To investigate the implications of forest fragmentation for conservation of leaf-litter lizards the importance of fragment size, corridors and forest structure was examined in 20 forest fragments and six localities within a continuous forest in the Atlantic Plateau of Sao Paulo state, Brazil. The fragments were 2-276 ha in area and had different degrees of connectivity depending on the presence or absence of corridors. Two species of lizards were dominant, Ecpleopus gaudichaudii and Enyalius perditus. Variation in forest structure among sites was important only in explaining the abundance of E. perditus. Regardless of variation in forest structure, lizard species composition, total lizard abundance, number of species and abundance of E. perditus were sensitive to fragmentation per se but not to fragment size or corridor linkage. The inhospitable matrix surrounding fragments is probably what determines the presence and abundance of E. perditus and the high er lizard richness in continuous forests. These conditions may have prevented lizard species from recolonizing the forest fragments. Our results emphasize that the conservation of this leaf-litter fauna depends on the maintenance of large tracts of continuous forests and not on the size of fragments or on the presence of forest connections. Strategies for conservation of leaf-litter lizards in such highly fragmented Atlantic Forest landscapes should consider the enlargement of landscape connectivity between fragments and continuous forest, allowing the latter areas to act as a source of individuals for fragments.

Two competing hypotheses have been suggested to explain thermal sensitivity of lizards to environmental conditions. These are the static and the labile hypotheses. The static hypothesis posits that thermal physiology is evolutionary conservative and consequently relatively insensitive to directional selection. Contrarily, the labile hypothesis states that thermal physiology does respond readily to directional selection in some lizard taxa. In this paper, we tested both hypotheses among species of Liolaemus lizards. The genus Liolaemus is diverse with about 200 species, being broadly distributed from central Peru to Tierra del Fuego at the southern end of South America. Data of field body temperature (T(b)) from Liolaemus species were collected from the literature. Based on the distributional range of the species we also collected data of mean annual ambient temperatures. We observed that both the traditional analysis and the phylogenetic approach indicate that in the genus Liolaemus T(b) of species varies in a manner that is consistent with ecological gradient of ambient temperature. The data suggest that the thermal physiology of Liolaemus lizards is evolutionarily flexible, and that this plasticity has been partially responsible for the colonization of a wide array of thermal environments. The order Scorpiones is one of the most cytogenetically interesting groups within Arachnida by virtue of the combination of chromosome singularities found in the 59 species analyzed so far. In this work, mitotic and meiotic chromosomes of 2 species of the family Bothriuridae were detailed. This family occupies a basal position within the superfamily Scorpionoidea. Furthermore, review of the cytogenetic data of all previously studied scorpions is presented. Light microscopy chromosome analysis showed that Bothriurus araguayae and Bothriurus rochensis possess low diploid numbers compared with those of species belonging to closely related families. Gonadal cells examined under light and in transmission electron microscopy revealed, for the first time, that the Bothriuridae species possess typical monocentric chromosomes, and male meiosis presented chromosomes with synaptic and achiasmatic behavior. Moreover, in the sample of B. araguayae studied, heterozygous translocations were verified. The use of techniques to highlight specific chromosomal regions also revealed additional differences between the 2 Bothriurus species. The results herein recorded and the overview elaborated using the available cytogenetic information of Scorpiones elucidated current understanding regarding the processes of chromosome evolution that have occurred in Bothriuridae and in Scorpiones as a whole.

In tetrapod squamates, the diversity of micro-ornamentations of the epidermis of the contact areas of hands and feet is generally associated with constraints and modalities related to locomotion. Polychrus acutirostris is a medium-sized lizard that occurs in open heterogeneous habitats in South America, such as the cerrados, caatingas, and fallow lands. It progresses slowly on branches of various diameters in its arboreal environment. It can also move more rapidly on the ground. The hands and feet are prehensile and may be considered an adaptation for grasping and climbing. Epidermal surfaces from the palmar and plantar areas of the hands and feet of P. acutirostris were prepared for SEM examination, and studied at various magnifications. They show three major levels of complexity: (1) scale types, organized in gradients of size and imbrication, (2) scalar ornamentations, organized by increasing complexity and polarity, and (3) presence of Oberhautchen showing typically iguanian honeycomb micro-ornamentations. The shape and surface structure of the scales with their pattern of micro-ornamental peaks, which improve grip, and the grasping hands and feet indicate that P. acutirostris is morpho-functionally specialized for arboreality. Kayotypes of four neotropical teiid lizard species (Tupinambinae) were herein studied after conventional as well as silver staining and CBG-banding: Crocodilurus amazonicus (2n = 34), Tupinambis teguixin (2n = 36), Tupinambis merianae and Tupinambis quadrilineatus (2n = 38). The karyological data for T. quadrilineatus as well as those obtained using differential staining for all species were unknown until now. The karyotypes of all species presented 12 macrochromosomes identical in morphology, but differed in the number of microchromosomes: 22 in C. amazonicus, 24 in T. teguixin and 26 in T. quadrilineatus and T. merianae. The Ag-NOR located at the secondary constriction at the distal end of pair 2 is shared by all species, contrasting with the variability observed for this character in species of the related Teiinae. CBG-banding revealed a species-specific pattern in T. quadrilineatus with conspicuous interstitial C-blocks at the proximal region of the long arm of pair 4 and the whole heterochromatic short arm of pair 6. The karyological data reported here corroborates the relationship hypothesis obtained for Tupinambis based on molecular characters. T. teguixin presents the putative ancestral karyotype for the genus with 2n = 36 whereas T. merianae and T. quadrilineatus exhibit 2n = 38, due to an additional pair of microchromosomes.

