Sewage sludge produced by the SABESP wastewater treatment plant (Companhia de Saneamento Básico do Estado de São Paulo), located in Barueri, SP, Brazil, may contain high contents of nickel (Ni), increasing the risk of application to agricultural soils. An experiment was carried out under field conditions in Jaboticabal, SP, Brazil, with the objective of evaluating the effects on soil properties and on maize plants of increasing rates of a sewage sludge rich in Ni that had been applied for 6 consecutive years. The experiment was located on a Typic Haplorthox soil, using an experimental design of randomized blocks with four treatments (rates of sewage sludge) and five replications. At the end of the experiment the accumulated amounts of sewage sludge applied were 0.0, 30.0, 60.0 and 67.5 t ha-1. Maize (Zea mays L.) was the test plant. Soil samples were collected 60 d after sowing at depths of 0-20 cm for Ni studies and from 0 to 10 cm and from 10 to 20 cm for urease studies. Sewage sludge did not cause toxicity or micronutrient deficiencies to maize plants and increased grain production. Soil Ni appeared to be associated with the most stable fractions of the soil organic matter and was protected against strong extracting solutions such as concentrated and hot HNO3 and HCl. Ni added to the soil by sewage sludge increased the metal concentration in the shoots, but not in the grain. The Mehlich 3 extractor was not efficient to evaluate Ni phytoavailability to maize plants. Soil urease activity was increased by sewage sludge only in the layer where the residue was applied. © 2006 Elsevier Ltd. All rights reserved.

The CMS detector is designed around a large 4 T superconducting solenoid, enclosed in a 12 000-tonne steel return yoke. A detailed map of the magnetic field is required for the accurate simulation and reconstruction of physics events in the CMS detector, not only in the inner tracking region inside the solenoid but also in the large and complex structure of the steel yoke, which is instrumented with muon chambers. Using a large sample of cosmic muon events collected by CMS in 2008, the field in the steel of the barrel yoke has been determined with a precision of 3 to 8% depending on the location. © 2010 IOP Publishing Ltd and SISSA.

Vários métodos de captura de mosquitos foram desenvolvidos, demonstrando que houve uma evolução dos tipos de armadilhas ao longo do tempo, porém nenhum se comparou à coleta direta por Atração Humana (AH). Este trabalho teve por objetivo, testar métodos alternativos, desenvolvidos para coletar mosquitos, incluindo o desenvolvimento de novas técnicas, que podem substituir o método tradicional de captura direta em humanos. O estudo foi desenvolvido próximo ao Parque Ambiental de Belém, Pará, Brasil. Os métodos utilizados foram: armadilhas CDC, CDC UV, Shannon, armadilha desenvolvida por J. A. Rocha, armadilha desenvolvida por I. S. Gorayeb e coleta por AH. As armadilhas foram instaladas em seis pontos, por seis noites consecutivas. Foram capturados 13.099 espécimes de Culicidae distribuídas em sete gêneros e 27 espécies, sendo Anopheles trianulatus a mais abundante. Os dados foram tratados estatisticamente pelos métodos ANOVA, Análise multivariada – distâncias euclidianas. A coleta por AH direta foi o método que coletou maior diversidade e abundância de mosquitos, seguidos da armadilha de Shannon para abundância e armadilha de Gorayeb para diversidade.

