Net photosynthesis (A) and transpiration rates (E), stomatal conductance (g), water use efficiency (WUE), intrinsic water use efficiency (IWUE) and internal leaf CO2 concentration (C) in response to different vapor pressure deficit (1.2 and 2.5 kPa) were investigated in 'Pera' sweet orange plants affected by citrus variegated chlorosis (CVC), a disease caused by Xylella fastidiosa. All plants were well watered and leaf water potential (Pw) was also measured by the psychrometric technique. Results showed that healthy plants responded to higher vapor pressure deficit (VPD), lowering its net photosynthesis and transpiration rates, and stomatal conductance. However, diseased plants presented no clear response to VPD, showing lower A, E and g for both VPDs studied and very similar values to these variables in healthy plants at the highest VPD. Internal leaf CO2 concentration also decreased for healthy plants when under the highest VPD, and surprisingly, the same pattern of response was found in plants with CVC. These results, the lower Psi(w) and higher WUE values for diseased plants, indicated that this disease may cause stomatal dysfunction and affect the water resistance through xylem vessels, which ultimately may play some role in photosynthetic metabolism. This paper reports the results obtained using the osmotic stress method applied to the purified cathodic and anodic hemoglobins (Hbs) from the catfish Hoplosternum littorale, a species that displays facultative accessorial air oxygenation. We demonstrate that water potential affects the oxygen affinity of H. littorale Hbs in the presence of an inert solute (sucrose). Oxygen affinity increases when water activity increases, indicating that water molecules stabilize the high-affinity state of the Hb. This effect is the same as that observed in tetrameric vertebrate Hbs. We show that both anodic and cathodic Hbs show conformational substrates similar to other vertebrate Hbs. For both Hbs, addition of anionic effectors, especially chloride, strongly increases the number of water molecules bound, although anodic Hb did not exhibit sensitivity to saturating levels of ATP. Accordingly, for both Hbs, we propose that the deoxy conformations coexist in at least two anion-dependent allosteric states, T-o and T-x, as occurs for human Hb. We found a single phosphate binding site for the cathodic Hb.

Conduziu-se um experimento na Fazenda São Manoel, localizada em São Manuel, SP, pertencente à Faculdade de Ciências Agronômicas da Universidade Estadual Paulista, com o objetivo de avaliar os efeitos de diferentes lâminas de irrigação no crescimento da cultura da batata (Solanum tuberosum ssp. tuberosum), cultivar Aracy. O ensaio foi instalado em um Latossolo Vermelho-Escuro, textura arenosa, sob uma cobertura de plástico. O delineamento experimental utilizado foi o de blocos ao acaso, com cinco tratamentos, cinco coletas de plantas para fins de análise de crescimento, e quatro repetições. Os tratamentos consistiam em irrigar a batata quando a tensão da água no solo atingia 15, 35, 55, 75 e 1.500 kPa. O aumento nas lâminas de irrigação induz incremento no índice de área foliar, na duração da área foliar, na taxa de crescimento relativo e na taxa assimilatória líquida.

Water and 3% NaCl intake were increased by the injection of 4 ng angiotensin II (ANG II) into the anteroventral third ventricle (AV3V) region of rats. Pretreatment with two specific ANG II receptor antagonists, [octanoyl-Leu8]ANG II and [Leu8]ANG II, significantly reduced ANG II-induced water and saline intake. This inhibition lasted approximately 30 min, with partial recovery at 60 min. In rats with electrolytic lesion of the bilateral ventromedial nucleus of hypothalamus (VMH), the effect of ANG II on water intake was not different from that observed in sham rats, but saline ingestion increased. In summary, the present results show that the AV3V region is an important central structure for ANG II-induced saline ingestion. Lesion of the VMH increases the response to ANG II, showing an interaction between the AV3V region and the VMH in the regulation of salt ingestion.

Central cholinergic mechanisms are suggested to participate in osmoreceptor-induced water intake. Therefore, central injections of the cholinergic agonist carbachol usually produce water intake (i.e., thirst) and are ineffective in inducing the intake of hypertonic saline solutions (i.e., the operational definition of sodium appetite). Recent studies have indicated that bilateral injections of the serotonin receptor antagonist methysergide into the lateral parabrachial nucleus (LPBN) markedly increases salt intake in models involving the activation of the renin-angiotensin system or mineralocorticoid hormones. The present studies investigated whether sodium appetite could be induced by central cholinergic activation with carbachol (an experimental condition where only water is typically ingested) after the blockade of LPBN serotonergic mechanisms with methysergide treatment in rats. When administered intracerebroventricularly in combination with injections of vehicle into both LPBN, carbachol (4 nmol) caused water drinking but insignificant intake of hypertonic saline. In contrast, after bilateral LPBN injections of methysergide (4 mug), intracerebroventricular carbachol induced the intake of 0.3 M NaCl. Water intake stimulated by intracerebroventricular carbachol was not changed by LPBN methysergide injections. The results indicate that central cholinergic activation can induce marked intake of hypertonic NaCl if the inhibitory serotonergic mechanisms of the LPBN are attenuated.

