No âmbito do projeto CV-DUST foi desenvolvida uma campanha de medição do aerossol atmosférico na Cidade da Praia (14° 55’ N, 23°29’ W), de janeiro de 2011 a janeiro de 2012. A concentração do aerossol foi determinada com base no método gravimétrico, com a amostragem feita em termos de PM10 e em frações granulométricas, usando impactores. Complementarmente, foi usado um contador ótico de partículas que permite a monitorização em contínuo e a classificação do número de partículas em 31 frações de tamanho na gama entre 0,25 e 32 μm. A composição química do aerossol foi determinada com incidência nos seguintes componentes: iões inorgânicos solúveis em água (Cl-, NO3-, SO42-, Na+, NH4+, K+, Mg2+ e Ca2+), carbonato total, elementos maioritários da crosta (Si, Na, Al, Fe, Ca, Mg, K, Ti e Mn) e elementos vestigiais (Ba, Zn, Zr, Pb, Cu, Ce, Ni, Cr, V, Co, Sc, As, Sm e Sb), assim como a fração carbonácea (carbono elementar – EC e o carbono orgânico - OC). Durante a campanha, a concentração de PM10 apresentou uma grande variabilidade temporal, com valores médios (à escala diária) situados entre 10 μg/m3 e 507 μg/m3, sendo a concentração média anual estimada em cerca de 59 μg/m3. As concentrações mais elevadas (tipicamente acima dos 100 μg/m3) foram registadas durante os eventos de poeira proveniente do Norte de África, sendo os mais intensos observados nos meses de janeiro, fevereiro e dezembro de 2011. Os registos do contador ótico, feitos em intervalos de 5 min, revelaram que durante os eventos de poeira as concentrações médias horárias das partículas PM10 e PM2.5 podem ultrapassar os 700 μg/m3 e 200 μg/m3, respetivamente. Com base nos resultados do método ótico, as contribuições das frações granulométricas PM1, PM(1-2.5) e PM(2.5-10) para a massa de PM10 foram estimadas em cerca de 11 %, 28 % e 61 %, respetivamente. A composição química do aerossol varia consideravelmente ao longo do ano e revela a predominância das partículas minerais e do sal marinho. Com base em cálculos do balanço mássico das espécies químicas, as contribuições dos dois constituintes maioritários para a massa de PM10 foram estimadas em cerca de 47 % (partículas minerais) e 17 % (sal marinho). O aerossol secundário (NO3-, NH4+ e fração não marinha do SO42) e o aerossol carbonáceo (EC + OC) contribuem cada um com cerca de 4 % e 3 %, respetivamente. A fração mássica restante (cerca de 29 %), corresponde aos constituintes não analisados, podendo a água ser a mais importante neste grupo. A análise química das amostras segregadas por tamanho revela a seguinte composição para as partículas PM1, PM(1-2.5) e PM(2.5-10): 5,2, 11,8 e 20,7 % (constituintes do sal marinho); 8,6, 3,7 e 3,1 % (iões secundários); 8,9, 1,5 e 1,3 % (EC + OC).

Based on the empirical comparative study between two churches from Pentecostal guidance - both located in Parnamirim/RN - and supported on a dialogic interaction between my interlocutors and theoretical references, I proposed me to reflect about how this protestant segment represents and articulates questions such as gender and power relationships, and the daily impact of that in their followers life. In other words, this dissertation aims to understand the reason of the asymmetry attributed to male and female, especially in what concerns the distribution of ecclesiastic works and the authority given to male, as well as the implication of this reality in the reconfiguration of morality and religious praxis in daily life of individuals and involved groups. From this perspective, this work was divided in three chapters, in which I investigate the tension/relationship between faith and secularism, for from this question on concessions and/or prohibitions related to the limits and involvement of the followers with the world and with the very Pentecostal ethos arise. I also analyze here aspects concerning to both ecclesiastic hierarchy and power, with the objective of elucidating how it occurs, what kind of criteria and implications they consider as well as about the nature of the religious labor division between men and women and, finally, I try to understand how the conversion/adhesion of members is reflected in the redefinitions of gender and its relationship between the ecclesiastical and domestic spaces. The diligence and energy spent in this work is in the hope that its fruits can corroborate in the expansion of anthropological knowledge which, in this particular case, involves the Brazilian Pentecostal phenomenon