We sequenced the small subunit (SSU) rRNA and glycosomal glyceraldehyde-3-phosphate dehydrogenase (gGAPDH) genes of two trypanosomes isolated from the Brazilian snakes Pseudoboa nigra and Crotalus durissus terrificus. Trypanosomes were cultured and their morphometrical and ultrastructural features were characterized by light microscopy and scanning and transmission electron microscopy. Phylogenetic trees inferred using independent or combined SSU rRNA and gGAPDH data sets always clustered the snake trypanosomes together in a clade closest to lizard trypanosomes, forming a strongly supported monophyletic assemblage (i.e. lizard-snake clade). The positioning in the phylogenetic trees and the barcoding based on the variable V7-V8 region of the SSU rRNA, which showed high sequence divergences, allowed us to classify the isolates from distinct snake species as separate species. The isolate from P. nigra is described as a new species, Trypanosoma serpentis n. sp., whereas the isolate from C. d. terrificus is redescribed here as Trypanosoma cascavelli.

Blood examination by microhaematocrit and haemoculture of 459 snakes belonging to 37 species revealed 24% trypanosome prevalence in species of Viperidae (Crotalus durissus and Bothrops jararaca) and Colubridae (Pseudoboa nigra). Trypanosome cultures from C. durissus and P. nigra were behaviourally and morphologically indistinguishable. In addition, the growth and morphological features of a trypanosome from the sand fly Viannaniyia tuberculata were similar to those of snake isolates. Cross-infection experiments revealed a lack of host restriction, as snakes of 3 species were infected with the trypanosome from C. durissus. Phylogeny based on ribosomal sequences revealed that snake trypanosomes clustered together with the sand fly trypanosome, forming a new phylogenetic lineage within Trypanosoma closest to a clade of lizard trypanosomes transmitted by sand flies dagger. The clade of trypanosomes from snakes and lizards suggests an association between the evolutionary histories of these trypanosomes and their squamate hosts. Moreover, data strongly indicated that these trypanosomes are transmitted by sand flies. The flaws of the current taxonomy of snake trypanosomes are discussed, and the need for molecular parameters to be adopted is emphasized. To our knowledge, this is the first molecular phylogenetic study of snake trypanosomes.

Contrary to previous research, training may improve exercise performance in a lizard, the brown anole. A brief, two-week training period resulted in increased performance speed and distance before exhaustion in trained lizards. Trained lizards were also able to more effectively use leg glycogen stores, however each of these improvements were not found in lizards treated with alcohol. Liver glycogen concentrations were also lower in alcohol-treated lizards, and patterns of liver glycogen concentrations during recovery indicate some hepatic lactate gluconeogenesis.

