The present paper studies the influence of different nutrients for the production of two cellulolytic enzymes: endo beta-1.4 glucanase and exo beta-1.4 glucanase by anaerobic fungi taken from cow rumen, that were fed a diet of corn silage and Brachiaria decumbens grass hay. During the enzymatic degradation assays, it was observed that the addition of some essential nutrients in the formulation of the culture medium contributed positively in the cellulolytic enzyme production, with exception of riboflavin. Such results contributed in the establishment of an effective method for the evaluation of enzymatic activities in anaerobic fibrolytic fungi. In this work, nutrients added to enrich the culture medium have successfully proven that they can be used as inoculating agents (inductors) in diets rich in ensilage with law nutritive value.

The stomatal density and index in compressed leaves of Glossopteris communis from two different roof shales from the Lower Permian in Parana Basin, Brazil (Western Gondwana) have been investigated to test the possible relationship with modeled global changes in atmospheric CO(2) during the Phanerozoic. The obtained parameters show that the genus Glossopteris from the Cool Temperate biome can be used as CO(2) -proxy, despite the impossibility of being compared with living relatives or equivalents. When confronted with already published data for the Tropical Summer Wet biome, the present results confirm the detection of low levels of atmospheric CO(2) during the Early Permian, as predicted by the modeled curve. Nevertheless, the lower stomatal numbers detected at the climax of the coal interval (Faxinal Coalfield, Sakmarian) when compared to the higher ones obtained in leaves from a younger interval (Figueira Coalfield, Artinskian) could be attributed to temporarily high levels of atmospheric CO(2). Therefore, the occurrence of an extensive peat generating event at the southern part of the basin and subsequent greenhouse gases emissions from this environment may have been enough to reverse regionally and temporarily the reduction trend in atmospheric CO(2). Additionally, the Faxinal flora is preserved in a tonstein layer, which is a record of volcanic activity that could also cause a rise in atmospheric CO(2). During the Artinskian, the scarce generation of peat mires, as revealed by the occurrence of thin and discontinuous coal layers, and the lack of volcanism evidence would be insufficient to affect the general low CO(2) trend.

Transgenic Citrus sinensis (L.) Osb. cv. Hamlin plants expressing the hrpN gene were obtained by Agrobacterium tumefaciens (Smith and Towns) Conn-mediated transformation. hrpN encodes a harpin protein, which elicits the hypersensitive response and systemic acquired resistance in plants. The gene construct consisted of gst1, a pathogen-inducible promoter, a signal peptide for protein secretion to the apoplast, the selection genes nptI1 or aacC1 and the Nos terminator. The function of gst1 in citrus was evaluated in transgenic C. sinensis cv. Valencia harboring the reporter gene uidA (gus) driven by this promoter. Histochemical analysis for gus revealed that gst1 is activated in citrus leaves by both wounding and inoculation with Xanthomonas axonopodis Starr and Garces pv. citri (Hasse) Vauterin et al. Genetic transformation was confirmed by Southern blot hybridization in eight cv. Hamlin acclimatized plants. RT-PCR confirmed hrpN gene expression in seven cv. Hamlin transgenic lines before pathogen inoculation. Some hrpN transgenic lines showed severe leaf curling and abnormal growth. Six hrpN transgenic lines were propagated and evaluated for susceptibility to X axonopodis pv. citri. RT-PCR confirmed gene expression in all six hrpN transgenic lines after pathogen inoculation. Several of the hrpN transgenic lines showed reduction in susceptibility to citrus canker as compared with non-transgenic plants. One hrpN transgenic line exhibited normal vegetative development and displayed very high resistance to the pathogen, estimated as up to 79% reduction in disease severity. This is the first report of genetic transformation of citrus using a pathogen-inducible promoter and the hrpN gene. Further evaluations of the transgenic plants under field conditions are planned. Nevertheless, the evidence to date suggests that the hrpN gene reduces the susceptibility of citrus plants to the canker disease. (C) 2009 Elsevier B.V. All rights reserved.