La Planeación Estratégica es una herramienta administrativa que mediante la formulación de estrategias permite alcanzar los objetivos y misión de la empresa, considerando los factores externos y recursos disponibles. En el diseño de un Plan Estratégico de Comercialización es importante que contenga el análisis de las cuatro variables controlables por las empresas: Producto, Precio, Plaza y Promoción. Las Asociaciones Cooperativas Productoras de Camarón Marino ubicadas en el Cantón Salinas del Potrero Jurisdicción de Jiquilisco departamento de Usulután, actualmente no cuentas con las herramientas necesarias para dar a conocer el producto, razón por la cual el diseño de un Plan Estratégico para la Comercialización del camarón marino sería de gran utilidad para contribuir al desarrollo económico sostenible de las Asociaciones Cooperativas. El planteamiento de los objetivos principales de la investigación son los siguientes: Presentar propuesta de Filosofía empresarial y estructura organizativa para el desarrollo eficiente de la administración. Presentar una mezcla de mercadotecnia que fortalezca las actividades de comercialización a través de estrategias que contribuyan al cumplimiento de los objetivos del Plan de Comercialización. Establecer el método de fijación de precios del Camarón Marino; para determinar el precio adecuado que permita solventar las necesidades económicas que las Asociaciones Cooperativas presentan. Presentar un canal de distribución del Camarón Marino a las Asociaciones Cooperativas del Cantón Salinas del Potrero; para contribuir a la correcta elección de un medio de colocación efectivo, que mejore los ingresos monetarios de los miembros de éstas. Se incluye la metodología de la de investigación utilizada para la recolección de la información, la cual se obtuvo de fuentes primarias a través de entrevistas a miembros de las Asociaciones Cooperativas, guías de pregunta a intermediarios y a miembros de las Asociaciones y observación directa; fuentes secundarias mediante consultas bibliográficas; permitiendo conocer un panorama de la situación actual en la que se encuentran las Asociaciones Cooperativas productoras de camarón marino en lo que a comercialización se refiere, de lo cual se concluyó que los asociados no cuentan con un Plan Estratégico para la Comercialización del Camarón marino. Así mismo no poseen un método para la fijación del precio que permita recuperar la inversión y obtener un margen de utilidad, no cuentan con un canal de distribución del producto que les permita hacer llegar el camarón hasta las manos del consumidor final.

No resulta en absoluto desconocido que en las primeras décadas del siglo XX, precisamente desde 1900 hasta 1939, en España se inicia una época caracterizada por el florecimiento cultural y artístico en la que la mujer se incorpora al ámbito laboral y empieza a adquirir ciertos derechos de los que hasta entonces no gozaba. Tales cambios la llevan a desempeñar un papel eficaz en el ámbito político, cultural y social, superando los impedimentos convencionales que la mantienen al margen de la sociedad, dedicada exclusivamente al entorno familiar y el sagrado deber conyugal y maternal. A partir de 1914, debido a la incorporación femenina al campo laboral, el asociacionismo y las reivindicaciones vinculadas a la emancipación, empieza un momento de gran esplendor y toda la sociedad española experimenta grandes transformaciones. No es de extrañar, pues que la nueva España brinde una oportunidad a la mujer para acceder al ámbito literario, tras estar sometida al rechazo e intolerancia con que el siglo anterior vincula la relación femenina con la pluma, permitiéndole por fin, ejercer la actividad intelectual, aunque con ciertas limitaciones, debido a que la sociedad estaba aún influenciada por los cánones decimonónicos. En este contexto, he de señalar que la participación femenina en el mundo de la literatura fue muy relevante, aunque muchas de las autoras que empezaron su trayectoria en aquel periodo han sido relegadas al olvido. Una de estas escritoras es Sara Álvarez Insúa Escobar a la que elegí como tema de esta tesis doctoral con el fin de poner al descubierto su obra junto a su prolífica labor literaria en una etapa difícil para el reconocimiento profesional de la mujer. Por ello, el principal objetivo de este trabajo tiene como fin contribuir a la recuperación actual de su prestigio literario. Durante la fase de investigación, descubrí que Sara Insúa se encuentra entre las destacadas escritoras de la época de la "Edad de Plata" que dejan su impronta en la literatura escrita por mujeres en aquel momento y también noté que su obra sufre una gran desatención por parte de la crítica. Por ello, como primera fase de elaboración de ese estudio, tuve que recuperar y reconstruir una gran parte de sus datos biográficos...

Os públicos estão em primeiro lugar, diz Sara Barriga Brighenti, coordenadora do recém-inaugurado Museu do Dinheiro (Lisboa). Esta afirmação pode parecer óbvia, mas nem sempre a conseguimos ver concretizada na maioria dos museus. Em visita guiada ao Museu do Dinheiro, Brighenti falou-nos dos desafios de programar um museu tendo em mente a perspectiva do visitante.

Characterized for the first time in erythrocytes, phosphatidylinositol phosphate kinases (PIP kinases) belong to a family of enzymes that generate various lipid messengers and participate in several cellular processes, including gene expression regulation. Recently, the PIPKIIα gene was found to be differentially expressed in reticulocytes from two siblings with hemoglobin H disease, suggesting a possible relationship between PIPKIIα and the production of globins. Here, we investigated PIPKIIα gene and protein expression and protein localization in hematopoietic-derived cells during their differentiation, and the effects of PIPKIIα silencing on K562 cells. PIPKIIα silencing resulted in an increase in α and γ globins and a decrease in the proliferation of K562 cells without affecting cell cycle progression and apoptosis. In conclusion, using a cell line model, we showed that PIPKIIα is widely expressed in hematopoietic-derived cells, is localized in their cytoplasm and nucleus, and is upregulated during erythroid differentiation. We also showed that PIPKIIα silencing can induce α and γ globin expression and decrease cell proliferation in K562 cells.