A abordagem da doença alérgica, nomeadamente da alergia respiratória, surge reforçada com o contributo da Aerobiologia no conhecimento, evolução e controlo da doença. Por seu turno, a caracterização bioquímica dos aeroalergénios de uma região pemite avaliar a sua composição e potencial alergénico, com particular interesse para espécies vegetais cuja introdução e/ou proximidade a áreas populacionais, potenciam o aparecimento ou o aumento de sensibilização a aeroalergénios. O trabalho apresentado nesta dissertação incidiu em duas vertentes de investigação distintas, porém complementares e interdependentes: numa primeira parte, no estudo da composição aerobiológica da atmosfera da cidade do Funchal no período 2002-2004 (Capítulo I), e numa segunda parte, na caracterização bioquímica dos alergénios polínicos mais frequentes detectados no referido período (Capítulo II). Assim, numa primeira parte, a monitorização aerobiológica (incluindo pólenes, esporos de fungos e demais partículas de origem biológica), foi realizada com um polinómetro do tipo volumétrico, tipo Hirst (Burkard). Os dados obtidos foram correlacionados com os parâmetros meteorológicos e avaliado o seu significado estatístico, permitindo antever a influência das variáveis ambientais em cada tipo particular de pólen e de fungo. Constataram-se algumas diferenças no conteúdo aerobiológico comparativamente ao restante território nacional. No espectro polínico dominaram as Poaceae e Urticaceae, plantas ornamentais (Asteraceae, Boraginaceae, Cupressaceae) e as representativas da faixa norte da cidade (Ericaceae, Myrtaceae e Pinaceae). Comparativamente a outras regiões do País, o espectro polínico foi no cômputo geral semelhante, embora se destaque para esta região, a expressividade do tipo Corylus no total polínico anual. A Primavera e o início de Verão, corresponderam às épocas de maior diversidade e concentração de pólenes. As diferenças encontradas no espectro polínico da cidade do Funchal são explicadas por variáveis intrínsecas desta região, tais como a composição vegetal, a localização da cidade, condições geo-climáticas inerentes, e a influência dos parâmetros meteorológicos, nomeadamente a temperatura e humidade relativa. Verificou-se a ocorrência de um maior número de pólenes quando a humidade se situa entre os 50 e 60 %, com a precipitação e a velocidade do vento a atingir valores mais baixos. A análise da variação intra-diurna observada revelou que há uma maior representação de pólenes entre as 11 e as 16 horas. Este estudo aerobiológico confere dados para o estabelecimento do primeiro calendário polínico da região e a definição de padrões de sazonalidade. Por seu turno, a fenologia dos principais tipos polínicos observados no Funchal permite definir um padrão anual de ocorrências polínicas. Relativamente à aeromicologia, verifica-se que, durante o período de estudo foram observados esporos de fungos sobretudo na Primavera (particularmente em Abril e Maio), início do Verão e no Outono. Os Deuteromicetes representaram a classe predominante, sendo Cladosporium o fungo mais abundante na atmosfera do Funchal, cujas concentrações mais elevadas ocorrem a humidades relativas de 40 a 70%. Tal como os pólenes, os esporos apresentam um dinâmica de variação intra-diurna particular: ocorreram em maior concentração entre as 13 e as 15 horas, surgindo igualmente nas primeiras horas da madrugada e da noite. O coberto vegetal do Funchal poderá afectar a aeromicologia local, na medida em que constitui um substrato importante para o crescimento de fungos, tal como as gramíneas que proliferam em quantidade e variedade ao longo de todo o ano. Constatou-se a existência de correlação entre a ocorrência de alguns esporos de fungos e taxa polínicos mais frequentes na atmosfera do Funchal. A análise de Spearman sugere a existência de correlação entre a ocorrência de Parietaria com a de Alternaria e Drechslera. Os esporos de fungos, incluindo esporos de fetos constituem uma fracção significativa das partículas na atmosfera do Funchal, sendo cerca de 11 vezes superior à dos pólenes. Na segunda parte do trabalho, o estudo bioquímico dos aeroalergénios polínicos implicou a optimização dos procedimentos de extracção, apurando-se três técnicas para a obtenção dos perfis proteicos de extractos de pólen desde a sua fonte natural. Esta análise permitiu detectar e identificar através das técnicas de SDS-PAGE-Imunoblotting, proteínas IgE específicas do pólen de plantas possivelmente relacionadas com a sensibilização alérgica. Em consonância com a monitorização aerobiológica, foram preparados extractos de pólen de 10 espécies de plantas. Os resultados em SDS-PAGE revelaram um elevado polimorfismo proteico em todos os extractos. Obtiveram-se extractos de pólen de 7 plantas acerca das quais não se conheciam estudos desta natureza: Acacia mearnsii, Avena barbata, Carduus squarrosus, Carlina salicifolia, Datura candida, Echium nervosum e Urtica membranosa. Por imunoblotting detectou-se no soro de dois pacientes IgE específica a proteínas de D. candida, com pesos moleculares entre 150,71 ± 0,05 e 58,92 ± 5,67 KDa. O soro de um deles reagiu igualmente com 5 alergénios de P. pinaster com 42,02 ± 0,05; 38,61 ± 0,46 ; 35,70 ± 7,78 ; 31,82 ± 2,11 e 27,45 ± 0,46 KDa. No soro de outro indivíduo foi detectada IgE específica para uma proteína de A. mearnsii com 66,66 ± 0,13 KDa, e outra de C. squarrosus,de 67,53 ± 0,29 KDa. É de destacar a sensibilidade e fiabilidade da técnica de Imunoblotting, e o interesse em incluila na metodologia de diagnóstico complementar da alergia respiratória. A ampla difusão de espécies como Ricinus communis, Urtica spp. ou A. mearnsii e sua proximidade à presença humana, reforçam, por um lado, a importância da vigilância aerobiológica, e por outro lado, requer uma definição do seu carácter alergénico para a população desta região.