This study investigated the effects of bilateral injections of serotonergic receptor ligands into the lateral parabrachial nucleus (LPBN) on the presser and dipsogenic responses induced by intracerebroventricular (icv) injection of angiotensin II (ANG II). Rats with stainless steel cannulas implanted bilaterally into the LPBN and into the left lateral ventricle were used to study icy ANG II-induced water intake and presser responses. Pretreatment with the serotonergic 5-HT1/5-HT2 receptor antagonist methysergide (1-8 mu g/200 nl) bilaterally injected into the LPBN increased the water intake induced by icv ANG II (50 ng/mu l) administered via the lateral ventricle, but pretreatment with methysergide (4 mu g/200 nl) did not change the presser response produced by icy ANG II. After bilateral injection of either serotonin (5-HT, 5 mu g/200 nl) or the serotonergic 5-HT2a/5-HT2c receptor agonist (+/-)-2,5-dimetoxy-4-iodoamphetamine hydrochloride (DOI; 0.5-10 mu g/200 nl) into the LPBN, the water intake induced by ANG II was significantly reduced. These results are consistent with other observations indicating that the LPBN is associated with inhibitory mechanisms controlling water intake induced by ANG II treatment and suggest that serotonergic pathways may be involved in this effect.

In this study we investigated: (a) the effects of intracerebroventricular (i.c.v.) injections of moxonidine (an alpha(2)-adrenergic and imidazoline receptor agonist) on the ingestion of water and NaCl induced by 24 h of water deprivation; (b) the effects of i.c.v. injection of moxonidine on central angiotensin II (ANG II)- and carbachol-induced water intake; (c) the effects of the pre-treatment with i.c.v, idazoxan (an alpha(2)-adrenergic and imidazoline receptor antagonist) and RX 821002 (a selective alpha(2)-adrenergic antagonist) on the antidipsogenic action of central moxonidine. Male Holtzman rats had stainless steel cannulas implanted in the lateral cerebral ventricle. Intracerebroventricular injection of moxonidine (5 and 20 nmol/1 mu l) reduced the ingestion of 1.5% NaCl solution (4.1 +/- 1.1 and 2.9 +/- 2.5 ml/2 h, respectively vs. control = 7.4 +/- 2.1 ml/2 h) and water intake (2.0 +/- 0.6 and 0.3 +/- 0.2 ml/h, respectively vs. control = 13.0 +/- 1.4 ml/h) induced by water deprivation, Intracerebroventricular moxonidine (5 nmol/1 mu l) also reduced i.c.v. ANG Ii-induced water intake (2.8 +/- 0.9 vs. control = 7.9 +/- 1.7 ml/1 h) and i.c.v. moxonidine (10 and 20 nmol/1 mu l) reduced i.c.v. carbachol-induced water intake (4.3 +/- 1.7 and 2.1 +/- 0.9, respectively vs. control = 9.2 +/- 1.0 ml/1 h). The pre-treatment with i.c.v. idazoxan (40 to 320 nmol/1 mu l) abolished the inhibitory effect of i.c.v, moxonidine on carbachol-induced water intake. Intracerebroventricular idazoxan (320 nmol/1 mu l) partially reduced the inhibitory effect of moxonidine on water deprivation-induced water intake and produced only a tendency to reduce the antidipsogenic effect of moxonidine on ANG Ii-induced water intake. RX 821002 (80 and 160 nmol/1 mu l) completely abolished the antidipsogenic action of moxonidine on ANG Ii-induced water intake. The results show that central injections c: moxonidine strongly inhibit water and NaCl ingestion. They also suggest the involvement of central alpha(2)-adrenergic receptors in the antidipsogenic action of moxonidine. (C) 1999 Elsevier B.V.