Bone marrow is organized in specialized microenvironments known as 'marrow niches'. These are important for the maintenance of stem cells and their hematopoietic progenitors whose homeostasis also depends on other cell types present in the tissue. Extrinsic factors, such as infection and inflammatory states, may affect this system by causing cytokine dysregulation (imbalance in cytokine production) and changes in cell proliferation and self-renewal rates, and may also induce changes in the metabolism and cell cycle. Known to relate to chronic inflammation, obesity is responsible for systemic changes that are best studied in the cardiovascular system. Little is known regarding the changes in the hematopoietic system induced by the inflammatory state carried by obesity or the cell and molecular mechanisms involved. The understanding of the biological behavior of hematopoietic stem cells under obesity-induced chronic inflammation could help elucidate the pathophysiological mechanisms involved in other inflammatory processes, such as neoplastic diseases and bone marrow failure syndromes.

Protocols for the generation of dendritic cells (DCs) using serum as a supplementation of culture media leads to reactions due to animal proteins and disease transmissions. Several types of serum-free media (SFM), based on good manufacture practices (GMP), have recently been used and seem to be a viable option. The aim of this study was to evaluate the results of the differentiation, maturation, and function of DCs from Acute Myeloid Leukemia patients (AML), generated in SFM and medium supplemented with autologous serum (AS). DCs were analyzed by phenotype characteristics, viability, and functionality. The results showed the possibility of generating viable DCs in all the conditions tested. In patients, the X-VIVO 15 medium was more efficient than the other media tested in the generation of DCs producing IL-12p70 (p=0.05). Moreover, the presence of AS led to a significant increase of IL-10 by DCs as compared with CellGro (p=0.05) and X-Vivo15 (p=0.05) media, both in patients and donors. We concluded that SFM was efficient in the production of DCs for immunotherapy in AML patients. However, the use of AS appears to interfere with the functional capacity of the generated DCs.

Previous results provided evidence that Cratylia mollis seed lectin (Cramoll 1,4) promotes Trypanosoma cruzi epimastigotes death by necrosis via a mechanism involving plasma membrane permeabilization to Ca(2+) and mitochondrial dysfunction due to matrix Ca(2+) overload. In order to investigate the mechanism of Ca(2+) -induced mitochondrial impairment, experiments were performed analyzing the effects of this lectin on T. cruzi mitochondrial fraction and in isolated rat liver mitochondria (RLM), as a control. Confocal microscopy of T. cruzi whole cell revealed that Cramoll 1,4 binding to the plasma membrane glycoconjugates is followed by its internalization and binding to the mitochondrion. Electrical membrane potential (∆Ψm ) of T. cruzi mitochondrial fraction suspended in a reaction medium containing 10 μM Ca(2+) was significantly decreased by 50 μg/ml Cramoll 1,4 via a mechanism insensitive to cyclosporine A (CsA, membrane permeability transition (MPT) inhibitor), but sensitive to catalase or 125 mM glucose. In RLM suspended in a medium containing 10 μM Ca(2+) this lectin, at 50 μg/ml, induced increase in the rate of hydrogen peroxide release, mitochondrial swelling, and ∆Ψm disruption. All these mitochondrial alterations were sensitive to CsA, catalase, and EGTA. These results indicate that Cramoll 1, 4 leads to inner mitochondrial membrane permeabilization through Ca(2+) dependent mechanisms in both mitochondria. The sensitivity to CsA in RLM characterizes this lectin as a MPT inducer and the lack of CsA effect identifies a CsA-insensitive MPT in T. cruzi mitochondria.

Leg ulcers represent a particularly disabling complication in patients with sickle cell disease (SCD). Platelet gel (PG) is a novel therapeutic strategy used for accelerating wound healing of a wide range of tissues through the continuous release of platelet growth factors. Here, we describe the use of PG preparation according to Anitua's PRGF (preparations rich in growth factors) protocol for treating chronic nonhealing ulcers in patients with SCD. A positive response occurred in 3 patients with an area reduction of 85.7% to 100%, which occurred within 7 to 10 weeks, and a 35.2% and 20.5% of area reduction in 2 other patients, who however, had large ulcers. After calcium chloride addition, the platelet-rich plasmas demonstrated enhanced platelet-derived growth factors-BB (P < .001), transforming growth factor-β1 (P = .015), vascular endothelial growth factors (P = .03), and hepatocyte growth factors (nonsignificant) secretion. Furthermore, calcium chloride addition induced a significant decrease in platelet number (P = .0134) and there was no leukocyte detection in the PG product. These results demonstrate that PG treatment might impact the healing of leg ulcers in sickle cell disease, especially in patients with small ulcers.