Pepper (Capsicum annuum L.) plants were grown aeroponically in a Singapore greenhouse under natural diurnally fluctuating ambient shoot temperatures, but at two different root-zone temperatures (RZTs): a constant 20 +/- 2 degrees C RZT and a diurnally fluctuating ambient (A) (25-40 degrees C) RZT, Plants grown at 20-RZT had more leaves, greater leaf area and dry weight than A-RZT plants. Reciprocal transfer experiments were conducted between RZTs to investigate the effect on plant growth, stomatal conductance (g(s)) and water relations. Transfer of plants from A-RZT to 20-RZT increased plant dry weight, leaf area, number of leaves, shoot water potential (Psi(shoot)), and g(s); while transfer of plants from 20-RZT to A-RZT decreased these parameters. Root hydraulic conductivity was measured in the latter transfer and decreased by 80% after 23 d at A-RZT. Transfer of plants from 20-RZT to A-RZT had no effect on xylem ABA concentration or xylem nitrate concentration, but reduced xylem sap pH by 0.2 units. At both RZTs, g(s) measured in the youngest fully expanded leaves increased with plant development. In plants with the same number of leaves, A-RZT plants had a higher g(s) than 20-RZT plants, but only under high atmospheric vapour pressure deficit. The roles of chemical signals and hydraulic factors in controlling g(s) of aeroponically grown Capsicum plants at different RZTs are discussed.

This study was conducted to determine the effect of pre-exposure of oocytes to Ricinus communis (RCA-1) lectin and osteopontin (OPN) in uterine tube fluid (UTF) on in vitro sperm-egg binding and fertilization. In vitro-matured bovine oocytes were incubated (39 degrees C, 5% CO(2) in air) for 2 h in the following treatments: (i) 500 mu l of fertilization medium (FM); (ii) 250 mu l of FM with 0.25 ml of non-luteal ampullary uterine tube fluid (NLAUTF); (iii) 250 mu l of FM with 250 mu l of NLAUTF and 4 mu l of RCA-1 lectin; (iv) 250 mu l of FM with 250 mu l of NLAUTF, a rabbit polyclonal antibody (1:200) against purified bovine milk OPN, and RCA-1 lectin; (v) 500 mu l of FM and RCA-1 lectin. Following incubation, oocytes were washed, placed in FM with 2 mu g heparin, and incubated with 1 x 10(5) frozen-thawed spermatozoa per 10 oocytes. Oocytes used to assess sperm binding were stained with Hoescht 33342, and the number of sperm bound per zona pellucida counted. The remaining oocytes were fixed in acid alcohol, stained with 1% acetate-orcein and observed to determine the presence of pronuclei. More sperm bound to the zona pellucida (mean +/- SEM) when oocytes were incubated in treatment 3 (59.0 +/- 5.5) than in treatments 2 (46.4 +/- 5.6), 4 (18.1 +/- 5.4), 5 (33.4 +/- 5.6) or 1 (32.5 +/- 5.6). More oocytes were fertilized when incubated in treatment 3 (91% +/- 3.0) than in 2 (84% +/- 3.0), 4 (40% +/- 3.0), 5 (77% +/- 3.0) or 1 (76% +/- 3.0). As in previous studies, this study suggests that RCA-1 lectin enhances binding of UTF-derived OPN to bovine oocytes, resulting in increased sperm-egg binding and fertilization in vitro and a possible role in fertilization.

Objective. The objective of this study was to evaluate the sealing ability of AH Plus, Epiphany, Acroseal, Endofill, and Polifil after active lateral condensation technique, by using a bacterial test, during 64 days. Study design. One hundred bovine incisors were cleaned and shaped; then they were filled with the endodontic sealers and adapted into a microcentrifuge tube. The setup root/microcentrifuge tube was added to glass flasks containing Brain Heart Infusion broth. A culture of Enterococcus faecalis was inserted into the upper chamber of each assembly. Daily leakage was evaluated through the broth turbidity. Results. The results were submitted to statistical analysis (Kaplan-Meier method, Kruskal-Wallis and Dunn tests). Conclusions. AH Plus and Endofill had the worst sealing ability when compared with Polifil, which showed the least leakage. Acroseal and Epiphany showed a tendency toward having an intermediate behavior; however, there was no significant difference among Acroseal, Epiphany, and the other sealers. The control of movement is predicated upon a system of constraints of musculoskeletal and neural origin. The focus of the present study was upon the manner in which such constraints are adapted or superseded during the acquisition of motor skill. Individuals participated in five experimental sessions, ill which they attempted to produce abduction-adduction movements of the index finger in time with an auditory metronome. During each trial, the metronome frequency was increased in eight steps from an individually determined base frequency. Electromyographic (EMC) activity was recorded from first dorsal interosseous (FDI), first volar interosseous (FVI), flexor digitorum superficialis (FDS), and extensor digitorum communis (EDC) muscles. The movements produced on the final day of acquisition more accurately matched the required profile, and exhibited greater spatial and temporal stability, than those generated during initial performance. Tn the early stages of skill acquisition, an alternating pattern of activation in FDI and FVI was maintained, even at the highest frequencies. Tn contrast, as the frequency of movement was increased, activity in FDS and EDC was either tonic or intermittent. As learning proceeded, alterations in recruitment patterns were expressed primarily in the extrinsic muscles (EDC and FDS). These changes took the form of increases in the postural role of these muscles, shifts to phasic patterns of activation, or selective disengagement of these muscles. These findings suggest that there is considerable flexibility in the composition of muscle synergies, which is exploited by individuals during the acquisition of coordination.