Coffea canephora plants (clone INCAPER-99) were submitted to low N (LN) or high N (HN) applications and two watering regimes (daily irrigation and irrigation every 5 days for a month). Although water potential was not altered significantly by N, HN plants showed higher relative water content than did LN plants under water deficit. Only HN plants exhibited some ability for osmotic adjustment. Plants from both N treatments increased their cell wall rigidity under drought, with a more pronounced augmentation in HN plants. In well-watered plants, carbon assimilation rate increased with increasing N while stomatal conductance did not respond to N supply. Under drought conditions, carbon assimilation decreased by 68-80% compared to well-watered plants, whereas stomatal conductance and transpiration rate declined by 35% irrespective of the N applications. Stable carbon isotope analysis, combined with leaf gas exchange measurements, indicated that regardless of the watering treatments, N increased the long-term water use efficiency through changes in carbon assimilation with little or no effect on stomatal behaviour.

In mature and young leaves of sunflower (Helianthus annuus L. cv. Catissol-01) plants grown in the greenhouse, photosynthetic rate, stomatal conductance, and transpiration rate declined during water stress independently of leaf age and recovered after 24-h rehydration. The intercellular CO 2 concentration, chlorophyll (Chl) content, and photochemical activity were not affected by water stress. However, non-photochemical quenching increased in mature stressed leaves. Rehydration recovered the levels of non-photochemical quenching and increased the F v/F m in young leaves. Drought did not alter the total Chl content. However, the accumulation of proline under drought was dependent on leaf age: higher content of proline was found in young leaves. After 24 h of rehydration the content of proline returned to the same contents as in control plants.

The present study had as its objective the assessment of the possible effects of hydric stress on the growth, physiological characteristics of two different genetic materials from Eucalyptus urograndis. The experiment was carried out in a greenhouse at Faculdade de Ciências Agronômicas of UNESP, campus Botucatu from March to July, 2005. The hydric management was established based on the soil water potential. Two water levels were established, doing the evapotranspired water replacement by pot weighing. Two clones were used, Eucalyptus urograndis 105 and 433, being the first one more resistant to the hydric deficit and the 433 more sensitive to stress. The study was made from a 2×2 factorial (two levels of water × two genetic materials). For the hydric management, the plants were irrigated when they reached a soil water potential of -0.03 MPa or -1.5 MPa. The assessments made were: diffusive water vapor of stomato, transpiration, leaf temperature and leaf water potential. The physiological evaluations throughout the day, in the end of the experiment. Treatments without hydric stress had a higher performance in all studied characteristics, but the clones had no influence. The stomatic resistance followed the potentials, showing higher values in the treatments submitted to hydric deficiency, more intensely for clone 433, being that this also happened with the leaf water potential. The transpiration also followed the leaf water potential and the stomatic resistance more intensely for clone 105 both comparing stressed plants and non-stressed plants. Consequently, the leaf temperatures had higher values for clone 433 on the stressed treatment. Thus, it can be concluded that there was a better performance in plants kept on a soil water potential of -0.03 MPa and a higher resistance to hydric stress for clone 105.

The objective of this research was to study the effects of five different treatments of grass (Brachiaria decumbens) straw mulch on common beans (Phaseolus vulgaris L.): 0% (0 t.ha-1), 25% (2,25 t.ha-1), 50% (4,5 t.ha-1), 75% (6,75 t. ha-1) and 100% (9,0 t/ha) designed by randomized blocks, with four replicates. The irrigation was applied when minimum soil water potential were reached about - 30kPa. The water management based on tensiometers and soil water characteristic curve. A microsprinkler irrigation system was used. The experiment was set up at the Experimental Station of Embrapa Rice and Bean (Empresa Brasileira de Pesquisa Agropecuária Arroz e Feijão) at Santo Antonio de Goiás, Brazil, in a Dark - Red Latosol soil. The results showed: the bean yield and his components were not affected by treatments, except grain number/pod,. The mulch increased the water use efficiency and, consequently, decreased the number of irrigations when the mulch reached more than 50% straw mulch. The treatment with 100% of mulching presented the largest leaf area index and dry matter accumulation was not affected by mulching.

The scope of this experiment is to study the influence of soil water potential on lettuce productivity, particularly in relation to deficit and excess of water. Four lettuce cultivars (Americana, Roxa, Crespa and Mimosa), four minimum soil water potential (-0,001, -0,005, -0,012 and 0,022 MPa) and three replicates in experimental randomized design. The results allowed concluding that the -0,012 MPa has the tendency to produce the highest green mass among her soil water potential applied. The Mimosa showed the tendency to produce the highest evapotranspiration among the cultivars. The cultivars Americana e -0,05 MPa was the best combination (148,33g) the worst was the Roxa and -0,022 MPa minimum soil water potential.