O ensaio imunoenzimático para identificação de repastos sangüíneos apresenta especificidade até nível de gênero, sensibilidade para identificação de repastos sangüíneos parciais e detecção de repastos múltiplos. Foram identificadas as fontes alimentares de 82% dos anofelinos coletados em campo. Foram testados seis anticorpos monoclonais (humano, suíno, cão, bovino, rato e galinha) e destes, apenas o anti- IgG bovino apresentou instabilidade. Obteve-se 55,7% (519/932) dos repastos positivos para sangue humano, o que demonstra, a preferência alimentar destes anofelinos por humanos. Dos 206 mosquitos que apresentaram repasto único, 27,6% foi em humanos. O Anopheles darlingi apresentou 41% dos repastos em humanos e o Índice de Sangue Humano (HBI) no intradomicílio foi de 0,71. An. marajoara apresentou 51,3% dos repastos em humanos, embora tenha sido encontrada em grande quantidade no ambiente extradomiciliar e apresentando HBI no intradomicílio de 0,76. O An. nuneztovari foi a espécie mais abundante, apresentando comportamento exofílico e antropofílico, com 53,8% de repastos em humano e HBI no intradomicílio de 0,65. O método ELISA Sanduíche está implantado, identificando a fonte alimentar das espécies de anofelinos coletadas em campo, exceto para bovino. É a primeira vez que esta técnica é utilizada para determinação de repastos sanguíneos em anofelinos na região Amazônica brasileira. É importante a determinação da fonte alimentar das espécies de anofelinos no sentido de caracterizar o comportamento antropofílico e assim associá-las ou não à transmissão de malária.

Rhodamine B is a dye that when ingested results in fluorescent bands in growing hair and whiskers of many mammals. Previous research at Wildlife Services’ (WS) National Wildlife Research Center (NWRC) found that rhodamine B is a successful biomarker in raccoon whiskers and that raccoons do not have a taste aversion to the dye when it comprises ≤ 3% of a bait. Our study assessed the ease of bait distribution, whisker collection, and evaluation of the biomarker for potential use in the Oral Rabies Vaccination (ORV) program administered by the WS National Rabies Management Program (NRMP). In collaboration with WS operations personnel from Ohio and Michigan, 750 fishmeal polymer baits each containing 150 mg of rhodamine B were hand distributed at NASA’s Plum Brook Station, Sandusky, Ohio in the summer of 2008. Four weeks after baits were distributed whiskers from 162 raccoons were collected. Wildlife Services biologists and technicians evaluated the whiskers for fluorescence using a handheld UV magnifying lamp. Biologists then sent the whiskers to the NWRC, Ft. Collins, Colorado for confirmation of fluorescence under a UV microscope. Results suggest a high level of agreement between the two methods of evaluation. Surveys completed by biologists confirmed that the ease of use, less invasive sampling techniques and promptness of results obtained through the use of rhodamine B are advantageous to the tetracycline biomarker presently used by the ORV program. All participants recommended further evaluation of rhodamine B for its inclusion in future efforts requiring biomarker evaluation.

Blue rayon (BR) in combination with the Salmonella/microsome assay was used to evaluate the mutagenicity of fish bile samples. Specimens of Mugil curema from two sites were collected over a 1-year period. Piacaguera channel contains high concentrations of total polycyclic aromatic hydrocarbons (PAHs) and other contaminants, while Bertioga channel was considered the reference sites in this study. Bile was extracted with BR and tested with TA98, TA100, and YG1041 strains with and without S9 in dose response experiments. PAH metabolite equivalents were analyzed using reverse-phase high performance liquid chromatography /fluorescence. Higher mutagenic responses were observed for the contaminated site; YG1041 with S9 was the most sensitive strain/condition. Mutagenicity ranged from 3,900 to 14,000 rev./mg at the contaminated site and from 1,200 to 2,500 rev./mg of BR at the reference site. The responses of YG1041 were much higher in comparison with the TA98 indicating the presence of polycyclic compounds from the aromatic amine class that cause frameshift mutation. TA100 showed a positive mutagenic response that was enhanced following S9 treatment at both sites suggesting the presence of polycyclic compounds that require metabolic activation. benzo(a)pyrene, naphthalene, and phenanthrene metabolite equivalents were also higher in the bile of fish collected at the contaminated site. It was not possible to correlate the PAH metabolite quantities with the mutagenic potency. Thus, a combination of the Salmonella/microsome assay with YG1041 with S9 from BR bile extract seems to be an acceptable biomarker for monitoring the exposure of fish to mutagenic polycyclic compounds. The present study aimed to evaluate the interactions of the pesticide Vertimec (R) 18EC in aquatic ecosystems. In this respect, soil plots were contaminated with Vertimec (R) 18EC at the concentration indicated for strawberry crops (0.125 L of solution m(-2)). After the contamination, torrential rainfall was simulated and the surface runoff was collected and transferred to mesocosm tanks in five treatments, run in triplicate: (1) control-C; (2) runoff from an uncontaminated plot-UR; (3) runoff from the plot contaminated with Vertimec (R) 18EC-CR; (4) direct application of Vertimec (R) 18EC in the water-V and (5) water samples gathered randomly to verify whether there was contamination between the mesocosms-RS. Water samples from these tanks were also submitted to ecotoxicological tests with Daphnia similis and analyses to evaluate the limnological characteristics, in five collection periods over 10 days (240 h). Physical and chemical differences were observed in the water samples, mainly related to increased turbidity, suspended solids and nutrients (nitrogen and phosphate forms). Acute toxicity was observed for the direct application treatment for the entire experimental period, and in some periods for the CR treatment (from 48 h to 168 h). The results obtained suggest that the pesticide did not fully degrade during the study period (10 days) in the direct application treatment, demonstrating that the presence of other substances in the commercial formulation contribute to the maintenance of toxicity. This represents a potential risk for aquatic ecosystems in areas adjacent to where the chemical is applied. (C) 2011 Elsevier Inc. All rights reserved.

