O interesse no comportamento dinâmico de estruturas metálicas vem crescendo nas últimas décadas no Brasil, em decorrência de acidentes com colapso total de algumas estruturas devido às vibrações ambientes em diversas regiões do país. Na região amazônica, por exemplo, onde esse tipo de estrutura deve vencer obstáculos como florestas e rios de grande largura, casos de colapso total de estruturas metálicas também são relatados. O foco principal dessa dissertação é o estudo do comportamento modal de estruturas metálicas submetidas às vibrações ambientes cuja magnitude das forças de excitação é desconhecida. Dois estudos de caso são apresentados: no primeiro deles, o comportamento modal de uma torre de linha de transmissão de energia elétrica é investigado; e no segundo caso, tanto o comportamento modal como os níveis de desconforto de uma ponte são estudados. Os estudos realizados neste último caso visam avaliar os níveis de desconforto da ponte quando submetida às excitações ambientes como rajadas de vento e o tráfego de veículo de acordo a norma brasileira NBR 8800 (1986). Em ambos os estudos de caso foram realizadas análises experimentais e computacionais. Na etapa experimental, ambas as estruturas foram monitoradas com emprego de um conjunto de acelerômetros de baixa freqüência e também de um sistema de aquisição apropriados para ensaios de vibração de estruturas civis. Como é muito difícil medir a magnitude das forças de excitação ambientes, foram utilizados os métodos de identificação estocásticos SSI-DATA e SSI-COV para extração de parâmetros modais de estruturas civis a partir somente dos dados de resposta coletados nos ensaios de vibração. Entre as atividades desenvolvidas nessa etapa, destaca-se a criação de um programa computacional com recursos do Graphical User Interface (GUI) da plataforma Matlab®, destinado à identificação modal de estruturas civis com o emprego dos referidos métodos estocásticos. Esse programa é constituído de três módulos: o primeiro é destinado ao processamento e tratamento dos sinais coletados nos ensaios de vibração; o segundo é utilizado para adicionar as informações do posicionamento dos acelerômetros utilizados nos arquivos dos sinais de resposta; e o terceiro e último módulo é destinado à identificação a partir dos arquivos de dados de resposta processados nos dois primeiros módulos. Na etapa das análises teóricas, foram criados modelos numéricos utilizando o método dos elementos finitos para simular o comportamento dinâmico das estruturas analisadas. Comparando os resultados obtidos em ambas as etapas de análise, verifica-se que resultados experimentais e teóricos apresentaram parâmetros bastante próximos entre si nos primeiros modos de vibração. Os resultados experimentais mostraram que ambos os métodos estocásticos foram muito eficientes na identificação das estruturas ensaiadas.

The present study was performed to investigate the effect of treatment with furosemide on the pressor response induced by intracerebroventricular (i.c.v.) injections of cholinergic (carbachol) and adrenergic (norepinephrine) agonists, angiotensin II (ANGII) and hypertonic saline (HS, 2 M NaCl). The changes induced by furosemide treatment on the pressor response to intravenous (i.v.) norepinephrine, ANGII and arginine vasopressin (AVP) were also studied. Rats with a stainless-steel cannula implanted into the lateral ventricle (LV) were used. Two injections of furosemide (30 mg/kg b.wt. each) were performed 12 and 1 h before the experiments. Treatment with furosemide reduced the pressor response induced by carbachol, norepinephrine and ANGII i.c.v., but no change was observed in the pressor response to i.c.v. 2 M NaCl. The pressor response to i.v. ANGII and norepinephrine, but not AVP, was also reduced after treatment with furosemide. These results show that the treatment with furosemide impairs the pressor responses induced by central or peripheral administration of adrenergic agonist or ANGII, as well as those induced by central cholinergic activation. The results suggest that the treatment with furosemide impairs central and peripheral pressor responses mediated by sympathetic activation and ANGII, but not those produced by AVP. © 1992.

