表明,复性后的重组蛋白能在DTT存在的条件下还原H2O2。

In India scientific commercial farming of Macrobrachium is yet to get a start, the major constraint being the lack of seed. Seed collection from the natural sources alone may not be sufficient, and in such cases seed production in hatcheries is inevitable. This necessitates knowledge of the reproductive biology of the species technology development for brood stock and larval rearing, and the optimum conditions required for larval development to undertake large scale seed production.The present study was taken up with the major objectives of developing a proper, simple and feasible technology for the larval rearing of Macrobrachium, with special emphasis on. mass larval rearing suitable for commercial level operations.

marine bacterium, Micrococcus MCCB 104, isolated from hatchery water, demonstrated extracellular antagonistic properties against Vibrio alginolyticus, V. parahaemolyticus, V. vulnificus, V. fluviallis, V. nereis, V. proteolyticus, V. mediterranei, V. cholerae and Aeromonas sp., bacteria associated with Macrobrachium rosenbergii larval rearing systems. The isolate inhibited the growth of V. alginolyticus during co-culture. The antagonistic component of the extracellular product was heat-stable and insensitive to proteases, lipase, catalase and α-amylase. Micrococcus MCCB 104 was demonstrated to be non-pathogenic to M. rosenbergii larvae

For establishing nitrification in prawn (non-penaeid, salinity 10–15 ppt) and shrimp (penaeid, salinity 30–35 ppt) larval production systems, a stringed bed suspended bioreactor (SBSBR) was designed, fabricated, and validated. It was fabricated with 5 mm polystyrene and low density polyethylene beads as the substrata for ammonia and nitrite oxidizing bacterial consortia, respectively, with an overall surface area of 684 cm2. The reactors were activated in a prototype activator and were transported in polythene bags to the site of testing. Performance of the reactors activated with the nitrifying bacterial consortia AMONPCU-1 (ammonia oxidizers for non-penaeid culture) and NIONPCU-1 (nitrite oxidizers for non-penaeid culture) was evaluated in a Macrobrachium rosenbergii larval rearing system and those activated with AMOPCU-1 (ammonia oxidizers for penaeid culture) and NIOPCU-1 (nitrite oxidizers for penaeid culture) in a Penaeus monodon seed production system. Rapid setting up of nitrification could be observed in both the static systems which resulted in a higher relative per cent survival of larvae