O objetivo deste trabalho foi avaliar os efeitos do óleo de mamona sobre a broca-grande do tomateiro, Helicoverpa zea, e sobre o parasitoide de ovos Trichogramma pretiosum. Foi determinada a mortalidade larval da broca, que foi submetida ao óleo de mamona por ingestão e contato às concentrações 0,5, 1,0, 1,5, 2,0, 2,5 e 3,0% (v v-1). A ação do óleo de mamona sobre T. pretiosum foi determinada mediante testes de seletividade e suscetibilidade. As maiores mortalidades de larvas foram observadas no tratamento por contato com as menores concentrações (0,5 e 1,0%), enquanto com as concentrações intermediárias (1,5; 2,0 e 2,5%), não houve diferença entre as vias de aplicação. À maior concentração testada (3,0%), o tratamento por ingestão proporcionou mortalidade superior àquela obtida por contato. O número de ovos parasitados por T. pretiosum, no teste de seletividade, foi afetado negativamente pelo óleo de mamona, contudo, os demais parâmetros avaliados quanto à seletividade e à suscetibilidade não foram afetados. O óleo de mamona reduz a sobrevivência de larvas de H. zea tanto por ingestão quanto por contato. Além disso, não prejudica o desenvolvimento de T. pretiosum, desde que as pulverizações sejam realizadas após as liberações do parasitoide.

A mamona (Ricinus communis) tem hábito de crescimento indeterminado, com expressiva desuniformidade de maturação dos grãos, o que dificulta sobremaneira a colheita mecanizada. O objetivo deste trabalho foi avaliar a eficiência do herbicida glifosato, isolado, ou, em mistura com 2,4-D, e também do paraquat, na dessecação em pré-colheita da mamona. Foram realizados dois experimentos de campo, no delineamento em blocos ao acaso, com quatro repetições, em Rancharia-SP, de março a setembro de 2005. No experimento 1, avaliou-se a mistura de doses de 2,4-D e de glifosato, e no experimento 2 a dessecação somente com paraquat. Foi determinado o teor de água da parte aérea de plantas de mamona em diferentes dias após a aplicação (DAA). A dessecação da mamona com a mistura de 2,4-D e glifosato tem efeito somente a partir de 20 DAA. O 2,4-D aplicado isoladamente tem expressiva capacidade de desidratação do dossel vegetal, porém, inferior à da mistura de 2,4-D e glifosato. A melhor dessecação é com 1,34 kg ha-1 de 2,4-D mais 1,42 kg ha-1 de glifosato. A dose de 1,2 kg ha-1 de paraquat reduz expressivamente o teor de água das plantas da mamona, porém, com desempenho inferior ao da mistura de 2,4-D e glifosato.

A finalidade do armazenamento do grão de pólen é conservar material para futura utilização, proporcionando-lhe condições ótimas, de forma a manter seu poder germinativo, vigor e integridade genética originais. O objetivo deste trabalho foi verificar o poder germinativo dos grãos de pólen de mamoneira (Ricinus communis L.), após armazenamento a baixas temperaturas. Para tanto, grãos de pólen dos cultivares IAC 80 e AL Guarany 2002 foram utilizados e armazenados em quatro temperaturas: -196, -72, -18 e 4 ºC, durante um período de oito semanas. A avaliação da viabilidade deste pólen foi realizada semanalmente, até completar cinco semanas, sendo realizada uma última avaliação após a oitava semana de armazenamento, por meio do teste de germinação in vitro. Meio de cultura, contendo 10 gL-1 de ágar, 100 gL-1 de sacarose, 0,004 gL-1 de ácido bórico e pH 6,0, foi preparado e depositado em placas de lâmina escavada, onde o pólen foi distribuído sobre a superfície do meio. As placas foram incubadas a 20 ºC em BOD. O delineamento experimental foi inteiramente casualizado, em arranjo fatorial 2 x 4 (2 cultivares x 4 temperaturas) para cada tempo de armazenamento, sendo analisados 100 grãos de pólen por repetição, totalizando seis de cada tratamento. Verificou-se diferença significativa entre os tratamentos para a interação cultivar x temperatura na quinta semana de observação (p < 0,05). Demais interações foram altamente significativas em todas as épocas de observação (p < 0,01). Portanto, a utilização de ultrabaixas temperaturas permite a manutenção da viabilidade de grãos de pólen de mamoneira, até a quinta semana de armazenamento.