Fungal and mycotoxin contamination was investigated in field samples of nuts, shells and pods of the Brazil nut collected during different periods in Itacoatiara, State of Amazonas, Brazil: day 0, samples still on the tree: days 5, 10 and 15, samples in contact with soil for 5, 10 and 15 days, respectively. The most prevalent fungi were Aspergillus flavus in fruit pods and nuts and Fusarium spp. in shells. Penicillium spp. and A. flavus were isolated from soil, and Fusarium spp. and Penicillium spp. from air. Aflatoxins and cyclopiazonic acid were not detected in any of the samples analyzed. The high frequency of isolation of aflatoxigenic A. flavus strains from soil and Brazil nuts increases the chance of aflatoxin production in these substrates. These findings suggest a possible contamination before drying and indicate soil as the main source of fungal contamination of Brazil nuts. (c) 2012 Elsevier Ltd. All rights reserved.

Many extractors are used to quantify available P in soils, but few studies have assessed the availability of P in soils of the wet tropics amended with high rates of biosolids. In this study, ion exchange resin, Mehlich-1 solution, and Fe-impregnated strips were used to quantify available P in samples from an Oxisol amended with surface-applied biosolids in a long-term field experiment. The soil's maximum capacity for P adsorption was also estimated. Experimental design consisted of randomized blocks, with four treatments and three replicates. Samples of biosolids were collected every year during the experiment, from 1999 to 2002. In 1999, two applications were made before growing maize (Zea mays L.) in austral summer and winter. Treatments were: Control (no biosolids added); B (biosolids added at rates based on their total N content); B2 (biosolids added at twice the rate of B), and B4 (biosolids added at four times the rate of B). Soil samples were collected at 0- to 0.1-, 0.1- to 0.2-, and 0.2- to 0.4-m depths. Biosolids were broadcast applied and incorporated into the soil to a depth of 0.2 m using a rotary hoe. The Oxisol had a high P-adsorption capacity (around 2450 mg kg(-1)) because of its high contents of clay and Fe and Al oxides. All the extractors were effective at assessing P availability and were positively correlated among themselves. Available P soil contents correlated positively with P content in maize leaves and grains, and the resin method yielded the highest correlation with P contents in leaves and grains.