Die vorliegende Dissertation beschreibt die Verschmelzung der Konzepte von konjugierten Polyelektrolyten und amphiphilen Kammpolymeren in Form von konjugierten, Poly(2,7-carbazol)-basierenden Polyelektrolytkammpolymeren mit Poly(L-lysin)seitenketten sowie Alkyl- oder Polyethylenglykolsubstituenten. Die Synthese wurde durch die Suzuki-Polykondensation von monodispersen Makromonomeren erreicht. Hierbei fand die Precursor-Synthesestrategie Anwendung. In diesem Ansatz war die ε-Aminofunktion des Lysins mit einer Benzoyloxycarbonylschutzgruppe geschützt. Der Aufbau der benötigten monodispersen Makromonomere erfolgte durch die Kupplung von Poly(L-lysin)ketten an den Carbazolbaustein mittels eines aktivierten Esters. Eine Besonderheit der hergestellten Kammpolymere lag in den konformativen Eigenschaften seiner einzelnen Komponenten. Dabei konnte die Konformation der Poly(L-lysin)seitenketten infolge ihres Polyelektrolyt- und Peptidcharakters gezielt mit Hilfe des pH-Wertes und der Ionenstärke variiert werden, wohingegen das konjugierte Rückgrat seine steife Konformation beibehielt. Infolge des Polyelektrolytcharakters zeigte sich zudem, dass die Polymere in sauren und neutralen, wässrigen Lösungen zu großen Teilen in Form von Domänenstrukturen auftraten, während im Basischen eine sofortige Aggregation eintrat. Ein weiteres Merkmal der vorgestellten Polyelektrolytkammpolymere war ihr amphiphiler Charakter. Diese Amphiphilie der in dieser Arbeit vorgestellten Polyelektrolytkammpolymere beeinflusste dabei maßgeblich ihre unstrukturierte Anordnung in Lösung, in der festen Phase sowie an Oberflächen.

Die in der vorliegenden Dissertation entwickelten organochemischen Protokolle und Konzepte erweitern die Bottom-Up-Synthese von atompräzisen Nanographenstreifen (GNR) um zwei fundamentale Bereiche. Zum einen die Dotierung der halbleitenden GNR mit Schwefel oder Stickstoffatomen und zum anderen ein Protokoll für eine lösungsbasierte Synthese von stickstoffdotierten Zickzack-Streifen.rnDie Dotierung von GNR beinhaltet die Synthese von monomeren Bausteinen bei denen, im Gegensatz zu ihren reinen Kohlenstoffhomologen, definierte Positionen am Rand mit zwei oder vier Stickstoff- beziehungsweise zwei Schwefelatomen ersetzt wurden. Die Synthese atompräziser GNR konnte mit verschiedenen experimentellen Methoden analysiert und anschaulich über STM visualisiert werden. Neben einer n-Dotierung gelang so auch erstmals eine Bottom-Up-Synthese von schwefeldotierten GNR. Eine mögliche Anwendung in der Nanoelektronik aufbauend auf dotierten GNR wurde bestätigt, indem durch Co-Polymerisation von stickstoffhaltigen mit reinen Kohlenstoffmonomeren Heteroschnittstellen zwischen dotierten und undotierten Bereichen hergestellt werden konnten. Solche Heteroschnittstellen sind fundamentale Grundlage von Dioden und damit Basis einer Vielzahl elektronischer Elemente wie Solarzellen oder Leuchtdioden.rnWährend für halbleitende GNR mit einer Armlehnen-Form ein breites Spektrum an organischen Syntheseprotokollen zur Verfügung stand, existierte zu Beginn dieser Arbeit keines für GNR mit Zickzack-Struktur. Innerhalb dieser Arbeit konnte eine Bottom-Up-Synthese zur Erschließung stickstoffdotierter GNR mit Zickzack-Randstruktur erarbeitet werden. Durch die Verwendung eines (2-Hydroxymethyl)phenylboronsäureesters werden Hydroxymethylsubsituenten entlang eines Polyphenylenrückgrats eingebaut, die nach Kondensation mit dem Stickstoffatom eine Zickzack-Kante ergeben. Innerhalb der synthetisierten Zielstrukturen kann das 9a-Azaphenalen als letztes, bislang nicht erschlossenes Isomer der Azaphenalene, als wiederkehrende Struktur, gefunden werden. Die Reaktivität der Zickzackkante konnte zudem zum Aufbau einer Vielzahl bislang unzugänglicher, polycyclischer Heteroaromaten über 1,3-dipolare Addition dieses polycyclischen Azomethin Ylides (PAMY) genutzt werden.rn