Coleodactylus natalensis Freire, 1999, an endemic species of Atlantic Forest fragments around the Natal municipality, Rio Grande do Norte State, Brazil, has as type locality the Parque Estadual Dunas de Natal (05º48 S to 05º53 S and 35º09 W to 35º12 W), one of the largest restinga (herb and shrub association on sand dunes along the Brazilian coastline) associate fragment, surrounded by urban zone, placed on setentrional Atlantic Forest limits. We made estimates on populational density, spatial distribution, habitat and microhabitat preferences and feeding ecological aspects like sazonal and sexual variations on diet, prey electivities and niche breadth. We randomly sampled ninety-six 50m2 quadrants in each of the four habitats identified in the study area. Were collected 49 specimens and their stomach contents were analyzed; prey items found were correlated with leaf-litter invertebrates from habitat samples. We found a 98,5 ± 75,5 individuals/ha density, in grouped distribution pattern on densest habitats and random distribution on others habitats. This species lives mostly on leaf-litter in forest habitats, in higher humidity points, with lower temperatures, deeper leaf litter and lower sea level elevations than the randomly chosen points in the study area. Isopoda and Aranae were the most important prey categories in numeric, frequency and volumetric terms. Niche breadth has an intermediate value and was variable in sexual and in habitat terms. There was no correlation between morfometric measures and prey size on diet. The C. natalensis population studied seems to be diet opportunist, although selects larger prey items. The Parque Estadual das Dunas do Natal has several indications of anthropic pressure from the surrounding urban area that may affects the local C. natalensis population. Thus, the fragility of this species calls for urgent conservation efforts

Sea anemones contain a variety of biologically active substances. Bunodosoma caissarum is a sea anemone from the Cnidaria phylum, found only in Brazilian coastal waters. The aim of the present work was to study the biological effects of PLA(2) isolated from the sea anemone B. caissarum on the isolated perfused kidney, the arteriolar mesenteric bed and on insulin secretion. Specimens of B. caissarum were collected from the Sao Vicente Channel on the southern coast of the State of São Paulo, Brazil. Reverse phase HPLC analysis of the crude extract of B. caissarum detected three PLA(2) proteins (named BcPLA(2)1, BCPLA(2)2 and BcPLA(2)3) found to be active in B. caissarum extracts. MALDI-TOF mass spectrometry of BcPLA(2)1 showed one main peak at 14.7 kDa. The N-terminal amino acid sequence of BcPLA(2)1 showed high amino acid sequence identity with PLA(2) group III protein isolated from the Mexican lizard (PA23 HELSU, HELSU, PA22 HELSU) and with the honey bee Apis mellifera (PLA(2) and 1POC_A). In addition, BcPLA(2)1 also showed significant overall homology to bee PLA(2). The enzymatic activity induced by native BCPLA(2)1 (20 mu g/well) was reduced by chemical treatment with p-bromophenacyl bromide (p-BPB) and with morin. BcPLA(2)1 strongly induced insulin secretion in presence of high glucose concentration. In isolated kidney, the PLA(2) from B. caissarum increased the perfusion pressure, renal vascular resistance, urinary flow, glomerular filtration rate, and sodium, potassium and chloride levels of excretion. BcPLA(2)1, however, did not increase the perfusion pressure on the mesenteric vascular bed. In conclusion, PLA(2), a group III phospholipase isolated from the sea anemone B. caissarum, exerted effects on renal function and induced insulin secretion in conditions of high glucose concentration. Intending to explain the extraordinary lizard coexistence levels found in Australian deserts, Morton & James (1988) figured out a hypothesis which defends that the termite diversity would bring about lizard radiation. This study aims to verify the validation of that hypothesis in Caatinga lizard assemblages. This study also objectives verifying if the termite defense mechanisms influence their consuming levels by lizards and if this pattern differs between different lizard lineages. Termites were collected using a standardized sampling protocol of termites. Besides using haphazard sampling, we collect lizards with 108 pitfall traps in each area. Intending to check the linkage between the termite and lizard assemblages, the lizard stomach contents were analyzed and then a canonical correspondence analysis was performed. The presence of nonrandom patterns of diet overlap among the lizard species was also examined. Aiming to check if the defense mechanisms of termite influence their consuming pattern by lizards it was performed a laboratory experiment where termite with different defense mechanisms were offered to lizards of two different lineages. We verified that lizard assemblages do not consume termites according to termite abundance in ecosystems. Furthermore, mean niche overlap lizard species did not differ significantly from that expected by chance. We found that termite chemical defense mechanism does influence the termite s pattern consuming by lizards. These results do not corroborate premises which support Morton & James hypothesis (1988) and point out that lizard do not chose termites based on their abundance, but, trying to avoid consuming termites which exhibit chemical defense mechanisms. This defense mechanism, however, may not be the only explanation to patterns of termite s consuming by lizards.

