The purpose of this research was to demonstrate the applicability of reduced-size STR (Miniplex) primer sets to challenging samples and to provide the forensic community with new information regarding the analysis of degraded and inhibited DNA. The Miniplex primer sets were validated in accordance with guidelines set forth by the Scientific Working Group on DNA Analysis Methods (SWGDAM) in order to demonstrate the scientific validity of the kits. The Miniplex sets were also used in the analysis of DNA extracted from human skeletal remains and telogen hair. In addition, a method for evaluating the mechanism of PCR inhibition was developed using qPCR. The Miniplexes were demonstrated to be a robust and sensitive tool for the analysis of DNA with as low as 100 pg of template DNA. They also proved to be better than commercial kits in the analysis of DNA from human skeletal remains, with 64% of samples tested producing full profiles, compared to 16% for a commercial kit. The Miniplexes also produced amplification of nuclear DNA from human telogen hairs, with partial profiles obtained from as low as 60 pg of template DNA. These data suggest smaller PCR amplicons may provide a useful alternative to mitochondrial DNA for forensic analysis of degraded DNA from human skeletal remains, telogen hairs, and other challenging samples. In the evaluation of inhibition by qPCR, the effect of amplicon length and primer melting temperature was evaluated in order to determine the binding mechanisms of different PCR inhibitors. Several mechanisms were indicated by the inhibitors tested, including binding of the polymerase, binding to the DNA, and effects on the processivity of the polymerase during primer extension. The data obtained from qPCR illustrated a method by which the type of inhibitor could be inferred in forensic samples, and some methods of reducing inhibition for specific inhibitors were demonstrated. An understanding of the mechanism of the inhibitors found in forensic samples will allow analysts to select the proper methods for inhibition removal or the type of analysis that can be performed, and will increase the information that can be obtained from inhibited samples.

Melanoma is a type of skin cancer and is caused by the uncontrolled growth of atypical melanocytes. In recent decades, computer aided diagnosis is used to support medical professionals; however, there is still no globally accepted tool. In this context, similar to state-of-the-art we propose a system that receives a dermatoscopy image and provides a diagnostic if the lesion is benign or malignant. This tool is composed with next modules: Preprocessing, Segmentation, Feature Extraction, and Classification. Preprocessing involves the removal of hairs. Segmentation is to isolate the lesion. Feature extraction is considering the ABCD dermoscopy rule. The classification is performed by the Support Vector Machine. Experimental evidence indicates that the proposal has 90.63 % accuracy, 95 % sensitivity, and 83.33 % specificity on a data-set of 104 dermatoscopy images. These results are favorable considering the performance of diagnosis by traditional progress in the area of dermatology

Planar cell polarity (PCP) occurs in the epithelia of many animals and can lead to the alignment of hairs, bristles and feathers; physiologically, it can organise ciliary beating. Here we present two approaches to modelling this phenomenon. The aim is to discover the basic mechanisms that drive PCP, while keeping the models mathematically tractable. We present a feedback and diffusion model, in which adjacent cell sides of neighbouring cells are coupled by a negative feedback loop and diffusion acts within the cell. This approach can give rise to polarity, but also to period two patterns. Polarisation arises via an instability provided a sufficiently strong feedback and sufficiently weak diffusion. Moreover, we discuss a conservative model in which proteins within a cell are redistributed depending on the amount of proteins in the neighbouring cells, coupled with intracellular diffusion. In this case polarity can arise from weakly polarised initial conditions or via a wave provided the diffusion is weak enough. Both models can overcome small anomalies in the initial conditions. Furthermore, the range of the effects of groups of cells with different properties than the surrounding cells depends on the strength of the initial global cue and the intracellular diffusion.