Seismic assessment and seismic strengthening are the key issues need to be figured out during the process of protection and reusing of historical buildings. In this thesis the seismic behaviors of the hinged steel structure, a typical structure of historical buildings, i.e. hinged steel frames in Shanghai, China, were studied based on experimental investigations and theoretic analysis. How the non-structural members worked with the steel frames was analyzed thoroughly. Firstly, two 1/4 scale hinged steel frames were constructed based on the structural system of Bund 18, a historical building in Shanghai: M1 model without infill walls, M2 model with infill walls, and tested under the horizontal cyclic loads to investigate their seismic behavior. The Shaking Table Test and its results indicated that the seismic behavior of the hinged steel frames could be improved significantly with the help of non-structural members, i.e., surrounding elements outside the hinged steel frames and infilled walls. To specify, the columns are covered with bricks, they consist of I shape formed steel sections and steel plates, which are clenched together. The steel beams are connected to the steel column by steel angle, thus the structure should be considered as a hinged frame. And the infilled wall acted as a compression diagonal strut to withstand the horizontal load, therefore, the seismic capacity and stiffness of the hinged steel frames with infilled walls could be estimated by using the equivalent compression diagonal strut model. A SAP model has been constructed with the objective to perform a dynamic nonlinear analysis. The obtained results were compared with the results obtained from Shaking Table Test. The Test Results have validated that the influence of infill walls on seismic behavior can be estimated by using the equivalent diagonal strut model.

The Agrobacterium tumefaciens VirB/D4 type IV secretion system (T4SS) delivers oncogenic T-DNA and effector proteins to susceptible plant cells. This leads to the formation of tumors termed Crown Galls. The VirB/D4 T4SS is comprised of 12 subunits (VirB1 to VirB11 and VirD4), which assemble to form two structures, a secretion channel spanning the cell envelope and a T-pilus extending from the cell surface. In A. tumefaciens, the VirB2 pilin subunit is required for assembly of the secretion channel and is the main subunit of the T-pilus. The focus of this thesis is to define key reactions associated with the T4SS biogenesis pathway involving the VirB2 pilin. Topology studies demonstrated that VirB2 integrates into the inner membrane with two transmembrane regions, a small cytoplasmic loop, and a long periplasmic loop comprised of covalently linked N and C termini. VirB2 was shown by the substituted cysteine accessibility method (SCAM) to adopt distinct structural states when integrated into the inner membrane and when assembled as a component of the secretion channel and the T-pilus. The VirB4 and VirB11 ATPases were shown by SCAM to modulate the structural state of membrane-integrated VirB2 pilin, and evidence was also obtained that VirB4 mediates extraction of pilin from the membrane. A model that VirB4 functions as a pilin dislocase by an energy-dependent mechanism was further supported by coimmunoprecipitation and osmotic shock studies. Mutational studies identified two regions of VirB10, an N-terminal transmembrane domain and an outer membrane-associated domain termed the antennae projection, that contribute selectively to T-pilus biogenesis. Lastly, characterization of a VirB10 mutant that confers a ‘leaky’ channel phenotype further highlighted the role of VirB10 in gating substrate translocation across the outer membrane as well as T-pilus biogenesis. Results of my studies support a working model in which the VirB4 ATPase catalyzes dislocation of membrane-integrated pilin, and distinct domains of VirB10 coordinate pilin incorporation into the secretion channel and the extracellular T-pilus.