Tropidurus semitaeniatus and Tropidurus hispidus are lizard species that are widely distributed in Brazil. The former is endemic to the Caatingas, where they live in syntopy with the latter, which is distributed in other ecosystems and biomes. In this work, diet was evaluated including seasonal variations as well as foraging behavior, to determine the importance of these factors in the structure of these populations and the coexistence of these species in syntopy. No significant difference was found in diet composition between the two species, although T. hispidus feed on larger preys than do T. semitaeniatus; this fact, along with the different foraging behavior in the rainy season, are ecological factors that contribute to the syntopic coexistence of these species in the Caatinga area studied

As part of a broader project, Diversity and Distribution Patterns of Floristic and Faunistic composition of remnants of Potiguar s Atlantic Forest, as subsidies to conservation , that subsidizes a group of institutional research, This study aimed to evaluate the structure of the assemblage of lizards a remnant of the of the northern Atlantic Forest, identifying ecological factor (s) that contribute to the coexistence of sympatric species. Additionally, we studied the thermal ecology and thermoregulatory behavior of umbrophily and heliophily species live the Parque Estadual Mata da Pipa (PEMP), a remnant of Atlantic forest located in the Tibau do Sul municipality of, Rio Grande do Norte State, Brazil. It is one of the largest remnants of the Atlantic Forest and has an area of approximately 290 ha. The study was performed by four excursions to the field for 20 days each, when active search and pitfalls traps were used to record and colleted specimens in different habitats of the area. We recorded the presence of 19 species of lizards, of which seven are typical of forest areas, three are endemic Atlantic Forest, these two northern and one are new record for the Rio Grande do Norte. The use of resources, the results showed that phylogenetically related species do not always use a similar way the resources available; the feeding niche was the segregated component of the species that overlapped extensively in the use of space and vice versa. To examine the thermal ecology and thermoregulatory behavior of Kentropyx calcarata and Coleodactylus natalensis, we recorded the clocal temperature (Tc), oh the substrate (Ts) and of the air (Ta) to investigate what of these are the source of heat more important to the temperature s body of these lizards. Behavioral observations were conducted to analyze strategies to optimize the acquisition of heat. The air temperature explained most strongly to variation in body temperature of K. calcarata, while the temperature of the substrate to C. natalensis. As for the behavioral observations, they confirmed that K. calcarata is an active thermoregulatory; C. natalensis is a passive thermoregulatory.

In the present work, we investigated behavioral changes associated with the increase in Zif268 protein expression within telencephalic areas of the tropical lizard Tropidurus hispidus that correspond to the mammalian hippocampus (HC). We used 13 male individuals of this species, collected at the Federal Agrotechnical School of Rio Grande do Norte, under SISBIO license number 19561-1. Four animals had their brains removed and were submitted to a Western blot with antibodies for the Zif268 protein. The remaining animals were separated in two different groups: a control group (n=4) and an exploration group (n=5). Animals from the exploration group were exposed to an enriched environment with many sensory cues novel to them. Control group animals stayed in the environment they were already habituated to. After 90 min from the onset of exposure to the new environment, animals from both groups were submitted to intracardiac perfusion with fixative, and the brains were removed, cryoprotected and frozen. After that, brains were sectioned at 20 μm and the sections were subjected to immunohistochemistry for the Zif268 protein. We verified that the Zif268 protein is likely conserved in the brain of T. hispidus, which showed antigenicity for the antibody anti-Zif268 made in mammals. In animals from the exploration group, we detected an increase of the Zif268 protein in the Septum, Striatum, Dorsoventricular Area and in cortical areas corresponding to the HC. This increase was proportional to the amount of environmental exploration, with maximum positive correlation in the hippocampal subareas Medial Cortex (R = 0.94 and p = 0.004) and Dorsomedial Cortex (R = 0.92 and p = 0.006). The data corroborate the notion that the reptilian hippocampus, as well as the mammalian HC, plays an important role in spatial exploration.