El bienestar animal surge del conocimiento de los animales como seres que experimentan dolor, sufrimiento o estrés, la importancia del impacto en su salud, su comportamiento y también en su eficiencia reproductiva. La evaluación del bienestar incluye el comportamiento, la salud y la forma en la que los animales responden al manejo y a la gestión de sus condiciones de vida (alojamiento y alimentación). Bajo este estudio se desarrolla e implementa la evaluación del bienestar de los sementales y yeguas de cría de un centro de reproducción estatal, así como los potros de recría, futuros reproductores, mediante el estudio del efecto de las condiciones de manejo, para determinar cómo influyen las condiciones de vida sobre los indicadores de bienestar basados en el animal. Se utilizó un protocolo de evaluación según el modelo Welfare Quality® adaptado a la especie equina que incluye un total de 45 indicadores basados en el animal y basados en el ambiente, de una población total conformada por 196 animales del Centro Militar de Cría Caballar de Écija (Sevilla). Se evaluaron y compararon inicialmente las dos subpoblaciones de equinos adultos con aptitud reproductora constituida por 49 sementales estabulados y 43 yeguas de cría en semilibertad. Posteriormente, se realizó la evaluación de bienestar de 104 potros de recría futuros reproductores que vivían en libertad. Los resultados revelaron diferencias significativas en el estado de bienestar de los animales adultos sometidos a distintas condiciones de manejo. La evaluación del alojamiento y la alimentación puso de manifiesto que en los animales estabulados el área disponible para cada animal en las instalaciones era insuficiente y existía riesgo de lesión, dejaban restos de alimento y la condición corporal estaba aumentada. Se observaron sólo comportamientos anormales en los animales estabulados. También fueron más propensos a mostrar curiosidad en los test de comportamiento y también desviaciones de salud como el rascado en la crin y la cola, cojera, heridas en la región de las extremidades y problemas dentales, como los más destacados. Por el contrario, en las yeguas que vivían en semilibertad, se dieron reacciones de temor en los test de comportamiento, heridas distribuidas fundamentalmente por el cuello, tronco, grupa y regiones genitales, así como escalas de condición corporal disminuida. El análisis de los factores de riesgo usando un modelo de regresión, demostró cómo el manejo en semilibertad se asociaba a la aparición de ciertos trastornos de salud y desviaciones de comportamiento. En la población joven, destacaron las respuestas de curiosidad en los test de comportamiento, la presencia de heridas en las extremidades, descarga nasal, dolor en el dorso, pelos rotos en crin y cola y problemas generalizados de piel, entre otros. La metodología seguida para la evaluación del bienestar mediante la selección de un panel adecuado de indicadores permite determinar la incidencia de problemas de salud y comportamiento de la población reproductora y cómo influye el sistema de manejo, base para intervenciones futuras en la mejora del bienestar animal y su productividad como reproductores.

The main objective of the research is to demonstrate new physiological characteristics receptors in the presence of mosquito larvae. 100 larvae of different species were collected and studied for a week in periods of 8-24 hrs. Larvae stages I, II, III and IV have photo-thermo receptors of light and heat housed in the body is divided into head, thorax and abdomen, perceive hot or cold environments, and have fibers in chest or hairs lining your body or abdomen, and a pair of antennae on the head. Stages II and III are more developed than the initial stages. They are attracted by the dark green at the bottom, a pair of eyes that perceive light and color. Have receptors proteins (RP55) that capture motion at a speed the slightest movement of waves in the water. Its nose is not well developed but have chemoreceptors. They adapt to changes in pH in alkaline media, are sensitive to chemical, thermal and mechanical changes nociceptors have electroreceptors or galvanoreceptores sensitive to electrical stimuli, have mechanoreceptors that are sensitive to touch, pain, pressure, gravity, sound. They have a GPS position that seems the guides. It is precisely in the fibers, mushrooms or bristles are recipients along with the micro villi in head, thorax and abdomen.
RESUMEN El objetivo principal de la investigación es demostrar nuevas características fisiológicas como la presencia de receptores en las larvas de mosquitos. Se recolectaron 100 larvas de diferentes especies y se estudiaron por una semana en periodos de 8 a 24 hrs. Las larvas de los estadios I,II,III y IV tienen foto-termo receptores de luz y calor alojados en el cuerpo que se divide en cabeza, tórax y abdomen, perciben ambientes fríos o calientes, así como tienen fibras en tórax o pelos que recubren su cuerpo, y un par de antenas en la cabeza. Los estadios II y III son más desarrollados que las etapas iniciales. Tienen receptores proteicos RP55. Les atrae el color verde oscuro en el fondo, un par de ojos que perciben la luz y color con fotoreceptores. Tienen receptores RP55 de movimiento que captan a una velocidad el más mínimo movimiento de ondas en el agua. Su olfato no está muy desarrollado pero tienen quimioreceptores. Se adaptan a cambios de pH en medios alcalinos, tienen nociceptores sensibles a cambios químicos, térmicos y mecánicos, tienen galvanoreceptores o electroreceptores sensibles a estímulos eléctricos, tienen mecanoreceptores que son sensibles al tacto, dolor, presión gravedad, sonido. Tienen un GPS de posición que pareciera las orienta.

The nitrogen-fixing bacterium Sinorhizobium meliloti must adapt to diverse conditions encountered during its symbiosis with leguminous plants. We characterized a new symbiotically relevant gene, emrR (SMc03169), whose product belongs to the TetR family of repressors and is divergently transcribed from emrAB genes encoding a putative major facilitator superfamily-type efflux pump. An emrR deletion mutant produced more succinoglycan, displayed increased cell-wall permeability, and exhibited higher tolerance to heat shock. It also showed lower tolerance to acidic conditions, a reduced production of siderophores, and lower motility and biofilm formation. The simultaneous deletion of emrA and emrR genes restored the mentioned traits to the wild-type phenotype, except for survival under heat shock, which was lower than that displayed by the wild-type strain. Furthermore, the ΔemrR mutant as well as the double ΔemrAR mutant was impaired in symbiosis with Medicago sativa; it formed fewer nodules and competed poorly with the wild-type strain for nodule colonization. Expression profiling of the ΔemrR mutant showed decreased expression of genes involved in Nod-factor and rhizobactin biosynthesis and in stress responses. Expression of genes directing the biosynthesis of succinoglycan and other polysaccharides were increased. EmrR may therefore be involved in a regulatory network targeting membrane and cell wall modifications in preparation for colonization of root hairs during symbiosis.