É communis opinio que a comunicação profissional enquadrada num contexto de globalização e de internacionalização dos saberes especializados – inerentes a qualquer atividade profissional do atual quadro societal – pressupõe o domínio multilingue de competências comunicativas especializadas. Qualquer plano de estudos marcado por um air du temps – em áreas ligadas à comunicação empresarial – oferece pelo menos duas línguas estrangeiras tidas como hegemónicas no panorama internacional. Sendo esta uma premissa comummente aceite não deixa, todavia, de ser curioso que o valor cognitivo, cultural e económico de uma língua como o português, uma das dez línguas mais faladas no mundo, tenha deixado de granjear o reconhecimento daqueles que dela dependem inelutavelmente para se afirmarem profissionalmente. A competência comunicativa especializada em língua materna constitui, do nosso ponto de vista, o mais poderoso instrumento de afirmação de identidade e de competitividade junto do mercado laboral, ao instituir-se como principal ferramenta cognitiva, analítica e performativa ao serviço da comunicação especializada. O trabalho em sala de aula é revelador de que uma percentagem significativa de alunos manifesta insegurança aquando das opções terminológicas e pragmático-discursivas inerentes à interação oral e à produção escrita especializadas. Para tal, torna-se imperativo investir no estudo exploratório de práticas pedagógicas em português – língua materna de especialidade – que conduzam à melhoria desses resultados, como procuraremos demonstrar através da análise de um caso prático. Se hoje, mais do que nunca, a complexidade semiótica da rede impõe uma gestão equilibrada do fluxo caleidoscópico de impressões (Whorf, 1957) – na formação académica – não pode ficar esquecido o potencial organizador, cognitivo e heurístico da língua materna para a construção de um perfil comunicativo especializado, competente e competitivo.

Introduction: Anatomical variations of the extensor tendons to the fingers are of great clinical interest, due to the relatively high frequency of tendon injury in clinical practice. Material and methods: During routine dissection of the right upper limb of a 67-year-old female preserved corpse, the extensor indicis proprius (EIP) muscle belly originated 3 independent tendons, each with a separate fascial sheath, forming a triple EIP tendon. There was a larger tendon, which occupied a central position, that represented the usual single EIP tendon. In addition, there were two thinner radial and ulnar accessory EIP tendons. The radial-EIP tendon crossed deep to the extensor digitorum communis (EDC) tendon to the index finger in the distal half of the dorsum of the hand to reach the radial side of the extensor expansion hood of the index finger. Discussion: According to the literature, the frequency of a triple EIP tendon ranges from 0%, to as high as 7%, although most authors do not acknowledge the presence of this variant in their series. This variant of the EIP tendon, in which the radial-EIP terminated laterally to the termination of the tendon of the EDC to the index finger, may be a source of confusion intraoperatively, as the EIP tendon has traditionally been identified on the basis of its ulnar location with respect to the EDC tendon. Conclusion: The possibility of a triple EIP tendon should certainly be born in mind by all surgeons when performing tendon repairs, tenoplasties or tendon transfers.

Introduction Aedes aegypti is responsible for the transmission of the dengue and yellow fever viruses. This study evaluated the effects of extracts from Cnidosculos phyllacanthus, Ricinus communis, and Coutarea hexandra on the developmental periods of A.aegypti larvae and pupae. Crude extracts of C. phyllacanthus and C. hexandra and oil from R. communis and C. phyllacanthus were used. Methods Bioassays of the larvicidal and pupicidal effects of these products at different concentrations and times of exposure were evaluated. The lethal and sublethal effects were determined using different concentrations in larvicidal tests. Mortality data were evaluated by Probit analysis to determine the LC50 and LC90 values. Results The vegetable oils from C. phyllacanthus and R. communis demonstrated greater efficiency for larval control with an LC50=0.28µl/mL and an LC90=1.48µl/mL and LC50=0.029µl/mL and a LC90=0.26µl/mL, respectively. In pupal tests toxic effects for all insects were verified after exposure to the products at significant LC50 and LC90 values for 24 and 48h. The effects of sublethal concentrations of C. phyllacanthus (oil) were more effective on the insects. Conclusions The vegetables oils from C. phyllacanthus and R. communis demonstrated greater potential from the control of different developmental periods in the life cycle of this insect.