This research aimed to evaluate the occurrence of Staphylococcus aureus isolates in milk and in the milking environment of 10 small-scale farms (<400 L/d) located in the regions of Franca and Ribeirao Preto, state of Sao Paulo, Brazil. Two-hundred twenty samples of milk were collected from individual cows, along with 120 samples from bulk tank milk, 389 samples from milking equipment and utensils (teat cups, buckets, and sieves), and 120 samples from milkers' hands. Fifty-six Staph. aureus strains were isolated from 849 analyzed samples (6.6%): 12 (5.5%) from milk samples of individual cows, 26 (21.7%) from samples of bulk tank milk, 14 (3.6%) from samples collected from equipment and utensils, and 4 (3.3%) from samples from milkers' hands. Pulsed-field gel electrophoresis typing of the 56 Staph. aureus isolates by SmaI restriction enzyme resulted in 31 profiles (pulsotypes) arranged in 12 major clusters. Results of this study indicate a low incidence, but wide distribution of Staph. aureus strains isolated from raw milk collected from individual cows and surfaces of milkers' hands and milking equipment in the small-scale dairy farms evaluated. However, the high percentage of bulk milk samples found with Staph. aureus is of public health concern because raw, unprocessed milk is regularly consumed by the Brazilian population.

The present study aimed determines lead (Pb), antimony (Sb) and barium (Ba) as the major elements present in GSR in the environmental air of the Ballistics Laboratory of the Sao Paulo Criminalistics Institute (I.C.-S.P.), Sao Paulo, SP, Brazil. Micro environmental monitors (mini samplers) were located at selected places. The PM2.5 fraction of this airborne was collected in, previously weighted filters, and analyzed by sector field inductively coupled plasma mass spectrometer (SF-HR-ICP-MS). The higher values of the airborne lead, antimony and barium, were found at the firing range (lead (Pb): 58.9 mu g/m(3); barium (Ba): 6.9 mu g/m(3); antimony (Sb): 7.3 mu g/m(3)). The mean value of the airborne in this room during 6 monitored days was Pb: 23.1 mu g/m(3); Ba: 2.2 mu g/m(3); Sb: 1.5 mu g/m(3). In the water tank room, the air did not show levels above the limits of concern. In general the airborne lead changed from day to day, but the barium and antimony remained constant. Despite of that, the obtained values suggest that the workers may be exposed to airborne lead concentration that can result in an unhealthy environment and could increase the risk of chronic intoxication. The continued growth of large cities is producing increasing volumes of urban sewage sludge. Disposing of this waste without damaging the environment requires careful management. The application of large quantities of biosolids (treated sewage sludge) to agricultural lands for many years may result in the excessive accumulation of nutrients like phosphorus (P) and thereby raise risks of eutrophication in nearby water bodies. We evaluated the fractionation of P in samples of an Oxisol collected as part of a field experiment in which biosolids were added at three rates to a maize (Zea mays L) plantation over four consecutive years. The biosolids treatments were equivalent to one, two and four times the recommended N rate for maize crops. In a fourth treatment, mineral fertilizer was applied at the rate recommended for maize. Inorganic P forms were extracted with ammonium chloride to remove soluble and loosely bound P; P bound to aluminum oxide (P-Al) was extracted with ammonium fluoride; P bound to iron oxide (P-Fe) was extracted with sodium hydroxide; and P bound to calcium (P-Ca) was extracted with sulfuric acid. Organic P was calculated as the difference between total P and inorganic P. The predominant fraction of P was P-Fe, followed by P-Al and P-Ca. P fractions were positively correlated to the amounts of P applied, except for P-Ca. The low values of P-Ca were due to the advanced weathering processes to which the Oxisol have been subjected, under which forms of P-Ca are converted to P-Fe and P-Al. The fertilization with P via biosolids increased P availability for maize plants even when a large portion of P was converted to more stable forms. Phosphorus content in maize leaves and grains was positively correlated with P fractions in soils. From these results it can be concluded that the application of biosolids in highly weathered tropical clayey soils for many years, even above the recommended rate based on N requirements for maize, tend to be less potentially hazardous to the environment than in less weathered sandy soils because the non-readily P fractions are predominant after the addition of biosolids.