Nos últimos anos um grande número de estudos utilizando a análise de imagens termográficas de animais na faixa do infravermelho vem sendo realizado. Além dos avanços tecnológicos que tornaram os sensores mais acessíveis, a preocupação com o bem estar e ambiência das instalações animais tem fomentado a utilização desse método não invasivo e de fácil execução. Neste trabalho foram utilizados 20 bovinos das raças Nelore e Pantaneiro. Após tricotomia de uma área sobre o músculo Longissimus dorsi próximo da 13ª costela, seguida de repouso por 24 horas, os animais foram avaliados por três dias consecutivos, seis vezes por dia. Foram aferidas as temperaturas em áreas com e sem pelos com uso de câmera termográfica. O experimento foi realizado em um Delineamento Inteiramente Casualizado (DIC), considerando o efeito de raça. As variáveis temperatura sem pelo (TSP) e a diferença de temperatura entre áreas com e sem pelo (DIF) não apresentaram efeito significativo para raça. Por outro lado, para a temperatura em áreas com pelos(TCP) o efeito de raça foi significativo, com os pantaneiros apresentando as maiores médias. Animais de ambas as raças encontravam-se em condições de conforto térmico durante o experimento. Os resultados mostram a necessidade de se olhar com cautela para trabalhos envolvendo imagens termográficas capturadas em superfícies recobertas por pelos em animais. Como as avaliações foram feitas na sombra e só houve diferença significativa entre áreas com pelos, é possível que as temperaturas registradas com termografia infra vermelha na superfície da pelagem dos animais representem, pelo menos em parte, características termofísicas dos pelos que as compõem (como a capacidade térmica, por exemplo), sendo tais temperaturas pouco representativas para a superfície do corpo dos indivíduos propriamente, como é comum se considerar.

A análise tricológica é um método não invasivo com potencial para a identificação de mamíferos por meio das características microscópicas de seus pelos. Permite identificar e diferenciar amostras ao nível de Ordem, Família, Gênero, Espécie e até mesmo Subespécies em alguns grupos zoológicos. A caracterização morfométrica de estruturas da cutícula em pelos de animais de raças bovinas criadas no Brasil poderia indicar a possibilidade de uso desta técnica como marcador racial. Um total de 120 animais (machos e fêmeas), pertencentes às raças Caracu, Curraleiro Pé-Duro e Nelore, provenientes de rebanhos de diferentes regiões geográficas foi amostrado. Os dados de área e perímetro das escamas cuticulares foram submetidos à análise de variância, sendo o efeito de raça significativo (P<0,001) e as médias das variáveis estudadas significativamente diferentes entre as raças (P<0,05). A técnica tricológica apresentou elevada acurácia na diferenciação das raças bovinas estudadas. O método, que tem como característica baixo custo de execução, se mostrou como uma ferramenta de grande utilidade para identificação não invasiva de bovinos, com potencial de uso e aplicação para outras espécies de interesse para a pecuária.

Yacon, Smallanthus sonchifolius, an Andean species. is a rich source of dictetíc oligofructans with low glucose content. proteins and phenolic compounds. These constituents have shown efficacy in the prevention of diet-related ehronic diseases, including gastroin-testinal disorders and diabetes |1,2|. Yacon is part of a research program at the National Center for Natural Products Research (NCNPR) and University of Mississippi Field Station to develop new alternative root crops for Mississippi while attempting to im-prove the diet of low incorne families. Yacon can be easily propa-gated by cultings. Virus and nematode infections have been re-ported on plants propagated by cuttings in Brazil. a country that hás adopted Yacon as specialty crop [3|. We have developed two culture systems. autotrophic and heterotrophic, to produce healthy plants. Herem we describe the presence of endophytic bactéria m micropropagated Yacon. In auxin free media, new roots were induced. Overa 15day period. the average root mduction per expiam was 5.45 to 8.75 under autotrophic and heterotrophic cul-tures, respectively. Root lenglh vaned between 3 and 60mrn. The presence of root hairs and lateral roots was noticed only in auto-trophic condilions. Linepithema micans Forel is a poorly studied species that is now considered a pest in vineyards in South Brazil. Larval characters have been used in a few phylogenetic studies, and their importance greatly depends in the amount of available information on different species. This study presents a complete panorama on the external morphology of the immatures of L. micans based on observations by light and scanning electron microscopy. The number of larval instars was estimated as three based on the frequency distribution of head widths of 965 larvae. Larvae of L. micans were similar to other Linepithema Mayr in the general format of body and mandibles, presenting a dorsal abdominal protuberance, nine pairs of spiracle, and unbranched hairs. On the other hand, L. micans was unique for having shorter hairs, predominantly denticulate, intraspecific variation in the number of antennal sensilla and in the types of sensilla on the labial palps were reported.