Native from south eastern Australia, Eucalyptus globulus is the main species in eucalypts plantations in Portugal. The most serious foliar disease in eucalypt plantations is linked to Mycosphaerella senso lato, which affects young trees in the juvenile phase foliage causing leaf necrosis. This disease results in reduced growth rate of the host and lower wood volume, thus causing significant productivity losses. The most common name for this disease was Mycosphaerella Leaf Disease that became inappropriate when most of the pathogens on eucalypts were re-distributed into several genera. The term "Eucalyptus Leaf Disease Complex" is now more appropriate. The overall aim of this thesis was to investigate the Eucalyptus Leaf Disease Complex in Portugal, focusing on species diversity, taxonomy and the role played by each species in the disease complex on Eucalyptus globulus. Literature on the Eucalyptus Leaf Disease Complex was reviewed and the species were distributed into several genera. A survey based on symptomatic leaves collected from several Eucalyptus globulus plantations and characterized by morphological and molecular tools provided an overview of species incidence and of the most frequent species in the disease complex. The present work reveals additional species of Mycosphaerella senso lato associated with eucalypt plantations in Portugal. Thus, five new records of Teratosphaeria and phylogenetically related species were added to the Iberian Peninsula, namely, Neodevriesia hilliana, for the first time on Myrtaceae; Quasiteratosphaeria mexicana, Teratosphaericola pseudoafricana, Teratosphaeria pluritubularis and Teratosphaeria lusitanica, a new species. Furthermore, new anamorphic structures were found and two new combinations were made. Regarding other genera, some species were observed for the first time, such as Cladosporium cladosporioides, Fusicladium eucalypti, Mycosphaerella madeirae, in the mainland. In addition to leave diseases, Teratosphaeria gauchensis was found causing a severe stem and trunk canker on Eucalyptus globulus. The aggressiveness of several species was compared to evaluate each species individually in the complex, permitting to distinguish different behaviours, from primary to secondary pathogens. Cladosporium cladosporioides, M. communis and M. lateralis, appeared to be more aggressive than Teratosphaeria nubilosa. In fact, contrary to the prevailing views on this disease complex, Teratosphaeria nubilosa is not the only species responsible for the disease, which clearly involves a complex of species acting together.

This paper is a joined publication of the Depts. of Genetics and of Technology, of the E. S. A. "Luiz de Queiroz", Universidade de São Paulo, and deals with the variation of the percentage oil content in the whole seeds, the embryos and the seed-coat of 28 varieties of castor-beans (Ricinus communis, L.). Primarily, the authors, as a justification of this paper, make reference to the applications which castor-oil has in industry, medicine, etc. In accordance with the weight of 100 seeds, the varieties of castor-beans were classified into 3 classes : small seeds (100 seeds less than 30 g), medium seeds (100 seeds between 30 g and 60) and large seeds (100 seeds more than 60 g). The percentage of oil in the seed, embryo and seed-coat, the dimensions of the seeds and the weight of 100 seeds are given for every variety in table 1. In order to obtain an estimate of the variability for the methods of determination of the oil percentage, in the 3 differents parts of the seeds and also in the 3 groups of seeds, the coefficient of variability was calculate (table 2). It is showed that the variation in the seed and embryo is low and that in the seed-coat is very high. The analysis of variance, with regard to the difference among the 3 types of seeds (small, medium and large), among the 3 parts of the seed (whole seed, embryo and seed-coat) and residual error, is given in table 3. Only, the oil content of whole seeds among types of seeds was significant at the 5% level. The t test among the correspondent means is not significant for the difference between medium and large seeds is significant between both these types (medium and large) and small seeds. The fiducial limits in relation to the mean of the oil percentage in the 3 differents types of seed, show that there is one variety (n. 1013-2), which has a percentage of oil, in the medium type of seed, significantly at the 5% level (table 4), higher than the general mean. Since the distribution of the percentage of oil in the seedcoat is discontinuous, 5 groups were established (table 5). All the differences between groups are significant (table 6). For practical purposes, when we have to remove the seed coat, one should eliminate those varieties which loose at least 3% of oil by this procedure. There is a significant linear correlation at 5% level between the percentage of oil in the seed and in the embryo, of the smali and medium type of seeds (table 7), and also, when taking the 3 types together (lower part of table 7), one finds that the same is true. Also, the correlation between the percentages of oil in the embryo and in the seed-coat of the 3 types together is significant at 5% level. According to the results obtained in relation to the percentage in 28 varieties studied, it can be recommended, for breeding purposes, to work only with those varieties which belong to the medium and the large types of seeds.