The formation of the vertebrate face is an extremely complex developmental process, which needs to coordinate the outgrowth of several facial primordia. Facial primordia are small buds made up of mesenchymal masses enclosed by an epithelial layer that surrounds the primitive mouth. The upper jaw is formed by the maxillary process, the lateral nasal process, and the frontonasal process while the mandibular process forms the lower jaw. Recent experiments using genetics in mice and bead implantation approaches have shown that the pitx2 homeobox gene and Bmp signaling play important roles in this complex developmental process. However, the molecular mechanisms underlying the function of pitx2 and Bmp in these events are still unclear. Here, we show that pitx2 is required for oral epithelium maintenance, and branchial arch signaling is pitx2 dosage sensitive by using pitx2 allelic combinations that encode varying levels of pitx2. Maintenance of fgf8 signaling requires only low pitx2 dosage while repression of Bmp signaling requires high pitx2 levels. Different incisor and molar phenotypes in low level pitx2 mutant embryos suggest a distinct requirement for pitx2 in tooth-type development. The results show that pitx2 is required for craniofacial muscle formation and expanded Bmp signaling results in excess bone formation in pitx2 mutant embryos. Fate-mapping studies show that ectopic bone results from excessive bone growth, instead of muscle transformation. Moreover, by using cre/loxp system we show that partial loss of Bmpr-IA in the facial primordia results in cleft lip/palate, abnormal teeth, ectopic teeth and tooth transformation. These phenotypes suggest that Bmp signaling has multiple functions during craniofacial development. The mutant palate shelves can fuse with each other when cultured in vitro, suggesting that cleft palate is secondary to the partial loss of Bmpr-IA. Furthermore, we prove that Bmp4, one of the ligands of Bmpr-IA, plays a role during lip fusion developmental process and partial loss of Bmp4 in the facial primordia results in the lip fusion delay. These results have provided insight to understand the complex signaling cascades that regulate craniofacial development. ^

RV POSEIDON cruise POS264 was carried out by the Institut für Meereskunde of the University of Hamburg and staff from the Niels Bohr Instituttet for Astronomi, Fysik og Geofysik of the University of Copenhagen also participated. The cruise had several objectives: - to educate undergraduate students in the handling of oceanographic instrumentation and in the collection and analysis of field data, - to map the cold overflow through the Faroe-Bank Channel from the Norwegian Sea into the Icelandic Basin and to study its short-time variability and - to quantify the contributions of the water masses which are involved in the mixing of the overflow plume with its ambient water. The planning and preparation of the cruise involved the participating students and was carried out during seminars, both at the Universities of Hamburg and Copenhagen. Following a review of the recent literature and an analysis of historical data the observational programme was designed. Hydrographic and current profiling stations were occupied along several sections crossing the overflow. The experiment was financed by the University of Hamburg. Temperature, salinity and dissolved oxygen data from CTD stations are presented. The temperature and salinity data were quality controlled and calibrated. Oxygen data are not calibrated as no oxygen samples were taken additionally during the cruise.

A fully integrated on-board electronic system that can perform in-situ structural health monitoring (SHM) of aircraft?s structures using specifically designed equipment for SHM based on guided wave ultrasonic method or Lamb waves? method is introduced. This equipment is called Phased Array Monitoring for Enhanced Life Assessment (PAMELA III) and is an essential part of overall PAMELA SHM? system. PAMELA III can generate any kind of excitation signals, acquire the response signals that propagate throughout the structure being tested, and perform the signal processing for damage detection directly on the structure without need to send the huge amount of raw signals but only the final SHM maps. It monitors the structure by means of an array of integrated Phased Array (PhA) transducers preferably bonded onto the host structure. The PAMELA III hardware for SHM mapping has been designed, built and subjected to laboratory tests, using aluminum and CFRP structures. The 12 channel system has been designed to be low weight (265 grams only), to have a small form factor, to be directly mounted above the integrated PhA transducers without need for cables and to be EMI protected so that the equipment can be taken on board an aircraft to perform required SHM analyses by use of embedded SHM algorithms. Moreover, the autonomous, automatic and on real-time working procedure makes it suitable for the avionic field, sending the corresponding alerts, maps and reports to external equipment.

This paper summarizes the work developed in order to establish a framework for seismic retrofitting of bridges. In this context, the first objetive is to find a numerical model to evaluate the damage induced in a structure, under seismic action, as an index of its vulnerability. The model used has the adventage that is based on concepts of fracture mechanics and concentrated plasticity. As a result, the work is based on basic principles. The performance of this model is being evaluated. Some results of the computer program developed for this purpose are shown.
